Feres, Tetrahedron Lett. 24, 3769 (1983)). 3-Hydroxy-3-methyl-glutarsäure selbst zeigt an der Ratte und im Humanversuch signifikante cholesterinsenkende Wirkung (Z. Beg, Experimentia 23, 380 (1967), ibid 24, 15 (1968), P. J. N2 bei 0°C aufgeschlämmt. Bei dieser Temperatur werden
101 ml (0.16 Mol) BuLi in Hexan zugetropft. Dabei bildet
sich ein tiefrotes Phosphorylid. Man rührt noch ca. 15 Min.,
und tropft dann 27.21 g (0.16 + 0.016 Mol) 0-Chlormethylbenzaldehyd
XVI (hergestellt nach G. Dreyfahl u. G. Plötner Chem.
Ber. 94, 907 (1961)) in 20 ml abs. THF gelöst innerhalb 30
Min. zu. Man rührt noch ca. 1 Std. nach, entfernt das Tetrahydrofuran
im Vakuum, extrahiert den Rückstand mehrmals mit
einer Mischung 500 ml Pentan und 300 ml H2O. Die vereinigten
Pentanextrakte werden mit MgSO4 getrocknet und eingeengt.
Der Rückstand wird unter Zusatz von Aktivkohle mit Äther
(300 ml) versetzt und erneut extrahiert. Der Ätherextrakt
wird abgetrennt und über Nacht im Kühlschrank aufbewahrt.
Der Äther wird abdekantiert vom Ungelösten, filtriert und
im Vak. eingeengt. Zurück bleibt ein hellgelbes Öl.
Ausbeute: 37.47 g (95% d. Th.) XVIIg

für ca. 3-6 Stunden erwärmt. Nach ca. 1/2 Stunde begann das
gebildete unlöstliche Phosphoniumsalz auszufallen. Der Reaktionsverlauf
wurde durch Dünnschichtchromatographie in
CH3OH/CH2Cl2 = 1/1 auf Kieselgelplatten verfolgt. Nach Beendigung
der Reaktion wurde abgekühlt, das feste Salz abgesaugt
und mit Diäthyläther gewaschen, i. Vak. bei 150°C getrocknet.
Ausbeute: 48.3 g weiße Kristalle (63% d. Th.) XVIIIg

Kieselgel wird abfiltriert, mit Methanol nachgewaschen.
Das Filtrat wird i. Vak. eingeengt. Das zurückbleibende
Öl wird in 50 ml Isopropanol gelöst und das Phosphoniumsalz
durch Zugabe von Diäthyläther ausgefüllt und abgesaugt,
anschließend wird im Vak. bei 150°C getrocknet.
Ausbeute: 23.3 g weiße Kristalle (91% d. Th.) IIg (Z = CH2-CH2)

Bei 0°C wurden 9.7 ml (15.5 mMol) Butyllithium in Hexan zugetropft. Man rührte ca. 30 Minuten bei 0°C. Dann wurden ebenfalls bei 0°C 6.12 g (15.35 mMol) 6S-Formyl- 4R(tert.-Butyldiphenyl)-silyloxy-2-methoxy-3,4,5,6-tetrahydro- 2H-pyran (Beispiel 12) in 15 ml abs. Tetrahydrofuran zugetropft. Nach 1 Stunde Rühren bei Raumtemperatur wurde THP im Vak. abgezogen, der Rückstand im Essigester aufgenommen und mehrmals mit Wasser extrahiert und über MgSO4 getrocknet, filtriert, eingeengt. Der Rückstand wurde auf einer Kieselgelsäule (z. B. Merck Lobar® Gr. C) mit Cyclohexan- Essigester 40 : 1 chromatographisch in die reinen E- bzw. - a) the phosphonium salts of formula II in which R 1 , R 2 , R 3 and Z have the meaning given for the formula I and X = Cl, Br, J, with the chiral aldehyde of the formula III, wherein R 5 is a protective group stable against bases and weak acids, to a compound of the formula wherein R 1 , R 2 , R 3 and Z have the meaning given for formula I, R 5 has the meaning given for formula III (and AB represents the (-CH = CH -) group),
- b) in a compound of the general formula IV, the methyl acetal acid hydrolyzes to a lactol of the formula V, wherein R 1 , R 2 , R 3 and Z have the formula I, R 5 has the meaning given for the formula III (and AB represents the (-CH = CH -) group),
- c) the compound of the general formula V oxidizes to a lactone of the formula VI, wherein R 1 , R 2 and R 3 and Z have the meaning given for formula I, R 5 has the meaning given for formula III (and AB represents the (-CH = CH -) group),
- d) in a compound of the general formula VI, the protective group R 5 is split off to give a compound of the formula I,
in which R 1 , R 2 , R 3 and Z have the meanings given for the formula I and AB represents the (-CH = CH -) group,
optionally a compound of general formula I in which AB represents a (-CH = CH -) group, hydrogenated to a compound of general formula I in which AB represents a (-CH 2 -CH 2 -) group, the Hydrogenation can also take place in the compounds of the formulas IV, V or VI to give corresponding compounds in which AB represents the (-CH 2 -CH 2 -) group, optionally converting a hydroxylactone I into the corresponding free hydroxy acids Ia or their salts or optionally represents the corresponding ester from the free hydroxy acids Ia or from the hydroxylactone I.
