Abstract
Description
Die Erfindung betrifft neue Gelformulierungen für thera-The invention relates to new gel formulations for thera-
peutische und diagnostische Zwecke.therapeutic and diagnostic purposes.
The disadvantage of these formulations is that In the process, lumps form that make it difficult to release the active ingredient or sagar preventri. In DE-OS 33 07 816 lyophilizates of alkali polyacrylates described, which are mixed with aqueous solutions of prostaglandins. the end With this formula, the active ingredient is released better, unsatisfactorily However, the partly inhomogeneous incorporation of the active ingredient still remains in such celes with a consistently high viscosity and, above all, the applicability of these gels by the user. The invention was based on the object of providing gel formulations for active ingredients with limited stability to develop in body cavities or to be used on body surfaces.
In CA 1072-413 (Derwent Publication No. 18422C / 11) the possible application of polyoxyethylene-polyoxypropylene block copolymers as carriers of pharmaceuticals for therapeutic or prophylactic use on the body surface or -n called body cavities. The percentage by weight of gel components is 15-50% specified. The preferred range is based on the exemplary embodiment with 30% by weight from: 20% Pluronic F 127 and 1 Pluronic L 44 in aqueous solution.
Weitere Angaben über die Verwendung von Pluronic F-127-Zubereitungen findet man nur noch für die ophthalmologische Praxis (DE-OS 27 08 152).Further information on the use of Pluronic F-127 preparations can only be found for ophthalmological practice (DE-OS 27 08 152).
It has now been found to be useful for therapeutic and diagnostic Use of Fluronic F-127 gel formulations in body cavities or on body surfaces the proportion of 1 to 19 percent by weight of Pluronic F-127 is particularly advantageous proves in these formulations.
Dieses reversible vol-Gel-Verhalten erlaubt es, den gelösten Wirkstoff leicht in eine gekühlte Pluronic F-127-Lösung homogen einzuarbeiten und in ein geeignetes Applikationssystem aufzuziehen (z.B.Aqueous Pluronic F-127 solutions with a Pluronic F-127 content of 18-19 percent by weight are easy to move at low and high temperatures Liquids with low viscosity and only at medium temperatures (20-500C) they form highly viscous celes. This reversible vol-gel behavior allows the the dissolved active ingredient can be easily incorporated homogeneously into a cooled Pluronic F-127 solution and put up in a suitable application system (e.g.
Immediately after application A highly viscous gel forms at the application site.
Figure 1 shows the temperature-dependent viscosity behavior of Pluronic F-127 solutions with different proportions of Pluronic F-127 graphically shown. It can be seen from this figure that an optimal viscosity for the body temperature range is reached with solutions whose proportion of Pluronic F-127 is between 18 and 19 percent by weight. A 20% Pluronic F-127 solution must already be cooled more deeply than one in order to bring the active substance into solution 18-19% solution. Figure 2 showing the percentage of Pluronic F-127 against the temperature was plotted also shows why the range 18-19% is a particularly preferred area for pharmaceutical preparations. All solvers with a proportion of 18-19% Pluronic F-127 are in the body temperature range of 37 ° C as a gel and does not have to be cooled below 170 ° C to dissolve the active ingredient.
Die Viskositäten wurden mittels eines Rotationsviskosimeters (Haake Viskotester VT02) gemessen. The viscosities were determined using a rotary viscometer (Haake Viscotester VT02) measured.
Pluronic F-127 formulations of the present invention versus the previously customary gels based on cellulose derivatives have the advantage that the active ingredient can be incorporated in a cool state without special cooling, that Pluronic F-127 can be sterilized without changing its gel behavior and that the active ingredient in Pluronic F-127 solutions has short-term stability.
All tluronic F-127 formulations are lightweight when cool can be applied and guarantee an intensive treatment before they are heated up and gel formation mucosal contrast fitting. Pluronic F-127 is a good mucous membrane compatible Gel former, which is toxicologically harmless and from the body after some time is absorbed.
The invention relates to gel formulations with prostaglandins, Steroid hormones, antifungal drugs, and antibiotics for therapeutic purposes as well Gel formulations with X-ray contrast media, ultrasound contrast media and FMR contrast media for diagnostic purposes.
The gel formulations according to the invention contain the active ingredient in the therapeutically or diagnostically effective dose that is relevant for the Active ingredients is well known. If necessary, the new formulations Additives that are known and customary in galenicals.
Vaginal use of sulproston (16-pheroxy-prostaglandin-E2-methanesulfonamide) -containing Pluronic F-127 gel in pregnant guinea pigs always resulted in rapid insertion of uterine contractions. The overwhelming number who aborted treated animals.
