Description
Die Erfindung bezieht sich auf ein Verfahren und Gerät zum Messen der Zuckerkonzentration, insbesondere in einer Körperflüssigkeit, z. B. Die dem Zuckerkranken zur Selbstbestimmung seines Blutzuckers angebotenen Urin-Färbeteststreifen ergeben auch nur einen äußerst groben Anhaltspunkt über den BJutzuckergehalt und sind daher als Grundlage für eine gezielte Therapie praktisch nicht geeignet Die genaue Bestimmung des Blutzuckers, die nur im Labor erfolgen kann, erfordert die Abnahme von Blut, wozu der Patient in die Klinik oder zum Arzt kommen muß, was natürlich wegen der Belästigung des Patienten und der anfallenden Kosten nicht häufig genug durchgeführt werden kann. Aus diesem Grunde hatte man also bereits den Einsäte von Glucosezellen bei der Messung der Zuckerkonzentration vorgeschlagen, die eine raschere Meßanzeige für den Konzentrationswert auch bei Anwendung durch technische Laien liefern sollten. Bei den zu diesem Zweck bisher vorgeschlagenen Glucosezellen (z. B. DT-OS 19 32 581. DT-OS 19 33 302, Nature. Vol. 214, 1967, S. 986 bis 988) handelt es sich jedoch durchweg um sogenannte Enzym-Elektroden, wo also dem Elektrolyten ein Enzym, z. B. Glucose-Oxidase oder Peroxidase, beigefügt ist. Dieses Enzym bewirkt, daß im Elektrolyten vor den eigentlichen Zellen-Elektroden Glucose mit Sauerstoff direkt chemisch reagiert, beispielsweise in dem Sinne, daß Glucose mit Sauerstoff zu Glucon-Säure und Wasserstoffperoxyd reagiert Durch diese chemische Reaktion ändern sich die Eigenschaften des Elektrolyten, beispielsweise im Hinblick auf die Sauerstoffkonzentration, den pH-Wert und/oder die Leitfähigkeit. Die sich jeweils einstellende Änderung eines dieser Werte dient dann als indirektes Maß für den Glucosegehalt der Lösung. Ein solches indirektes Meßverfahren ist jedoch wenig exakt und sehr störanfällig, da z. B. aufgrund von Störungen verursachte Änderungen der Sauerstoffkonzentration, des pH-Wertes und/oder der Leitfähigkeit im Elektrolyten immer in die Messung mit eingehen und demnach das Meßergebnis auch verfälschen können. Neben den Enzym-Elektroden sind ferner auch noch solche Meßeinrichtungen bekanntgeworden (z. B. GB-PS 7 96 674), die den Zuckergehalt einer Zuckerlösung über eine elektrische Leitfähigkeitsmessung bestimmen. Zu diesem Zweck werden in die Zuckerlösung Elektroden eingeführt und durch Anlegen eines Wechselstromes die Stromleitfähigkeit der Lösung zwischen den Elektroden ermittelt. Leitender Bestandteil ist dabei jedoch nicht der Zucker; die Leitfähigkeit wird vielmehr bestimmt durch die Beweglichkeit der Ionen sonstiger Bestandteile der Lösung, die mit zunehmender Zuckerkonzentration entsprechend abnimmt. Die Zucl;erkonzentration wird also, wie auch schon bei den Enzym-Elektroden nur indirekt ermittelt und es ergeben sich damit dieselben Nachteile, die auch der indirekten Messung mit Enzym-Elektroden anhaften. Zu beachten ist außerdem, daß das Leitfähigkeits-Meßverfahren keine chemischen Umsetzreaktionen am Zucker bewirkt. Gemäß der Erfindung reagiert vielmehr die Glucose nach Antransport an die Oberfläche der Glucoseelektrode direkt elektrochemisch, d.h. das Glucosemolekül ist der elektroaktive Teil. Die Reaktion unmittelbar an den Elektroden verläuft dabei in der Weise, daß an der einen Elektrode (Glucoszelektrode) die Glucose oxidiert, an der anderen Elektrode (Sauerstoffelektrode) hingegen Sauerstoff reduziert wird. Da an der Glucoseelektrode jedoch die Glucose der Körperflüssigkeit direkt umgesetzt wird, ergibt sich über den gemessenen Elektrodenstrom oder die gegebenenfalls gemessene Elektrodenspannung bzw. Transitionszeit auch ein direktes Maß für den Zuckergehalt in der Körperflüssigkeit. Dieses Maß ist jedoch wesentlich exakter als das beispielsweise mit Enzym-Elektroden gewonnene indirekte Maß für den Zuckergehalt, da beispielsweise aufgrund von Störung hervorgerufene Änderungen, z. B. der Sauerstoffkonzentration, des pH-Wertes und/oder der Leitfähigkeit im Elektrolyten in die Messung nicht eingehen und damit für das Meßergebnis ohne Bedeutung sind. Dieselben Vorteile ergeben sich jedoch auch gegenüber jenen Meßverfahren, die mit Leitfähigkeitsmessung arbeiten. Mit der Erfindung erhält man also exakte direkte Konzentrationswerte des Zucker^ die als elektrisches Signal sofort anfallen, d. h. der jeweils ermittelte exakte Konzentrationswert kann dann wie erwünscht sofort an einem geeigneten Signalanzeigegerät abgelesen werden. Das Arbeiten nach dem erfindungsgemäßen Verfahren erfordert praktisch keine besondere technische Erfahrung, d. h. die Ermittlung des Konzentrationswertes kann auch durch einen technischen Laien erfolgen. Geräte zur Durchführung des erfindungsgemäßen Verfahrens sind in den Unteransprüchen 2 bis 11 beschrieben. Die bei diesen Geräten eingesetzten Glucosezellen können auf kleinstem Raum aufgebaut werden und eignen sich daher auch für die Implantation im Körper eines Patienten. Damit ergibt sich insbesondere für einen Zuckerkranken die einfache Möglichkeit, sich laufend ausreichend genau über seine Blutzuckerkonzentration zu informieren (z. Jede dieser Zellen 22 besteht aus zwei für die Glucose durchlässige Sauerstoffelektroden 23, 24 sowie einer zwischen diesen Elektroden angeordneten Glucoseelektrode 25 (die Elektroden sind untereinander sowie zur Körperflüssigkeit hin durch Trennwände aus hydrophilem Material, vorzugsweise Ionenaustauscher, abgetrennt). Die Sauerstoffelektroden 23, 24 bestehen jeweils aus einem Silbernetz mit aufgebrachter Kohle als Katalysator. Die Glucoseelektrode 25 besteht aus platiniertem Platin.
