Description
- Die Erfindung bezieht sich auf neue Antitumormittel und insbesondere auf Chinazolin-Derivate, die eine Antitumoraktivität besitzen.
- Eine Gruppe von Antitumormitteln umfaßt die Antimetaboliten, welche Antagonisten von Folsäure sind, wie z.B. Aminopterin und Methotrexat. Eine neuere Verbindung dieser Type, die sich in klinischen Versuchen als vielversprechend erwiesen hat, ist als CB3717 bekannt und in der GB-PS 20 65 653 B beschrieben. Trotz der guten Aussichten zeigt aber CB3717 beim Menschen Symptome von Toxizität, insbesondere hinsichtlich der Leber und der Nieren.
- Es wird angenommen, daß Verbindungen dieser Type dadurch als Antitumormittel wirken, weil sie das Enzym Thymidylatsynthetase inhibieren. Ihre Antitumoraktivität kann in vitro dadurch ermittelt werden, daß man ihre Inhibierungswirkung auf dieses Enzym bestimmt. Eine Bestimmung ist auch in Zellenkulturen möglich, und zwar aufgrund ihrer Inhibierungswirkung auf die Zellenlinie L1210.
- In der europäischen Patentanmeldung 0 204 529 sind gewisse 6-Arylaminomethyl-4-oxochinazolin-Derivate angegeben, die in der 2-Stellung unsubstituiert sind und von denen festgestellt wird, daß sie als Inhibitoren des Enzyms Thymidylatsynthetase wirken und daß sie eine Antikrebsaktivität besitzen. In der europäischen Patentanmeldung 0 239 362, die lediglich unter Artikel 54(3) EPÜ einen Stand der Technik darstellt, sind 6-Arylaminomethyl-4-oxochinazolin-Derivate beschrieben, die in der 2-Stellung einen Substituenten tragen, wie z.B. eine Alkyl-Gruppe, z.B. eine Methyl-Gruppe, und von denen ebenfalls festgestellt wird, daß sie als Inhibitoren des Enzyms Thymidylatsynthetase wirken. Die angegebenen Chinazolin- Derivate sind in jeder der 5-, 7-, 8- und 9-Stellungen unsubstituiert (wobei die 9-Stellung üblicherweise der Methyl- Gruppe in der 6-Stellung am Chinazolin-Ring zugeordnet wird).
- Es wurde nunmehr gefunden, daß gewisse 6-Arylaminomethyl-4- oxochinazolin-Derivate, die in einer oder mehreren der 5-, 7-, 8- und 9-Stellungen substituiert sind, Antitumormittel darstellen. Einige dieser Chinazolin-Derivate sind beträchtlich aktiver als CB3717, und weiterhin sind sie besser wasserstabil als jene Verbindung, was klinisch wichtig ist, um die Leichtigkeit der Ausscheidung durch die Niere zu steigern und damit Toxizitätsymptome zu verringern.
- Gegenstand der Erfindung ist also ein Chinazolin der Formel
- worin
- R¹ für Alkyl, Cycloalkyl, Alkenyl, Alkinyl, Alkoxy oder Alkylthio mit jeweils bis zu 6 Kohlenstoffatomen steht; oder
- R¹ für Aryl, Aryloxy, Arylthio oder Arylalkyl mit jeweils bis zu 10 Kohlenstoffatomen steht; oder
- R¹ für Halogeno, Hydroxy oder Mercapto steht; oder
- R¹ für Alkyl mit bis zu 3 Kohlenstoffatomen steht, das einen oder mehrere Substituenten trägt, die ausgewählt sind aus Halogeno, Hydroxy, Amino, Alkoxy, Alkanoyloxy, Alkylthio, Alkylamino, Dialkylamino und Alkanoylamino mit jeweils bis zu 6 Kohlenstoffatomen und Arylthio, Aroyloxy und Aroylamino mit jeweils bis zu 10 Rohlenstoffatomen; oder
- R¹ für Alkoxy mit bis zu 3 Kohlenstoffatomen steht, das einen oder mehrere Substituenten trägt, die ausgewählt sind aus Hydroxy und Alkoxy mit bis zu 6 Kohlenstoffatomen;
- R² für Wasserstoff, Alkyl, Alkenyl, Alkinyl, Hydroxyalkyl, Alkoxyalkyl, Mercaptoalkyl, Alkylthioalkyl, Halogenoalkyl, Cyanoalkyl, Aminoalkyl, Alkylaminoalkyl, Dialkylaminoalkyl, Alkanoylalkyl, Carboxyalkyl, Carbamoylalkyl oder Alkanoyl mit jeweils bis zu 6 Kohlenstoffatomen oder Aroylalkyl mit bis zu 10 Kohlenstoffatomen steht;
- Ar für Phenylen, Naphthylen oder Heterocyclen steht, das unsubstituiert ist oder das einen oder mehrere Substituenten trägt, die ausgewählt sind aus Halogeno, Phenyl, Cyano, Nitro, Hydroxy, Amino und Carbamoyl sowie aus Alkyl, Alkoxy, Halogenoalkyl, Alkanoylamino, Alkylthio und Alkoxycarbonyl mit jeweils bis zu 6 Kohlenstoffatomen;
- R³ für eine solche Gruppierung steht, daß R³-NH&sub2; eine Aminosäure ist;
- R&sup4; für Wasserstoff oder Alkyl mit bis zu 4 Kohlenstoffatomen steht;
- R&sup5; für Wasserstoff oder Alkyl mit bis zu 4 Kohlenstoffatomen steht; und
- R&sup6;, R&sup7; und R&sup8; jeweils für Wasserstoff, Hydroxy, Alkyl, Alkoxy, Alkylthio, Alkylamino oder Dialkylamino mit jeweils bis zu 4 Kohlenstoffatomen; oder für Phenyl, Halogeno, Nitro, Cyano oder Amino; oder für Alkyl, Alkoxy oder Alkylthio mit jeweils bis zu 4 Kohlenstoffatomen steht, das einen oder mehrere Substituenten trägt, die ausgewählt sind aus Halogeno, Hydroxy, Amino, Alkoxy, Alkylamino und Dialkylamino mit jeweils bis zu 4 Kohlenstoffatomen;
- mit der Maßgabe, daß mindestens eines der Symbole R&sup4;, R&sup5;, R&sup6;, R&sup7; und R&sup8; für etwas anderes als Wasserstoff steht; oder ein pharmazeutisch zulässiges Salz oder ein pharmazeutisch zulässiger Ester davon.
- Es ist zu beobachten, daß ein erfindungsgemäßes Chinazolin ein oder mehrere asymmetrische Kohlenstoffatome besitzen und damit in racemischen und optisch aktiven Formen existieren kann. Es ist darauf hinzuweisen, daß die Erfindung jede racemische Form des Chinazolins und jede optisch aktive Form desselben umfaßt, die eine Antitumoraktivität besitzt. Es ist allgemein bekannt, wie eine racemische Verbindung in seine optisch aktiven Formen getrennt werden kann.
- Ein geeigneter Wert für R¹, R², R&sup4;, R&sup5;, R&sup6;, R&sup7; oder R&sup8;, wenn es für Alkyl steht, oder für einen Alkyl-Substituenten in Ar, ist beispielsweise Methyl, Ethyl, Propyl, Isopropyl oder t-Butyl.
- Ein geeigneter Wert für R¹, wenn es für Cycloalkyl steht, ist beispielsweise Cyclopropyl, Cyclopentyl oder Cyclohexyl.
- Ein geeigneter Wert für R¹ oder R², wenn es für Alkenyl steht, ist beispielsweise Prop-2-enyl, But-2-enyl, But-3- enyl, 2-Methylprop-2-enyl, Hex-2-enyl, Hex-5-enyl oder 2,3- Dimethylbut-2-enyl.
- Ein geeigneter Wert für R¹ oder R², wenn es für Alkinyl steht, ist beispielsweise Prop-2-inyl, But-2-inyl, But-3-inyl, Pent- 2-inyl, 3-Methylpent-4-inyl, Hex-2-inyl oder Hex-5-inyl.
- Ein geeigneter Wert für R¹, R&sup6;, R&sup7; oder R&sup8;, wenn es für Alkoxy oder Alkylthio steht, oder für einen Alkoxy- oder Alkylthio-Substituenten in Ar, ist beispielsweise Methoxy Ethoxy, Isopropoxy, t-Butoxy, Methylthio oder Isopropylthio.
- Ein geeigneter Wert für R¹, wenn es für Aryl oder Arylalkyl steht, ist beispielsweise Phenyl, Tolyl, Benzyl, α-Methylbenzyl oder Phenethyl.
- Ein geeigneter Wert für R¹, wenn es für Aryloxy oder Arylthio steht, ist beispielsweise Phenoxy, Tolyloxy, Phenylthio oder Tolylthio.
- Ein geeigneter Wert für R¹, R&sup6;, R&sup7; oder R&sup8; wenn es für Halogeno steht, ist beispielsweise Fluoro, Chloro, Bromo oder Jodo.

- Ein geeigneter Wert für R², wenn es für Aroylalkyl steht, ist beispielsweise Phenacyl oder 2-Benzoylethyl.
- Ein geeigneter Wert für Ar, wenn es für Heterocyclen steht, ist beispielsweise ein 5gliedriges oder 6gliedriges aromatisches (d.h. vollständig ungesättigtes) Heterocyclen- Diradikal mit bis zu 2 Heteroatomen, die aus Sauerstoff, Stickstoff und Schwefel ausgewählt sind, wie z.B. Thienylen, Pyridylen, Pyrimidinylen, Thiazolylen oder 0xazolylen.
- Ein geeigneter Halogeno-, Halogenoalkyl-, Alkanoylamino- oder Alkoxycarbonyl-Substituent in Ar ist beispielsweise Fluoro, Chloro, Bromo, Jodo, Fluoromethyl, Difluoromethyl, Trifluoromethyl, Acetamido, Propionamido, Isopropionamido, Methoxycarbonyl, Ethoxycarbonyl oder Isobutoxycarbonyl.
- Ein geeigneter Wert für R³ ist eine solche Gruppierung, daß R³-NH&sub2; eine natürlich vorkommende Aminosäure ist, wie z.B. L-Asparaginsäure, L-Glutaminsäure, L-Alanin, L-Phenylalanin, L-Serin, Glycin, L-Ornithin, L-Valin, L-Leucin oder L-Isoleucin. Alternativ kann R³ für eine solche Gruppierung stehen, daß R³-NH&sub2; L-2-Aminobuttersäure oder eine Poly-L- glutaminsäure der Formel
- ist, worin n für eine Ganzzahl Von 1 bis 10 steht.
- Alternativ kann R³ für eine solche Gruppierung stehen, daß R³-NH&sub2; L-Norvalin, L-Alloisoleucin, L-2-Phenylglycin oder L- tert-Leucin ist.
- Um Zweifel zu vermeiden, wird auf folgendes hingewiesen:
- L-0rnithin ist L-2,5-Diaminopentansäure,

- Ein geeignetes pharmazeutisch zulässiges Salz eines erfindungsgemäßen Chinazolins ist beispielsweise ein Säureadditionssalz mit z.B. einer anorganischen oder organischen Säure, wie z.B. Salz-, Bromwasserstoff-, Trifluoroessig- oder Maleinsäure, oder ein Alkalimetall-, beispielsweise Natrium-, Erdalkalimetall-, beispielsweise Calcium-, oder Ammonium-, beispielsweise Tetra(2-hydroxyethyl)ammonium-,-Salz.
- Ein geeigneter pharmazeutisch zulässiger Ester eines erfindungsgemäßen Chinazolins ist beispielsweise ein Ester mit einem aliphatischen Alkohol mit bis zu 6 Kohlenstoffatomen, beispielsweise ein Methyl-, Ethyl- oder t-Butyl-Ester.
- Es wird darauf hingewiesen, daß, wenn R³ zwei Carbonsäure- Reste enthält, d.h., wenn es sich beispielsweise von Asparagin- oder Glutaminsäure ableitet, ein Salz oder ein Ester ein Monosalz oder -ester oder ein Disalz oder -ester sein kann.
- Ein besonderes erfindungsgemäßes Chinazolin ist ein solches mit der obigen Formel,

- Ein besonders bevorzugtes erfindungsgemäßes Chinazolin ist ein solches mit der obigen Formel,

- worin R² für Wasserstoff, Methyl, Ethyl, Prop-2-enyl, Prop- 2-inyl, 2-Hydroxyethyl, 3-Hydroxypropyl, 2-Fluoroethyl, 2-Bromoethyl, Cyanomethyl oder Acetonyl steht;
- worin Ar für 1,4-Phenylen, Thien-2,5-diyl, Pyrid-2,5-diyl oder Thiazol-2,5-diyl steht, das unsubstituiert ist oder das einen oder zwei Substituenten trägt, die ausgewählt sind aus Fluoro, Chloro, Hydroxy, Amino und Methyl;

- In Fällen, bei denen Ar für Pyrid-2,5-diyl oder Thiazol-2,5-diyl steht, ist zu beobachten, daß zwei erfindungsgemäße isomere Chinazoline möglich sind, wobei die Gruppe -CONHR³ sich entweder in der 2- oder 5-Stellung befindet. Es wird darauf hingewiesen, daß die Erfindung alle diese isomeren Formen umfaßt, die eine Antitumoraktivität besitzen.
- Ein weiteres besonders bevorzugtes erfindungsgemäßes Chinazolin ist ein solches mit der obigen Formel,
- worin R¹ für Methyl, Ethyl, Methoxy, Fluoromethyl oder Hydroxymethyl steht;

- N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl) amino]-2-fluorobenzoyl]-L-glutaminsäure, N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-ethylamino]-2-thenoyl]-L-glutaminsäure, N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-methylamino]picolinoyl]-L-glutaminsäure, N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-methylamino]thiazol-2-carbonyl]-L-glutaminsäure, N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl) amino]benzoyl]-L-valin und N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl) amino]-2-fluorobenzoyl]-L-valin.
- Ein erfindungsgemäßes Chinazolin kann durch irgendein Verfahren hergestellt werden, von dem bekannt ist, daß es sich für die Herstellung von chemisch verwandten Verbindungen eignet.
- Ein bevorzugtes Verfahren zur Herstellung eines erfindungsgemäßen Chinazolins besteht darin, daß eine Verbindung der Formel
- mit einer Verbindung der Formel
- HNR²-Ar-CONHR³
- umgesetzt wird, wobei in diesen Verbindungen R¹, R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8; und Ar die oben angegebenen Bedeutungen besitzen, mit der Maßgabe, daß bei Vorliegen einer Hydroxy-Gruppe in R¹, R³, R&sup6;, R&sup7;, R&sup8; oder Ar, bei Vorliegen einer Hydroxyalkyl-Gruppe in R¹, R², R&sup6;, R&sup7; oder R&sup8; bei Vorliegen einer Hydroxyalkoxy-Gruppe in R¹, R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Hydroxyalkylthio-Gruppe in R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Amino-Gruppe in R³, R&sup6;, R&sup7;, R&sup8; oder Ar, bei Vorliegen einer Aminoalkyl-Gruppe in R¹, R², R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Alkylaminoalkyl-Gruppe in R¹, R², R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Alkylamino-, Alkylaminoalkoxy-, Alkylaminoalkylthio-, Aminoalkoxy- oder Aminoalkylthio-Gruppe in R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Carboxy- oder Carboxyalkyl-Gruppe in R² oder R³ oder bei Vorliegen einer Mercapto- oder Mercaptoalkyl-Gruppe in R¹, R² oder R³ jede Amino-, Carboxy- und Mercapto-Gruppe durch eine übliche Schutzgruppe geschützt ist und jede Hydroxy- Gruppe durch eine übliche Schutzgruppe geschützt sein kann oder auch nicht;
- R&sup9; für Wasserstoff oder eine Schutzgruppe steht; und Z für eine Austrittsgruppe steht;
- worauf jede unerwünschte Schutzgruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar oder jede Schutzgruppe R&sup9; durch übliche Maßnahmen entfernt wird
- Eine geeignete Schutzgruppe für eine Hydroxy-Gruppe ist beispielsweise eine veresternde Gruppe, wie z.B. eine Acetyl- oder Benzoyl-Gruppe, die durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, entfernt werden kann. Wenn R¹ und R² keine Alkenyl- oder Alkinyl-Gruppe enthalten, dann kann die Schutzgruppe beispielsweise eine α-Arylalkyl-Gruppe, wie z.B. eine Benzyl-Gruppe, sein, die durch Hydrierung über einen Katalysator, beispielsweise Palladium-auf-Holzkohle, entfernt werden kann.
- Eine geeignete Schutzgruppe für eine Amino-Gruppe ist beispielsweise eine Alkoxycarbonyl-Gruppe, wie z.B. eine t- Butyloxycarbonylgruppe, die durch Behandlung mit einer organischen Säure, wie z.B. Trifluoroessigsäure, entfernt werden kann. Sie kann beispielsweise auch eine Benzyloxycarbonyl- Gruppe sein, die durch Behandlung mit einer Lewis-Säure, wie z.B. Bor-tris(trifluoroacetat), entfernt werden kann.
- Eine geeignete alternative Schutzgruppe für eine primäre Aminogruppe ist beispielsweise eine Phthaloyl-Gruppe, die durch Behandlung mit einem Alkylamin, wie z.B. Dimethylaminopropylamin, oder mit Hydrazin entfernt werden kann.
- Eine geeignete Schutzgruppe für eine Carboxy-Gruppe ist beispielsweise eine veresternde Gruppe, wie z.B. eine Methyl- oder Ethyl-Gruppe, die durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, entfernt werden kann, oder beispielsweise eine t-Butyl-Gruppe, die durch Behandlung mit einer organischen Säure, wie z.B. Trifluoroessigsäure, entfernt werden kann.
- Eine geeignete Schutzgruppe für eine Mercaptogruppe ist beispielsweise eine veresternde Gruppe, wie z.B. eine Acetyl- Gruppe, die durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, entfernt werden kann.
- Ein geeigneter Wert für R&sup9;, wenn es für eine Schutzgruppe steht, ist beispielsweise eine Pivaloyloxymethyl-Gruppe, die durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, entfernt werden kann.
- Z kann beispielsweise eine Halogeno- oder Sulfonyloxy-Gruppe sein, wie z.B. eine Chloro-, Bromo-, Methansulfonyloxy- oder Toluol-p-sulfonyloxy-Gruppe.
- Die Schutzgruppen für die verschiedenen Carboxy-Gruppen in R³ können veresternde Gruppen sein, so daß das Produkt nach der Entfernung von irgendwelchen unerwünschten Schutzgruppen in R¹, R², R³, R&sup6;, R&sup7;, R&sup8;und Ar und irgendeiner Schutzgruppe R&sup9; in die Definition der erfindungsgemäßen Chinazoline fällt. In einem solchen Fall können die Carboxy-Schutzgruppen in R³ entfernt oder beibehalten werden. Alternativ kann eine unterschiedliche Schutzgruppe verwendet werden, die später entfernt wird.
- Ein weiteres bevorzugtes Verfahren zur Herstellung eines erfindungsgemäßen Chinazolins besteht darin, daß eine Säure der Formel
- oder ein reaktives Derivat davon mit einer Verbindung der Formel R³-NH&sub2; umgesetzt wird, wobei in diesen Verbindungen R¹, R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Ar die oben angegebenen Bedeutungen besitzen und jede Mercapto-, Amino-, Alkylamino- und Carboxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt ist und jede Hydroxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt sein kann oder auch nicht; worauf die Schutzgruppen durch übliche Maßnahmen entfernt werden.
- Ein geeignetes reaktives Derivat einer Säure der obigen Formel kann beispielsweise ein Acylhalogenid, wie z.B. ein Acylchlorid, das durch Umsetzung der Säure mit einem anorganischen Säurechlorid, wie z.B. Thionylchlorid, gebildet wird; ein gemischtes Anhydrid, wie z.B. ein Anhydrid, das durch Umsetzung der Säure mit einem Chloroformiat, wie z.B. Isobutylchloroformiat, gebildet wird; ein Acyl-azid, wie z.B. ein Azid, das durch Umsetzung der Säure mit einem Azid, wie z.B. Diphenylphosphoryl-azid, gebildet wird; oder das Produkt der Umsetzung der Säure mit einem Carbodiimid, wie z.B. Dicyclohexylcarbodiimid, sein.
- Die als Ausgangsmaterial verwendete Carbonsäure kann erhalten werden durch Umsetzung einer Verbindung der Formel,
- worin R¹, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Z die oben angegebenen Bedeutungen besitzen, mit einer Verbindung der Formel,
- HNR²-Ar-CO&sub2;R¹&sup0;
- worin R² und Ar die oben angegebenen Bedeutungen besitzen und R¹&sup0; für eine Schutzgruppe steht, die unter Bildung einer Carbonsäure entfernt werden kann.
- R¹&sup0; kann beispielsweise eine Methyl- oder Ethyl-Gruppe sein, die durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, entfernt werden kann, oder R¹&sup0; kann beispielsweise eine t- Butyl-Gruppe sein, die durch Abspaltung mit einer organischen Säure, wie z.B. Trifluoroessigsäure, entfernt werden kann.
- Die Schutzgruppe für die Carboxy-Gruppe in R¹&sup0; kann beispielsweise eine veresternde Gruppe sein, die entfernt werden kann, während die Schutzgruppe für irgendwelche Mercapto-, Amino-, Carboxy- und Hydroxy-Gruppen in R¹, R², R&sup6;, R&sup7;, R&sup8; und Ar beibehalten wird.
- Ein weiteres bevorzugtes Verfahren zur Herstellung eines erfindungsgemäßen Chinazolins, worin R¹ für Alkoxy, Aryloxy oder Alkoxy mit bis zu 3 Kohlenstoffatomen steht, welches einen oder mehrere Substituenten trägt, die ausgewählt sind aus Hydroxy und Alkoxy, besteht darin, daß eine Verbindung der Formel
- mit einer Verbindung der Formel
- HNR²-Ar-CONHR³
- umgesetzt wird, wobei in diesen Verbindungen R¹ die vorstehend zuletzt angegebene Bedeutung besitzt, R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8; und Ar die oben angegebenen Bedeutungen besitzen, mit der Maßgabe, daß jede Mercapto-, Amino-, Alkylamino- und Carboxy-Gruppe in R², R³, R&sup6;, R&sup7;, R&sup8;, und Ar durch eine übliche Schutzgruppe geschützt ist und jede Hydroxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt sein kann oder auch nicht, und Z für eine Austrittsgruppe steht; worauf die Schutzgruppen durch übliche Maßnahmen entfernt werden und die Gruppe R¹, die in der 4-Stellung des Chinazolin-Rings vorliegt, durch Hydrolyse mit einer Base, wie z.B. Natriumhydroxid, abgespalten wird, um ein erfindungsgemäßes Chinazolin herzustellen.
- Ein weiteres bevorzugtes Verfahren zur Herstellung eines ererfindungsgemäßen Chinazolins, worin R¹ für Mercapto, Alkylthio, Arylthio, Alkylthioalkyl oder Arylthioalkyl steht, besteht darin, daß ein Chinazolin der Formel
- worin R¹ für Halogeno oder Halogenoalkyl steht und R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Ar die oben angegebenen Bedeutungen besitzen, mit der Maßgabe, daß jede Mercapto-, Amino-, Alkylamino-, Carboxy- und Hydroxy-Gruppe in R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt sein kann oder alternativ jede Amino-, Alkylamino-, Carboxyund Hydroxy-Gruppe auch nicht geschützt sein muß, mit Thioharnstoff umgesetzt wird, um eine Verbindung herzustellen, worin R¹ für Mercapto steht, oder mit einem Alkyl- oder Arylthiol umgesetzt wird, um eine Verbindung herzustellen, worin R¹ für Alkylthio, Arylthio, Alkylthioalkyl oder Arylthioalkyl steht;
- worauf die Schutzgruppen durch übliche Maßnahmen entfernt werden, wie dies oben angegeben ist.
- Ein weiteres bevorzugtes Verfahren zur Herstellung eines erfindungsgemäßen Chinazolins, worin R1 für Alkylthio steht, besteht darin, daß ein Chinazolin der Formel
- worin R¹ für Mercapto steht und R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Ar die oben angegebenen Bedeutungen besitzen, mit der Maßgabe, daß jede Mercapto-, Amino-, Alkylamino-, Carboxy- und Hydroxy-Gruppe in R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt sein kann, wie dies oben festgestellt wurde oder alternativ jede Amino-, Alkylamino-, Carboxyl- und Hydroxy-Gruppe nicht geschützt sein muß, mit einer Base, wie z.B. Ammoniumhydroxid, umgesetzt wird;
- worauf das erhaltene Thiolat-Salz mit einem Alkylhalogenid, wie z.B. Methyljodid, alkyliert wird;
- worauf die gegebenenfalls anwesenden Schutzgruppen durch übliche Maßnahmen entfernt werden, wie dies oben festgestellt wurde.
- Wie oben bereits festgestellt, besitzen die erfindungsgemäßen Chinazoline eine Antitumoraktivität. Sie können selbst aktiv sein oder eine Promedizin darstellen, die in vivo in eine aktive Verbindung umgewandelt wird. Wie bereits festgestellt, wird angenommen, daß die erfindungsgemäßen Chinazoline als Antitumormittel dadurch wirken, daß sie das Enzym Thymidylatsynthetase inhibieren. Diese Antitumoraktivität kann in vitro dadurch gemessen werden, daß man die Inhibierungswirkung auf dieses Enzym bestimmt, aber sie kann auch in Zellkulturen durch die Inhibierungswirkung auf die Mäuse-Leukämie-Zellenlinie L1210 bestimmt werden (GB-PS 20 65 653 B).
- Im allgemeinen besitzen diejenigen erfindungsgemäßen Chinazoline je nach der Struktur eine Inhibierungswirkung auf Thymidylatsynthetase in den folgenden Konzentrationen: IC&sub5;&sub0; im Bereich von beispielsweise 0,01 bis 1 um. Die Inhibierungseigenschaften auf die L1210-Zellenlinie liegen bei den folgenden Konzentrationen: IC&sub5;&sub0; im Bereich von beispielsweise 0,01 bis 50 um.
- Im allgemeinen besitzen diejenigen erfindungsgemäßen Chinazoline, die besonders bevorzugt sind, eine Inhibierungswirkung auf Thymidylatsynthetase in den folgenden Konzentrationen: IC&sub5;&sub0; im Bereich von beispielsweise 0,01 bis 1 uM. Die Inhibierungseigenschaften auf die L1210-Zellenlinie liegen bei den folgenden Konzentrationen: IC&sub5;&sub0; im Bereich von beispielsweise 0,01 bis 5 um.
- So besitzt beispielsweise das Chinazolin N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2-inyl)- amino]benzoyl]-L-glutaminsäure einen IC&sub5;&sub0; von 0,03 uM gegen Thymidylatsynthetase und einen IC&sub5;&sub0; von 0,18 uM gegen die L1210-Zellenlinie.
- Ein erfindungsgemäßes Chinazolin kann an einen Warmblüter, einschließlich einen Menschen, in Form einer pharmazeutischen Zusammensetzung verabreicht werden, die das Chinazolin gemeinsam mit einem pharmazeutisch zulässigen Verdünnungs- oder Trägermittel enthält.
- Die Zusammensetzung kann eine für orale Verabreichung geeignete Form, wie z.B. eine Tablette oder Kapsel, oder insbesondere eine für parenterale Injektion geeignete Form, wie z.B. eine sterile Lösung, Suspension oder Emulsion, oder eine für topische Verabreichung geeignete Form, wie z.B. eine Salbe oder Creme, oder eine für rektale Verabreichung geeignete Form, wie z.B. ein Suppositorium, aufweisen.
- Die Zusammensetzungen können zusätzlich zum erfindungsgemäßen Chinazolin ein oder mehrere Antitumorstoffe enthalten, die beispielsweise ausgewählt sind aus mitotischen Inhibitoren, wie z.B. Vinblastin; Alkylierungsmitteln, wie z.B. cis-Platin, Carboplatin und Cyclophoshamid; anderen Antimetaboliten, wie z.B. 5-Fluoroacil, Cytosin-arabinosid und Hydroxyharnstoff; intercalierenden Antibiotika, wie z.B. Adriamycin und Bleomycin; Enzymen, wie z.B. Asparaginase; Topoisomerase-Inhibitoren, wie z.B. Etoposid, und biologischen Ansprechmodifiziermitteln, wie z.B. Interferon.
- Das Chinazolin wird normalerweise an einen Warmblüter in einer Dosis im Bereich von 50 bis 5000 mg je m² Körperfläche des Warmblüters verabreicht.
- Die Erfindung wird durch die folgenden Beispiele näher erläutert:
- Die Strukturen aller erfindungsgemäßen Verbindungen wurden durch magnetische Protonenresonanz und Massenspektroskopie sowie durch Elementaranalyse bestätigt. Die magnetischen Protonenresonanzspektren wurden unter Verwendung eines Jeol FX 90Q- oder eines Bruker AM200-Spektrometers bestimmt, der mit einer Feldstärke von 200 MHz arbeitete. Die chemischen Verschiebungen sind in ppm Downfield von Tetramethylsilan als innerer Standard (δ-Skala) angegeben, und Mehrfachspitzen sind wie folgt angegeben: s = Singlett; d = Dublett; d von d's = Dublett von Dubletten; t = Triplett; m = Multiplett. Schnellatomenbombardement-Massenspektraldaten (FAB) wurden unter Verwendung eines VG Analytical MS9-Spektrometers und Xenongas erhalten, und, sofern zweckmäßig, sind auch entweder positive Ionendaten oder negative Ionendaten aufgeführt.
- Die Kolonnenchromatographie wurde unter Verwendung des Silicagels Merck Art 9385 ausgeführt.
- Ein Gemisch aus 6-Bromomethyl-3,4-dihydro-2,7-dimethyl- chinazolin-4-on (1,07 g), N-[p-(Prop-2-inyl)aminobenzoyl]- L-glutaminsäure-diethylester (GB-PS 20 65 653 B; 1,44 g), 2,6-Lutidin (1,4 ml) und Dimethylformamid(8 ml) wurde 24 h unter einer Argonatmosphäre bei 80ºC gerührt. Das Gemisch wurde auf Labortemperatur abgekühlt, eine Mischung aus Eis und Wasser (10 ml) wurde zugegeben, und das erhaltene Gemisch wurde 2 h gerührt und filtriert. Der erhaltene Feststoff wurde getrocknet und durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei Ethylacetat als Eluiermittel verwendet wurde.
- Ein Gemisch aus dem so erhaltenen Produkt (0,4 g), Ethanol (3,5 ml) und wäßriger N-Natriumhydroxid-Lösung (2,2 ml) wurde 20 h bei Labortemperatur gerührt. Das Gemisch wurde zur Trockne eingedampft, der Rückstand wurde in entsalztem Wasser aufgelöst, und die Lösung wurde durch Zugabe von wäßriger 2N Salzsäure auf pH 2,5 angesäuert. Das Gemisch wurde zentrifugiert, und der feste Rückstand wurde mit Wasser (4 x 20 ml) gewaschen und getrocknet. Auf diese Weise N-[p-[N-(3,4- Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2- inyl) amino]benzoyl]-L-glutaminsäure (enthaltend 1 ,5 Equivalente Wasser; 0,2 g) erhalten, Fp 18º C (Zersetzung). NMR-Spektrum: (CD&sub3;SOCD&sub3;) 2.0 (m, 2H, CH&sub2;), 2.35 (breit t, 2H, CH&sub2;CO&sub2;H) , 2.35 (s, 3H, 2-CH&sub3;) , 2.45 (s, 3H, 7-CH&sub3;) , 3.17 (t, 1H, C CH, J=2 Hz), 4.28 (breit s, 2H, CH&sub2;C CH), 4.35 (m, 1H, NHCH) , 4.67 (s, 2H, CH&sub2;N) , 6.82 (d, 2H, aromatisch, J=9 Hz) , 7.45 (s, 1H, 8-H) , 7.75 (s, 1H, 5-H) , 7.77 (d, 2H, aromatisch, J=9 Hz), 8.23 (d, 1H, NH, J=8 Hz);

- Das als Ausgangsmaterial verwendete Chinazolinon wurde wie folgt erhalten:
- Ein Gemisch aus 3,4-Dimethylacetanilid (16,3 g), Ethylcarbamat (14 g), Phosphorpentoxid (30 g) und Xylol (55 ml) wurde heftig mit einem mechanischen Rührer unter einer Argonatmospähre gerührt. Das Gemisch wurde langsam auf annähernd 60ºC erhitzt, worauf eine sichtbare Reaktion unter Wärmeentwicklung und unter einer Zunahme der Viskosität des Gemischs eintrat. Die Temperatur des Gemischs wurde während 90 min auf 150ºC angehoben, und das Gemisch wurde 2 h bei dieser Temperatur gerührt, währenddessen weiteres Phosphorpentoxid (12 g insgesamt) portionsweise zugegeben wurde. Das Gemisch wurde abgekühlt, und das Xylol wurde abdekantiert. Ein Gemisch aus Eis und Wasser (250 ml) wurde zum Rückstand zugegeben, und das Gemisch wurde 30 min gerührt und filtriert. Der Feststoff wurde durch Dünnschichtchromatographie unter Verwendung von Ethylacetat als Lösungsmittel analysiert, und wenn er irgendein Produkt enthielt, dann wurde dieses durch Chromatographie gereinigt, wie es unten für das erhaltene Harz beschrieben ist. Das Filtrat wurde im Eisbad auf eine Temperatur von weniger als 5ºC abgekühlt, und die Acidität der Lösung wurde durch Zugabe einer konzentrierten wäßrigen Natriumhydroxidlösung auf pH 5 verringert. Das Gemisch wurde mit Ethylacetat (2 x 50 ml) extrahiert, und die vereinigten Extrakte wurden über Magnesiumsulfat getrocknet, filtriert und eingedampft. Der Rückstand wurde durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei ein 1:1-Gemisch V/V aus Methylenchlorid und Ethylacetat als Eluiermittel verwendet wurde. Auf diese Weise wurden in der Reihenfolge der Elution 3,4-Dihydro-2,6,7-trimethylchinazolin-4-on (2 g) und 3,4-Dihydro-2,5,6-trimethylchinazolin-4-on (2 g) erhalten.
- Ein Gemisch aus der ersteren Verbindung (0,94 g), N-Bromosuccinimid (0,9 g), Benzoylperoxid (10 mg) und Chloroform (35 ml) wurde 28 h auf Rückfluß erhitzt. Das Gemisch wurde auf Labortemperatur abgekühlt und filtriert. Der Feststoff wurde mit Chloroform und mit einer warmen Lösung von Ethylacetat gewaschen. Auf diese Weise wurde 6-Bromomethyl-3,4- dihydro-2,7-dimethylchinazolin-4-on (0,56 g), Fp 340ºC, erhalten.
- Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, außer daß N-[2-Fluoro-4-(prop-2-inyl)aminobenzoyl]-L-glutaminsäure-diethylester (hergestellt gemäß GB 21 88 319 A) anstelle von N-[p-(prop-2-inyl)aminobenzoyl]-L-glutaminsäure-diethylester verwendet wurde, Auf diese Weise wurde N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2-inyl)amino]-2-fluorobenzoyl]-L-glutaminsäure (enthaltend 1 Euivalent Wasser), Fp 240-245ºC, erhalten.
- Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, wobei das entsprechende 6-Bromomethylchinazolinon anstelle von 6-Bromomethyl-3,4-dihydro-2,7-dimethylchinazolin-4-on und das entsprechende Diethyl-L-glutamat als Ausgangsmaterialien verwendet wurden. Auf diese Weise wurden die in der folgenden Tabelle beschriebenen Verbindungen erhalten, deren Struktur durch magnetische Protonenresonanz und Massenspektroskopie sowie durch Elementaranalyse bestätigt wurde.
- Das entsprechende 6-Bromomethylchinazolinon wurde unter Verwendung des in Beispiel 1 bei der Herstellung von Ausgangsmaterialien beschriebenen Verfahrens hergestellt, außer daß das entsprechende Acetanilid anstelle von 3,4-Dimethylacetanilid. - Anmerkung (1): 2-Chloro-4-methylacetanilid (18 g) wurde als Ausgangsmaterial verwendet, und nur ein Isomer, nämlich 8-Chloro- 2,6-dimethylchinazolin-4-on (0,6 g) wurde beim ersten Teil des in Beispiel 1 beschriebenen Verfahrens, der die Herstellung von Ausgangsmaterialien betrifft, erhalten. Im zweiten Teil dieses Verfahrens wurde das Gemisch aus Chinazolinon, N-Bromosuccinimid, Benzoylperoxid und Chloroform 66 h auf Rückfluß erhitzt.
- Anmerkung (2): N-[p-Methylaminobenzoyl]-L-glutaminsäure-diethylester (Journal of Heterocyclic Chemistry, 1975, 12, 1283) wurde verwendet.

- Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, wobei 6-(1-Bromoethyl)-3,4-dihydro-2-methylchinazolin-4-on als Ausgangsmaterial anstelle von 6-Bromomethyl-3,4-dihydro- 2,7-dimethylchinazolin-4-on verwendet wurde. Auf diese Weise wurde N-[p-[N-[1-(3,4-Dihydro-2-methyl-4-oxochinazolin-6-yl)- ethyl]-N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure als Monohydrat, Fp 157ºC, erhalten.
- Das als Ausgangsmaterial verwendete Chinazolin wurde unter Verwendung des die Herstellung von Ausgangsmaterialien betreffenden Teils von Beispiel 1 erhalten, außer daß p-Ethylacetanilid anstelle von 3,4-Dimethylacetanilid verwendet wurde. Es wurde nur ein Isomer, nämlich 6-Ethyl-2-methylchinazolin-4-on, aus dem ersten Teil des Verfahrens erhalten.
- Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, außer daß 6-(1-Bromoethyl)-3,4-dihydro-2-methylchinazolin- 4-on und N-[p-Methylaminobenzoyl]-L-glutaminsäure-diethylester als Ausgangsmaterialien verwendet wurden. Auf diese Weise wurde N-[p-[N-[1-(3,4-Dihydro-2-methyl-4-oxochinazolin- 6-yl)ethyl]-N-methylamino]benzoyl]-L-glutaminsäure (enthaltend 1,5 Equivalente Wasser), Fp 165-185ºC, erhalten.
- Das in Beispiel beschriebene Verfahren wurde wiederholt, wobei das entsprechende 6-Bromomethylchinazolin-4-on und das entsprechende Diethyl-L-glutamat als Ausgangsmaterialien verwendet wurden. Sofern nichts anderes angegeben ist, wurde das entsprechende L-Glutamat erhalten, wie es in GB 21 88 319 A beschrieben ist. Auf diese Weise wurden die in der folgenden Tabelle beschriebenen Verbindungen erhalten, deren Struktur durch magnetische Protonenresonanz und Massenspektroskopie sowie durch Elementaranalyse bestätigt wurde. - Anmerkung (1): Die -CONH- Gruppe befindet sich in der 2-Stellung des Pyridin-Ringes.
- Ein Gemisch aus 6-Bromomethyl-3,4-dihydro-2,8-dimethyl-3- pivaloyloxymethylchinazolin-4-on (0,63 g), N-[p-(Prop-2-inyl)- aminobenzoyl]-L-glutaminsäure-diethylester (0,62 g), 2,6- Lutidin (0,6 ml) und Dimethylformamid (6 ml) wurde 19 h unter einer Argonatmosphäre bei 80ºC gerührt. Das Gemisch wurde auf Labortemperatur abgekühlt, ein Gemisch aus Eis und Wasser (10 ml) wurde zugegeben, und das erhaltene Gemisch wurde 2 h gerührt und filtriert. Der erhaltene Feststoff wurde getrocknet und durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei Ethylacetat als Eluiermittel verwendet wurde.
- Ein Gemisch aus dem so erhaltenen Produkt (0,56 g), Ethanol (4,0 ml) und wäßriger N Natriumhydroxid-Lösung (3,4 ml) wurde 5 h bei Labortemperatur gerührt. Das Gemisch wurde zur Trockne eingedampft, der Rückstand wurde in entsalztem Wasser aufgelöst, und die Lösung wurde durch Zugabe von wäßriger 2N Salzsäure auf pH 2,5 angesäuert. Das Gemisch wurde zentrifugiert, und der feste Rückstand wurde mit Wasser (5 x 20 ml) gewaschen und getrocknet. Auf diese Weise wurde N-[p-[N- (3,4-Dihydro-2,8-dimethyl-4-oxochinazolin-6-ylmethyl)-N- (prop-2-inyl) amino]benzoyl]-L-glutaminsäure (enthaltend 2 Equivalente Wasser; 0,13 g), Fp 165-173ºC, erhalten.

- Das als Ausgangsmaterial verwendete Chinazolinon wurde unter Verwendung des ersten Teils des in Beispiel 1 beschriebenen Verfahrens, der die Herstellung von Ausgangsmaterialien betrifft, erhalten, außer daß 2,4-Dimethylacetanilid anstelle von 3,4-Dimethylacetanilid verwendet wurde. Es wurde nur ein Isomer, nämlich 3,4-Dihydro-2,6,8-trimethylchinazolin-4-on, erhalten. Natriumhydrid (2,39 g) wurde zu einer gerührten Suspension des so erhaltenen Produkts (9,3 g) in Dimethylformamid (100 ml) zugegeben, und das Gemisch wurde 1 h bei Labortemperatur gerührt. Eine Lösung von Chloromethylpivalat (8,23 ml) in Dimethylformamid (20 ml) wurde zugegeben, und das Gemisch wurde 18 h bei Labortemperatur gerührt und dann auf ein Gemisch aus Eis und Wasser (350 ml) geschüttet. Die Ausfällung wurde abfiltriert, getrocknet und durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei Methylenchlorid als Eluiermittel verwendet wurde. Auf diese Weise wurde 3,4-Dihydro-2,6,8-trimethyl-3-pivaloyloxymethylchinazolin-4-on (8,7 g) erhalten.
- Ein Gemisch aus dem so erhaltenen produkt (7,8 g), N-Bromosuccinimid (4,6 g) und Tetrachlorkohlenstoff (200 ml) wurde mit einer 275 Watt-Sonnenlampe 5 h bestrahlt und dann 17 h bei 4ºC gelagert. Die Ausfällung wurde abfiltriert, getrocknet und durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei Ethylacetat und Petrolether (Kp 60-80ºC, 1;4 V/V) als Eluiermittel verwendet wurde. Auf diese Weise wurde 6-Bromomethyl-3,4-dihydro-2,8-dimethyl-3-pivaloyloxymethylchinazolin-4-on (0,67 g) und 6,8-Di(bromomethyl)- 3,4-dihydro-3-pivaloyloxymethylchinazolin-4-on (0,36 g) erhalten.
- Das in Beispiel 5 beschriebene Verfahren wurde wiederholt, wobei das entsprechende 6-Bromomethylchinazolinon anstelle von 6-Bromomethyl-3,4-dihydro-2,8-dimethyl-3-pivaloyloxymethylchinazolin-4-on und das entsprechende Diethyl-L- glutamat als Ausgangsmaterialien verwendet wurden. Sofern nichts anderes angegeben ist, wurde das entsprechende Diethyl-L-glutamat so erhalten, wie es in GB 21 88 319 A beschrieben ist. Auf diese Weise wurden die in der folgenden Tabelle beschriebenen Verbindungen erhalten, deren Struktur durch magnetische Protonenresonanz und Massenspektroskopie sowie durch Elementaranalyse bestätigt wurde. Das entsprechende 6-Bromomethylchinazolinon wurde unter Verwendung des in dem Teil von Beispiel 5 beschriebenen Verfahrens, der die Herstellung von Ausgangsmaterialien betrifft, hergestellt, außer daß das entsprechende Acetanilid anstelle von 2,4-Dimethylacetanilid verwendet wurde. - Anmerkung (1): 3-Methoxy-4-methylacetanilid (25,1 g) wurde anstelle von 3,4-Dimethylacetanilid als Ausgangsmaterial im ersten Teil des in Beispiel 1 beschriebenen Verfahrens, der die Herstellung von Ausgangsmaterialien betrifft, verwendet, wobei 2,6-Dimethyl-7-methoxychinazolin-4-on (18,6 g) erhalten wurde.
- Ein Gemisch aus 6-Bromomethyl-3,4-dihydro-2,7-dimethylchinazolin-4-on (0,27 g), N-[5-Methylaminothiazol-2-carbonyl]- L-glutaminsäure-diethylester (0,56 g), Calciumcarbonat (0,2 g) und Dimethylformamid (4 ml) wurde 2,5 h unter einer Argonatmosphäre bei 95ºC gerührt. Das Gemisch wurde abgekühlt und filtriert, und das Filtrat wurde zur Trockne eingedampft. Der Rückstand wurde durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei zunächst Methylenchlorid als Eluiermittel verwendet wurde und dann die Polarität des Lösungsmittels stufenweise bis zu einem 25:2 V/V Gemisch aus Methylenchlorid und Ethanol gesteigert wurde.
- Ein Gemisch aus dem so erhaltenen Produkt (0,37 g), Ethanol (8 ml), Wasser (8 ml) und wäßriger N-Natriumhydroxid-Lösung (4,2 ml) wurde unter einer Argonatmosphäre 8 h bei Labortemperatur gerührt. Das Gemisch wurde bis zu einem Volumen von annähernd 5 ml eingedampft und in ein Zentrifugenröhrchen filtriert. Das Filtrat wurde mit 2N Salzsäure auf pH 3 angesäuert. Die erhaltene Ausfällung wurde durch Zentrifugieren isoliert, viermal mit Wasser gewaschen und getrocknet. Auf diese Weise wurde N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-N-methylamino]thiazol-2-carbonyl]- L-glutaminsäure (enthaltend 1,25 Equivalente Wasser; 0,23 g), Fp 185-190ºC, erhalten.

- Der als Ausgangsmaterial verwendete N-[5-Methylaminothiazol- 2-carbonyl]-L-glutaminsäure-diethylester wurde wie folgt erhalten:
- 0xalylchlorid (13 ml) wurde tropfenweise zu einer gerührten Suspension von 5-Nitrothiazol-2-carbonsäure (12,9 g; Chem. Ber. 1973, 106, 722) in einem Gemisch aus Methylenchlorid (80 ml) und Dimethylformamid (20 ml) zugegeben. Das Gemisch wurde bei Labortemperatur 30 min gerührt und zur Trockne eingedampf, wobei 5-Nitrothiazol-2-carbonylchlorid erhalten wurde.
- Das so erhaltene Produkt wurde in Methylenchlorid (150 ml) aufgelöst und zu einem Gemisch Von Diethyl-L-glutamat-hydrochlorid (35,4 g), Triethylamin (51,2 g) und Methylenchlorid (250 ml), welches in einem Bad aus Eis und Wasser gekühlt wurde, zugegeben. Die Zugabegeschwindigkeit wurde dabei so eingestellt, daß die Temperatur des Reaktionsgemischs unter 15ºC blieb. Das Gemisch wurde auf Raumtemperatur erwärmen gelassen und 16 h gerührt. Das Gemisch wurde mit Wasser (2 x 300 ml) gewaschen, über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft. Der Rückstand wurde durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei zunächst Methylenchlorid als Eluiermittel verwendet wurde und hierauf die Polarität des Lösungsmittels stufenweise bis zu einem 19:1 V/V-Gemisch aus Methylenchlorid und Ethylacetat gesteigert wurde. Auf diese Weise wurde N-[5- Nitrothiazol-2-carbonyl]-L-glutaminsäure-diethylester als Gummi (24,3 g) erhalten.
- Ein Gemisch aus dem so erhaltenen Produkt (4 g), Eisenpulver (10 g) und Eisessig (44 ml) wurde heftig bei 80ºC 30 min gerührt. Das Gemisch wurde auf Labortemperatur abgekühlt, in Wasser (100 ml) geschüttet und mit Methylenchlorid (2 x 200 ml) extrahiert. Die vereinigten organischen Extrakte wurden mit Wasser (1 x 100 ml) gewaschen, über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft, wobei N-[5-Aminothiazol-2-carbonyl]-L-glutaminsäure-diethylester als Gummi (3,6 g) erhalten wurde.
- Ein Gemisch aus dem so erhaltenen Produkt (3,6 g), Methyljodid (2 ml) und Dimethylformamid (10 ml) wurde bei 60ºC 1 h unter einer Argonatmosphäre gerührt. Eine zweite Portion Methyljodid (1 ml) wurde zugegeben, und das Gemisch wurde dann noch 1 h bei 60ºC gerührt. Das Gemisch wurde abgekühlt, in eine gesättigte wäßrige Natriumbicarbonat-Lösung (50 ml) geschüttet und mit Ethylacetat (3 x 30 ml) extrahiert. Die vereinigten Extrakte wurden mit Wasser gewaschen, über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft. Der Rückstand wurde durch Chromatographie auf einer Silicagelkolonne gereinigt, wobei zunächst Methylenchlorid als Eluiermittel Verwendet wurde und hierauf die Polarität des Lösungsmittels stufenweise bis zu einem 10:1 V/V-Gemisch aus Methylenchlorid und Ethylacetat gesteigert wurde. Auf diese Weise wurden N-[5-Dimethylaminothiazol-2-carbonyl]-L- glutaminsäure-diethylester (0,84 g) und N-[5-Methylaminothiazol-2-carbonyl]-L-glutaminsäure-diethylester (1,1 g) erhalten.
- Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, außer daß N-[p-(Prop-2-inyl)aminobenzoyl]-L-valin-ethylester anstelle von N-[p-(Prop-2-inyl) aminobenzoyl]-L-glutaminsäure-diethylester verwendet wurde. Auf diese Weise wurde N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl)amino]benzoyl]-L-valin (enthaltend ein Equivalent Wasser), Fp 250-255ºC, erhalten.

- Der als Ausgangsmaterial verwendete N-[p-(Prop-2-inyl)aminobenzoyl]-L-valin-ethylester wurde aus Ethylvalinat-hydrochlorid in analoger Weise hergestellt, wie es für die Herstellung von N-[p-(Prop-2-inyl) aminobenzoyl]-L-glutaminsäurediethylester aus Diethylglutamat-hydrochlorid in J. Med. Chem., 1986, 29, 1114, beschrieben ist.
- Das in Beispiel 8 beschriebene Verfahren wurde wiederholt, außer daß N-[2-Fluoro-4-(prop-2-inyl) aminobenzoyl]-L-valin- methylester anstelle von N-[p-(Prop-2-inyl)aminobenzoyl]-L- valin-ethylester verwendet wurde. Auf diese Weise wurde N- [4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl)amino]-2-fluorobenzoyl]-L-valin-methylester, Fp 237-240ºC, und dann nach der zweiten Stufe des Verfahrens N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)- N-(prop-2-inyl)amino]-2-fluorobenzoyl]-L-valin (enthaltend 0,5 Equivalente Wasser), Fp 255-257ºC (Zersetzung), erhalten.
- Der als Ausgangsmaterial verwendete N-[2-Fluoro-4-(prop-2- inyl)aminobenzoyl]-L-valin-methylester wurde aus Methylvalinathydrochlorid in einer ähnlichen Weise erhalten, wie es für die Herstellung von N-[2-Fluoro-4-(prop-2-inyl)aminobenzoyl]- L-glutaminsäure-diethylester aus Diethylglutamat-hydrochlorid in GB-PS 21 75 903 und 21 88 319 A beschrieben ist.
- Diphenylphosphoryl-azid (0,41 g) und Triethylamin (0,55 ml) wurden aufeinanderfolgend zu einem Gemisch aus P-[N-(3,4- Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2- inyl)amino]benzoesäure (als Trifluoroessigsäuresalz; 0,48 g) und Dimethylformamid (20 ml) zugegeben, und das Gemisch wurde 18 h bei Labortemperatur gerührt. Das Gemisch wurde filtriert. 1,8-Diazabicyclo[5,4,0]undec-7-en (0,6 ml) wurde zu einem Gemisch aus dem so erhaltenen Feststoff, L-tert-Leucin (0,26 g) und Dimethylformamid (20 ml) zugegeben, und das Gemisch wurde 18 h bei Labortemperatur gerührt. Das Gemisch wurde auf ein Volumen von annähernd 5 ml eingedampft, in ein Gemisch aus Eiswasser (50 ml) geschüttet, mit wäßriger 0,2 N-Salzsäure auf pH 3 angesäuert und zentrifugiert. Der feste Rückstand wurde mit Wasser (5 x 20 ml) gewaschen und getrocknet. Auf diese Weise wurde N-[p-[N-(3,4-Dihydro-2,7- dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2-inyl)amino]- benzoyl]-L-tert-leucin (enthaltend 1,5 Equivalente Wasser, 0,33 g), Fp 233-236ºC, erhalten.

- Elementaranalyse: gefunden C, 64,4; H, 6,3; N, 11,0; C&sub2;&sub7;H&sub3;&sub0;N&sub4;O&sub4;.1,5H&sub2;O erfordert C, 64,6; H, 6,6; N, 11,2 %.
- Die als Ausgangsmaterial verwendete p-[N-(3,4-Dihydro-2,7- dimethyl-4-oxochinazolin-6-ylmethyl)-N-(prop-2-inyl) amino]- benzoesäure (als ihr Trifluoroessigsäuresalz) wurde unter Verwendung des ersten Teils des in Beispiel 1 beschriebenen Verfahrens erhalten, außer daß p-(Prop-2-inyl) aminobenzoesäure-t-butylester (erhalten nach der Vorschrift von GB-PS 21 88 319 A) anstelle von N-[p-(Prop-2-inyl)aminobenzoyl]- L-glutaminsäure-diethylester verwendet wurde, und daß hierauf ein Gemisch aus dem so erhaltenen Produkt (2,8 g) und Trifluoroessigsäure (20 ml) bei Labortemperatur 15 min zusammen gerührt und zur Trockne eingedampft wurde.
- Das in Beispiel 10 beschriebene Verfahren wurde wiederholt, wobei L-Phenylglycin (d.h. (S) -α-Aminophenylessigsäure) anstelle von L-tert-Leucin verwendet wurde. Auf diese Weise wurde N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6- ylmethyl)-N-(prop-2-inyl)amino]benzoyl]-L-phenylglycin (enthaltend 0,75 Equivalente Wasser), Fp 199-201ºC, erhalten.
Claims (10)
1. Chinazolin der Formel
worin
R¹ für Alkyl, Cycloalkyl, Alkenyl, Alkinyl, Alkoxy oder
Alkylthio mit jeweils bis zu 6 Kohlenstoffatomen steht; oder
R¹ für Aryl, Aryloxy, Arylthio oder Arylalkyl mit jeweils
bis zu 10 Kohlenstoffatomen steht; oder
R¹ für Halogeno, Hydroxy oder Mercapto steht; oder
R¹ für Alkyl mit bis zu 3 Kohlenstoffatomen steht, das einen
oder mehrere Substituenten trägt, die ausgewählt sind aus
Halogeno, Hydroxy, Amino, Alkoxy, Alkanoyloxy, Alkylthio,
Alkylamino, Dialkylamino und Alkanoylamino mit jeweils bis
zu 6 Kohlenstoffatomen und Arylthio, Aroyloxy und Aroylamino
mit jeweils bis zu 10 Kohlenstoffatomen; oder
R¹ für Alkoxy mit bis zu 3 Kohlenstoffatomen steht, das einen
oder mehrere Substituenten trägt, die ausgewählt sind aus
Hydroxy und Alkoxy mit bis zu 6 Kohlenstoffatomen;
R² für Wasserstoff, Alkyl, Alkenyl, Alkinyl, Hydroxyalkyl,
Alkoxyalkyl, Mercaptoalkyl, Alkylthioalkyl, Halogenoalkyl,
Cyanoalkyl, Aminoalkyl, Alkylaminoalkyl, Dialkylaminoalkyl,
Alkanoylalkyl, Carboxyalkyl, Carbamoylalkyl oder Alkanoyl
mit jeweils bis zu 6 Kohlenstoffatomen oder Aroylalkyl mit
bis zu 10 Kohlenstoffatomen steht;
Ar für Phenylen, Naphthylen oder Heterocyclen steht, das
unsubstituiert ist oder das einen oder mehrere Substituenten
trägt, die ausgewählt sind aus Halogeno, Phenyl, Cyano,
Nitro, Hydroxy, Amino und Carbamoyl sowie aus Alkyl, Alkoxy,
Halogenoalkyl, Alkanoylamino, Alkylthio und Alkoxycarbonyl
mit jeweils bis zu 6 Kohlenstoffatomen;
R³ für eine solche Gruppierung steht, daß R³-NH&sub2; eine
Aminosäure ist;
R&sup4; für Wasserstoff oder Alkyl mit bis zu 4 Kohlenstoffatomen
steht;
R&sup5; für Wasserstoff oder Alkyl mit bis zu 4 Kohlenstoffatomen
steht; und
R&sup6;, R&sup7; und R&sup8; jeweils für Wasserstoff, Hydroxy, Alkyl,
Alkoxy, Alkylthio, Alkylamino oder Dialkylamino mit jeweils
bis zu 4 Kohlenstoffatomen; oder für Phenyl, Halogeno,
Nitro, Cyano oder Amino; oder für Alkyl, Alkoxy oder
Alkylthio mit jeweils bis zu 4 Kohlenstoffatomen steht, das einen
oder mehrere Substituenten trägt, die ausgewählt sind aus
Halogeno, Hydroxy, Amino, Alkoxy, Alkylamino und
Dialkylamino mit jeweils bis zu 4 Kohlenstoffatomen;
mit der Maßgabe, daß mindestens eines der Symbole R&sup4;, R&sup5;,
R&sup6;, R&sup7; und R&sup8; für etwas anderes als Wasserstoff steht;
oder ein pharmazeutisch zulässiges Salz oder ein
pharmazeutisch zulässiger Ester davon.
2. Chinazolin nach Anspruch 1, worin
R¹ für Methyl, Ethyl, Isopropyl, Cyclopropyl, Prop-2-enyl,
Prop-2-inyl, Methoxy, Ethoxy, Methylthio, Phenyl, Tolyl,
Phenoxy, Benzyl, Phenethyl, Chloro, Bromo, Hydroxy, Mercapto,
Fluoromethyl, Difluoromethyl, Trifluoromethyl,
2-Fluoroethyl, Chloromethyl, Hydroxymethyl, 2-Hydroxyethyl,
Aminomethyl, Methoxymethyl, Acetoxymethyl, Methylthiomethyl,
Methylaminomethyl, Dimethylaminomethyl, Acetamidomethyl,
2-Hydroxyethoxy, 2-Methoxyethoxy oder 2-Ethoxyethoxy steht;
R² für Wasserstoff, Methyl, Ethyl, Propyl, Prop-2-enyl,
But-2-enyl, Prop-2-inyl, But-2-inyl, 2-Hydroxyethyl, 3-
Hydroxypropyl, 2-Methoxyethyl, 3-Methoxypropyl,
2-Mercaptoethyl, 2-Methylthioethyl, 2-Fluoroethyl, 2-Chloroethyl, 2-
Bromoethyl, 3-Fluoropropyl, Cyanomethyl, 2-Cyanoethyl, 2-
Aminoethyl, 2-Methylaminoethyl, 2-Dimethylaminoethyl,
Acetonyl, Carboxymethyl, Carbamoylmethyl oder Acetyl steht;
Ar für 1,4-Phenylen, Thienylen, Pyridylen, Pyrimidinylen,
Thiazolylen oder 0xazolylen steht, das unsubstituiert ist
oder das einen oder zwei Substituenten trägt, die
ausgewählt sind aus Fluoro, Chloro, Bromo, Phenyl, Cyano, Nitro,
Hydroxy, Amino, Carbamoyl, Methyl, Ethyl, Methoxy, Ethoxy
Fluoromethyl, Difluoromethyl, Trifluoromethyl und
Acetamido;
R³ für eine solche Gruppierung steht, daß R³-NH&sub2;
L-Glutaminsäure, Glycin, L-Alanin, L-Phenylalanin, L-Serin, L-0rnithin,
L-Asparaginsäure, L-Valin, L-Leucin, L-Isoleucin,
L-2-Aminobuttersäure, L-Norvalin, L-Alloisoleucin, L-2-Phenylglycin
oder L-tert-Leucin ist;
R&sup4; für Wasserstoff, Methyl oder Ethyl steht;
R&sup5; für Wasserstoff, Methyl oder Ethyl steht; und
R&sup6;, R&sup7; und R&sup8; jeweils für Wasserstoff, Hydroxy, Methyl, Ethyl,
Methoxy, Ethoxy, Methylthio, Methylamino, Dimethylamino,
Phenyl, Fluoro, Chloro, Bromo, Nitro, Cyano, Amino,

3. Chinazolin nach Anspruch 1, worin
R¹ für Methyl, Ethyl, Methoxy, Fluoromethyl oder
Hydroxymethyl steht;
R² für Wasserstoff, Methyl, Ethyl, Prop-2-enyl, Prop-2-inyl,
2-Hydroxyethyl, 3-Hydroxypropyl , 2-Fluoroethyl,
2-Bromoethyl, Cyanomethyl oder Acetonyl steht;
Ar für 1,4-Phenylen, Thien-2,5-diyl, Pyrid-2,5-diyl oder
Thiazol-2,5-diyl steht, das unsubstituiert ist oder das einen
oder zwei Substituenten trägt, die ausgwwählt sind aus Fluoro,
Chloro, Hydroxy, Amino und Methyl;
R³ für eine solche Gruppierung steht, daß R³-NH&sub2;
L-Glutaminsäure, L-Valin, L-Leucin, L-Isoleucin, L-2-Aminobuttersäure,
L-Norvalin, L-Alloisoleucin, L-2-Phenylglycin oder L-tert-
Leucin ist;
R&sup4; für Wasserstoff oder Methyl steht;
R&sup5; für Wasserstoff steht;
R&sup6; für Wasserstoff, Methyl, Methoxy, Fluoro oder Chloro
steht;
R&sup7; für Wasserstoff, Methyl, Methoxy, Fluoro oder Chloro
steht; und
R&sup8; für Wasserstoff, Methyl, Methoxy, Fluoro oder Chloro
steht;
mit der Maßgabe, daß mindestens eines der Symbole R&sup4;, R&sup6;,
R&sup7; und R&sup8; für etwas anderes als Wasserstoff steht.
4. Chinazolin nach Anspruch 1, worin
R¹ für Methyl, Ethyl, Methoxy, Fluoromethyl oder
Hydroxymethyl steht;
R² für Wasserstoff, Methyl, Ethyl, Prop-2-enyl, Prop-2-inyl,
2-Hydroxyethyl, 3-Hydroxypropyl, 2-Fluoroethyl oder Acetonyl
steht;
Ar für 1,4-Phenylen, Thien-2,5-diyl, Pyrid-2,5-diyl oder
2-Fluoro-1,4-phenylen steht;
R³ für eine solche Gruppierung steht, daß R³-NH&sub2;
L-Glutaminsäure ist;
R&sup4; für Wasserstoff oder Methyl steht;
R&sup5; für Wasserstoff steht;
R&sup6; für Wasserstoff oder Chloro steht;
R&sup7; für Wasserstoff, Methyl, Fluoro oder Chloro steht; und
R&sup8; für Wasserstoff, Methoxy oder Chloro steht;
mit der Maßgabe, daß mindestens eines der Symbole R&sup4;, R&sup6;,
R&sup7; und R&sup8; für etwas anderes als Wasserstoff steht.
5. Chinazolin nach Anspruch 1, worin
R¹ für Methyl steht;
R² für Wasserstoff, Methyl, Ethyl, Prop-2-inyl oder
2-Fluoroethyl steht;
Ar für 1,4-Phenylen oder Thien-2,5-diyl; oder für Pyrid-2,5-
diyl oder Thiazol-2,5-diyl, wobei sich die Gruppe -CONHR³
jeweils in der 2-Stellung befindet; oder für 2-Fluoro-1,4-
phenylen, wobei sich die Gruppe -CONHR³ in der 1-Stellung
befindet, steht;
R³ für eine solche Gruppierung steht, daß R³-NH&sub2;
L-Glutaminsäure, L-Valin, L-2-Phenylglycin oder L-tert-Leucin ist;
R&sup4; für Wasserstoff oder Methyl steht;
R&sup5; für Wasserstoff steht;
R&sup6; für Wasserstoff oder Chloro steht;
R&sup7; fur Wasserstoff, Methyl, Methoxy, Fluoro oder Chloro
steht; und

6. Die Verbindung
N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure,
N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-methylamino]benzoyl]-L-glutaminsäure,
N-[p-[N-(7-Fluoro-3,4-dihydro-2-methyl-4-oxochinazolin-6-
ylmethyl-N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure,
N-[p-[N-(7-Chloro-3,4-dihydro-2-methyl-4-oxochinazolin-6-
ylmethyl)-N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure,
N-[p-[N-[1-(3,4-Dihydro-2-methyl-4-oxochinazolin-6-yl)-
ylmethyl)-N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure,
N-[p-[N-[1-(3,4-Dihydro-2-methyl-4-oxochinazolin-6-yl)-
ethyl]-N-(prop-2-inyl)amino]benzoyl]-L-glutaminsäure oder
N-[p-[N-[1-(3,4-Dihydro-2-methyl-4-oxochinazolin-6-yl)-

7. Die Verbindung
N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-(prop-2-inyl)amino]-2-fluorobenzoyl]-L-glutaminsäure,
N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-ethylamino]-2-thenoyl]-L-glutaminsäure,
N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-methylamino]picolinoyl]-L-glutaminsäure,
N-[5-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-methylamino]thiazol-2-carbonyl]-L-glutaminsäure,
N-[p-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-
N-(prop-2-inyl)amino]benzoyl]-L-valin oder
N-[4-[N-(3,4-Dihydro-2,7-dimethyl-4-oxochinazolin-6-ylmethyl)-

8. Verfahren zur Herstellung eines Chinazolins nach
Anspruch 1 oder eines pharmazeutisch zulässigen Salzes oder
Esters davon, bei welchem
(a) eine Verbindung der Formel
mit einer Verbindung der Formel
HNR²-Ar-CONHR³
umgesetzt wird, wobei in diesen Verbindungen R¹, R², R³, R&sup4;,
R&sup5;, R&sup6;, R&sup7;, R&sup8; und Ar die in Anspruch 1 angegebenen
Bedeutungen besitzen, mit der Maßgabe, daß bei Vorliegen einer
Hydroxy-Gruppe in R¹, R³, R&sup6;, R&sup7;, R&sup8; oder Ar, bei Vorliegen
einer Hydroxyalkyl-Gruppe in R¹, R², R&sup6;, R&sup7; oder R&sup8;, bei
Vorliegen einer Hydroxyalkoxy-Gruppe in R¹, R&sup6;, R&sup7;
oder R&sup8;, bei
Vorliegen einer Hydroxyalkylthio-Gruppe in R&sup6;, R&sup7; oder R&sup8;,
bei Vorliegen einer Amino-Gruppe in R³, R&sup6;, R&sup7;, R&sup8; oder Ar,
bei Vorliegen einer Aminoalkyl-Gruppe in R¹, R², R&sup6;, R&sup7;
oder R&sup8;, bei Vorliegen einer Alkylaminoalkyl-Gruppe in R¹,
R², R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer Alkylamino-,
Alkylaminoalkoxy-, Alkylaminoalkylthio-, Aininoalkoxy- oder
Aminoalkylthio-Gruppe in R&sup6;, R&sup7; oder R&sup8;, bei Vorliegen einer
Carboxy- oder Carboxyalkyl-Gruppe in R² oder R³ oder bei
Vorliegen einer Mercapto- oder Mercaptoalkyl-Gruppe in R¹,
R² oder R³ jede Amino-, Carboxy- und Mercapto-Gruppe durch
eine übliche Schutzgruppe geschützt ist und jede Hydroxy-
Gruppe durch eine übliche Schutzgruppe geschützt sein kann
oder auch nicht;
R&sup9; für Wasserstoff oder eine Schutzgruppe steht; und
Z für eine Austrittsgruppe steht;
worauf jede unerwünschte Schutzgruppe in R¹, R², R³, R&sup6;,
R&sup7;, R&sup8; und Ar oder jede Schutzgruppe R&sup9; durch übliche
Maßnahmen entfernt wird;
(b) eine Säure der Formel
oder ein reaktives Derivat davon mit einer Verbindung der
Formel R³-NH&sub2; umgesetzt wird, wobei in diesen Verbindungen
R¹, R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Ar die in Anspruch 1
angegebenen Bedeutungen besitzen und jede Mercapto-, Amino-,
Alkylamino- und Carboxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8;
und Ar durch eine übliche Schutzgruppe geschützt ist und
jede Hydroxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch
eine übliche Schutzgruppe geschützt sein kann oder auch nicht;
worauf die Schutzgruppen durch übliche Maßnahmen entfernt
werden;
(c) zur Herstellung eines erfindungsgemäßen Chinazolins,
worin R¹ für Alkoxy, Aryloxy oder Alkoxy mit bis zu 3
Kohlenstoffatomen steht, das einen oder mehrere Substituenten
trägt, die ausgewählt sind aus Hydroxy und Alkoxy, eine
Verbindung der Formel
mit einer Verbindung der Formel
HNR²-Ar-CONHR³
umgesetzt wird, wobei in diesen Verbindungen R¹ die
vorstehend zuletzt angegebene Bedeutung besitzt, R², R³, R&sup4;,
R&sup5;, R&sup6;, R&sup7;, R&sup8; und Ar die in Anspruch 1 angegebenen
Bedeutungen besitzen, mit der Maßgabe, daß jede Mercapto-, Amino-,
Alkylamino- und Carboxy-Gruppe in R², R³, R&sup6;, R&sup7;, R&sup8; und Ar
durch eine übliche Schutzgruppe geschützt ist und jede
Hydroxy-Gruppe in R¹, R², R³, R&sup6;, R&sup7;, R&sup8; und Ar durch eine
übliche Schutzgruppe geschützt sein kann oder auch nicht,
und Z für eine Austrittsgruppe steht;
worauf die Schutzgruppen durch übliche Maßnahmen entfernt
werden und die Gruppe R¹, die in der 4-Stellung des
Chinazolin-Rings vorliegt, durch Hydrolyse mit einer Base
abgespalten wird;
(d) zur Herstellung eines erfindungsgemäßen Chinazolins,
worin R¹ für Mercapto, Alkylthio, Arylthio, Alkylthioalkyl
oder Arylthioalkyl steht, ein Chinazolin der Formel
worin R¹ für Halogeno oder Halogenoalkyl steht und R², R³,
R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;, R&sup9; und Ar die in Anspruch 1 angegebenen
Bedeutungen besitzen, mit der Maßgabe, daß jede Mercapto-,
Amino-, Alkylamino-, Carboxy- und Hydroxy-Gruppe in R², R³,
R&sup6;, R&sup7;, R&sup8; und Ar durch eine übliche Schutzgruppe geschützt
sein kann oder alternativ jede Amino-, Alkylamino-, Carboxy-
und Hydroxy-Gruppe auch nicht geschützt sein muß, mit
Thioharnstoff umgesetzt wird, um eine Verbindung herzustellen,
worin R¹ für Mercapto steht, oder mit einem Alkyl- oder
Arylthiol umgesetzt wird, um eine Verbindung herzustellen,
worin R¹ für Alkylthio, Arylthio, Alkylthioalkyl oder
Arylthioalkyl steht;
worauf die Schutzgruppen durch übliche Maßnahmen entfernt
werden; oder
(e) zur Herstellung eines erfindungsgemäßen Chinazolins,
worin R¹ für Alkylthio steht, ein Chinazolin der Formel
worin R¹ für Mercapto steht und R², R³, R&sup4;, R&sup5;, R&sup6;, R&sup7;, R&sup8;,
R&sup9; und Ar die in Anspruch 1 angegebenen Bedeutungen besitzen,
mit der Maßgabe, daß jede Mercapto-, Amino-, Alkylamino-,
Carboxy- und Hydroxy-Gruppe in R², R³, R&sup6;, R&sup7;, R&sup8; und Ar
durch eine übliche Schutzgruppe geschützt sein kann oder
alternativ jede Amino-, Alkylamino-, Carboxyl- und Hydroxy-
Gruppe nicht geschützt sein muß, mit einer Base umgesetzt
wird;
worauf das erhaltene Thiolat-Salz mit einem Alkylhalogenid
alkyliert wird und worauf gegebenenfalls anwesende
Schutzgruppen durch übliche Maßnahmen entfernt werden.
9. Pharmazeutische Zusammensetzung, welche in Chinazolin
nach Anspruch 1 oder ein pharmazeutisch zulässiges Salz oder
einen pharmazeutisch zulässigen Ester davon gemeinsam mit
einem pharmazeutisch zulässigen Verdünnungs- oder
Trägermittel enthält.
10. Die Verwendung eines Chinazolins nach Anspruch 1
oder eines pharmazeutisch zulässigen Salzes oder Esters davon
zur Herstellung eines Medikaments für die Behandlung von
Tumoren im menschlichen oder tierischen Körper.
