Um zu prüfen, ob aus hitzedenaturierten Fleischproben DNA gewonnen werden kann, wurden an jeweils 100 mg nach Haus frauenart zubereitetem Schweine- bzw. Rinderschnitzeln und an frischem Fleisch von Rind und Schwein DNA-Präparationsverfahren angewandt. Aliquots der Präpa rationen und Kontroll-DNA aus Lymphozyten wurden elektro phoretisiert (Fig. 1). Es zeigt sich, daß sowohl aus rohem, als auch zubereitetem Fleisch DNA isoliert werden kann. Bei rohem Fleisch (Fig. 1, Spur 5 und 7) liegt die DNA größtenteils, bei den Kontrollen (Fig. 1, Spur 1 bis 4) ausschließlich in hochmolekularer Form vor. Bei den hitzedenaturierten Proben (Fig. 1, Spur 6 und 8) er scheint die DNA überwiegend als Schlier in den nieder molekularen Bereich. Um zu testen, ob die Tierartidenti fizierung an rohen und zubereiteten Fleischproben durch DNA-Hybridisierung möglich ist, wurden jeweils 1500 ng der daraus isolierten DNAs und jeweils 250 ng Kontroll-DNAs von Rind, Schwein, Mensch und Huhn mit radioaktiv markierter Schweine-DNA hybridisiert. Das Ergebnis zeigt Fig. 2. Man erkennt, daß bei den Kontrol len lediglich die Schweine-DNA mit der Sonde reagiert. Bei den Fleischproben bindet die Sonde an isolierte DNA aus rohem und gebratenem Schweinefleisch. Mit der DNA aus rohem und zubereitetem Rindfleisch wurde keine Hybridi sierung beobachtet.In order to check whether DNA can be obtained from heat-denatured meat samples, 100 mg pork or beef schnitzel prepared in the manner of a woman and in fresh meat from beef and pork DNA preparation methods were used. In the heat-denatured samples ( Fig. 1, lanes 6 and 8 ), the DNA appears predominantly as a streak in the low molecular weight range. In order to test whether the identification of animal species on raw and prepared meat samples by DNA hybridization is possible, 1500 ng of the DNAs isolated therefrom and 250 ng of control DNA from cattle, pork, human and chicken were hybridized with radioactively labeled pig DNA . The result is shown in FIG. 2. It can be seen that only the swine DNA reacts with the probe in the controls. In the meat samples, the probe binds to isolated DNA from raw and roasted pork. No hybridization was observed with the DNA from raw and prepared beef.
In einem weiteren Experiment wurden parallel 3 Dot blot- Hybridisierungen durchgeführt, wobei als Sonden Rind (S R ), Schwein (S S ) bzw. Huhn (S H )-DNA eingesetzt wurden. Auf jede der 3 Membranen wurden definierte Mengen Kontroll-DNA (1 ug) und unbekannte DNA-Mengen aus 10 ver schiedenen Fleischproben aufgetragen (Fig. 3). Es zeigte sich, daß menschliche DNA mit keiner der verwendeten Tier-DNA-Sonden reagiert. Rind-Kontroll-DNA und Huhn-Kontroll-DNA reagierte stark mit S R bzw. S H, Schweine-Kontroll-DNA reagierte stark mit S S und schwach mit S R . Aus der densitometrischen Auswertung dieser beiden Spots errechnete sich eine ca. 23 × höhere Hybridi sierungsstärke der Schweine-Kontroll-DNA mit S S als mit S R . Die Fleischproben 1 mit 10 konnten eindeutig Rind, Schwein bzw. Huhn zugeordnet werden. Jedoch zeigten die Proben 8 mit 10 geringe Kreuzreaktionen. Die densitome trische Auswertung dieser Spots ergab, daß, wie bei der Schweine-Kontroll-DNA, DNA aus denaturiertem und rohem Schweinefleisch mit S S ca. 20 × stärker reagiert als mit S R (Probe 8 und 10). Die Rind-DNA aus Probe 9 reagierte mit S R ca. 35 × stärker als mit S S . In keinem Fall wurde eine Kreuzhibridisierung von Rind- bzw. Schweine-DNA mit S H beobachtet. Diese Ergebnisse zeigen, daß die Methode geeignet ist, rohes und hitzedenaturiertes Fleisch von Rind, Schwein und Huhn eindeutig zu differenzieren. Ferner zeigt sich (Proben 3 und 8), daß eine Spezies-Identifizierung auch am Mageninhalt möglich erscheint.In a further experiment, 3 dot blot hybridizations were carried out in parallel, using beef (S R ), pig (S S ) or chicken (S H ) DNA as probes. The meat samples 1 with 10 could be clearly assigned to beef, pork or chicken. However, samples 8 with 10 showed minor cross-reactions. The densitometric analysis of these spots showed that, as with the pig control DNA, DNA from denatured and raw pork reacts with S S about 20 × more than with S R (samples 8 and 10 ). The bovine DNA from sample 9 reacted with S R approximately 35 times more strongly than with S S. In no case was cross-hibridization of bovine or porcine DNA with S H observed. These results show that the method is suitable for clearly differentiating raw and heat-denatured meat from beef, pork and chicken. It also shows (samples 3 and 8 ) that species identification also appears possible on the stomach contents.
