Description
- Die Erfindung betrifft Mittel, die einen physiologisch aktiven Wirkstoff und ein substituiertes Azacycloalkan in einer Menge, die zur Steigerung der Penetration des physiologisch aktiven Wirkstoffs durch die Haut oder eine andere Körpermembran eines Tieres wirksam ist, enthält.
- Viele physiologisch aktiven Wirkstoffe werden zur Erzielung gewünschter Ergebnisse am besten topisch angewendet. Die topische Anwendung kann, im Gegensatz zur systemischen Anwendung, die metabolische Zersetzung des Wirkstoffs verhindern, sie verhindert weitgehend Nebenwirkungen der Wirkstoffe und erlaubt hohe lokale Konzentrationen der Wirkstoffe.
- Das größte Problem bei der Anwendung physiologisch aktiver Wirkstoffe auf topischem Wege ist, daß die Haut eine sehr wirksame Penetrationsbarriere darstellt. Die Epidermis der Haut besitzt eine äußere Schicht aus toten Zellen, die Stratum Corneum genannt wird, die fest zusammengepreßt und ölig ist, und die eine wirksame Barriere gegen gasförmige, feste oder flüssige chemische Wirkstoffe, ob allein oder in Wasser oder in öligen Lösungen verwendet, bereitstellt. Wenn ein physiologisch aktives Wirkstoff das Stratum Corneum penetriert, kann es sofort durch die basale Schicht der Epidermis in die Dermis treten.
- Obwohl die Effektivität des Stratum Corneum als Barriere einen großen Schutz bereitstellt, macht sie auch Anstrengungen zur Anwendung nützlicher Wirkstoffe direkt auf lokale Körperflächen zunichte. Die Unfähigkeit physiologisch aktiver Wirkstoffe das Stratum Corneum zu durchdringen, verhindert ihre wirksame Verwendung zur Behandlung solcher Erkrankungen, wie Entzündung, Akne, Psoriasis, Herpes Simplex, Ekzeme, Pilz-, Virus- oder andere Mikroorganismus-Infektionen oder andere Störungen oder Erkrankungen der Haut oder der Schleimhäute oder Erkrankungen unterhalb der äußeren Hautoberfläche oder der Schleimhäute. Das Stratum Corneum verhindert die Absorption und Zurückhaltung kosmetischer Stoffe, wie Sonnenschutzmittel, Parfüme, Insektenschutzmittel und dergleichen durch die Haut.
- Physiologisch aktive Wirkstoffe können auf lokal betroffene Körperteile durch das hier beschriebene Vehikelsystem angewendet werden. Vehikel, wie USP Cold Cream, Ethanol und verschiedene Salben, Öle, Lösungsmittel und Emulsionen wurden bereits zuvor verwendet, um physiologisch aktive Inhaltsstoffe lokal anzuwenden. Die meisten solcher Vehikel sind zum Transport signifikanter Mengen physiologisch aktiver Wirkstoffe durch die Haut nicht wirksam. Ein solches Vehikel ist Dimethylsulfoxid. Die 1-Niedrigalkyl-substituierten Azacyclopentan-2-one, die 1 bis 4 Kohlenstoffatome in der Alkylgruppe besitzen, sind dafür bekannt, daß sie die perkutane Absorption von Chemikalien, beispielsweise Medikamenten, leicht verstärken. Früher wurde erkannt, daß es wünschenswert wäre, denselben oder einen höheren perkutanen Absorptionsspiegel mit wesentlich niedrigeren Konzentrationen der penetrationsfördernden Verbindung zu erhalten. Darum wurden verschiedene N-substituierte Azacycloalkan-2-one mit den gewünschten Eigenschaften erfunden. Diese neuen penetrationsfördernden Wirkstoffe sind in den US-Patentschriften 3989815, 3989816, 3 991 203, 4 122 170, 4316893, 4 405 616, 4 415 563, 4 423 040, 4 424 210 und 4 444 762 beschrieben.
- Erfindungsgemäß sollen neue penetrationsverstärkende Wirkstoffe mit den gewünschten Eigenschaften der Verstärkung der perkutanen Absorption physiologisch aktiver Wirkstoffe bei Konzentrationen, die niedriger sind als die 1-Niedrigalkyl-substituierten Azacyclopentan-2-one, bereitgestellt werden.
- Erfindungsgemäß sollen weiterhin penetrationsverstärkende Wirkstoffe, die den zuvor erwähnten neuen penetrationsverstärkenden Wirkstoffe, die in den obigen US-Patentschriften beschrieben sind, äquivalent sind, bereitgestellt werden.
- Weitere erfindungsgemäße Aufgaben und Vorteile gehen bei sorgfältigem Lesen der untenstehenden Beschreibung hervor.
- In der Beschreibung umfaßt der Term tierisch Menschen ebenso wie andere formen tierischen Lebens und insbesondere domestizierte Tiere und Haustiere.
- Die Erfindung betrifft Mittel zum Transport physiologisch aktiver Wirkstoffe durch Körpermembranen, wie Haut, und das Zurückhalten dieser Mittel in Körpergeweben. Insbesondere betrifft die Erfindung Mittel, die bei der topischen Verabreichung eines physiologisch aktiven Wirkstoff an Mensch oder Tier verwendbar sind, die den Wirkstoff und eine wirksame, nichttoxische Menge einer durch die allgemeine Formel
- dargestellten Verbindung, worin X und Z für Sauerstoff-, Schwefel- oder 2 Wasserstoffradikale stehen können; Y für -CH&sub2;-, -NH- oder Sauerstoff stehen kann; n und m ganze Zahlen von 1 bis 3 sind; p eine ganze Zahl von 3 bis 17 Ist; q 2, 4, 6 oder 8 ist; und R' aus H und Niedrigalkylgruppen mit 1 bis 4 Kohlenstoffatomen oder aus einer Verbindung, ausgewählt aus 2,6-Dimethyl-4-(1-oxohexadec-10-enyl)-morpholin 2,6-Dimethyl-4-(1-oxooctadec-10-enyl)-morpholin 2,6-Dimethyl-4-(1-oxododec-10-enyl)-morpholin 2,6-Dimethyl-4-(1-oxotridec-11-enyl)-morpholin 2,6-Dimethyl-4-(1-oxotetradec-12-enyl)-morphol in 2,6-Dimethyl-4-(1-oxoundec-10-enyl)-morpholin 2,6-Dimethyl-4-(1-oxodec-17-enyl)-morpholin 2,6-Dimethyl-4-(1-oxohexadec-10-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxooctadec-10-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxododec-10-enyl)-azac ycloheptan 2,6-Dimethyl-4-(1-oxotridec-11-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxotetradec-12-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxoundec-10-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxodec-17-enyl)-azacycloheptan 2,6-Dimethyl-4-(1-oxoh exadec-10-enyl)-azacyclohept-2-on 2,6-Dimethyl-4-(1-oxooctadec-10-enyl)-azacyclohept-2-on 2,6-Dimethyl-4-(1-oxododec-10-enyl)-azacyclohept-2-on 2,6-Dimethyl-4-(1-oxotrldec-11-enyl)-azacyclohept-2-on 2,6-Dimethyl-4-(1-oxotetradec-12-enyl)-az acyclohept-2-on 2,6-Dimethyl-4-(1-oxoundec-10-enyl)-azacyclohept-2-on und 2,6-Dimethyl-4-(1-oxodec-17-enyl)-azacyclohept-2-on ausgewählt wird, enthält.
- Vorzugsweise ist R' H, X ist Sauerstoff und q ist 2. Bei einer bevorzugteren Ausführungsform der Erfindung ist p 4 bis 16, z. B. 10.
- Es wurde gefunden, daß die physiologisch aktiven Wirkstoffe durch die obigen penetrationsverstärkenden Wirkstoffe durch Körpermembranen transportiert werden und im Körpergewebe zurückgehalten werden.
- Die ungesättigten 1-Oxohydrocarbyl-substituierten Derivate von erfindungsgemäßen Azacycloalkanen umfassen Verbindungen, die durch die allgemeinen Formeln:
- worin r eine ganze Zahl von 2 bis 6 ist und p und q die oben gegebene Bedeutung besitzen, dargestellt werden.
- Diese 1-Oxohydrocarbyl-substituierten Derivate, die als penetrationsverstärkende Zusätze in den erfindungsgemäßen Mitteln verwendbar sind, können durch unten beschriebene Verfahren hergestellt werden. Typische Beispiele von Verbindungen, die durch die obige allgemeine Formel dargestellt werden, schließen ein:

- Viele der Verbindungen, die durch die obige allgemeine Formel dargestellt werden, sind gut bekannt. Zusätzlich erläutern die Beispiele unten Herstellungsverfahren für viele der Verbindungen, die durch die obige allgemeine Formel dargestellt werden. Die Menge des 1-Oxohydrocarbyl-substituierten Derivats, die gemäß der vorliegenden Erfindung verwendet werden kann, ist eine wirksame, nichttoxische Menge zur Verstärkung der perkutanen Absorption. Im allgemeinen liegt diese Menge im Bereich zwischen etwa 0,01 bis etwa 5 und vorzugsweise etwa 0,1 bis 2 Gew.-% des Mittels.
- Die betreffenden Mittel können mit vielen physiologisch aktiven Wirkstoffen, die in den offenbarten Vehikeln löslich sind, Verwendung finden.
- Fungistatische und fungizide Wirkstoffe, wie beispielsweise Thiabendazol, Chloroxin, Amphotericin B, Candicidin, fungimycin, Nystatin, Chlordantoin, Clotrimazol, Miconazolnitrat, Pyrrolnitrin, Salicylsäure, Fezation, Tolnaftat, Triacetin und Zink- und Natriumpyrithion, können in den penetrationsverstärkenden Wirkstoffen, die hier beschrieben sind, aufgelöst und topisch auf betroffene Hautpartien angewendet werden. Beispielsweise werden so angewendete fungistatische oder fungizide Wirkstoffe durch das Stratum Corneum transportiert und behandeln so erfolgreich durch Pilze verursachte Hautprobleme. Die so angewendeten Wirkstoffe penetrieren nicht nur schneller, als wenn sie in den herkömmlichen Vehikeln angewendet werden, sondern treten in das tierische Gewebe zusätzlich in hohen Konzentrationen ein und werden im wesentlichen für längere Zeiträume zurückgehalten, wodurch eine weitaus erfolgreichere Behandlung bewirkt wird.
- Beispielsweise können die erfindungsgemäßen Mittel auch bei der Behandlung von Pilzinfektionen auf der Haut, die durch Candida und Dermatophyten, die zu Athletenfuß und zu Thineapedis führen, verursacht werden, durch Auflösen von Thiabendazol oder ähnlichen Antifungus-Wirkstoffen in einem der oben beschriebenen penetrationsverstärkenden Mittel und durch Ihre Anwendung auf die betroffene Fläche, eingesetzt werden.
- Die erfindungsgemäßen Mittel sind ebenfalls bei der Behandlung von Hautproblemen, beispielsweise Herpes Simplex, die mit einer Lösung von Ioddesoxyuridin, aufgelöst in einem der penetrationsverstärkenden Mittel, behandelt werden können oder solchen Problemen wie Warzen, die mit Mitteln, wie Podophyllin, aufgelöst in einem der penetrationsverstärkenden Mittel, behandelt werden können, verwendbar. Hautprobleme, wie Psoriasis, können durch topische Anwendung einer Lösung eines herkömmlichen topischen Stetoids in einem der penetrationsverstärkenden Mittel oder durch Behandlung mit Theophyllin oder Antagonisten von β-adrenergen Blockern, wie Isoproterenol, In einem der penetrationsverstärkenden Mittel behandelt werden. Kopfhauterkrankungen, wie Alopecia Areata, können wirksamer durch Auftragen von Steroiden, wie Triamcinolonacetonid, aufgelöst in einem der erfindungsgemäßen penetrationsverstärkenden Mittel, direkt auf die Kopfhaut, behandelt werden.
- Die erfindungsgemäßen Mittel sind ebenfalls für die Behandlung von milden Ekzemen, beispielsweise durch Auftragen einer Lösung von Fluocinolonacetonid oder seinen Derivaten, Hydrocortison, Triamcinolonacetonid, Indomethacin, oder Phenylbutazon, aufgelöst in einem der penetrationsverstärkenden Mittel, auf die betroffene Fläche verwendbar.
- Beispiele anderer physiologisch aktiver Stetoide, die mit den Vehikeln verwendet werden können, schließen Corticosteroide, wie beispielsweise Cortison, Cortodoxon, Flucetonid, Fluorcortison, Difluorsondiacetat, Flurandrenolonacetonid, Medryson, Amcinafel, Ancinafid, Betamethason und seine Ester, Chlorprednison, Clocortelon, Descinolon, Desonid, Dexamethason, Dichlorison, Defluprednat, Flucloronid, Flumethason, flunisolid, Fluocinonid, Flucortolon, Fluormethalon, Fluperolon, Fluprednisolon, Meprednison, Methylmeprednisolon, Paramethason, Prednisolon und Prednison ein.
- Die erfindungsgemäßen Mittel sind ebenfalls bei der antibakteriellen Chemotherapie, beispielsweise bei der Behandlung von Hauterkrankungen, die mit pathogenen Bakterien zusammenhängen, verwendbar. Typische antibakterielle Wirkstoffe, die erfindungsgemäß verwendet werden können, umfassen Sulfonamide, Penicilline, Cephalosporine, Penicillinase, Erythromycine, Lincomycine, Vancomycine, Tetracycline, Chloramphenicole und Streptomycine. Typische Beispiele der zuvor genannten Verbindungen schließen Erythromycin, Erythromycinethylcarbonat, Erythromycinestolat, Erythromycinglucepat, Erythromycinethylsuccinat, Erythromycinlactobionat, Lincomycin, Clindamycin, Tetracyclin, Chlortetracyclin, Demeclocyclin, Doxycyclin, Methacyclin, Oxytetracyclin und Minocyclin ein.
- Die erfindungsgemäßen Mittel sind ebenfalls als Schutz der hochempfindlichen Haut oder sogar der normalempfindlichen Haut vor Beschädigung oder Unwohlsein durch Sonnenbrand, verwendbar. Somit kann eine Dermatitis Actinica durch Auftragen eines Sonnenschutzes, wie Paraaminobenzoesäure oder seiner gut bekannten Derivate, die in einem der oben beschriebenen penetrationsverstärkenden Mittel aufgelöst werden, auf die oberflächlichen Hautpartien, die der Sonne ausgesetzt werden sollen, vermieden werden und die schützende Paraaminobenzoesäure oder ihre Derivate werden erfolgreicher In das Stratum Corneum transportiert und werden darum auch bei Exposition gegenüber Wasser oder beim Waschen für einen im wesentlichen längeren Zeitraum zurückgehalten, als wenn sie in herkömmlichen Vehikeln auf die Haut aufgetragen werden. Die Erfindung ist insbesondere in herkömmlichen Sonnenschutzlotionen, die bei Aktivitäten, zu denen Schwimmen gehört, verwendet werden, verwendbar, weil die Ultraviolett-abschirmenden Inhaltsstoffe in den herkömmlichen Trägerstoffen beim Eintauchen in Wasser von der Haut abgewaschen werden.
- Die erfindungsgemäßen Mittel können auch bei der Behandlung von Narbengewebe durch Auftragen von Mitteln, die Collagen welch machen, wie Aminopropionitril oder Penicillamin, aufgelöst in einem der erfindungsgemäßen penetrationsverstärkenden Mittel, topisch auf das Narbengewebe, Verwendung finden.
- Wirkstoffe, die normalerweise als Augentropfen, Ohrentropfen oder Nasentropfen angewendet werden, sind wirksamer, wenn sie in den erfindungsgemäßen penetrationsverstärkenden Mitteln aufgelöst werden.
- Wirkstoffe, die bei der Diagnose verwendet werden, können wirksamer verwendet werden, wenn sie aufgelöst in einem der penetrationsverstärkenden erfindungsgemäßen Mittel angewendet werden. Pflastertests zur Diagnose von Allergien können sofort, ohne Anritzen der Haut, oder das Abdecken der einem Allergen ausgesetzten Fläche, durchgeführt werden, wenn die Allergene in einem der erfindungsgemäßen penetrationsverstärkenden Mittel angewendet werden.
- Die erfindungsgemäßen Mittel sind ebenfalls für die topische Anwendung von kosmetischen oder ästhetischen Wirkstoffen verwendbar. Beispielsweise werden Verbindungen, wie das Melanin-stimulierende Hormon (MSH) oder Dihydroxyaceton und dergleichen wirksamer zur Förderung einer Sonnenbräune auf der Haut angewendet, wenn sie in einem der erfindungsgemäßen penetrationsverstärkenden Mittel aufgelöst werden. Der Wirkstoff wird schneller und in größeren Mengen in die Haut transportiert, wenn er erfindungsgemäß angewendet wird. Haarfärbemittel dringen ebenfalls vollständiger und wirksamer ein, wenn sie in einem der erfindungsgemäßen penetrationsverstärkenden Mittel aufgelöst werden.
- Die Wirksamkeit solcher topisch angewendeten Stoffe als Insektenabwehrmittel oder Geruchsstoffe, wie Parfüme und Eau de Colognes, kann verlängert werden, wenn solche Mittel, die in einem der erfindungsgemäßen penetrationsverstärkenden Mittel aufgelöst werden, angewendet werden.
- Es ist zu betonen, daß das Vorausgegangene einfach Beispiele physiologisch aktiver Wirkstoffe einschließlich therapeutischer und kosmetischer Wirkstoffe mit bekannten Wirkungen für bekannte Krankheiten sind, die wegen ihrer bekannten Eigenschaften erfindungsgemäß wirksamer verwendet werden können.
- Zusätzlich können die erfindungsgemäßen penetrationsverstärkenden Mittel auch zur Erzeugung therapeutischer Wirkungen, die zuvor noch nicht bekannt waren, verwendet werden. Das heißt, durch Verwendung der penetrationsverstärkenden Mittel, die hier beschrieben sind, können therapeutische, bisher unbekannte, Wirkungen erreicht werden.
- Als Beispiel für das Vorausgegangene, ist Griseofulvin als das Behandlungsmittel der Wahl für Pilzinfektionen der Haut und der Nägel bekannt. Bisher war die Verabreichungsform von Griseofulvin oral. Jedoch war es bereits seit langem bekannt, daß die orale Behandlung wegen der Nebenwirkungen, die davon herrühren, daß der gesamte Körper dem Griseofulvin ausgesetzt ist und der Tatsache, daß nur die äußeren Schichten der betroffenen Haut behandelt werden müssen, nicht bevorzugt ist. Darum wäre es vorteilhaft, da Pilzinfektionen im allgemeinen Infektionen der Haut und der Nägel sind, Griseofulvin topisch anzuwenden. Jedoch wurde Griseofulvin trotz einer lang bemerkten Notwendigkeit für ein topisches Griseofulvin bisher zur Behandlung topischer Pilzerkrankungen oral verwendet, weil früher kein Präparat bekannt war, das topisch verabreicht werden konnte und durch das eine ausreichende Retention von Griseofulvin In der Haut erfolgt wäre, um therapeutisch verwendbar zu sein.
- Jedoch wurde nun gefunden, daß Griseofulvin im Bereich therapeutischer Konzentrationen zwischen etwa 0,1 % und etwa 10 % wirksam topisch verwendet werden kann, wenn es mit einem der hier beschriebenen penetrationsverstärkenden Mittel kombiniert wird.
- Ein weiteres Beispiel ist Akne, deren Name allgemein auf jede entzündliche Krankheit der Talgdrüsen angewendet wird, ebenso Akne Vulgaris. Der für die Akneinfektion typischerweise verantwortliche Mikroorganismus Ist Corynebacterium Aknes. Verschiedene therapeutische Verfahren zur Behandlung der Akne sind bereits versucht worden, einschließlich topischer antibakterieller Mittel, z. B. Hexachlorophen, und systemische Antibiotika wie Tetracyclin. Während die Behandlungen mit systemischen Antibiotika als teilweise wirksam bekannt sind, sind die topischen Behandlungen im allgemeinen nicht wirksam.
- Seit langem ist bekannt, daß die systemische Behandlung der Akne wegen den Nebenwirkungen, die von der Exposition des gesamten Körpers gegenüber Antibiotika und der Tatsache herrühren, daß nur die betroffene Haut behandelt werden muß, nicht bevorzugt ist. Jedoch wurden im allgemeinen Antibiotika trotz eines lang vorhandenen Bedürfnisses für eine topische Aknebehandlung nur dazu verwendet, die Akne systemisch zu behandeln, weil bisher kein antibakterielles Präparat, das topisch verwendet werden konnte und das bei der Behandlung von Akne therapeutisch wirksam ist, bekannt war. Jedoch wurde nun gefunden, daß Antibiotika, insbesondere die aus der Lincomycin- und Erythromycinfamilie der Antibiotika, zur topischen Behandlung von Akne verwendet werden können, wenn sie mit einem der hier beschriebenen penetrationsverstärkenden Mittel kombiniert werden.
- Das so angewendete Antibiotikum wird in therapeutisch wirksamen Mengen in und durch die Epidermis und tieferen Hautschichten ebenso wie in Follikel und Comedonen (talgverstopfte Follikel, die C. Aknes enthalten) transportiert und können dadurch erfolgreich zur zeitweisen Eliminierung der Anzeichen und Symptome der Akne verwendet werden.
- Der Term "physiologisch aktiver Wirkstoff" wird hier verwendet, um eine umfangreiche Klasse verwendbarer chemischer und therapeutischer Wirkstoffe, die physiologisch aktive Stetoide, Antibiotika, Antimykotika, antibakterielle Wirkstoffe, Antineoplastika, Allergene, Antihistaminika, entzündungshemmende Mittel, Ultraviolettschutzmittel, Diagnostika, Parfüme, Insektenabwehrmittel und Haarfärbemittel umfassen, zu bezeichnen.
- Die Darreichungsformen für die topische Anwendung können Nasalspraylösungen, Lotionen, Salben, Cremes, Gele, Suppositorien, Sprays und Aerosole einschließen. Typische inerte Trägerstoffe, mit denen die zuvor genannten Darreichungsformen aufbereitet werden, schließen Wasser, Aceton, Isopropylalkohol, Freone, Ethylalkohol, Polyvinylpyrrolidon, Propylenglycol, Geruchsstoffe, gelerzeugende Stoffe, Mineralöl, Stearylalkohol, Stearinsäure, Spernaceti, Sorbitanmonooleat, "Polysorbate", "Tweens", Sorbit und Methylcellulose ein.
- Die Menge des Mittels und somit der zu verabreichende physiologisch aktive Wirkstoff darin ist offensichtlich eine für das gewünschte, davon erwartete Ergebnis eine wirksame Menge. Dies wird selbstverständlich von jedem gewöhnlichen Anwender festgestellt. Aufgrund der erreichten verstärkten Aktivität kann die Dosis des physiologisch aktiven Wirkstoffes oft, gegenüber der im allgemeinen angewendeten, verringert werden. In Übereinstimmung mit den allgemeinen vernünftigen Formulierungsgewohnheiten kann anfangs eine Dosis nahe am unteren Ende des betreffenden physiologisch aktiven Wirkstoffes angewendet werden und die Dosis, wie aus der beobachteten Antwort ersichtlich, erhöht werden, wie bei den ärztlichen Routineverfahren.
- Die Erfindung wird weiterhin in den folgenden Beispielen erläutert, die verschiedene Gesichtspunkte der Erfindung erläutern und nicht zur Limitierung des Umfangs der Erfindung, wie durch die beigefügten Patentansprüche definiert, gedacht sind. Beispiel 1
- Herstellung von N-(1-Oxoundec-10-enyl)-azacycloheptan. Zu einer Lösung von Hexamethylenimin (10,0 g, 0,101 md) in 200 ml Toluol wurden 10 ml Triethylamin hinzugegeben, gefolgt von einer tropfenweisen Zugabe von 10-Undecenoylchlorid (20,7 g; 0,101 mol). Das Reaktionsgemisch wurde bei Raumtemperatur 1 h lang gerührt. Nach der Aufarbeitung wurde das Rohprodukt einer Destillation unterworfen und 9,0 g reines Produkt wurden bei 187 bis 190ºC (1 mm Hg) Isoliert. Beispiel 2
- Herstellung von N-(1-Oxoundec-10-enyl)-azacycloheptan-2-on. Beispiel 1 wurde unter Verwendung von 10,0 g (0,088 mol) von ξ-Caprolactam wiederholt und 7,0 g des Rohprodukts wurden bei 187 bis 190ºC (1 mm Hg) isoliert. Beispiel 3
- Herstellung von N-(1-Oxoundec-10-enyl)-morpholin. Beispiel 1 wurde unter Verwendung von 10,0 g (0,115 mol) Morpholin wiederholt und 5,5 g reines Produkt wurden bei 170ºC (1 mm Hg) isoliert.
- Die Bespiele 1, 2 und 3 wurden unter Verwendung von Natriumhydrid In THF (Tetrahydrofuran), das Triethylamin und Toluol ersetzte, wiederholt, um dieselben Verbindungen zu erhalten.
- Die Verbindungen aus den Beispielen 1 bis 3 wurden als penetrationsverstärkende Mittel gemäß der unten angegebenen Verfahrensweise getestet:
- Die Haut von weiblichen haarlosen Mäusen, 4 bis 6 Wochen alt, wurde vom Tier entfernt und über Penetrationscavitäten mit normaler Salzlösung, mit der das Corium umspült wurde, angebracht. Ein Plastikzylinder mit einem Durchmesser von 1,4 cm wurde auf der epidermalen Seite auf jedes Stück geklebt. 0,1 % ³H-Triamcinolonacetonid wurde auf die epidermale Oberfläche über den Zylinder von 1,4 cm Durchmesser gegeben (0,01 cc). Die Haut wurde bei Raumtemperatur und Umgebungsfeuchtigkeit Inkubiert.
- Nach 6 Stunden und 24 Stunden wurden 2 cc aus dem 10 cc-Reservoir der normalen Salzlösung, mit der das Corium umspült wurde, entfernt. Die entfernten 2 cc normale Salzlösung wurden nach der 6stündigen Probe mit 2 cc normaler Salzlösung ersetzt.
- Die 2 cc Aliquote wurden in Scintillationsflüssigkeit gegeben und die Radioaktivität in einem Scintillationszähler bestimmt. Die eindringende Menge wurde als Prozent der eingesetzten Dosis berechnet.
- Bei jedem Experiment wurde das ³H-Triamcinolonacetonid in Ethanol aufgelöst und das zu testende penetrationsverstärkende Mittel wurde bis zu der gewünschten Konzentration zugegeben.
- Die Kontrolle war 1-n-Dodecylazacycloheptan-2-on, eine Verbindung, die in den US-Patentschriften, die oben angegeben sind, mit überlegenen penetrationsverstärkenden Eigenschaften beschrieben werden. Fünf getrennte Tests wurden für jede Verbindung und die Kontrolle durchgeführt und aus den Resultaten der Mittelwert gebildet.
- Die Ergebnisse, die in der Tabelle unten angegeben sind, zeigen, daß die Verbindungen aus den Beispielen 1 und 2 penetrationsverstärkende Eigenschaften besitzen, die mindestens zu 1-n-Dodecylazadodecylazacycloheptan-2-on äquivalent sind. TABELLE

- Eine Aerosolform der Formulierung aus Beispiel 5 wurde durch Zubereitung des folgenden Gemisches hergestellt:

- Die folgenden Lösungsformulierungen wurden hergestellt: Clindamycingrundlage Clindamycinphosphorsäure Natriumhydroxid 1,0 M Chlorwasserstoffsäure Dinatriumedetat · 2H&sub2;O Geruchsstoff N-(1-Oxoundec-10-enyl)-azacycloheptan-2-on Gereinigtes Wasser Isopropanol
- Diese Lösungen sind bei der Behandlung von Akne bei Menschen wirksam.
- Die folgende Lösungsformulierung wurde hergestellt:

- Die folgende Lotionformulierung kann so hergestellt werden, daß sie etwa 0,001 bis 1 %, vorzugsweise 0,1 % Fluocinolonacetonid enthält:

- Natriumlaurylsulfat 15,0
- N-(1-Oxoundec-10-enyl)-azacycloheptan-2-on 1 ,0 Wasser (auf 100 % auffüllen).
- Das Steroid wurde in dem Vehikel aufgelöst und zu einer gerührten Kühlungsschmelze der anderen Inhaltsstoffe gegeben. Die Zubereitung ist Insbesondere bei der Behandlung von entzündeten Dermatosen bei topischer Anwendung auf der betroffenen Hautpartie verwendbar. Die Menge und Häufigkeit der Anwendung erfolgt nach der Standardpraxis für die topische Anwendung dieses Steroids. Die Penetration des Steroids in das entzündete Gewebe wird verstärkt und eine therapeutische Konzentration wird schneller erreicht und für eine längere Dauer aufrechterhalten, als wenn das Steroid in herkömmlichen Formulierungen angewendet wird.
- Die Beispiele 6 bis 14 werden wiederholt, außer daß N-(1-Oxoundec- 10-enyl)-azacycloheptan-2-on durch die Verbindungen aus den Beispielen 1 und 3 ersetzt wird. Es werden vergleichbare Ergebnisse erzielt.
Claims (16)
1. Mittel, dadurch gekennzeichnet, daß
es eine wirksame Menge eines physiologisch aktiven Wirkstoffs
und eine nichttoxische, wirksame, penetrierende Menge einer
Verbindung der allgemeinen Formel
worin X und Z für Schwefel-, Sauerstoff- oder 2
Wasserstoffreste stehen; Y für -CH&sub2;-, -NH- oder Sauerstoff stehen kann;
n und m ganze Zahlen von 1 bis 3 sind; p eine ganze Zahl von
3 bis 17 ist; q 2, 4, 6 oder 8 ist; und R' aus H und einer
Niedrigalkylgruppe mit 1 bis 4 Kohlenstoffatomen oder einer
Verbindung, ausgewählt aus
2,6-Dimethyl-4-(1-oxohexadec-10-enyl)-morpholin
2,6-Dimethyl-4-(1-oxooctadec-10-enyl)-morpholin
2,6-Dimethyl-4-(1-oxodedec-10-enyl)-morpholin
2,6-Dimethyl-4-(1-oxotridec-11-enyl)-morpholin
2,6-Dimethyl-4-(1-oxotetradec-12-enyl)-morph olin

2. Mittel nach Anspruch 1, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
antibakterielles Mittel ist.
3. Mittel nach Anspruch 2, dadurch
gekennzeichnet, daß der antibakterielle Wirkstoff ein Antibiotikum
ist.
4. Mittel nach Anspruch 3, dadurch
gekennzeichnet, daß das Antibiotikum aus der Gruppe, bestehend aus
Lincomycin, Clindamycin, Erythromycin und pharmazeutisch
anwendbaren Salzen davon ausgewählt ist.
5. Mittel nach Anspruch 1, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
physiologisch aktives Steroid ist.
6. Mittel nach Anspruch 1, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
Antimekotikum ist.
7. Mittel nach Anspruch 1, dadurch
gekennzeichnet, daß der physiologisch aktive
Wirkstoffloddesoxyuridin ist.
8. Mittel nach Anspruch 1, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff 5-Fluoruracil
ist.
9. Mittel zur topischen Verabreichung eines physiologisch
aktiven Wirkstoffs an einen Menschen oder ein Tier, dadurch
gekennzeichnet, daß es eine wirksame Menge
eines physiologisch aktiven Wirkstoffs und eine nichttoxische,
wirksame, penetrierende Menge einer Verbindung der
allgemeinen Formel
worin r eine ganze Zahl von 2 bis 6 ist, p eine ganze Zahl
von 3 bis 17 ist und q 2 ist, enthält.
10. Mittel nach Anspruch 9, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
antibakterielles Mittel ist.
11. Mittel nach Anspruch 10, dadurch
gekennzeichnet, daß der antibakterielle Wirkstoff ein Antibiotikum
ist.
12. Mittel nach Anspruch 11, dadurch
gekennzeichnet, daß das Antibiotikum aus der Gruppe, bestehend aus
Lincomycin, Clindamycin, Erythromycin und pharmazeutisch
anwendbaren Salzen davon ausgewählt ist.
13. Mittel nach Anspruch 9, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
physiologisch aktives Steroid ist.
14. Mittel nach Anspruch 9, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff ein
Antimekotikum ist.
15. Mittel nach Anspruch 9, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff
Ioddesoxyuridin ist.
16. Mittel nach Anspruch 9, dadurch
gekennzeichnet, daß der physiologisch aktive Wirkstoff 5-Fluoruracil
ist.
