Abstract
Description
Novel Coumarin Prevents and Methods for Making Them The Invention relates to new coumarin compounds and a process for their preparation.
No substances have yet been described which were all previously desirable Have properties for sunscreens, namely: (1) stability, (2) none Odor, (3) no causing skin irritation and the like, (4) no causing a sensitization of the human skin to sunlight, (5) oil compatibility and (6) protection against both the scorching and tanning rays of the Sun Sunscreens have already been developed that do one provide satisfactory protection against the sun's scorching rays but let the tanning rays pass through. In sunlight can be found in an approximate Range from about 2 900 to 3 100 R the rays with the most harmful burning ones Wavelengths. Rays with these wavelengths affect human skin the development of severe burns, redness) blistering and edema formation. These phenomena are characteristic of many sunburns. These appearances are not only undesirable from an aesthetic point of view, they are they also represent a serious impairment to health. For elimination the effect of these scorching rays of the sun are therefore already chemical compounds been used to filter out these rays. Because of the general attitude the public that tanning is a desirable phenomenon, which goes hand in hand with a good state of health, one has so far numerous products used, which filter out the burning rays of the sun, but the tanning ones Let rays through.
In recent years it has been widely accepted that, although the tanning has some beneficial effects, especially in terms of protecting the skin Skin, from a further burn, nevertheless the tan or Tanning with a general destruction of the skin due to the influence of actinic Radiation is connected. Although the majority of the population through such Browning or tanning is not greatly impaired, but there are a large number by people who are so sensitive even to the sun's tanning rays are that exposure to radiation at this wavelength poses serious health hazards brings with it. Other people again do not share the general Opinion that the tan is beautifying and prefer to remain untanned. Thus there is for those persons who need protection against both the burning as well as need the tanning rays of the sun, and for those who don't want to get tanned, a need for sunscreens with a wide range of effects, which provide adequate skin protection. These funds should be considered in the conditions of use stable enough to remain effective for several hours, and they should be easily applied to the skin. You would have to continue with the various oily cosmetic preparations that are currently on the market, be compatible. After all, they should neither irritate nor sensitize the skin cannot be easily removed by simply washing with water. The need for the final property is obvious as it becomes a false one A feeling of security would result if the material was readily available, for example when bathing, would be removed.
The term "non-sensitizing" as used herein is intended to mean that the compounds according to the invention reduce the sensitivity of the skin to light exposure to ultraviolet light in the range of about 2900 R to about 7400 2 should not reinforce.
In order to achieve the objects of the present invention it is therefore necessary to provide connections that (1) focus on the sun's rays in the range of about 2 900 R to about 5 400 R exert an excellent filtering effect. This The range includes both the burning and the tanning wavelengths of sunlight. The connections should continue to (2) odorless and does not irritate or sensitize the skin.
It is also necessary / that the connections with the above mentioned properties are either essentially oil-miscible or that there is to it is able to produce a non-granular smooth suspension in oils (e.g. in those as used in the usual cosmetic sunscreens for the skin to be able to form, and with the various currently common cosmetic Be compatible with preparations.
It is also necessary that the absorption properties for the ultraviolet radiation of the compounds having the aforementioned properties are not changed by changes in pH in the environment.
According to the present invention, it has now been found that (1) a class of compounds described below have excellent filtering properties for the burning and tanning rays of the sun, especially for the ultraviolet ones Has rays with about 2,900 to about 3,400 R, (2) that these compounds have a have unexpectedly good lightfastness and stability when exposed to ultraviolet radiation ()) that these compounds also retain their filtering properties maintained in the event of changes in pH in the vicinity of these compounds, (4) that these compounds are mixed with oily mixtures suitable for application to the are intended for human skin, are compatible (i.e.
daß sie stabile Suspensionen bilden oder damit mischbar sind) und (5) daß diese Verbindungen geruchlos sind und die menschliche Haut nicht reizen und nicht sensibilisieren.that they form stable suspensions or are miscible with them) and (5) that these compounds are odorless and do not irritate human skin and not sensitize.
It has been found that known agents, e.g.
4-Methyl-7-hydroxycumarin (vergleiche britische Patentschrift 464 732 und Gregoire et al, Ann. Pharm. Pharm. Franc., 9, 493-513 (1951)) all do not have the advantages mentioned above. These connections have none Stability that their filtering or sieving properties even in the case of pH changes the environment. They are also not considered compatible with considered oily preparations which are used on the human skin. This also applies to other known compounds, such as 4-methyl-7-lower alkoxy coumarin or asculin (6,7-dihydroxycoumarin-6-glucoside).
The compounds which have been found to be suitable according to the invention and which are used in the agents according to the invention have the general formula: wherein R1 is alkadienyl, cycloalkadienyl or cycloalkenyl.
Suitable groups R1 are, for example: 1. Alkadienyl with 5 to 20 carbon atoms, 2. Cycloalkadienyl with 6 to 10 carbon atoms, 3. Cycloalkenyl with 5 to 8 carbon atoms, The general procedure for making those contemplated by the present invention Connections sees the Implementation of a suitable halide with 7-hydroxy-coumarin (umbelliferone) or 7-hydroxy-4-lower-alkyl-coumarin.
Als Ausgangsstoffe geeignete Halogenide sind z.B.: 1. Cycloalkenylchloride oder -bromide mit 3 bis 15 Kohlenstoffatomen, 2. Alkadienylchloride oder -bromide mit 5 bis 20 Kohlen-. stoffatomen, 3. Cycloalkadienylchloride oder -bromide mit 6 bis 10 Kohlenstoffatomen.Halides suitable as starting materials are, for example: 1. Cycloalkenyl chlorides or bromides with 3 to 15 carbon atoms, 2. alkadienyl chlorides or bromides with 5 to 20 coal. material atoms, 3. cycloalkadienyl chlorides or bromides with 6 to 10 carbon atoms.
The condensation takes place in the presence of bases, for example of Alkali metal carbonates or bicarbonates, such as potassium carbonate or sodium carbonate, and in an organic solvent compatible with the reactants is. Alkyl or alicyclic ketones, such as acetone or cyclohexanone, are preferred. In view of the simplicity and economy of the reaction, the The reaction is preferably carried out at atmospheric pressure and under reflux. Higher Reaction temperatures and consequently also higher pressures reduce the reaction time. It is advisable to add a catalyst such as potassium or sodium iodide, to increase the reaction speed. This varies at higher temperatures Reaction time from about 10 hours to about 30 hours depending on the particular one used Halide and the yield of product that is desired. At the end of the response period the reaction product becomes in the usual manner by crystallization or distillation isolated.

Umbelliprenine with the structure: The range of application for the compounds considered according to the invention in oily sunscreens is about 1% to about 10% (based on the weight of the ready-to-use preparation). The preferred range is about 1% to about 6% by weight. Any oily carrier suitable for application to human skin can be used. Examples are: lanolin, cetyl alcohol, wheat germ oil, sesame oil, polyethylene glycol stearate (in which the polyethylene glycol portion has a molecular weight of less than 400), other fatty acid esters of polyethylene glycol (in which the polyethylene glycol portion has a molecular weight of less than 400) and the like. After the preparation has been produced, it can be applied to the human skin in any arbitrary and easy-to-use manner. An example of the amount to be applied is an amount of 5 to about 200 mg (of the active ultraviolet absorption compound = coumarin ether) per square meter of skin. In the individual case it will depend on the environment of exposure and the nature of the skin. For example, high-intensity ultraviolet radiation in the case of highly sensitive skin would require the preparations to be applied with concentrations in the upper part of the range.

Examples 3 to 5 In the following examples, as in example 1 procedure, with the exception that instead of myrcene hydrochloride the following Halides were used in molar equivalent amounts: Example halide reaction product Ultraviolet values (E at 3,220) 3 allyl chloride 7-allyloxy-4-methyl-17,500 coumarin 4 prenyl chloride 7- (3-methyl-2-butene-1-17,600 oxy) -4-methyl-coumarin 5 benzyl chloride 7-benzoxy-4-methyl-17,650 coumarin Examples 6 to 8 The compounds Examples 3 through 5 each became a separate sample from that in Example 2 given cream. Using the compounds of Examples 3 to 5 in place of the compounds of Example 1 gave essentially the same results obtain. There was only the exception that the compounds of Examples 3 to 5 formed suspensions rather than miscible systems with the oily sunscreens. Even so, the products of Examples 3 to 5 were compatible with the oily carrier.
