Der Aufschluß erfolgte in 1.5 ml Eppendorfgefäßen. Die Eppendorfgefäße wurden mit 1.2 g Glasperlen (0,5 mm Durchmesser) und 0.6 ml 40%iger Zellsuspension gefüllt und für 15 min bei einer Frequenz von 1800/sec geschüttelt. Die Ergebnisse zeigten, daß bezüglich der Aminkomponente anscheinend nur TMA umgesetzt wird, inaktiv waren: Dimethylamin, Methyl amin, Dimethylethylamin, Trimethylamin-N-oxid, Cholinchlorid, Betain, Carnitin und D,L-Carnitinamid. Auch bezüglich des Elektronenakzeptors ist das Enzym anscheinend hochspezifisch, TMA wird nicht umgesetzt, wenn PES/DCPIP durch folgende Verbindungen ersetzt wird: Methylenblau, K3(Fe(CN)6), Wurster′s Blau, PES, DCPIP, NAD⁺ oder NADP⁺; lediglich mit der struktur analogen Verbindung Phenazinmethosulfat findet eine Reaktion statt (98%iger Umsatz im Vergleich zu PES). Daneben wurden einige der oben erwähnten Amin-Verbin dungen getestet, ob sie den Umsatz von TMA inhibie ren. Folgende Verbindungen wurden in jeweils äqui molarer Konzentration zu TMA dem kompletten Enzymtest zugesetzt (in Klammern sind als Ergebnis die jeweili gen Enzymaktivitäten angegeben): Dimethylamin (96%), Cholin (102%), Betain (90%) oder Carnitin (110%). Werden dem kompletten Testansatz mit PES,DCPIP und TMA noch das Coenzym NAD⁺ oder NADP⁺ zugesetzt, ist keine Steigerung des Umsatzes nachweisbar.For the preparation of a substrate spectrum in the test (see Example 2.D.)
