Abstract
Description
The invention relates to a method and a Vorrich to influence growth, division and differences adornment of cells.
Experimental work by the Institute of Molecular Physical Physiology in Hanover (Director: Dr. Dr. Herbert Athenstaedt) have a new, interdisciplinary Research area in the border area between biomedicine and Opened up physics; there were fundamentally new Er knowledge, especially in cell and development biology. These basic scientific results possible useful applications for biotech nology, medicine and biology.
The investigations showed that two properties that have long been known in physics for certain crystals and synthetic polymers, namely spontaneous polarization and pyroelectricity ( 1 , 2 ), also occur in living organisms. They have been properly detected in the tissues and organs of humans, animals and plants as well as in living cells. Both properties play a fundamental role in the growth processes (morphogenesis) and in numerous physiological functions of the organisms ( 3-10 ).
The molecular requirements for the polarity of living cells have so far not been clarified ( 11, 12 ). Pyroelectric measurement methods were used to demonstrate spontaneous polarization, ie a permanent electrical dipole moment, in the direction of the cell's main axis (longitudinal polarization of the cell); there is also polar, ie pyroelectric, behavior in the radial axial directions of the cell (radial polarization of the cell ( 3 , 4 ).
Die Epidermis hat somit die physikalischen Eigen schaften eines empfindlichen pyroelektrischen Detektors und Transducers (3, 4, 7-9).Further experimental series showed that tissues and organs, which are made up of polar single cells, also show spontaneous polarization in defined axis directions. The epidermis, the outermost cellular layer that surrounds the animal and plant organism almost on its entire surface, has a permanent electrical dipole moment perpendicular to the epidermis surface. The epidermis thus has the physical properties of a sensitive pyroelectric detector and transducer ( 3 , 4 , 7-9 ).
Spontaneous polarization combined with pyroelectric behavior has also been demonstrated in the central nervous system (brain, spinal cord) ( 3-6 ), in sensory receptors ( 3-5 ), in the endothelium of the cardiovascular system ( 4 ), in all essential structures (bones, cartilage , Intervertebral discs, ligaments, tendons) of the vertebrate skeletal system ( 10 ) and in the conduction tissues (phloem, xylem) of the plants.
It has been shown that the growth direction in Cells and tissues match the direction (the Vector) of an inherent spontaneous polarization, i.e. so a permanent electrical dipole moment.
The object of the present invention is a method and an apparatus for influencing Specify growth, division and differentiation of cells.
This task is solved procedurally in that the electrical dipole moments of the cells in terms of strength and / or change direction.
The invention uses the knowledge that cells are a show spontaneous polarization. Showed by the invention itself in a surprising way that growth, division and Differentiation of cells can be influenced by Change in the electrical dipole moments of the cells.
Because of the spontaneous polarization of the individual cells tissues, organs and organisms also show polar properties. The higher architectural levels (tissues, organs) of the Organisms arise through a coordinated three-dimensional sional orientation of cellular electrical dipoles. The invention is an artificial control of the Growth and differentiation of animal and plant structures possible, by modification the dipole texture.
This means that biological systems of all kinds, ie human, animal and plant single cells, as well Protozoa (including bacteria, viruses), human, animal and plant tissues and organs, as well as animal ones and plant organisms are affected.
Under the influence of growth according to the present Invention will be an artificial reinforcement or Ab weakening of growth and / or differentiation, a standstill of growth and / or differentiation, however also understood an unlimited increase.
In particular, cultures can be bio technologically or biomedically important single cell, Ge weave cells, tissues and organs with a relatively low host economic effort are produced. You can also advantageous biosensors are manufactured.
Advantageous and expedient embodiments of the task solution according to the invention are in subclaims featured.
The dipole texture in biological systems can be changed by using direct electric currents or direct electric fields, the biological objects being subjected to suitable direct current electric currents or direct electric fields by using electrodes of suitable size and shape, cf. Claims 4 and 5. Homogeneous or inhomogeneous DC fields can be used. The dipole structure can also be changed by bodies, substrates, materials, biological structures or the like with a defined electrical surface charge, cf. Claims 6 to 8. The DC currents, DC fields and surface electric charges can be made variable. This enables a fine adjustment to the apparently very differentiated charge conditions on the surfaces of human, deep and plant tissue cells as well as of individual cells (for example of certain bacteria or viruses). The development of cells and cell organelles can be controlled much more effectively. An adaptation to the particular characteristics of the specific cell type or a very specific cultivation project can be carried out optimally by changing the surface potential from negative potential values over the potential zero to positive potential values at any time during a culture process. The change can be exactly determined with the help of measuring devices. The change in potential can take place continuously or in stages. With this method it is possible to optimize a cell culture process and to precisely control the required level of potential - even during the course of the process - at any time according to the desired standards.
A device for influencing growth, division and differentiation of cells is given in claim 17. Advantageous and practical developments of this Vorrich tion include further claims 18 to 29.

As far as reasonably reasonable, the figures of the Drawing for the same components same reference numerals ver turns.
Fig. 1 shows an apparatus 2 in which a DC electric field for influencing growth, division and differentiation of cells is used. The device consists of a ver with a lid 3 culture container 4 , which is located in the electrical, practically homogeneous DC field between two plate-shaped electrodes 6 and 8 , which are connected to the poles of a DC voltage source 9 . The culture container 4 , in which a cell culture 5 is located, stands on the, for example, negatively charged, electrode 6 designed as a base plate. This made of metal (eg stainless steel) floor plate 6 is designed so that any number of culture containers can be parked on it. The upper electrode 8 is preferably formed as a transparent conductor layer on the preferably also transparent cover 3 of the culture container to ensure an effortless observation of the cell culture. A changeover switch (not shown) enables a polarity reversal of the electrodes 6 and 8 and thus a reversal of the direction of the constant field between the electrodes. The field strength within the culture container or within the cell culture 5 can be precisely regulated by a measuring instrument 10 and a variable resistance R. The contact to the upper conductor layer or to the upper electrode can be realized, for example, by a resilient, pivotable metal bracket (not shown).
Fig. 2 shows a device 12 for influencing growth, division and differentiation of cells in which the culture container 4 shaped plates on an array of two, in the manner of a plate capacitor are arranged spaced electrodes 14 and 16 is. The culture container 4 is located in the field lines of the upper electrode 14 . The relatively weak and non-homogeneous field compared to the device of FIG. 2 can be controlled by the electrical voltage applied to the electrodes in such a way that a reaching effect in the cell culture 5 is achieved. In the apparatus of Fig. 2, the upper electrode 14 need not be made of transparent material.
Fig. 3 shows an apparatus 20 with a receptacle 21 for a variety of devices 2 of the type shown in Fig. 1 (in the present case only five devices with culture containers A , B , C , D and E are shown.) The parallel switched devices 2 are in the large receptacle 21 made of an insulating material, for example plastic, through the bottom 24 of which the lower plate-shaped electrodes are connected to a pole of a DC voltage source 26 . The upper electrodes 8 are connected to the other pole of the DC voltage source. For each device, a certain field strength is adjustable with the help of an adjustable resistor R 1 , R 2 , R 3 , R 4 , R 5 , which is arranged in the parallel branch of each device 2 . With the help of measuring instruments 28 , 28 ', the respective voltage present between the electrodes and thus the field strength can be read off. The measurement can be done via the electrodes 6 and 8 (measuring instrument 28 ) or via the resistors (measuring instrument 28 '). In order to prevent mutual interference of the fields, the individual devices 2 are separated from one another by shields 30 .
The device according to FIG. 3 makes it possible to determine in a short test period which field strength is optimally suitable for carrying out a specific cultivation program. This requires that all Kulturbe containers are loaded with the same test material (for example, a cell type to be examined for the first time).
In order to illustrate the function of such an apparatus 20 , it is assumed that in the culture container No. A the field strength, for example, the value 100 of any unit, No. B the value 200 , No. C the value 300 and so on, so that a potential series is created. This opens up new possibilities and improvements for an optimized influence on the development of biological systems.
FIGS. 4 and 5 show an apparatus 40 and a means of apparatus 42 which uses a variety of gene Vorrichtun 40. In the device 40 or the apparatus 42 , the cell cultures are not exposed to a direct field, but rather to a direct current.
The devices 40 have culture containers 44 of any size made of electrically insulating material. The culture containers can have a rectangular or square base. The culture containers 44 are provided on two opposite inner wall surfaces with electrodes 46 , 48 (cathode and anode), which are connected to a DC voltage source 50 . The bottom of the culture container can be neutral or consist of an inorganic or organic material which is permanently polarized between the electrodes in the direction from one electrode to the other. Inorganic materials for this are, for example, the ferroelectric lithium niobate (LiNbO 3 ) or the pyroelectric tourmaline (here the disks would have to be cut parallel to the polar Z axis). If the bottom surface of the vessel is composed of several adjacent sections of such a polar material (such as, for example, a room floor made of parallel boards), care must be taken that the polar axes of all sections are parallel to each other.
It has proven to be advantageous if the polar bottom surface of the vessels is provided with parallel grooves 52 in the direction of the anode - cathode (for example as in the case of the parallel scratching of an optical diffraction grating). The grooves cause the cells to accumulate in the grooves and align them particularly well. Instead of grooves, longitudinally polarized thin rods can also be placed close to one another on the bottom of the culture container. Such rods can for example consist of collagen fibers, which show a relatively strong longitudinal polarization. The directional effect of a weak direct current, which is passed between the electrodes through the culture container, can be enhanced in this way by the directional effect of the polar soil material.
How a design according to FIGS. 4 and 5 looks in section has been drawn in dashed lines for the culture container A in FIG. 3.
The devices 40 in the apparatus 42 are connected in parallel. The parallel circuits have adjustable resistances Ra, Rb, Rc for changing the current in the parallel circuits. The respective current strength can be measured with the aid of measuring instruments 54 .
The cells in the cell cultures of the devices according to FIGS. 4 and 5 have to be immobilized in order to prevent the electrophoretic migration. This can be done by adding gelatin or agar.
In the immobilized living cells are through the direct current (depending on the direction and strength of the direct current) an amplification or weakening of the causes inherent dipole moments.
It is also possible to pass through the inherent dipole moment a correspondingly dosed direct current to - zero bring (neutralization of the dipole moment). Likewise a change in direction or reversal of the dipole moments achievable as long as the direct current acts, whereby after-effects are to be expected according to Ab switching the direct current. A constant change of direction Turnaround may also be achievable.
The devices according to FIGS. 1, 2 and 4 are those for large-scale production. The culture containers themselves can have any size and any shape and form. Polystyrene is particularly well suited as a material for the culture containers.
The lid electrode 8 can be made of a transparent metal oxide. It is also possible to dispense with an additional cover electrode if the cover itself consists of an electrically conductive glass or a transparent, electrically conductive material, for example a transparent metal oxide.
