1994, 35, 2679]. Bekannt sind auch 2-Alkyl-3-hexadecyloxypropan-1-yl-phosphocholine mit biologischer Aktivität [H.-P. Kertscher et al., Pharmazie 1985, 40, 702].2 '- (Trimethylammonio) ethyl-3-alkylmercapto-2-methoxymethyl-prop-1-yl-phosphate and their oxidation products with 3-alkylsulfinyl or 3-alkylsulfonyl groups are the subject EP 050327 and DE 39 06 952. In particular the connection with a hexadecyl mercaptorest (ilmofosin) and the analogues with heptadecylmercapto- and octadecyl mercaptorest and its oxidation products have pronounced anti-tumor properties. Oxygen analogs of ilmofosin have also been presented. They have an inhibitory effect on the cell growth of certain cancer cell lines [R. Bittman et al., Tetrahedron Lett. 1994, 35, 2679]. 2-Alkyl-3-hexadecyloxypropan-1-yl-phosphocholines are also known biological activity [H.-P.
The previously known comparable compounds have the disadvantage that they are in their biological effects are limited or less effective. You can also in Racemize the body and thus cause undesirable side effects. Furthermore are these substances are easily metabolizable and therefore have a comparatively short duration Duration of action. Are fluorinated compounds of the structure claimed in this invention so far unknown.
The object of the invention is to address the disadvantages of the prior art described so far to overcome and new fluorinated ether lipids with specific biological effects synthesize and make them available as active ingredients.
The main advantages of the invention are that with the new fluorinated compounds A more specific spectrum of action is achieved that the compounds in the body are not racemize that their metabolism difficult and thus increases the duration of action becomes. It is thus possible to reduce the dose required for therapy. Furthermore the compounds are easily accessible through chemical reactions that have been tested per se.
It has now been found that the previously unknown 2 '- (trimethylammonio) ethyl-3-alkoxy-2- fluoro-2-methyl-prop-1-yl-phosphate and 2 '- (trimethylammonio) ethyl-3-alkoxy-2-fluoro-2- methoxymethyl-prop-1-yl-phosphate compared to e.g. B. the Ilmofosin or cis-platinum show modified antitumor effect or are able to platelet formation influence.

in denen der Rest R1 eine kurze gesättigte Alkylkette oder eine Alkoxymethylgruppe und der Rest R2 eine lange gesättigte Alkylkette bedeuten sowie deren Stereoisomere.in which the radical R 1 is a short saturated alkyl chain or an alkoxymethyl group and the radical R 2 is a long saturated alkyl chain and their stereoisomers.
In particular, they are compounds in which R 1 is a methyl or methoxy methyl group and R 2 is a saturated unbranched alkyl radical having 14 to 20 carbon atoms, preferably hexadecyl and octadecyl radicals.

The invention relates to all stereoisomeric compounds of the formula I and their pharmacologically acceptable salts. The compounds of general formula I are produced by known synthetic steps.
a) compounds of the general formula I in which the radical R 1 is a methyl group and the radical R 2 is an alkyl group of the chain length given above are preferably prepared starting from methallyl alcohol (1),
by alkylating it to 2 under the conditions of a Williamson ether synthesis with a corresponding haloalkane.
The corresponding alkyl methallyl ethers 2 are then subjected to halofluorination, in which the halofluorides 3 are formed as a racemate in accordance with the Markovnikov rule. I, Br, Cl can be used as halogen atoms X.
Acetate 4 entstehen.
The terminal halogen substituent in 3 is then exchanged for an acetate residue in a nucleophilic substitution reaction, the racemic fluorine-substituted acetates 4 being formed.
Saponification of the ester function gives the fluorohydrins 5 as racemates from 4.
The hydrolysis of the racemic acetates 4 can also be carried out by using hydrolases in aqueous buffer systems take place, a kinetic resolution occurs and only an enantiomer of 5 is obtained while an enantiomer of acetates 4 is unchanged remains. Another possibility of the kinetic resolution is in Implementation of the - as described above - obtained by saponification racemic Fluorhydrine 5 with an acetylating agent and the same enzyme in an organic Solvent. The other enantiomer of acetates 4 is formed, while the oppositely configured fluorohydrins 5 are not implemented.
The racemic fluorohydrins 5 or each of their enantiomers are now separated with a phosphoric acid derivative of the general formula 6,
in der X und Y jeweils Chlor oder Brom oder zusammen ein Sauerstoffatom bedeuten können, in Gegenwart eines säurebindenden Hilfsstoffes umgesetzt und ggf. ohne Isolierung eines Zwischenproduktes mit Trimethylamin zu I quarternisiert, wobei R2 ein Alkylrest der oben angegebenen Kettenlänge ist und das Produkt I in racemischer oder optisch aktiver Form entsteht.in which X and Y can each represent chlorine or bromine or together represent an oxygen atom, reacted in the presence of an acid-binding auxiliary and quaternized with trimethylamine to I without isolation of an intermediate, where R 2 is an alkyl radical of the chain length indicated above and the product I in racemic or optically active form.
Synthesis 1982, 924; K. C. Reddy et al. Tetrahedron Lett. 1994, 35, 2676].

Villieras et al., Synthesis 1982, 924; KC Reddy et al. Tetrahedron Lett. 1994, 35, 2676].
Bisether 8 gebildet.
The bisethers 8 are formed from 7 by a Williamson ether synthesis with a methylating agent.

The halofluorination of 8 gives the halofluorides 9 regioselectively, which are isolated as racemates and in which X can be the halogens I, Br and Cl.
The terminal halogen substituent in 9 is then exchanged for an acetate residue by nucleophilic substitution, and the fluorinated acetates 10 are thus obtained in racemic form.
The hydrolysis of the racemic acetates 10 can be carried out by alkaline saponification and thus gives the racemic fluorohydrins 11. On the other hand, analogously to the procedures described under a), enzyme-catalyzed can carry out a kinetic resolution of 10, one enantiomer not being attacked, while the other is hydrolyzed to the respective optically active fluorohydrins 11.
The two other enantiomers of acetates 10 and fluorohydrins 11 can be separated starting from the racemic fluorohydrins 11, likewise analogous to that under a) described procedure, by enzyme-catalyzed acetylation in an organic Represent solvents.
The racemic fluorohydrins 11 or each of the enantiomers separately are now reacted with a phosphoric acid derivative 6 and the products formed, if appropriate without isolation, as described under a), with trimethylamine and the compounds I are thus obtained in racemic or optically active form, where R 2 is an alkyl radical of the chain length given above.
Halogenen I, Br und Cl entspricht.
12 in racemischer Form gebildet.
c) Compounds of the general formula I in which the radical R 1 is a methoxymethyl group and the radical R 2 is an alkyl group of the chain length mentioned above can alternatively to the process described under b) also be prepared in the following way:
The 2-alkoxymethylprop-2-enols 7 are subjected to halofluorination, where X corresponds to the halogens I, Br and Cl. Only one of the possible regioisomers 12 is formed in racemic form.
The terminal halogen substituent is then exchanged for an acetate residue in a nucleophilic substitution, with predominantly 13 being formed as a racemate.
14 erhalten.
The racemic carbamates 14 are obtained by reacting the monoacetates 13 with phenyl isocyanate.
Subsequent hydrolysis of the acetate function of 14 can be accomplished by alkaline saponification and provides the racemic fluorohydrins 15.
Under the conditions of a Williamson ether synthesis, the hydroxyl function is alkylated to 16 in a corresponding haloalkane in racemic form.
The carbamate group of 16 can be eliminated under basic conditions, the fluorohydrins 11 being obtained as racemates.
The racemic fluorohydrins 11 can, as described under b), in the target compounds of the general formula I are transferred.
