Description
Die vorliegende Erfindung betrifft neue Piperazinderivate
sowie Verfahren zu deren Herstellung und Arzneimittel, die diese Verbindungen enthalten.
Die z.Z. praktisch angewendeten Antiarrhythmika besitzen
unerwünschte Nebenwirkungen wie z.B. einen negativen Einfluss auf die Kontraktionskraft des Herzens. Der Einsatz
dieser Präparate ist daher nicht problemlos (vgl. z.B. Mürtz et al., Med. Mschr. 2J., 239 - 245 (1970),· Bleifeld et al.,
Dtsch. Med. Wschr. £6, 67I - 68O (1971))· Darüberhinaus weisen
sie nur eine kurze Wirkdauer auf, so dass durch die erforderliche häufige Einnahme in kurzen Zeitabständen zusätzliche
Sicherheitsrisiken für den Patienten entstehen. Es ist daher wünschenswert, über Substanzen zu verfügen,
die diese Nachteile nicht aufweisen.
Es wurde nun gefunden, dass bestimmte Piperazinderivate gut zur Behandlung von Herzkrankheiten geeignet sind. Gegenstand
der Erfindung sind Piperazinderivate der allgemeinen Formel I

Gegenstand der Erfindung ist weiter ein Verfahren zur Herstellung der Verbindungen der allgemeinen Formel I und
deren Salze mit physiologisch verträglichen Säuren, welches darin besteht, dass man Verbindungen der allgemeinen

worin R, , R„, R1, und R . die obengenannte Bedeutung besitzen
mit einer metallorganischen Verbindung reduziert und anschliessend, falls R-, und/oder R„ in den so erhaltenen
Produkten Wasserstoff ist, gegebenenfalls die Substituenten
am Stickstoff einführt und/oder freie Hydroxygruppen daran verestert, und falls R„ und/oder R. Alkylgruppen
sind, diese gegen Wasserstoff oder einen anderen Alkylrest
austauscht und die so erhaltenen Verbindungen gegebenenfalls in ihre Salze mit physiologisch verträglichen
Säure überführt.
Schliesslich betrifft die Erfindung noch Arzneimittel, welche Verbindungen der allgemeinen Formel I und deren
Salze mit physiologisch verträglichen Säuren enthalten. Als physiologisch verträgliche Säuren kommen u.a. in Betracht:
Salzsäure, Schwefelsäure, Phosphorsäure, Essig-

säure, Malonsäure, Bernsteinsäure, Zitronensäure, Weinsäure, Milchsäure, Diamidosulfönsäure.
Die Reduktion der Piperazinonderivate zu den entsprechenden
Piperazinen lässt sich mit komplexen Hydriden wie z.B. Lithiumaluminiumhydrid
oder Dibutylaluminiumhydrid in Äthern, vorzugsweise Diäthyl- oder Diisopropyläther oder cyclische
Äther wie Tetrahydrofuran oder Dioxan, durchführen. Es empfiehlt
sich, bei höheren Temperaturen vorzugsweise den Siedetemperaturen der verwendeten Lösungsmittel zu arbeiten.
Die Alkylierung des Piperazin-Ring-Systems mit substituierten
oder unsubstituierten Diphenylmethylhalogeniden
findet spezifisch am Stickstoff in Position 1 statt. Als Halogenide finden vorzugsweise die Bromide und Chloride
Verwendung, als Lösungsmittel vorzugsweise aromatische Kohlenwasserstoffe wie Benzol, Toluol und Xylol oder niedrigsiedende
Ketone wie Aceton, Methyläthylketon, Diisobutylketon. Geeignet sind auch z.B. Dimethylformamid und Hexamethylphosphorsäuretriamid;
die Temperaturen liegen vorzugsweise zwischen 25 bis 130°C. Es empfiehlt, sich der Zusatz
von basischen Kondensationsmitteln, wie tertiäre organische Basen, oder Alkalicarbonat, wie Kalium- oder Natriumcarbonat.

Eine Alkylierung an den Stickstoffatomen kann entsprechend
erfolgen. Bei Verwendung von AlkylChloriden bzw. -bromiden empfiehlt sich jedoch der Zusatz von Natrium- oder Kaliumjodid
und Anwendung eines geringen Überdrucks von etwa 1,5 bis 10 atü.
Eine Methylgruppe kann auch an die Stickstoffatome gebracht
werden, in dem man die Piperazine in geeigneten Lösungsmitteln, wie z.B. aromatische Kohlenwasserstoffe oder Halogenkohlenwasserstoffe,
in Gegenwart einer Base vorzugsweise Triäthylamin mit Chlorameisensäüreäthylester bei niedrigen
Temperaturen umsetzt. Das so erhaltene Acylierungsprodukt lässt sich sehr leicht in bekannter Weise mit komplexen Hydriden
reduzieren.
Die neuen Verbindungen verfügen über eine gute antiarrhythmische
Wirksamkeit bei Herzfunktionsstörungen unterschiedlicher Genese und Lokalisation.
Sie wirken in sehr geringen Dosen, zeigen also eine hohe Wirksamkeit.
Sie zeichnen sich durch eine ausserordentlich lange Wirkdauer bei oraler und parenteraler (intravenöser) Applikation
aus.

Sie werden gut resorbiert, können also auch oral prophylaktisch verabreicht werden.
Sie wirken weniger stark negativ inotrop als einige bekannte Antiarrhythmika.
Sie wirken in antiarrhythmischen Dosen praktisch nicht blutdrucksenkend
.
Sie antagonisieren die Effekte von vasokonstriktorisch wirkenden biogenen Aminen.
Die für die Reaktion benötigten Ausgangsmaterxalxen sind bislang nicht in der Literatur beschrieben worden. Deren
Herstellung wird im folgenden angegeben.

Herstellung der Ausgangsmaterialien
1.
A) 126,6 g 3)4-Dimethoxybenzyl-a-alanin-inethylester werden
in 25O ml Toluol gelöst und mit 58,3 g frisch destilliertem
Benzaldehyd versetztj die Reaktionslösung wird unter Rückfluss am Wasserabscheider erhitzt. Nach einer Stunde
wird das Lösungsmittel im Vakuum abgedampft, der rohe Benzalester in 300 ml Methanol gelöst und mit Raney-Nickel
hydriert. Nach 2 Stunden wird der Katalysator abfiltriert und das Filtrat im Vakuum zur Trockne eingeengt. Unter
Rühren und Eiskühlung wird mit I67 ml 3 η-Salzsäure versetzt.
Nach kurzer Zeit beginnt das Hydro'chlorid des N-Benzyl-3*4-dimethoxybenzyl-a-alanin-methylesters
zu kristallisieren (F = I85 C). Das Hydrochlorid wird mit I70 ml wässriger
Ammoniumhydroxid-Lösung und 250 ml Toluol versetzt und
gerührt. Die organische Phase wird abgetrennt, mit Wasser' halogenfrei gewaschen und über Magnesiumsulfat getrocknet.
Nach Entfernen des Lösungsmittels erhält man 154 S
(90 % der Theorie) N-Benzyl-( 3,4-dimethoxybenzyl)-oc-alanxnmethylester
als Öl, welches kristallin erstarrt. F 43 - 45°C. Bei Verwendung von optisch aktivem Ausgangsmaterial erhält
man in gleicher Ausbeute D-3?4-Dimethoxybenzyl-a-alaninmethylester.

B) 18,6 g N-Benzyl-3,4-dimethoxybenzyl-a-alanin-methylester
werden in 26 ml Eisessig gelöst, auf 5°C abgekühlt und mit 3,6 g Eis^versetzt. Innerhalb einer Stunde läßt man eine
Lösung von 4?5 g Kaliumcyanid in 8,8 ml Wasser bei 5 bis
10 C zutropfen. Nach einer Stunde wird der Reaktionsansatz
achtzehn Stunden lang bei 40°C gerührt und danach auf etwa 0 C abgekühlt. Die ausgefallenen Kristalle werden abfiltriert,
mit 20 ml Eiswasser gewaschen und getrocknet. Man erhält 18,9 g (95,3 % der Theorie) N-Benzyl-N-cyanomethyl-3,4-dimethoxybenzyl-oc-alanin-methylester
vom Schmelzpunkt 94 96OC (Methanol).
Entsprechend erhält man in gleicher Ausbeute (D)-N-Benzyl-N-cyanomethyl-3j
4-dimethoxyphenyl-a-alanin-methylester

C) In einem Autoklaven werden 38,, 2 g N-Benzyl-N-cyanomethyl-3,
4-dimethoxybenzyl-a-alanin-methylester, 200 ml Toluol sowie
400 ml mit Ammoniakgas gesättigtes Methanol und ein Teelöffel wasserfreier Raney-Cobalt auf 40°C erhitzt. Dann
legt man einen Druck von 100 atü Wasserstoff an. Nach 1-2 Stunden ist die Reaktion beendet. Danach wird filtriert und
das Filtrat eingeengt. Der Rückstand wird aus Methanol umkristallisiert. Man erhält 32,5. g (91,7 % der Theorie)
3-Methyl-3-(3,4-dimethoxybenzyl)-4-benzyl-piperazinon-(2).

D) 13*4 g 3-Methyl-3(3,4-dimethoxybenzyl)-4-benzylpiperazinon-(2)
werden in 80 ml Eisessig suspendiert und mit Palladiumkohle Wasserstoff bei Raumtemperatur hydriert. Nach
Minuten wird durch Filtration vom Katalysator befreit und das Lösungsmittel unter vermindertem Druck abdestilliert.
Der Rückstand wird in 25 ml Chloroform gelöst und mit 20%iger Ammoniumhydroxid-Lösung bis zur stark alkalischen Reaktion
versetzt. Nach Abtrennen der organischen Phase wird diese dreimal mit jeweils 10 ml Wasser extrahiert. Die Chloroformphase
wird unter vermindertem Druck zur Trockene eingedampft und durch Zugabe von Toluol und Abdestillieren derselben im
Vakuum vom restlichen Wasser befreit. Es werden 9*9 g
(99j 5 % der Theorie) 3~Methyl-3-(3,4-dimethoxybenzyl)-piperazinon-(2)
erhalten.

werden in 77 ml trockenem Dimethylformamid suspendiert, mit 8,2g trockenem Kaliumcarbonat versetzt und unter
Rühren eine Lösung von 4>3 g Methyljodid in 8 ml Dimethylformamid zugetropft. Nach weiterem zwölfstündigem Rühren wird filtriert und das Filtrat eingedampft. Der Rückstand wird mit 30 ml Methylenchlorid gelöst und filtriert. Die Lösung wird mit Wasser halogenfrei gewaschen und das Lösungsmittel abdestilliert. Es bleibt ein Öl zurück, welches in der Siedehitze in 80 ml Diisopropyläther gelöst wird. Beim Abkühlen kristallisieren 6,6 g fSl,6 % der Theorie) 3-Methyl-3-(3j4-dimethoxybenzyl)-4-methyl-piperazinon-(2) aus (F 95°C).

A) In 100 ml trockenem Dimethylformamid werden 4>S g 50%iges
Natriumhydrid in Mineralöl suspendiert und bei Raumtemperatur unter Rühren 35*4 g 3- (3j 4-Dimethoxybenzyl)-4-benzyl-3-niethylpiperazinon-(2),
gelöst in 100 ml trockenem Dimethylformamid zugetropftj wobei sich Wasserstoff entwickelt. Es wird 10 Minuten
bei 40 C weitergerührt. Im Verlauf von 20 Minuten wird
eine Lösung von 25 g Diphenylmethylbromid in 100 ml trockenem
Dimethylformamid zugegeben. Nach oOstündigem Rühren bei Raumtemperatur
wird das entstandene Natriumbromid abfiltriert. Das Filtrat wird eingedampft, in Benzol aufgenommen und filtriert.
Das Filtrat wird eingeengt und der Rückstand aus Diisopropyläther umkristallisiert. Man erhält 49^5 g (95 % der
Theorie) l-Diphenylmethyl-3-methyl-(3,4-dimethoxybenzyl)-4-benzyl-piperazinon-(2).
(F I58 - 159° C)
Analog erhält man in gleicher Ausbeute: (D)-l-Diphenylmethyl-3-methyl-3-(3jr4-dimethoxybenzyl)-4-benzyl-piperazinon-(2)
und (L)-l-Diphenylmethyl-3-methyl-3-(3,4-dimethoxybenzyl)-4-benzyl-piperazinon-(2).
Beide Substanzen werden nicht rein isoliert, sondern als Rohprodukt weiterverarbeitet.
B) 36 g l-Diphenylmethyl-J-methyl-3-(3,4-dimethoxybenzyl)-4-benzyl-piperazinon-(2)
werden in 200 ml Eisessig gelöst, mit 2 g lO^iger Palladiumkohle versetzt und bei Raumtemperatur

l) werden in 150 ml Aceton gelöst, mit 27,6 g Kaliumcarbonat und O, 5 g Kaliumiodid versetzt und unter Rühren
zum Sieden erhitzt. Zur Reaktionslösung werden 27,6 g Diphenylmethylbromid in 50 ml Aceton zugegeben. Danach wird fünf Stunden unter Rückfluss erhitzt. Das anorganische Salz wird abfiltriert und das Filtrat eingeengt. Der Rückstand wird in 100 ml Toluol aufgenommen und dreimal mit; 10 ml

um, so erhält man nach zwölfstündiger Reaktionszeit das 1-(p-Chlorphenyl-phenylmethyl)-3-methyl-3-(3,
4-dimethoxybenzyl)-piperazin in 80%iger Ausbeute. Das
Hydrochlorid hat den Schmelzpunkt von 235 - 237 C (isopropanol)
Analog erhält man in gleicher Ausbeute das (D)-I-(p-Chlorphenyl-phenylmethyl)-3-methyl-(3,4-dimethoxybenzyl)-piperazin
als glasartig erstarrtes Hydrat. Laut Wasserbestimmung
nach Karl Fischer enthält das Produkt 10 Mol Wasser.

6,4 g 1-Diphenylmethyl-3-methyl-3-(3*4-dimethoxybenzyl)-piperazin
(vgl. Beispiel 2) werden in 60 ml Aceton gelöst und mit 4)2 g trockenem Kaliumcarbonat versetzt. Dann wird
unter gutem Rühren bei 25 C eine Lösung von 2,2 g Methyljodid
in 20 ml Aceton getropft. Das Reaktionsgemisch wird zwölf Stunden lang bei Raumtemperatur gerührt. Es wird filtriert,
das Filtrat eingeengt, der Rückstand mit 50 ml Toluol aufgerührt
und filtriert. Die toluolische Lösung wird mit Wasser halogenfrei gewaschen und zur Trockene eingedampft.
Es bleibt ein honiggelbes Öl zurück, welches in 50 ml
Diäthyläther gelöst wird. Nach Einleiten von Chlorwasserstoff wird das entstandene Dihydrochlorid abgesaugt, zweimal
mit 20 ml Diäthyläther gewaschen, getrocknet und aus Äthylalkohol umkristallisiert. Man erhält 6,2 g (80 % der Theorie)
1-Diphenylmethyl-3-methyl-3- ( 3 j 4-dimethoxybenzyl)-4-methylpiperazin
(F 193°C).

50 g l-Diphenylmethyl-3-methyl-3-(3,4-dimethoxybenzyl)-piperazin
(vgl. Beispiel 2) werden in 200 ml Benzol und 400 ml Methanol gelöst und 48 Stunden in einem Druckgefäss
unter Zusatz von 22 g Äthylenoxid auf 60°C erhitzt. Anschliessend wird axe Lösung zur Trockene eingedampft. Der
Rückstand wird in 500 ml Diethylether gelöst, mit 60 ml 2n-HCl versetzt und kräftig gerührt. Die organische Phase
wird abgetrennt und mit Wasser halogenfrei gewaschen. Der wässrig saure Auszug wird mit Ammoniumhydroxidlösung alkalisiert,
mit Diethylether extrahiert, dieser getrocknet, filtriert und das Lösungsmittel abdestilliert. Es werden
54 g (98 % der Theorie) l-Diphenylmethyl-S-methyl-S-(3,4-dimethoxybenzyl)-4-hydroxyäthyl-piperazin
als farbloses Harz erhalten.

4,3 g 1-Diphenylmethyl-3-methyl-3-(3r4-dimethoxybenzyl)-4-hydroxyäthyl-piperazin
(vgl. Beispiel 7) werden unter Rühren in eine Suspension von 0,44 S Natriumhydrid in Mineralöl
in 20 ml trockenem Dimethylformamid zu'getropft und auf
80°C erwärmt. Nach einer Stunde wird eine Lösung von 2,1 g 3,4,S-T^imethoxybenzoylchlorid in 10 ml trockenem Dimethylformamid
zugetropft und die Suspension 2 Tage bei Raumtemperatur gerührt. Nach Abdestillieren des Lösungsmittels
wird der Rückstand in 100 ml Toluol gelöst, mit wenig Wasser
halogenfrei gewaschen und eingeengt. Der ölige Rückstand wird in 100 ml Diäthyläther gelöst, mit Chlorwasserstoff das
Dihydrochlorid hergestellt, filtriert und aus Isopropanol umkristallisiert. Man erhält 5 g (74,4 % der Theorie)
1-Diphenylmethyl-3-methyl-3-(3,4-dimethoxybenzyl)-4-ß-(3,4,5-trimethoxybenzoyl-oxyäthyl)-piperazin-Dihydrochlorid
(F 202°C).

Zu einer Suspension von 11,6 g Lithiumaluminiumhydrid in
l600 ml trockenem Tetrahydrofuran wird unter Rühren in der
Siedehitze eine Lösung von 55?6 g 3-Methyl-3-(3*4-dimethoxybenzyl)-4-methyl-piperazinon-(2)
(vgl. Beispiel 2) in 300 ml trockenem Tetrahydrofuran innerhalb von 2 Stunden zugetropft
und die Reaktionslösung eine weitere Stunde unter Rühren zum
Sieden erhitzt. Nach vorsichtigem Zugeben von Wasser wird vom Unlöslichen filtriert und das Filtrat zur Trockene eingedampft
und destilliert. Bei 103-165°C/5"10~ Torr gehen

13,3 g 3-Methyl-3-(3,4-dimethoxybenzyl)-4-methyl-piperazin
(vgl. Beispiel 10) werden mit 13,8 g trockenem Kaliumcarbonat sowie 0,1 g Kaliumiodid in 75 ml trockenem Aceton zum Sieden
erhitzt und unter Rückfluss und Rühren mit 12,3 g Diphenylmethylbromid
in 25 ml trockenem Aceton versetzt. Nach 5 Stunden wird abgekühlt, filtriert und das Filtrat zur Trockene eingedampft.
Der ölige Rückstand wird in I50 ml Toluol aufgenommen und mit Wasser halogenfrei gewaschen. Die organische Phase wird
eingedampft und der ölige Rückstand in 120 ml Diäthyläther gelöst.
Nach Einleiten von Chlorwasserstoff wird das entstandene Dihydrochlorid abfiltriert, mit Diäthyläther gewaschen und
aus wenig Äthylalkohol umkristallisiert. Es werden 13,7 g (85 % der Theorie) l-Diphenylmethyl-S-methyl-S-(3,4-dimethoxybenzyl
)-4-methyl-piperazin-dihydrochlorid (F 193°C) erhalten.

(vgl.3-c) ^verden in 80 ml absolutem Tetrahydrofuran gelöst und in der Siedehitze innerhalb
zwei Stunden in eine gerührte Suspension von 2,3 g Lithiumaluminiumhydrid
in 80 ml Tetrahydrofuran getropft. Die Suspension wird weitere sechs Stunden unter Rückfluß und
Rühren erhitzt. Nach vorsichtiger Zugabe von Wasser und
Filtration wird das Lösungsmittel abdestilliert. Es wird ein Öl erhalten, welches aus Diäthyläther umkristallisiert,
wird. Man erhält 7,7 g (92,5 % der Theorie) 1-Diphenylmethyl-3-methyl-3-(3>4-dimethoxybenzyl)-piperazin.

Stunden in eine siedende Suspension von 5*75 g Lithiumaluminiumhydrid
in 400 ml trockenem Tetrahydrofuran eingetropft und weitere drei Stunden zum Sieden erhitzt. Nach vorsichtiger
Zugabe von Wasser wird filtriert und das Filtrat eingedampft. Das zurückbleibende Öl wird analog Beispiel 4
weiter verarbeitet. Man erhält 20 g (80 % der Theorie) 1-Diphenylmethyl-3-methyl-3-(3,4-dimethoxybenzyl)-4-methyl-
