Abstract
Description
The invention relates to a means for stimulating the Tei developmental activity of animal and human cells and a Procedure for using this agent. It is both in the basic biological-medical research as well Biotechnology and the pharmaceutical industry can be used.
Both for the elucidation of basic biological processes as well as for the use of animal cells for biotechno logical purposes, cells are required that can be used for any duration can be cultivated in artificial media. The unbe limited cell division ability is a criterion of tumor cell len. But cells in the "pre-malignant" state can also to be mortal (immortal). In principle, the development is based development of an immortal cell for genetic changes, to change the function of one or more proteins lead that we for the negative regulation of cell division are substantial.
Immortalized cell lines can either be from certain Tu mor can be obtained directly or via detours from primary cell len arise, which were isolated from certain tissues. The spontaneous emergence of an immortal cell line is an ex Extremely rare event because there are probably at least two muta tions are required for this. In the past hence tumor viruses are often inducers of immortalization used. However, these had the disadvantage that they were also often undesirable tumorigenic changes in the cells called. In DD-WP 2 65 165 has already been proposed to Improve the immortalization process of the recombinants with the gene for the large T antigen of the polyomavirus the. This raised the problem of effective immortalization in principle without simultaneous tumorigenic change solved. However, the rate of cell division usually not affected by this procedure.
Many animal and human cells have an approx. 50- 60 cell divisions limited division potential without immorta lization. Longer viability is often desirable worth it, but not essential. However, that is Realization of the actual division potential also with difficult for many cell types, as this depends on an optimal ver supply with growth factors is dependent. So with vie len cell types limit the division activity in vitro found on just a few cycles. A pressing problem Position for the biotechnological use of cell cultures therefore consists in a principle solution for the establishment of Zel len of different organs with maximum exhaustion their natural division potential and, if possible, additional stimulation of cell division with regard to its speed efficiency.
The aim of the invention is to create and use a For general stimulation of division activity animal and human cells in the culture both obviously the exploitation of the natural division potential as well as the speed of the cell cycle. The agent according to the invention is characterized in that it as an active substance one or more nucleotide sequences with complementary sequence (antisense) to certain areas the mRNA of tumor suppressor genes or other negative contains genes acting on cell division activity. The Antisense nucleotide sequence preferably consists of one synthetic oligonucleotide at least 15 in length Nucleotides, with a length of 13-21 nucleotides before is moving. The oligonucleotide must be in nuclease resistant form are present, the phosphothioate form being particularly expedient is.
Above all, sequences are used as antisense nucleotide sequences used that are complementary to a region of the 5'-end, the region around the start codon, a region of an exon or a region at an intron-exon junction of that Gens is. Complete sequences to the tumor are preferred suppressor gene retinoblastoma gene, p53 gene or the E-cadherin selected. Examples of concrete sequences of the effective Substance in the agent according to the invention are 3'-CAGTACGGCGGGTTTT GG-5 ′ and 3′-AGCAAGTGAAAATGACTC-5 ′. To make the Mit The antisense nucleotide sequence according to the invention brought in solution, preferably in aqueous solution.
To use the agent to stimulate cell division ac The following details are given: Oligonucleotides are synthesized by known methods siert.
To stimulate a cell culture in its cell division activity lations, they will preferentially adhere to plastic Treat cells with the selected antisense oligonucleotides delt. To do this, the oligonucleotides are precipitated by alcohol sterilized, in sterile dist. Dissolved water and in one Concentration of 5-30 µM in the culture medium without serum given. The oligonucleotides can additionally with lipofectin can be mixed to make the absorption even more effective To reach cells. After an hour, the medium can be serum be added. The cells are labeled with the oligonucleotide incubated medium for several days. The cell number is compared daily in parallel approaches to an unrelated one acted culture determined.
In primary human lung fibroblasts (HEL) after three days there is a faster increase in the number of cells in the Comparison to control. This tendency stops at one time Use of oligonucleotides for at least 7 days. Within 10 days is at least 3 times this cell type Number of cells with treatment compared to the nonbe acted cells achieved. Analysis of the specific Rb mRNA by means of hybridization clearly shows that the mRNA in the Area of pairing with the antisense oligonucleotide degra is dated. The immunological protein detection shows that at the latest 3 days after the start of treatment with Oligonu kleotiden completely stopped the synthesis of pRb is.
Allow the treated culture to conflict for more than 7 days state, then there are dense islands of cells attached to it remember transformed foci, but actually no trans formed cells included. After about 14 days the Kul are door vessels evenly with a "multilayer" of cells to grow. The cells are passaged and again with Treated oligonucleotide (5-30 µM), then it keeps accelerated growth further until the natural limit of about 60 cell divisions is reached. In individual cases, too about 80 divisions can be achieved. Since the treated But dividing cells more than twice as fast as that Control cells, they end their lifespan significantly early forth.
Comparison of different oligonucleotides with complements Rity to different areas of the Rb mRNA shows that with many an almost identical effect can be achieved. In general, however, an oligonucleotide with the following has been found Sequence 3'-CAGTACGGCGGGTTTTGG-5 ', which is complementary to the region around the start codon is particularly effective. This means tet that at a relatively low concentration (10 µM) Oligonucleotide a maximum stimulation effect and full permanent deactivation of the pRb synthesis can be achieved. A Combination of two oligonucleotides, e.g. B. against the region of the start codon and against an exon sequence proves to be half the concentration (5 µM each) as similarly effective. On the other hand are against other regions when using oligonucleotides higher concentrations (30 µM) of the mRNA are required.
Frisch angelegte Zellen anderer Organe, z. Freshly created cells from other organs, e.g. B. the liver, the can only undergo 1-2 cell divisions without treatment after treatment with an Rb antisense oligonucleotide signi fictionally longer cultivated, whereby the cell division indicates is stimulated.
The agent according to the invention enables the number of cell parts lungs per unit of time. Its application also enables the cultivation of cells that would not otherwise are cultivable. This is used both in biotechnology fabric production as well as in biological-medical fundamental research makes great progress possible. Another embodiment of the invention is that Antisense nucleotide sequence to stimulate cell growth not to clog, but in the cell to be cultivated generate yourself. This was achieved by a re combined with expressible antisense sequence by trans is introduced into the cells. In this case be the preferred sequence is 30-400 nucleotides. The following information is given on the use of the agent: A recombinant is made, a selected one Sequence, preferably 50-400 nucleotides long, from the subject find tumor suppressor gene in antisense orientation behind a suitable promoter is cloned. This recombinant is known along with a selection marker transfected into the relevant cells. To Selection for the presence of the marker (e.g. neo) Get cells whose division activity is clearly stimulated is.
The invention is to nä by embodiments forth be explained.
An oligonucleotide with the sequence 3'-CAGTACGGCGGGTTTTGG-5 'is synthesized and dried as a phosphothioate by a known method on the automatic synthesis apparatus (Applied Biosystems). Approx. 100 OD units are dissolved in 100 µl H 2 O, adjusted to 0.3 M Na acetate and precipitated with 10 volumes of 96% ethanol at -70 ° C. The precipitate was centrifuged off and dissolved in 0.5 ml of sterile water. After measuring the concentration, dilute to 1 µg / µl by adding water. 60 µl of this solution are diluted in 1 ml cell culture medium, which corresponds to a concentration of 10 µM. This solution is applied to the cells previously sown in a 5 cm culture dish. In the example, HEL cells with a density of 5 × 10 4 cells per dish are used. There are 8 dishes with oligonucleotide and without (control). After one hour, 1 ml of medium with 10% serum is added. The following day, the cells are harvested from one dish each and counted in a counting chamber. The results are shown graphically (see Fig. 1) and show a significantly faster increase in the number of cells from the 2nd to the 10th. Day. After 10 days, a subculture is created, which is treated again with oligonucleotide in the manner described above. The renewed daily cell count results in basically the same curve, whereby the difference after the 1st and 2nd day is more clearly recognizable than in the first round.
An oligonucleotide with the sequence 3'-AGCAAGTGAAAATGACTC-5 'is prepared as described under 1, prepared for use and added to the cell culture. The inclusion of a growth curve by daily cell counting results in the same curve as in Fig. 1, whereby after 10 days there are not 3.5 times, but only 2.5 times more cells.
The two oligonucleotides of Examples 1 and 2 are shown in Combination at a concentration of 5 µM each sets, and the cell count is determined daily. Here results there is a steeper curve increase with a 5-fold cell number compared to the control cells after 10 days.
