6)-Homo-prostaglandin-E -heptylester6) Homo-prostaglandin-E -heptyl ester
The invention relates to the cü-Horaoprostaglandin-E-j ^ -heptyl ester and a method as a new compound its manufacture. The invention also relates to the use of the CJ homoprostaglandin Ej ^ -heptyl ester as an active ingredient directed in pharmaceutical preparations which are used for the prophylaxis or therapy of thrombosis.
It is also known that 6MlOmO-PGE 1 and prostaglandin E 1 (hereinafter referred to as PGE 1 ) have a strong effect on the contraction of the smooth muscles and a strong antihypertensive effect and inhibit the agglomeration of blood platelets. OJ-Homo-PGE , differs only slightly in chemical structure from PGE 1 , since & J-HOmO-PGE 1

Daher ist <U -Homo-PGE. als zweitbesseres Thromboseheilmittel anzusehen als PGE1.over PGE, has only one more carbon atom in the u- position, but uJ-homo-PGE .. is much stronger in the above-mentioned pharmacological effects than prostaglandin E 1 . For example, UJ-HOmO-PGE 1 inhibits platelet aggregation four times more than PGE ,. It is also as effective as PGE 1 in lowering blood pressure and intestinal contraction. In general, the antihypertensive effect and the effect on the intestinal contraction are considered undesirable side effects of thrombotic drugs. Hence <U -Homo-PGE . to be regarded as the second better thrombosis remedy than PGE 1 .
Es hat sich jedoch gezeigt, daß (J-HOmO-PGE1 die folgenden Nachteile in der Therapie hat:From the above point of view, the applicant synthesized Cu- homo-PGEi by a new method and investigated its use as a remedy for thrombosis. However, it has been shown that (J-HOmO-PGE 1 has the following disadvantages in therapy:
1. Its antihypertensive effects are still too strong, and so are the hypotensive effects and the intestinal contraction To establish sina at an effective dose.
The inhibitory effect on thrombus formation in vivo is transient and short-term.
To change 1 so that it is suitable as a thrombosis remedy, the applicant synthesized a wide variety of PGE 1 analogs in search of a compound that does not have the disadvantages mentioned above. As a result, it was found that the CJ -HOmO-PGE 1 -heptyl ester does not suffer from these disadvantages

The inhibition of thrombus formation is less with the (J-homo-PGE, -heptyl ester than with CJ-homo- PGE, but with the effective dose of ω-Hoaio-PGE 1 -heptyl ester no antihypertensive effect or induction of intestinal contractions is found , and the ester has the lo-facne duration of action of W-Homo-PGE «in vivo.

Agents were added to each of rabbit platelet rich plasma (PRP). The ADP, trombin or serotonin was dissolved in a physiological saline solution adjusted to pH 7Λ. The W-HOmO-PGE 1 heptyl ester was dissolved in ethanol at a concentration of 12 mg / ml. Each solution was then diluted to the appropriate concentration. The aggregation of the platelets was measured with an aggrenometer (Evans Model EEL 169). The result was recorded on a recorder as a function of time. The W-PGE ^ heptyl ester was added to the plasma and pre-incubated for 5 minutes before the addition of the agglomerating agent.

II. Hemmende Wirkung auf die Thrombusbildung II. Inhibitory effect on thrombus formation
Der Einfluss des W-HOmO-PGE1-Heptylesters auf die Thrombusbildung in Mesenteriumgefäßen wurde nach der Methode von Honour und Mitarbeitern ( Brit.J.Exp.Pathol. 45 175 1964)) untersucht. The influence of the W-HOmO-PGE 1 heptyl ester on thrombus formation in mesenteric vessels was investigated using the method of Honor and coworkers (Brit.J.Exp.Pathol. 45 175 1964)). A mesentery of a rabbit anesthetized with urethane was placed under a stereomicroscope and bleeded by injuring the arteriolar wall using a sharp needle about 50 µm in diameter. After the bleeding stopped, white stoppers formed. The phenomenon that the flow of blood was stopped by the white plug was frequently observed. The "forming" or "already formed" stoppers could, however, be completely dissolved by intravenous injection or infusion of J-homo-PGE.-heptyl ester.
The long duration of action of the Uj-HOmO-PGE 1 -Heptylester in vivo can also be seen in a comparison of the duration of the blood pressure lowering in Table 4. It is believed that in the case of the heptyl ester, the affinity of the prostaglandin-deactivating enzyme for the ester is reduced, so that the heptyl ester is stable.
