Die Erfindung betrifft Penicilline, die in C^-Stellung der Acylamino-Seitenkette
mit einem heterocyclischen Ring substituiert und als antibakterielle Mittel von Bedeutung sind.
In der GB-PS 1 130 445 sind Penicilline der Formol A und deren
nicht-toxische Salze beschrieben

in der R ein Phenyl- oder Thienylrest, R1 ein gegebenenfalls
substituierter heterocyclischer Rest und η 0 oder 1 ist.
In dieser Klasse von Penicillinen sind verschiedene Untergruppen beschrieben worden (vgl. GB-PSen 1 407 566 und 1 409 177 sov/ie
US-PS 3 864 329), die am heterocyclischen Ring eine Sauerstofffunktion haben (v/ie ein Keton oder eine gegebenenfalls veresterte
oder verätherte Hydroxylgruppe).
Es v/urde nun festgestellt, dass Penicilline, die in <X-Stellung
der Acylaiainoseitenkette mit einem heterocyclischen Ring substituiert sind, der eine Aminogruppe trägt, ein breites antibakterielles Spektrum haben, insbesondere gegen Pseudomonas-Organismen.
Die Erfindung betrifft daher Penicilline der Formel I, deren pharmakologisch verträgliche Salze und in vivo hydrolysierbare

Alkoxycarbonyloxyalkylester, wie der Äthoxycarbonyloxymethyl- und C^-Äthoxycarbonyloxyäthylester,
sowie Lactone, Thiolactone und Dithiolactone, d„he Verbindungen

mit einem niederen Alkoxyrest, Halogenatom oder einer Nitrogruppe substituiert ist.
Bevorzugt ist der gegebenenfalls substituierte Phthalidylester, z.B. der 3,4-Dimethoxyphthalidylester.
Geeignete Salze der Verbindungen der Formel I sind Metallsalze, z.B. Aluminiumsalze, Alkalimetallsalze, v/ie Natrium- und Kaliumsalze,
Erdalkalimetallsalze, wie Calcium- oder Magnesiumsalze, sowie Ammonium- und substituierte Ammoniumsalze, z.B. Salze mit
niederen Alky!aminen, wie Triäthylamin, niederen Hydroxyalkylaminen,
v/ie 2-Hydroxyäthylamin, Bis-(2-hydroxyäthyl)-amin, Tris-(hydroxymethyl)-amin
oder Tris-(2-hydroxyäthyl)-amin, Cycloaminen, v/ie Bicyclohexylamin, oder mit Procain, Dibenzylamin, N,N-Dlbenzyläthylendiamin,
1-Ephenamin, N-ivthylpiperidin, N-Benzyl-ßphenäthylamin,
Dehydroabietylamin, N,N'-Bis-dehydroabietyläthylendiamin oder mit Pyridinbasen, wie Pyridin, Collidin oder
Chinolin, oder mit anderen Aminen, die mit Penicillinen Salze bilden.
Pharmakologisch verträgliche Säureadditionssalze der Verbindungen der Formel I sind z.B. Salze mit anorganischen Säuren, v/ie das
Sulfat, Nitrat, Phosphat, Borat, Hydrohalogenide, z.B. das Hydrochlorid, Hydrobromid oder Hydrojodid, und mit organischen

Die Erfindung betrifft ferner ein Verfahren zur Herstellung der Penicilline der Formel I, das dadurch gekennzeichnet ist,
dass man eine Verbindung der Formel III oder deren N-geschütztes Derivat

Esterrest oder eine Carboxylschutzgruppe ist, mit einem N-acylierenden
Derivat einer Säure der Formel IV umsetzt

in der X, Z, R , R und R sowie die gestrichelte Linie wie
in Formel I definiert sind und vorhandene freie Hydroxyl- oder Aminogruppen gegebenenfalls geschützt sind,
und anschliessend gegebenenfalls einen oder mehrere der
folgenden Schritte durchführt:
1. Entfernen der N-schützenden Gruppen durch Hydrolyse
oder Alkoholyse,
2. Abspalten der Carboxylschutzgruppen,
3. Entfernen der Schutzgruppen für Amino- oder Hydroxylgruppen , und
4. Umwandeln in ein Salz oder einen Ester.
Beispiele für "N-geschützte Derivate" der Verbindungen der Formel III sind N-Silyl-Derivate und N-phosphorylierte Verbindungen.
Unter "N-Silyl-Derivat" der Verbindungen der Formel III wird
das Umsetzungsprodukt zwischen der 6-Aminogruppe der Verbindung der Formel III und einem silylierenden Mittel, wie
einem Halogensilan oder einem Silazan, verstanden. Im allgemeinen werden Silylchloride, insbesondere Trimethylchlorsilan
und Dimethyldichlorsilan, bevorzugt.
Unter "N-phosphoryliertes" Derivat der Verbindungen der
Formel III werden Verbindungen der Formel III verstanden, in der die 6-Aminogruppe mit einem Rest der Formel

substituiert ist, in der R und R, gleich oder verschieden sind

und Alkyl-, Halogenalkyl-, Aryl-, Arylalkyl-, Alkoxy-,
Halogenalkoxy-, Aryloxy-, Arylalkoxy- oder Dialkylaminoreste
sind, R ausserdem ein Halogenatom bedeuten kann oder R und

R. zusammen einen Ring bilden.
Geeignete Carboxy1-Schutzgruppen für den Rest der Formel
-CO2RX in Formel III sind Salze und Esterderivate der Carbonsäure,
vorzugsweise solche, die zu einem späteren Zeitpunkt leicht abgespalten werden können. Geeignete Salze sind
Salze mit tertiären Aminen, wie Trialky!aminen, N-Sthylpiperidin,
2,6-Lutidin, Pyridin, N-Methy!pyrrolidin oder Dimethylpiperazin.
Ein bevorzugtes Salz ist das Triäthylaminsalz.
/mit einer Aus diesen Estern kann man in herkömmlicher Weise, Z0B0 durch/
Säure, Base oder enzymatisch katalysierte Hydrolyse,, die freie
Säure erhalten.
Die Wahl des N-acylierenden Derivats einer Säure der Formel IV
hängt von der chemischen Natur der in der Säure vorhandenen Substituenten ab. Ein geeignetes N-acylierendes Derivat ist
das Säurehalogenid, vorzugsweise das Säurechlorid oder -bromid. Die Acylierung mit dem Säurehalogenid kann in Gegenwart eines
säurebindenden Mittels erfolgen, z.B„ eines tertiären Ämin„
wie Triäthylamin oder Dime thy !anilin,, einer anorganischen Base,
wie Calciumcarbonat oder NatriumbicarbonatjOder eines Oxirane,
das den während der Reaktion freigesetzten Halogenwasserstoff

bindet. Als Oxiran wird ein 1,2-Alkylenoxid mit 2 bis 6
Kohlenstoffatomen bevorzugt, z.B. Äthylenoxid oder Propylenoxid. Die Reaktion erfolgt im allgemeinen bei einer Temperatur
von -50 bis +50 C,vorzugsweise -20 bis +30 C, in einem
wässrigen oder nicht wässrigen Medium, wie wässrigem Aceton, Äthylacetat, Dimethylacetamid, Dimethylformamid, Acetonitril,
Dichlormethan, 1,2-Dichloräthan oder in Gemischen dieser
Lösungsmittel. Man kann die Reaktion aber auch in einer nicht stabilen Emulsion eines mit Wasser nicht mischbaren
Lösungsmittels durchführen, z.B. in einem aliphatischen Ester oder Keton » wie Isobuty!keton oder Butylacetat.
Das Säurehalogenid wird durch Umsetzen der Säure der Formel IV
oder deren Salz mit einem Halogenierungsmittel, z.B. einem chlorierenden oder bromierenden Mittel, wie Phosphorpentachlorid,
Thionylchlorid oder Oxalylchlorid, hergestellt.
Als N-acylierendes Derivat der Säure der Formel IV kann man
aber auch ein symmetrisches oder gemischtes Anhydrid verwenden. Geeignete gemischte Anhydride sind Alkoxyameisensäure-anhydride
oder Anhydride von z.B. Kohlensäuremonoestern, Trimethylessigsäure, Thioessigsäure, Dipheny!essigsäure, Benzoesäure,
oder
Phosphorsäuren,wie Phosphorsäure /phosphorige Säure, Schwefelsäure
aliphatischen oder aromatischen Sulfonsäuren, wie p-Toluolsulfonsäure. Die gemischten oder symmetrischen Anhydride
können in situ hergestellt werden, z.B. kann man ein Gemisch des Anhydrids unter Verwendung von N-Äthoxycarbonyl-2-äthoxy-1,2-dihydrochinolin
herstellen. Wird ein symmetrisches Anhydrid

verwendet, so kann die Reaktion in Gegenwart von 2,4-Lutidin
als Katalysator erfolgen.

Andere geeignete N-acyIierende Derivate der Säure der Formel
IV sind Säureazide oder aktivierte Ester, ζ.Β» Ester mit 2-Mercaptopyridin,
Cyanomethanol, p-Nitrophenol, 2,4-Dinitrophenol,
Thiophenole Halogenphenol, wie Pentachlorphenol,
oder Monome thoxy phenol oder 8-Hydroxychinolin / Amide j, wie N- Acyl=
saccharine/ oder N-Acylphthalimide oder ein Alkylideniminoester,
der durch Umsetzen der Säure der Formel IV mit einem Oxim hergestellt werden kann.
Diese aktivierten Ester, z.B„ der Ester, der mit 1-Hydroxy-

benzotriazol oder N-Hydroxysuccinimid entsteht, werden zweckmässigerweise in situ durch Umsetzen der Säure mit
der entsprechenden Hydroxylgruppen enthaltenden Verbindung in Gegenwart eines Carbodiimids, vorzugsweise Dicyclohexylcarbodiimid,
hergestellt.
Andere reaktive N-acylierende Derivate der Säure der Formel
IV sind die reaktiven Zwischenverbindungen, die in situ durch Umsetzen mit einem Kondensationsmittel entstehen.
Als Kondensationsmittel geeignet ist ein Carbodiimid, z.B.
oder
N,N-Diäthyl-, Dipropyl-./Diisopropylcarbodiimid, N,Nf-Cyclohexylcarbodiimid oder N-Ä'thyl-N1- # -dimethylaminopropylcarbodiimid, eine geeignete Carbonylverbindung, z.B. Ν,Ν'-Carbonyldiimidazol oder Ν,Ν'-Carbonylditriazol, ein Isoxazoliniumsalz, z.B. N-Ä*thyl-5-phenylisoxazolinium-3-sulfonat oder N-tert,-Butyl-5-methylisoxazoliniumperchlorat, ein N-Alkoxycarbonyl-2-alkoxy-1,2-dihydrochinolin, wie N-Ä'thoxycarbönyl-2-äthoxy-1,2-dihydrochinolin. Andere Kondensationmittel sind Lewis-Säuren,z.B. eine Verbindung der Formel BBr. - CJ oder ein Phosphorsäure enthaltendes Kondensationsmittel, wie Diäthylphosphorylcyanid. Die Kondensationsreaktion erfolgt vorzugsweise in einem organischen Medium, wie Methylenchlorid, Dimethylformamid, Acetonitril, Alkohol, Benzol, Dioxan oder Tetrahydrofuran.
N,N-Diäthyl-, Dipropyl-./Diisopropylcarbodiimid, N,Nf-Cyclohexylcarbodiimid oder N-Ä'thyl-N1- # -dimethylaminopropylcarbodiimid, eine geeignete Carbonylverbindung, z.B. Ν,Ν'-Carbonyldiimidazol oder Ν,Ν'-Carbonylditriazol, ein Isoxazoliniumsalz, z.B. N-Ä*thyl-5-phenylisoxazolinium-3-sulfonat oder N-tert,-Butyl-5-methylisoxazoliniumperchlorat, ein N-Alkoxycarbonyl-2-alkoxy-1,2-dihydrochinolin, wie N-Ä'thoxycarbönyl-2-äthoxy-1,2-dihydrochinolin. Andere Kondensationmittel sind Lewis-Säuren,z.B. eine Verbindung der Formel BBr. - CJ oder ein Phosphorsäure enthaltendes Kondensationsmittel, wie Diäthylphosphorylcyanid. Die Kondensationsreaktion erfolgt vorzugsweise in einem organischen Medium, wie Methylenchlorid, Dimethylformamid, Acetonitril, Alkohol, Benzol, Dioxan oder Tetrahydrofuran.

Bedeuten X, R , R oder R in den oben angebenen Formeln eine Hydroxylgruppe, so kann diese vor der Acylierung in herkömmlicher Weise geschützt werden, z.B. durch Verestern oder

Acylieren. Im allgemeinen ist im erfindungsgemässen Verfahren
jedoch eine Schutzgruppe für die Hydroxylgruppe nicht notwendig.
Auch die Äminogruppe des heterocyclischen Rings in Verbindungen
der Formel IV kann geschützt werden, Z0B0 mit
einem Rest, der unter so milden Bedingungen abgespalten werden kann, dass das Molekül nicht angegriffen wird, oder
mit einem Rest, der ebenfalls unter milden Bedingungen,in
eine Äminogruppe umgewandelt werden kann.
Beispiele für geschützte Aminogruppen sind die protonierte Äminogruppe (NH-), die nach der Acylierung durch einfaches
Neutralisieren in die freie Äminogruppe umgewandelt werden kann^ und der β,β,β-Trichloräthoxycarbonylaminorest, der
durch Reduktion mit Zink in Essigsäure in die freie Äminogruppe umgewandelt wird.
Geeignete Gruppen, die durch katalytische Hydrierung die
Äminogruppe rückbilden, sind Benzyloxycarbonylaminoreste,
die in p-Stellung mit einem Halogenatom, insbesondere einem Chloratom, einer Nitro- oder Methoxygruppe substituiert
sein können, ferner Triphenylmethyl-, Azido- oder Nitrogruppen. Die Hydrierung erfolgt vorzugsweise bei Raumtemperatur
bei atmosphärischem oder etwas höherem Druck. Bevorzugte Katalysatoren sind Edelmetalle, wie Palladium oder
Platin^oder Raney-Nickel. Man kann diese Reste aber auch durch
elektrolytische Reduktion in Aminogruppen tiberführen.

Andere Schutζgruppen, die durch milde saure Hydrolyse in

Beispiel a) 4-Azido-3-carbäthoxychinolin
1/8 g (7,6 mMol) 3-Carbäthoxy-4-chlorchinolin werden in 15 ml
trockenem Dimethylformamid bei Raumtemperatur gelöst und mit 0,8 g (12 mMol) Natriumazid versetzt. Das Gemisch wird bei
Raumtemperatur 24 Stunden gerührt, dann mit 250 ml Diäthyläther und 25 ml Wasser versetzt. Die Phasen werden getrennt,
die wässrige Phase wird zweimal- mit je 25 ml Diä thy lather
extrahiert. Die Ätherextrakte werden vereinigt, gut mit gesättigter Natriumchloridlösung gewaschen,über wasserfreiem
Magnesiumsulfat getrocknet und filtriert. Das Lösungsmittel wird unter vermindertem Druck entfernt. Man erhält 1,77 g
(96 %) eines weissen Feststoffs, Fp. 52 bis 53°C.

werden abfiltriert, mit Wasser gewaschen und an der Luft getrocknet.
Das IR-Spektrum zeigt, dass es sich hierbei um einen
aktivierten Ester handelt.
Das Filtrat wird mit 5 η Salzsäure auf pH 2,5 angesäuert. Das'
ausfallende Produkt (0,6 g = 57 %) wird abfiltriert, mit Wasser gewaschen und über Phosphorpentoxid unter vermindertem Druck

werden zu 300 ml stark am Rückfluss siedendem Diphenyläther
gegeben. Das Gemisch wird 20 Minuten am Rückfluss erhitzt, dann auf Raumtemperatur abgekühlt. Das Produkt wird abfiltriert,
gut mit Ligroin (Siedebereich 40 bis 60°C) gewaschen, und in siedendem Methanol gelöst. Die Lösung wird 30 Minuten mit
Kohle am Rückfluss erhitzt. Die Kohle wird durch Diatomeenerde abfiltriert, das Methanol wird unter vermindertem Druck
bis zur Trockene entfernt. Der zurückbleibende gelbe Feststoff wird in Chloroform gerührt. Das Produkt wird abfiltriert, mit
Chloroform gewaschen und an der Luft getrocknet. Ausbeute: 10,5 g (26 %), Fp. 270 bis 271°C (Zersetzung).
c) 3-Carbäthoxy-4-chlor-7~rcethyl-1,8-naphthyridin "
3,8 g (16 mMol) 3-Carbäthoxy-4-hydroxy-7-methyl-1,8-nap.hthyridin
werden in 46 ml (450 mMol) Phosphoroxychlorid suspendiert und
4 Stunden auf 70 bis 80 C erhitzt. Die Lösung wird unter vermindertem Druck eingeengt, der Rückstand wird vorsichtig in
zerstossenes Eis gegossen. Die entstandene Lösung wird mit lOprozentiger Natronlauge auf pH 6 eingestellt und mit Diäthyl-

Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Trockenmittel wird abfiltriert,
das Filtrat wird durch Sieden am Rückfluss mit Kohle entfärbt, durch Diatomeenerde filtriert und unter vermindertem Druck
zur Trockene eingedampft. Man erhält 3,7 g (92 %) der gewünschten Verbindung, Fp. 92 bis 93°C (Zersetzung).
Durch Chromatographie über Kieselgel mit Chloroform/Methanol im Verhältnis 9t1 als Eluiermittel erhält man eine analytisch

0,7 g (2,8 mMol) 4-Azido-7-chlorchinolin-3-carbonsäure werden
bei 0 bis 5°C in 25 ml trockenem Dimethylformamid gelöst, gerührt und mit 0,33 g (2,8 mMol) N-Hydroxysuccinimid und 0,63 g
(3 mMol) NjN'-Dicyclohexylcarbodiimid versetzt. Das Gemisch wird
1 Stunde bei 0 bis 5°C gerührt, dann nach Erreichen der Raumtemperatur
über Nacht bei dieser Temperatur-gerührt. Das Gemisch
wird dann mit 1,0g (2,8 mMol) Natriumampicillin bei
0 bis 5°C versetzt, 1 Stunde bei 0 bis 5°C gerührt. Nach Erreichen der Raumtemperatur im Verlauf von 30 Minuten wird
das) Reaktionsgemisch in 2 Liter schnell gerührten, trockenen
Diäthyläther gegossen. Der Niederschlag wird abfiltriert, mit trockenem Diäthyläther gewaschen und sofort in 50 ml Wasser
gelöst. Das wässrige Gemisch wird filtriert, der pH-Wert des Filtrats wird mit 5 η Salzsäure -in^Gegenwart von 50 ml Kthylacetat
auf 2,5 eingestellt. Die Phasen werden getrennt, die wässrige Phase wird zweimal mit je 50 ml Äthylacetat extrahiert.
Die Extrakte werden vereinigt, bei pH 2 zweimal mit je 25 ml Wasser,' dann mit 25 ml gesättigter Natriumchloridlösung gewaschen
und über wasserfreiem Magensiumsulfat getrocknet.

Das Lösungsmittel wird unter vermindertem Druck entfernt, der
Rückstand wird mit trockenem Diäthyläther verdünnt. Das Produkt wird abfiltriert, mit trockenem Diäthyläther gewaschen und

0,57 g (2,3 mMol) 4-Azico-7-chlorchinolin-3-carbonsäure werden
bei 0 bis 5 C in 15 ml trockenem Dimethylformamid gelöst, gerührt
und mit 0,26 g (2,3 mMol) N-Hydroxysuccinimid und 0,52 g
(2,5 mMol) N^'-Dicyclohexylcarbodiimid versetzt. Das Gemisch
wird bei 0 bis 5 C 1 Stunde gerührt. Nach Erreichen der Raumtemperatur wird das Gemisch über Nacht bei Raumtemperatur
stehengelassen. Dann wird es wieder auf 0 bis 5 C abgekühlt und mit 1,0 g (2,1 mMol) 6-/D- Ctf-Amino-Ctf-(4-hydroxyphenyl)-acetamidoT-penicillansäure-triäthylammoniumsalz
versetzt. Das Ger misch wird 1 Stunde bei 0 bis 5°C gerührt. Nach Erreichen der
Raumtemperatur im Verlauf von 30 Minuten wird das Gemisch in 2 Liter schnell gerührten trockenen Diäthyläther filtriert.
Der Niederschlag wird abfiltriert, mit trockenem Diäthyläther gewaschen und sofort in 50 ml Wasser gelöst. Das Gemisch wird
abfiltriert, der pH-Wert des Filtrats wird mit 5 η Salzsäure auf 2,8 eingestellt. Der Niederschlag wird abfiltriert, mit
