Description
The invention relates to a biochemically active matrix, which contains at least one biochemically active enzyme, that ge covalently to the surface of a support matrix is bound, which is formed by a cross-linked protein det and on a microporous membrane bil the carrier is adsorbed, a process for producing provision of such a matrix and the use for extracorporeal enzyme treatment.
Such a biochemically active matrix is from the U.S. Patent 4,438,183 known. With this well-known matrix is the protein fibrin that forms the carrier matrix, which s by reaction of fibrinogen with thrombin forms a soft gel. As a carrier material Glass or steel or plastics, such as polyolefins beaten. They can be used as plates, balls or tubes or inserted as permeable or semipermeable membranes be set.
Patients suffering from an imbalance caused by En zyme degradable blood ingredient suffer there treated by that their blood is an extracorporeal En undergoes symbology. Such enzyme treatments can be carried out more effectively if they are not on Whole blood, but done on blood serum. That's what he is for required, first in an upstream plasma pheresis unit the cellular components of the serum to separate. After the enzyme treatment, the blood components merged again.
(Vgl. Klein, M.D. und Lange, R., TIBTECH, July 1986, S. 179-186, Elsevier Sc. Publ., Amsterdam). There are a number of differences for immobilization Licher method, the biocompatibility of the enzyme carrier matrix plays an important role. In addition to the ver application of asparaginase, described in detail here the method used is also for immobilization other enzymes suitable such. B. the bilirubin oxidase to lower the serum bilirubin concentration Liver disease, heparinase to remove heparin from blood or serum, carboxypeptidase G 1 to break down Methotrexate, which is used in tumor treatments and the phenylammonia lyase to break down phenylalanine. Benefits of using immobilized enzymes are especially little or no immunological reactions with prolonged use or reuse Reactors. (See Klein, M.D. and Lange, R., TIBTECH, July 1986, pp. 179-186, Elsevier Sc. Publ., Amsterdam).
Enzymes can be used in extracorporeal blood treatment be used. The type of treatment corresponds approximately to that Plasmapheresis or the procedure in kidney dialysis.
By the assignment described in US Pat. No. 4,438,183 a microporous membrane with fibrin for the purpose of Creation of a carrier matrix for the active enzyme the transmembrane flow is reduced to less than 10% of the trans membrane flow of the microporous membrane decreased because the pores are closed by the carrier matrix. There such a biochemically active matrix is only on their flooded surface effective.
The object of the present invention was to have the effect improve the biochemical matrix and enzyme treatments on the serum of the Blu tes without performing the cellular component to have to separate le of the blood.
This task is solved with a generic bio chemically active matrix, which is characterized by that the support matrix covers the surface of the open pores passages and was formed by albumin.
In this biochemically active matrix according to the invention the pores are not closed, so that for the Formation of the matrix used microporous membrane even after the carrier matrix and the binding of the enzyme for the filtration of protein substances remains suitable.
Such a microporous membrane is preferred used, whose average pore diameter 0.05 to Is 5 µm. The pore area is selected so that the blood ingredients that are enzymatically altered or to be broken down with the filtrate through the membrane permeate pores.
Vorzugsweise beträgt die Molekulargewichtsausschlußgren ze 100 000 bis 3 Millionen. The molecular weight exclusion limit is preferably ze 100,000 to 3 million.
Above all, favorable transmembrane flow values can then targets if the mean specific pore volume 0.5 to 0.9 cm³ / g. The trans is preferably membrane flow through the biochemically active matrix little at least 80% of the transmembrane flow of the microporous membrane bran.
Der mittlere Porendurchmesser wird mittels der Methode nach Marshall, J.C. und Meltzer, T.H., veröffentlicht in Bull. Parenteral Drug Assoc., 30, 214 (1976) bestimmt. The mean pore diameter is determined using the method after Marshall, J.C. and Meltzer, T.H. in Bull. Parenteral Drug Assoc., 30, 214 (1976).
The specific pore volume of the microporous membrane is determined using the mercury intrusion method Horold, E. and Skan, E.L., Science 120 (3124), 805 (1954) certainly.
Die Molekulargewichtsausschlußgrenze (Cutoff) wird nach Baker, R.W. und Strathmann, H., wie sie in Journal of Applied Polymer Science Vol. 14, 1197 (1970) beschrieben ist, ermittelt. The molecular weight cutoff is reduced to Baker, R.W. and Strathmann, H., as described in the Journal of Applied Polymer Science Vol. 14, 1197 (1970) is determined.
The transmembrane flow is measured on a module with approx. 125 cm² exchange area with isopropanol after the ASTM regulation F 317-72.
For cross-linking of proteins, for example, are suitable as cyanuric chloride, cyclic anhydrides with 4 to 9 Koh Oil atoms such as succinic anhydride, maleic acid anhydride or adipic anhydride or active dicarboxylic acid esters such as bis-salicylic succinates, bis (3,5-dibromosalicyl) - Fumarate or bis (N-hydroxy-succinimido) adipate.
Preferably, however, according to the invention for the cross ver Network dialdehydes, such as. B. terephthalaldehyde set. The best results are in terms of Enzyme activity achieved when glutardialdehyde for the Cross-linking of the protein is used.
According to the invention, the biochemically active matrix is produced using a method which is characterized in that the following liquids, if appropriate in a circuit, are passed through a hydrophilic and / or hydrophilized microporous membrane: a solution of 0.05 to 1% by weight % albumin in a Puf fergemisch at a constant pH value, water, a solution containing a crosslinking agent for albumin, a solution of the enzyme in a buffer mixture at a constant pH value, and water again.
In a preferred embodiment of the invention, the solution of the enzyme (d) and the water (e) are each passed through from the opposite side of the microporous membrane. The solution (c) containing a Vernetzungsmit tel for albumin, is preferably a 1 to 20% dialdehyde solution in a buffer mixture at a constant pH value. This p H value is expediently 4.0. A preferred dialdehyde as a crosslinking agent for the albumin is glutardialdehyde.
The microporous membrane is preferably in the form of a Capillary bundle used, although modules with Flat membranes appear suitable. The biochemically active matrix according to the invention is proposed preferably for the extracorporeal enzyme treatment of Blood and has proven to be excellent here, because the activity of the enzymes is particularly effective sam can be brought to bear that the matrix for the non-cellular components of the blood or essential components of the blood are permeable.
The invention is illustrated by examples purifies.
A module with 100 microporous poly serves as the carrier propylene threads (Plasmaphan® Type PIL) with a top area of 125 cm². The hydrophobic polypropylene membrane is treated with isopropyl alcohol and the isopropyl then alcohol out with distilled water to wash.
Nach einer Spülung mit 200 ml destilliertem Wasser wird das adsorbierte Albumin mit einer 10%igen Glutardialdehydlösung in 50 mM Citrat puffer pH 4,0 30 minutenlang behandelt.115 mg of albumin were dissolved in 100 ml of 50 mM citrate buffer pH 4.0 loosened and through the polypropylene hollow threads from the inside filtered outwards, this filtration being 10 minutes was continued in the cycle. After a rinse with The adsorbed albumin becomes 200 ml of distilled water with a 10% glutardialdehyde solution in 50 mM citrate buffer pH 4.0 treated for 30 minutes.
L-asparaginase (E. coli) was used as a biochemically active enzyme. (83 U / mg) used. 106 mg (8800 IU) were added 50 ml of 50 mM citrate buffer pH 4.0 dissolved and 16 hours at 21 ° c in the circuit from the inside out through the module filtered. Then with 2 l of distilled water rinsed. There were 75 mg of albumin on the polypropylene adsorbed membrane and then 2024 IU L-asparaginase immobilized. The activity of the immobilized L-Asparaginase was 622 IU, which corresponds to 31% of the immobilized enzyme.
The transmembrane flow of the starting support material was 7.80 ± 0.50 ml / min · bar · cm² measured with isopropanol. After the support was coated with albumin, the Transmembrane flow 7.05 ± 0.45 ml / min · bar · cm² measured with Isopropanol. The biochemically active according to the invention Matrix showed a corresponding measurement with Iso propanol has a transmembrane flow of 7.10 ± 0.45 ml / min · bar · cm² on.
There was a corresponding module and treated as in Example 1 before then in place of the albumin solution with a solution of 150 mg of fibrinogen into 50 ml of phosphate buffer at pH 7.35 for 10 minutes in circulation through the module from NEN filtered out. It was then washed with 100 ml of distilled water. The adsorbed fibrinogen was treated for several minutes with 10% glutaraldehyde in phosphate buffer p H 7.35 30th Then it was washed again with distilled water. According to Example 1, a solution of L-asparaginase was circulated through the module from the inside out. Then washed flat with 2 liters of distilled water. 80 mg of fibrinogen were adsorbed onto the support and 4335 IU L-asparaginase were immobilized. The recovered activity was 992 IU, which corresponds to 23% of the immobilized enzyme. The transmembrane flow measured with isopropanol was 0.16 ± 0.02 ml / min · bar · cm².
It was a further module, as above, for example in the same Ver already described, treated, but was added to the solution of 150 mg of fibrinogen in 150 ml of phosphate buffer p H, optionally 7.35 3 IU thrombin. Otherwise, the module was treated in the same way as already described. However, it was found that after just 30 seconds, filtration of the fibrin-thrombin solution through the membrane was no longer possible, since the membrane was blocked by the fibrin formed. It is therefore not possible to fix L-asparaginase in the pore system of the polypropylene membrane. A transmembrane flow of 0.06 ± 0.01 ml / min · bar · cm² was measured with isopropanol.
