Abstract
Description
The invention relates to the method specified in claims 4 to 6 for the production of interferon-γ and the plasmid DNA DSM-6237 according to claim 1, with which the interferon-γ is expressed, and the method for the production of this plasmid DNA according to claims 2 and 3.
Interferon-γ from bacterial cells has been known for some time. This is how EP 00 describes it 87 686 interferon-γ and a process for its preparation. The gene for that Interferon γ protein is expressed in bacterial cells, i.e. H. this protein will synthesized in bacterial cells and must be purified from these cells. The Interferon-γ is expressed in bacterial cells. The yield in the expression achieved with this method is approximately 10% of the total protein.
In addition to the low yields in expression, cleaning also plays a role a decisive one via cation exchange processes in the known method Role. The purification step through cation exchange process is usually carried out according to the prior art with column chromatography, d. that is Interferon-γ is after the renaturation step in the presence of z. B. guanidinium chloride and a phosphate buffer on a column with the Pumped cation exchange material. The cation exchange material binds due to the high binding capacity for the interferon-γ the interferon-γ with a correspondingly high concentration in the upper area of the column. Because of that, because the high concentration of bound interferon-γs, the elution yields very much low. This means that only a small part of the bound interferon-γs can be combined with the corresponding ones Saline solutions are detached from the cation exchange material.
However, due to the great importance of interferon-γ for therapy, it is extreme Desirable, the method for producing interferon-γ in the way improve that both the expression yield and the yield in Cleaning step is enlarged.
It is therefore an object of the invention to provide an improved process for the production of Interferon-γ indicate, both an improvement in the expression rate the aim is to improve the cleaning as well.
The object is achieved in that a plasmid DNA is proposed with which the interferon-γ is expressed with a value 4 times higher, the DNA is characterized in that it is both a shortening of the 3 ' translated area as well as a shortening of the 5 'non-translated area having. According to the invention, the cleaning process is further proposed in this way to modify that the cleaning is carried out in a so-called batch process. This batch process is characterized in that an interferon-γ solution with the cation exchange material is evenly distributed, so that a To achieve even occupancy of the entire material, this is the case with Cleaning yield achieved a further increase in yield by a factor of 4.
In the following, the preparation of the plasmid DNA as well as the purification step is used Production of INF-γ described in detail.

Fig. 1 shows the exact sequence of the starting cDNA. The cDNA contains the entire coded area, as well as 5 'and 3' untranslated sequences. This cDNA was obtained from human cells using standard methods. The sequence coding for the mature interferon (179-607) is printed in bold. Signal sequence (110-178) and stop London (608-610) are underlined. This cDNA was sequenced and is long without polyA 1194 base pairs (bp). Nucleotides 182 to 629 were used for the construction of the expression plasmid.
The exact sequence which leads to the shortening of the cDNA is shown in FIG. 2.
The 5 ′ untranslated region (1-109) and the leader sequence of the protein (110- 181) were cleaved with the restriction endonuclease Ava II (Recognition sequence GGA / TCC. 181-185) removed. The initiation codon ATG (for Met) and the codon for the first amino acid were determined by synthetic DNA Pieces (commercially available linkers) added to the 5 'end of the cDNA. The one for that necessary individual steps are general state of the art.
The 3 'untranslated region of the c-DNA was partially cleaved by the Restriction endonuclease Ssp I (recognition sequence AATATT. 627-632) removed. The shortened cDNA was obtained by ligation of a Bam HI linker and a Auxiliary sequence (see attachment) inserted in the express convector.
The plasmid pKK 233-2 was constructed for the expression of interferon-γ in E. coli by E. Amann and J. Brosius (Gene 40 (1985) 183-190). It owns the inducible trc promoter, a multiple cloning site that contains the start codon (ATG) contains as well as terminators for the RNA polymerase.
This plasmid DNA (accession number DSM 6237), in which the 3 'does not translated region except for a residue of 19 nucleotides was expressed Interferon-γ with a factor 4 higher efficiency. The expression is thereby increased from 10 to 40% of the total protein.
According to the invention, it is further proposed to additionally increase the yield increase that the cleaning process is now carried out in a so-called batch process becomes.
The complete 143 amino acid human interferon-γ with an additional one Methionine at the NH₂ terminus is - as shown - at a rate of 40% of the total Expressed in a bacterial cell when its gene is in a construct, in the - as already shown - the 3 ′ non-coding area except for a remainder of 19 Nucleotides was cut out. With this high expression rate, more will be deposited as 90% of the interferon-γs in the form of "inclusion body", which the Present raw material for cleaning.
The inclusion bodies and thus the interferon-γ are replaced by a Denaturing step with guanidinium chloride in solution; in one The interferon-γ is renaturation step by dilution in a phosphate buffer folded into the biologically active form. The result is more than 90% clean interferon γ, is concentrated by a cation exchange process in the "batch process" and further cleaned and reaches another through a further gel filtration step Purity level of more than 95%.
This cation exchange process is usually carried out using column chromatography carried out, d. H. the interferon-γ after the renaturation step in Presence of 0.2 M guanidinium chloride and a phosphate buffer on a column pumped the cation exchange material and binds because of the high Binding capacity of the cation exchange material for the interferon-γ with accordingly high concentration in the upper part of the column. Because of the high Concentration of the bound interferon-γ, the elution yields are very low, i.e. H. only a small part of the bound interferon-γ can with corresponding Saline solutions are detached from the cation exchange material.
The batch process avoids this high concentration on the column. The procedure runs according to the invention as an example: the cation exchange material is evenly so in an interferon-γ solution with 0.1 mg protein per ml 0.2 M Guanidinium chloride stirred in phosphate buffer that the interferon-γ evenly distributed to all cation exchange material and binds the interferon-γ- Protein concentration not more than 2 mg / ml packed cation exchange material is. This process is known as the batch process. That with interferon-γ loaded cation exchange material is also batched with phosphate buffer washed and the interferon-γ then eluted in a batch with 2 M NaCl in phosphate buffer.
Through the batch process, the yield of 10% in the same enrichment Column process increased to 40% in the process according to the invention.
Fig. 3 shows the protein and DNA sequence of human interferon-γ, as obtained with the inventive method.
Die anderen Aminosäuren (1-143) entsprechen denen des natürlichen Interferon-γs.The interferon-γ therefore contains 144 amino acids. The first amino acid, methionine in position zero, is additionally present and was by protein sequencing proven. The other amino acids (1-143) correspond to those of the natural one Interferon-γs.
The method according to the invention therefore yields the same Fermentation conditions 16 times more clean IFN-γ (in mg) than in the prior art Technology. This is the first time that a procedure is available that allows Interferon-γ inexpensively. This can create new applications and a broader therapy can be developed.
