Abstract
Description
The invention relates to systems for temperature-regulated gene expression, especially for overexpression of cold-adapted, thermolabile Proteins from psychrophilic organisms.
Cold-adjusted, so-called psychrophilic organisms can still with grow extremely low temperatures. You can find these organisms among other things in the Arctic or Antarctic ice and sea water as well in the deep sea. Enzymes of these organisms are characterized by a high catalytic activity at very low temperatures, e.g. B. 4 ° C. This Is characterized by a compared to enzymes of heat-loving Organisms realized more flexible protein structure. This bigger one Thermoflexibility has the consequence that usual large-scale Production processes that work at 37 ° C to create partial lead denatured products, the so-called inclusion body in the Cells can be accumulated, and - if at all - only with less Yield can be renatured.
Feller et al. (Appl. Environ. Mikrobiol. Environ. Mikrobiol. 64 (1998), 1163-1165) the overexpression of a cold adapted amylase from an Antarctic Bacteria in the mesophilic expression host Escherichia coli. To Expression at 37 ° C no amylase activity could be measured. It was therefore proposed, after overexpression at 18 ° C or Incubate the fermenter culture at 4 ° C overnight. Through this a long drop in temperature could at least restore one Part of the overproduced amylase can be achieved. However, one can proteolytic degradation of the recombinant cold proteins by the cell's own Proteases occur during the long incubation period. Besides, that is Process unsuitable for large-scale production.
This system is based on the regulatory sequences of the main cold shock protein from E. coli, Csp. Furthermore, a cold-inducible expression system is described, in which a mutated promoter of the E. coli phage λ (pL) is used. This promoter is characterized by low temperatures of less than 20 ° C active and thus allows a selective expression of recombinant proteins under these conditions. The expression systems described are intended correct folding of recombinant proteins at temperatures below 20 ° C enable. A disadvantage of this method is the limitation to that Gram-negative bacterium E. coli, which is only a low temperature shows very poor growth.
The economic potential of cold-adapted proteins, e.g. B. enzymes, is very high. Especially in food processing and These proteins are widely used in molecular biological diagnostics Range of applications, but so far due to the high Production costs were limited. There is therefore a need for systems and to develop processes that are inexpensive and large-scale Enable the production of thermolabile proteins.
B. Saccharomyces cerevisiae, Pichia pastoris u. a. und Pilzen z. B. Aspergillus niger u. a., oder prokaryontischen Zellen, z. B. Escherichia coli, Lactobacillus lactis, Staphylococcus carnosis, Bacillus licheniformis, Bacillus brevis u. a., insbesondere in Gram-positiven Bakterien, z. This task is accomplished by providing a new one temperature-adjustable expression system solved, which preferably on an expression control sequence for a cold shock protein coding gene, in particular the cspB gene from B. subtilis and its regulation is carried out by an antisense RNA mechanism. The System enables cold-induced overexpression in a variety of organisms, e.g. B. eukaryotic cells such as yeasts, e.g. B. Saccharomyces cerevisiae, Pichia pastoris u. a. and mushrooms e.g. B. Aspergillus niger u. a., or prokaryotic cells, e.g. B. Escherichia coli, Lactobacillus lactis, Staphylococcus carnosis, Bacillus licheniformis, Bacillus brevis and the like. a., especially in Gram-positive bacteria, e.g. B. in Bacillus subtilis, but also in gram-negative bacteria. The system is preferably based on one Promoter that is active at low temperatures but cannot be induced by cold is. But cold-inducible promoters can also be used. The Use of the Gram-positive microorganism B. subtilis as Expression host is still preferred because it is better at temperatures of 20 ° C is adapted as E.coli, and better compared to E.coli Secretion of recombinant proteins in the extracellular medium is capable. Because of these characteristics, the inventive Expression system a more effective fermentation process at low Temperatures and more efficient and less expensive cleaning of the desired proteins after overexpression.
temperaturregulierten Genexpression umfassend
The present invention thus relates to a system for temperature-regulated gene expression
- a) eine Genexpressionseinheit enthaltend eine erste Expressions kontrollsequenz mit einem Promotor und einer ribosomalen Bindungsstelle, unda) a gene expression unit containing a first expression control sequence with a promoter and a ribosomal Binding site, and
- b) eine Regulationseinheit enthaltend eine Nukleinsäure, die bei Transkription eine zur ersten Expressionskontrollsequenz komplementäre Antisense-RNA ergibt, in operativer Verknüpfung mit einer temperaturregulierbaren zweiten Expressionskontrollsequenz.b) a regulatory unit containing a nucleic acid, which at Transcription to the first expression control sequence complementary antisense RNA results in operational linkage with a temperature-controllable second expression control sequence.
The second expression control sequence is preferably cold-regulatable, d. H. at a high temperature of e.g. B. 37 ° C is the transcription of Antisense RNA mediated by the second expression control sequence allowed, causing translation by one through the first Expression control sequence generated transcripts due to binding the antisense RNA is at least partially repressed to the transcript. On in this way a fermentation process is made possible in which first phase, the so-called growth phase at higher Temperatures, rapid growth of the host cells takes place while at under these conditions no or only very weak expression of the desired protein takes place. As soon as a sufficiently high cell density is reached, the second phase of fermentation, the so-called Start the production phase. In this phase the expression system induced by temperature reduction and the recombinant protein overproduced.
The temperature regulation mediated by antisense RNA can basically realized by two different embodiments become. In a first embodiment, the transcription takes place Antisense RNA only at high temperatures, e.g. B. 37 ° C. A bond this antisense RNA to the through the first expression control sequence generated mRNA upstream and / or in the region of the ribosomes binding site prevents initiation of translation of the foreign protein. The temperature-regulated transcription of the antisense RNA can by Use of temperature-sensitive repressors, z. B. of temperature-sensitive repressor CI587 of the E.coli phage λ, which only at Temperatures below 37 ° C are folded correctly and are therefore active. this has with the result that the repressor turns on when the temperature drops to 20 ° C its operator region binds and thus a transcription of the antisense DNA mediated by the second expression control sequence prevented. In order to no new antisense RNA is formed at this temperature. New from the mRNA molecules generated in the first expression control sequence are no longer present blocked and can attach to the ribosomes, a translation of the Foreign proteins are started.
In a second embodiment of the invention, an antisense RNA constructed in such a way that when the temperature drops to 20 ° C. Secondary structure forms so that they no longer adhere to the target mRNA can accumulate. With the help of appropriate computer programs corresponding mRNA secondary structures are calculated in advance. So can start from a given sequence by stepwise Corresponding antisense RNA molecules are generated that change at 20 ° C have a stable secondary structure that no longer matches the Target mRNA binds. In this embodiment, the expression of Antisense RNA by a preferably weak constitutive Promoter or by reducing the temperature to 20 ° C switchable promoter realized.
The antisense RNA is preferably up to 200 nucleotides in length shorter antisense RNA molecules are also possible.
The length of the complementary sequence section is preferably Antisense RNA molecules 20 to 100 nucleotides. The sequence of the antisense DNA is chosen so that the RNA generated by it through transcription Molecules at least partially the ribosomal binding site from that of the first Block expression unit of transcribed mRNA.
The gene expression unit of the system according to the invention contains one first expression control sequence, which is preferably for expression thermolabile proteins is suitable, d. H. at temperatures of ≦ 20 ° C one enables efficient transcription and translation. The first Expression control sequence therefore conveniently includes the promoter or / and the ribosomal binding site of a cold shock gene, for example the cspB gene from B.subtilis.
In addition, the first expression control sequence can be downstream of the ribosomal binding site a translatable nucleotide sequence, in particular an efficiently translatable at low temperatures Contain nucleotide sequence that is used to improve translation efficiency the desired proteins. This translatable nucleotide sequence can e.g. B. code for the N-terminus of cold shock proteins, e.g. B. for the first 10 to 20 amino acids of CspB. At the end of the translatable Nucleotide sequence can be located a stop codon, so that the translation in the form of a cistron with two separate coding areas (Translation-improving peptide or polypeptide and the desired one recombinant protein). Alternatively, the one you want recombinant protein also as a fusion protein with the translation-enhancing peptide or polypeptide are expressed, wherein in this case a gap point, e.g. B. a proteolytic cleft between the two domains of the fusion protein mentioned can.
The gene expression unit can also be a cloning site in operatively linked to the expression control sequence to enable the cloning of a desired target gene. In a Another embodiment of the invention can use the gene expression unit already a structural gene, which is preferably for a thermolabile protein encoded, in operative association with the expression control sequence contain.
The gene expression system according to the invention can be based on one or two Vectors or be localized on the chromosome of a host cell. The Vectors are preferably prokaryotic vectors, i. H. Vectors that are capable of propagation in a prokaryotic host cell. Examples for such vectors are plasmid vectors, bacteriophages, cosmids, etc. Vectors are preferably used which are used for propagation in Gram positive prokaryotic host cells, especially B. subtilis are. The vectors have a suitable one for the respective host cell Origin of replication and preferably an antibiotic resistance gene, to enable selection.
The invention further relates to a cell that has an inventive Expression system contains. Preferably the cell is a Gram positive Cell, especially a B. subtilis cell.

Culturing the cell in step (ii) of the method according to the invention is preferably carried out in such a way that until it is reached a predetermined cell density the expression of that for the desired Gene encoding polypeptide is largely repressed, i.
