In Fig. 1 ist eine erfindungsgemäße Vorrichtung zur Durchführung von Messungen mit einer erfindungsgemäßen biosensorischen Schicht (Biosensor) schematisch dargestellt. Die Vorrichtung besteht aus einer äußeren Meßkammer 1 aus Polytetrafluorethylen und einer inneren Meßkammer 2 aus demselben Material. In der Meßkammer 1 befindet sich ein Flüssigkeitsreservoir 3, wogegen die Meßkammer 2 mit einem Flüssigkeitsreservoir 4 gefüllt ist. In der Meßkammer 2 befindet sich eine Öffnung 5 mit einem Durchmesser von 0,6 mm. Diese Öffnung 5 wird mit einem Biosensor verschlossen, der aus einer löchrigen Membran 6 besteht, in deren Löcher 29 sich Lipiddoppelschichten 30 mit integrierten Ionenkanälen 32 befinden. Die löchrige Membran 6 ist dabei mit einem schlecht leitenden Silikonklebstoff 7 an der Meßkammer 2 befestigt. An der Unterseite der Meßkammer 1 ist ein handelsübliches Mikroskopdeckgläschen 8 mit einer Dicke von 0,2 mm befestigt. Dieses läßt die Beobachtung mit einem Mikroskop zu, dessen Strahlengang 9 angedeutet wurde. Zur Perfusion befindet sich an der äußeren Meßkammer 1 ein Einströmkanal 10 und ein Ausströmkanal 11. Zum Zweck der Injektion von Applikationslösungen 14 in das Flüssigkeitsreservoir 3 befindet sich im Einströmkanal 10 ein aus der Gas- und der Flüssigkeitschromatographie bekanntes Teflon- bzw. Gummiseptum 12. Durch dieses können mit einer Mikroliterspritze 13 die Applikationslösungen 14 injiziert werden. Über den Entlüftungsspalt 15 werden mögliche Luftbläschen 16 aus der einströmenden Lösung befreit. Die Injektion von Applikationslösungen 17 in das Flüssigkeitsreservoir 4 kann über eine Mikropipette 18 durch Anlegen eines kurzzeitigen Überdrucks 19 mittels eines handelsüblichen Mikroinjektors erfolgen. Für elektrophysiologische Messungen befindet sich im Flüssigkeitsreservoir 3 eine Silberchloridelektrode 20 und im Flüssigkeitsreservoir 4 eine Silberchloridelektrode 21. Einerseits zum Schutz der Oberfläche der Elektrode 21 vor Verunreinigungen durch Stoffe aus dem Flüssigkeitsreservoir 4 und andererseits zum Schutz der in der Lipidschicht in der löchrigen Membran 6 integrierten Ionenkanäle 32 vor einer möglichen Inhibierung durch Silberionen befindet sich im dargestellten Ausführungsbeispiel die Silberelektrode 21 in einer Mikropipette 22, die mit einer Pipettenlösung 23 gefüllt ist. Durch den kleinen Innendurchmesser der Pipettenspitze 24 von etwa einem Mikrometer wird eine Diffusionsbarriere für den Konzentrationsausgleich der im Flüssigkeitsreservoir 4 und in der Pipettenlösung 23 befindlichen Stoffe geschaffen.In Fig. 1, an apparatus according to the invention for performing measurements with an inventive biosensing layer (Biosensor) is shown schematically. A commercially available microscope cover slip 8 with a thickness of 0.2 mm is attached to the underside of the measuring chamber 1 . This permits observation with a microscope, the beam path 9 of which has been indicated. An inflow channel 10 and an outflow channel 11 are located on the outer measuring chamber 1 for perfusion. For the purpose of injection of application solutions 14 in the liquid reservoir 3 is located in the inlet conduit 10 a well-known from the gas and the liquid teflon or rubber septum 12th Through this, the application solutions 14 can be injected with a microliter syringe 13 . On the one hand, to protect the surface of the electrode 21 from contamination by substances from the liquid reservoir 4 and, on the other hand, to protect the ion channels 32 integrated in the lipid layer in the perforated membrane 6 from possible inhibition by silver ions, the silver electrode 21 is located in a micropipette 22 in the exemplary embodiment shown , which is filled with a pipette solution 23 . The small inside diameter of the pipette tip 24 of approximately one micrometer creates a diffusion barrier for the concentration compensation of the substances in the liquid reservoir 4 and in the pipette solution 23 .
In der Fig. 2 ist eine Ausführung einer erfindungsgemäßen Vorrichtung, bestehend aus einem Mikroskopadapter 25 mit den dazugehörigen Meßkammern 1 und 2 in demontierten Zustand dargestellt. Der abgebildete Mikroskopadapter 25 dient zur Befestigung der Meßkammern 1 und 2 an einem Mikroskoptisch eines inversen Mikroskops. Alle drei dargestellten Teile sind aus Polytetrafluorethylen hergestellt. Die Meßkammer 1 enthält zwei zylindrische Bohrungen 26, durch welche Kühlflüssigkeit zur Thermostatierung der Vorrichtung geleitet werden kann. Ferner ist in der abgebildeten Meßkammer 1 eine Halterung 27 für eine Silberchloridelektrode zu erkennen. In der Meßkammer 2 ist eine Nut 28 für einen laminaren Fluß der Lösung von Flüssigkeitsreservoir 3 eingearbeitet.In FIG. 2 is an embodiment of a device according to the invention, shown consisting of a microscope adapter 25 with the associated measurement chambers 1 and 2 in the disassembled state. Fig. 3 zeigt eine rasterelektronenmikroskopische Aufnahme einer löchrigen Membran 6 aus Polyethylenterephthalat, in die durch Ionenbeschuß mit anschließender Ätzung annähernd kreisrunde Löcher 29 mit einem mittleren Durchmesser von 400 nm eingebracht wurden. Die Membrandicke der abgebildeten Polyesterfolie beträgt 10 µm und die Porosität 10%. Ein Loch 29 dieser Polymermembran 6 wird durch eine Lipiddoppelschicht 30 verschlossen, die in horizontaler Richtung über die Lipidreservoire 31 mit der Polymermembran 6 verbunden ist. In vertikaler Richtung grenzt die Lipiddoppelschicht 30 einerseits an das Flüssigkeitsreservoir 3 und andererseits an das Flüssigkeitsreservoir 4. In der Lipiddoppelschicht 30 sind in der schematischen Darstellung zwei Ionenkanäle 32 integriert. Die elektrophysiologische Messung in Fig. 5 wurde mit einem erfindungsgemäßen Biosensor mit integrierten Gramicidin A-Kanälen bei einer Kommandospannung von -150 mV und einer Temperatur von 21°C erhalten. Für den Aufbau der Lipiddoppelschicht wurde eine 3% (w/v)-igen-Dekanlösung von einem natürlichen Sojabohnenextrakt und Cholesterin, welche im Molverhältnis 60 : 40 zueinander standen, verwendet. In den Flüssigkeitsreservoiren befanden sich jeweils 1 M CsCl-Lösungen. Als löchrige Membran 6 wurde eine 10 µm dicke Polyethylenterephthalatfolie eingesetzt, die Löcher mit einem mittleren Durchmesser von 10 µm und eine Porosität von 15% besaß. Aus der elektrophysiologischen Einzelkanalaufnahme dieses Ausführungsbeispiels kann man entnehmen, daß bei einer Stromstärke von ca. 3 pA kein Ionenkanal, bei einer Stromstärke von ca. 11 pA jeweils genau ein integrierter Gramicidin A-Kanal, bei einer Stromstärke von ca. 19 pA jeweils genau zwei integrierte Gramicidin A-Kanäle und bei einer Stromstärke von ca. 27 pA jeweils genau drei integrierte Gramicidin A-Kanäle gleichzeitig geöffnet sind.The electrophysiological measurement in FIG. 5 was obtained with a biosensor according to the invention with integrated gramicidin A channels at a command voltage of -150 mV and a temperature of 21 ° C. In Fig. 6 ist das Ergebnis einer elektrophysiologische Messung dargestellt, bei deren Aufnahme sich in beiden Flüssigkeitsreservoiren CsCl und CaCl2 in der Konzentration von jeweils 1 mol l-1 befanden. Ansonsten galten die gleichen Bedingungen wie bei der elektrophysiologischen Aufnahme in Fig. 5. Zu erkennen ist, wie die Gramicidin A-Kanäle durch die zweiwertigen Ca2+-Ionen blockiert werden. Im Unterschied zu der Aufnahme in Fig. 5 sind bei der Messung in Fig. 6 deutlich kürzere mittlere Kanalöffnungszeiten und kleinere mittlere Stromamplituden der integrierten Ionenkanäle zu beobachten. Das Ausführungsbeispiel in Fig. 7 zeigt schließlich das Ergebnis einer elektrophysiologische Messung, die mit einem erfindungsgemäß hergestellten Biosensor mit nikotinischen Acetylcholinrezeptoren, einem ligandengesteuerten Ionenkanal mit Acetylcholin als Agonisten, aufgenommen wurde. Die Kommandospannung betrug +200 mV und die Arbeitstemperatur lag bei 21°C. Für den Aufbau der Lipiddoppelschicht wurde eine 3% (w/v)-ige Squalenlösung von einem natürlichen Sojabohnenextrakt und Cholesterin, welche im Molverhältnis 60 : 40 zueinander standen, verwendet. Im Flüssigkeitsreservoir 3 befand sich eine Pufferlösung mit 1,3.10-1 mol l-1 CsCl, 2.10-3 mol l-1 CaCl2, 10-2 mol l-1 Ethylenbis(oxyethylennitrilo)- tetraessigsäure und 10-2 mol l-1 [4-(2-Hydroxyethyl)-piperazino]-ethansulfonsäure, die mit 1 M CsOH-Lösung auf pH = 7,3 eingestellt war. Im Flüssigkeitsreservoir 4 war eine Pufferlösung mit 10-6 mol l-1 Acetylcholin, 1,24.10-1 mol l-1 NaCl, 2.10-3 mol l-1 CaCl2, 2.10-3 mol l-1 MgCl2, 3,25.10-3 mol l-1 D-Glukose und 10-2 mol l-1 [4-(2-Hydroxyethyl)-piperazino]-ethansulfonsäure, die mit 1 M NaOH-Lösung auf pH = 7,4 eingestellt war. Als löcherige Membran 6 wurde eine 10 µm dicke Polyethylenterephthalatfolie eingesetzt, die Löcher mit einem mittleren Durchmesser von 10 µm und eine Porosität von 15% besaß. Bei einer Stromstärke von ca. 3,5 pA waren alle Ionenkanäle geschlossen, wogegen bei einer Stromstärke von ca. 9 pA jeweils genau ein Ionenkanal geöffnet war.
