Abstract
Description
The present invention relates to conjugates of cytostatic and Nα-modified Amino acids or peptides, process for their preparation and their use as a medicine, especially in the context of cancer.
Chemotherapy for tumors is accompanied by mostly serious Side effects due to the toxicity of chemotherapeutic agents to proliferate cells of other tissues. For many years, scientists have been involved with the problem of improving the selectivity of active ingredients used. On often followed approach is the synthesis of prodrugs, more or less selectively in the target tissue, for example by changing the pH (eg Tietze et al., DE 42 29 903) by enzymes (eg glucuronidases, Jacquesy et al., EP 511,917; Bosslet et al., EP 595 133) or by antibody-enzyme conjugates (Bagshaawe et al., WO 88/07378; Senter et al., U.S. Patent 4,975,278; Bosslet et al., EP 595 133) are released. The problem with these approaches is u. a. the lacking Stability of the conjugates in other tissues and organs and in particular the ubiquitous drug distribution, which is related to extracellular drug release in the Tumor tissue connects.
Die Realisierung des vielver sprechenden Potentials in der Klinik scheiterte jedoch an Toxizitäts- und Löslich keitsproblemen.20 (S) -camptothecin (3) is a pentacyclic alkaloid reported by Wall et al. was isolated (J. Amer. Chem. Soc. 88 (1966) 3888). It has a high anti-tumor Potential for action in numerous in vitro and in vivo tests. The realization of the vielver however, the potential for speech in the clinic failed due to toxicity and solubility keitsproblemen.
Upon opening of the E-ring lactone and formation of the sodium salt, a water-soluble compound obtained in a pH-dependent equilibrium with the ring-closed shape stands. Clinical studies have not led to this to success.
About 20 years later it was found that the biological activity on an enzyme Inhibition of topoisomerase I is due. Since then, the research Increased activity to provide more tolerable and in vivo effective camptothecin To find derivatives.
To improve the water solubility z. Salts of A-ring and B-ring modified camptothecin derivatives and of 20-O-acyl derivatives with ionizing Baren groups described (Vishnuvajjala et al., US 4,943,579). The latter Prodrug concept was later also applied to modified camptothecin derivatives (Wani et al., WO 96/02546). The described 20-O-acyl prodrugs have however, in vivo a very short half-life and become very fast for Split main body.

The conjugates thus obtained are synthetically readily accessible and show in in vitro a high activity against various tumor cell lines and tumor xenografts.
- Depending on the composition of the Nα-modified amino acids show the conjugates of the invention compared to those basic cytostatic drugs have significantly improved solubility properties.
- - Bedingt durch die α-Modifizierung sind sie in extrazellulärem Medium und in Blut wesentlich stabiler als die zuvor beschriebenen reinen Aminosäure- Prodrugs von Camptothecin-Derivaten.- Due to the α-modification they are in extracellular medium and in Blood is much more stable than the previously described pure amino acid Prodrugs of camptothecin derivatives.
In the case of 20-O-acylations of camptothecin derivatives, the ester-like linkage of the carrier residues with the 20-hydroxy group of for the effect stabilized important lactone ring.
