The invention relates to the manufacture and construction of microstructured analysis systems. The method according to the invention enables the production of plastic analysis systems which have a liquid- and gas-tight channel structure in which thin-film electrodes can be located at any point.
Description
The invention relates to the manufacture and construction of miniaturized Analysis systems, especially those with control and measuring devices for electrical conductivity.
Miniaturized analysis systems, especially those with microfluidic Channel structures are becoming increasingly important. In particular Miniaturized analysis systems, the possibilities for provide electrophoretic separation and analysis of samples.
Analysis units that are used for such applications can usually consist of a base plate (substrate) and a Cover, between which there are microchannel structures, electrodes and others required functionalities such as detectors, reactors, valves etc. are located.
To the demands placed on a microfluidic analysis system sufficient stability regarding mechanical, chemical, electrical and thermal effects. For the channel structures mean mechanical stability in particular Dimensional and volume stability, which is an important requirement for z. B. is a quantitatively reproducible sample task. Even inner ones Pressure stability of the microchannels is important with regard to the use of e.g. B. Pumps required to fill the microchannels. The used Of course, materials must be chemically inert to that in the Channels can be transported medium. So far electrodes in the channel are introduced, these should be in high accuracy (a few µm) in the channel can be positioned to z. B. when used as a detector to be able to deliver electrode reproducible results. This is also Prerequisite that the contact areas within the channel are free of Impurities are. The electrodes should also be small Allow internal resistance and a potentially high current flow. This applies in particular to so-called power electrodes with which in Depending on the medium used within the channels an electro kinetic flow can be generated. Ultimately, the electrodes should be light be connectable.
Often serves as material for the production of such analysis units Silicon or glass. The disadvantage of these materials, however, is that they are not suitable for inexpensive mass production of the analysis systems. Plastic-based materials are much more suitable for this. The Components, such as substrate and lid, that form the actual microstructures can then contain by known methods such as hot stamping, Injection molding or reaction molding can be produced inexpensively.
For closing the resulting open microstructures with Lids, on the other hand, have so far not been available for plastic components production-ready techniques. This applies in particular to such microphones channel structures, in which additional metallic electrodes on any Positions are to be positioned within a closed channel structure.
Die Oberfläche des Kanals besteht somit aus dem thermoplastischen Klebstoff.EP 0 738 306 describes a method for closing microchannels structures described, with a dissolved thermoplastic on the structured polymer substrate is spun. This solved thermo plast has a lower melting temperature than the parts to be glued. The lid and substrate are thermally bonded at 140 ° C. The The surface of the channel thus consists of the thermoplastic adhesive.
No. 5,571,410 produces microfluidic structures with laser ablation in Kapton TM and welded with a KJ®-coated Kapton TM film.

All of the above-mentioned methods make it possible by connecting one They allow the creation of substrate with a cover of microchannel structures but not the integration of electrodes which are in direct contact with the Have medium in the channels.
In EP 0 767 257 ist ein Verfahren zur Integration von Elektroden in Mikrostrukturen beschrieben, doch erlaubt dieses Verfahren nicht eine flüssigkeitsisolierte Kontaktierung, da zum photochemischen Abscheiden des Metalles in den Kanälen diese mit Metallsalzlösungen gespült werden müssen.EP 0 767 257 describes a method for integrating electrodes in Microstructures are described, but this method does not allow one liquid-insulated contacting, as for photochemical deposition of the metal in the channels, these are rinsed with metal salt solutions have to.
A method for the integration of electrodes anywhere inside half of a microstructured channel with the possibility of liquid isolated contacting of the electrodes was by Fielden et al. (P.R.- Fielden, S.J. Baldock, N.J. Goddard, L.W. Pickering, J.E. Prest, R.D. Snook, B. J.T. Brown, D.I. Vaireanu, "A miniaturized planar isotacho phoresis separation device for transition metals with integrated conductivity detection ", Proceedings Micro Total Analysis Systems '98, 323-326, Banff, Canada). The authors have a microfluidic channel Structure molded in silicone (PDMS) and press them mechanically against a circuit board provided with electrodes (copper). The channels are thus limited by two different materials. To the resulting Keeping ducts closed requires constant mechanical pressure be maintained. The pressure on the silicone cushion occurs this system easily deforms the channel structures.
The object of the present invention is therefore an improved provide microfluidic analysis system, its substrate and cover consist of polymeric organic materials and firmly together are connected, and in the electrodes at any point with Possibilities for liquid-insulated contacting are introduced can. If electrodes are to be integrated into the analysis system, an additional task is that the electrodes on each can be integrated anywhere in the duct system and not through the bonding process is damaged or detached.
It has been found that the combination of a new process for Manufacture of adhesive noble metal layers on plastic surfaces with a special bonding technique for joining two plastics components makes it possible to use microfluidic analysis systems with the Technology and properties discussed in the task to manufacture.

Fig. 2 and 3 show two options for contacting the electrodes.
As microfluidic analysis systems with measuring and control devices for According to the invention, electrical conductivity applies to systems in which Joining at least two components, such as. B. substrate and Cover, microchannel structures can be generated, the liquid and / or can be closed gas-tight. Substrate and lid are firmly connected to each other. In addition, these systems can contain electrodes anywhere in the duct system, free contact to the inside of the channel.
The microfluidic analysis systems can be varied by different Parameters such as the channel structure, the connection of other systems, such as pumps, supply lines etc., any arrangement of electrodes etc. can be adapted for different applications. The analysis systems according to the invention are particularly preferred for Applications in the field of electrophoretic separation and analysis, for example for capillary electrophoresis or isotachophoresis and for micro-preparative synthesis or derivatization of substances.
The components of the systems preferably consist of commercially available thermoplastics, such as PMMA (polymethyl methacrylate), PC (Polycarbonate) or PMP (polymethylpentene), cycloolefinic Copolymers or thermosetting plastics, such as Epoxy resins. All components preferably exist, i. H. Substrates and Lid, a system made of the same material.
The components can be produced by methods known to the person skilled in the art. Components that contain microstructures can be produced, for example, by established processes such as hot stamping, injection molding or reaction molding. Components which can be reproduced using known techniques for mass production are particularly preferably used. Microstructured components can have channel structures with cross-sectional areas between 10 and 250000 µm 2 .
The electrodes used in the analysis systems according to the invention are typically used to generate a river used by ions or for detection purposes. You need one have sufficient adhesive strength on the plastic components. This is for the assembly of the individual components as well as for the later use of the analysis systems is important.
B. Edelmetallen (Platin, Gold) bestehen. For the choice of the electrode material, the one that is primarily planned Decisive use of the analysis system. Because systems with Microchannel structures and integrated electrodes essentially in In the field of analytics, the electrodes should be made chemically inert materials, such as. B. Precious metals (platinum, gold) consist.
The choice of such materials and methods of application are known to the person skilled in the art. The metallization of plastic surfaces is typically carried out by electrochemical deposition of metals from metal salt solutions. For this, it is common practice to first pretreat the plastic surface chemically or mechanically in a multi-stage process, to apply a discontinuous primer and finally to carry out the electrochemical deposition. Descriptions of these metallization techniques can be found e.g. B. in US 4,590,115, EP 0 414 097, EP 0 417 037 and Wolf and Gieseke (GD Wolf, H. Gieseke, "New method for the full-surface and partial metallization of plastics," Galvanotechnik 84 , 2218-2226 , 1993 ). The wet chemical processes have in common that relatively complex pretreatment processes are necessary in order to achieve sufficient adhesive strengths.
Demnach müssen die Mischungen Polyarylensulfide, Polyimide oder einen aromatischen Polyester enthalten.DE 196 02 659 describes the adhesive application of copper to more phase polymer mixtures by means of vapor deposition or sputtering described. The composition is the cause of good adhesion called the polymer blends. Accordingly, the mixtures Contain polyarylene sulfides, polyimides or an aromatic polyester.
The influence of plasma pretreatments to achieve better detention properties of metals on plastic surfaces is described by Friedrich (J. Friedrich, "Plasma treatment of polymers", stick & seal 41, 28-33, 1997) using the example of various commercially available thermoplastics summarized. The general goal of plasma pretreatment is polar Generate functional groups on the polymer surface, so that a increased adhesive strength of metallic layers results. The example is Effect of chrome as an adhesive layer in the metallization of plastics described. As the cause of the good adhesion of chrome z. B. becomes a Interaction of polar groups, such as. B. carbonyl or ester groups, with 3d orbitals of chrome called.
The electrode structures on the plastic are particularly preferred components using a new two-layer technology. This will According to the invention, first an adhesion-promoting layer made of chromium oxide generated. It has been shown that chromium oxide is in contrast to precious metals has excellent adhesive properties on plastic surfaces. In addition is chromium oxide in contrast to elemental chromium and others Transition metals much more resistant to redox processes. Then the noble metal, such as for example, platinum or its alloys or gold.
The selective application of chromium oxide and the ones to be deposited thereon Precious metal layer on plastic substrates is preferably done in the lift-off Process or by means of the so-called shadow mask technique or Structuring of metallic surfaces initially applied over the entire surface Layers. These process technologies are standard processes of the micro structural engineering. The following are those for the two-layer technique necessary steps for the procedures mentioned briefly described.
Lift-off process: The plastic component to be selectively metallized is coated with a photoresist. This photoresist must not or only slightly dissolve the plastic part to be metallized. For PMMA z. For example, a photoresist of the company Allresist, Berlin (AR 5300/8) proved to be suitable. After exposure and development of the structures to be metallized, the metallic layers are applied in a sputtering system. The chromium oxide layer is applied during the sputtering process by introducing oxygen into the argon plasma typically used in the sputtering system. A conventional chrome target is used as the sputtering target. Typical chromium oxide layer thicknesses are 20-50 nm. Alternatively, a chromium oxide target can be used directly. The sputtering of platinum or its alloys or of gold is then carried out directly under standard conditions, ie in argon plasma. In the actual lift-off process, the still existing photoresist and with it the metal layer on the lacquer are detached from the plastic component in a developer from Allresist (AR 300-26 ).
Shadow mask technology: The plastic part to be selectively metallized is covered with a so-called shadow mask. This has to do with metalizing areas recesses. Be through this the metal layers sputtered in analogy to the lift-off process.
Structuring of flat metallic layers: First, a metal layer is applied to the entire surface of a plastic part to be selectively metallized, in analogy to the sputtering process already described. This is done in subsequent process steps, either by selective removal using z. B. laser ablation (gold and platinum) or z. B. structured by selective wet chemical etching. For structuring by means of wet chemical etching, a photoresist (Hoechst AG, Germany; AZ 5214 ) is first applied, exposed and developed on the metal layer. Gold is then stripped off in cyanide solution in the exposed areas.
The adhesive strength of with chrome as well as with chrome oxide as an adhesive layer Electrodes manufactured using sputtering technology were developed using Tear tests checked. The adhesive strength of the chrome oxide layers is clear greater. They are also suitable for ultrasound treatment in alkaline solution Metal layers which were produced with chromium oxide as an adhesive layer, compared to metal layers made with chrome as an adhesive layer became significantly more stable.
After production and preparation of the individual components, these are assembled by the method according to the invention. More preferred wise is a component, the substrate, microstructured and with back Bores for filling the channels and / or contacting the electrodes Mistake. Furthermore, the use of a so-called Sealing lip, d. H. one completely enclosing the channel structures Elevation on the substrates with heights between typically 0.5 to 5 µm, proved to be very advantageous with regard to the bonding process. The other component, the lid, is used for covering and is z. B. in electro provide the electrodes with phoretic analysis systems. In this In this case, the cover is referred to as an electrode cover according to the invention. There the method according to the invention not only relates to the production of the Measuring and control device related to the analysis systems can certain applications of the systems one of these preferred Arrangement require different functionalization of the components. In in this case, for example, more than two components, e.g. B. two Lid and a substrate etc. are put together to overlap to generate horizontal channel structures, or other functionalities, such as Detection systems, reaction chambers etc., integrated in the components become. According to the invention, all parts of the analysis systems that are joined together by means of a bonding process as components designated. They can be micro-structured, can be provided with electrodes or have other functionalities. A subdivision of the components into Substrates and lids or electrode lids, if the corresponding one Component is provided with electrodes, only serves the closer Description of the embodiment of the special components and does not constitute Restriction regarding further properties of the components, such as Microstructuring etc., or their combination with each other.
To assemble the components, an adhesive is preferably first applied to the microstructured component at the locations where there is no structuring. The layer thickness is between 0.5 and 10 µm, preferably between 3 and 8 µm. Typically, the application takes place by means of a flat roller application known from printing technology. The adhesive used must not or only very slightly dissolve the surface of the components so that the electrodes are not detached or interrupted by the adhesive during the bonding process. The product NOA 72 , thiol acrylate from Norland, New Brunswick, NJ, USA is therefore preferably used as the adhesive. This adhesive is cured photochemically. However, other types of adhesives that meet the above requirements can be used for the process.
After applying the adhesive, the second component with the Thin-film electrodes, for example, on an exposure machine appropriately positioned and pressed onto the substrate. The is preferred Use of strong glass plates as a pressing surface, so that the photochemical curing of the adhesive by irradiation with a mercury Lamp (emission wavelength 366 nm) can be carried out.
The positioning of the lid on the substrate can be used for the adhesive process typically visually under manual control, passive mechanically with the help of a locking device, optically mechanically under With the help of optical alignment marks or electrically mechanical with the help of electrical brands (contacts).
In another preferred embodiment, that with the Electrode-provided component on the areas that come together do not place the two components over a duct or electrically must be contacted with the adhesive. For this, for example a method known in printing technology (pad printing) used. The component with the channel structures is then appropriately positioned and pressed onto its counterpart. The Curing takes place as described above.
geschlossenen Mikrokanalstrukturen, in denen Elektroden an beliebigen
Stellen innerhalb der Kanäle positioniert werden können. Strukturierte
The method according to the invention makes it possible for the first time to produce closed microchannel structures in which electrodes can be positioned anywhere in the channels. Structured components (substrates) can be provided in a liquid-tight and gas-tight manner, for example with electrode covers. By using mostly commercially available plastics and simple processing steps, the analysis systems according to the invention can be produced inexpensively and in large numbers. By means of the method according to the invention for joining or bonding, the components are wetted with adhesive in such a way that no adhesive gets into the interior of the channel system after joining, ie into the channels, the walls or electrodes or other devices projecting into the channel system.
