Description
The invention relates to a method and an apparatus for digestion of biological cells for the extraction and analysis of their cell contents, in particular of nucleic acids.
For the examination of biological cells and in particular the analysis of the cellin hold, in particular proteins and nucleic acids (DNA and RNA), for example For medical diagnostics, the cell walls and membranes need to be opened be closed to ex. the cell interior in a suitable form from the cells to be able to
-auflösung unterscheiden:A number of known methods are suitable for this, which are described in the contribution by Carl A. Schnaitman, "Cell Fractionation" in Manual of Methods for General Bacteriology, American Society for Microbiology, Washington, DC 20006, 1981, Chapter 5, page 52-61, are described in an overview. The be in this post Methods written can be divided into different groups, which are differentiate according to their type of cell wall destruction or dissolution:
Mechanical processes are based on the generation of local pressure fluctuations on Location of the cells to be disrupted, making cell walls and membranes literally be broken up. Because of the strong necessary for cell wall destruction Pressure fluctuations, however, require appropriately stable devices as described, for example, in the article by J.R. Raper and E.A. Hyatt, "Modified Press For Disruption Of Microorganisms ", J. of Bacteriology, V. 85, pp. 712-713, be are written. Other devices for generating such large pressure or Shear forces are so-called ball mills or the so-called French press, which in "Manual of Industrial Microbiology and Biotechnology", American Society for Micro biology, Washington, D.C. 1986 in Chapter 9.3.6. on pages 103-104.
The devices mentioned above require a very large techni and mechanical effort that must be operated to such a large Generate pressure forces and master them safely, plenty of space and can be over difficult to automate this. Another disadvantage of the known pressure treatment biological cells also consists in the fact that the cell contents are ins especially the long-chain DNA molecules are meant to be highly fragmented, so that complete DNA analysis is difficult.
Ultrasonic digestion methods are also known in which the cell walls and Membranes are literally torn open by cavitation effects, so that the cellin nere can get outside. In the case of ultrasound digestion however a Liche sound power dissipated in the sample to be examined, causing the sample is heated unnecessarily. Cooling precautions are therefore necessary. Through the appearances However, temperature fluctuations are a reasonably accurate process control difficult. In addition, the known apparatus for using the High performance ultrasound difficult to clean, creating the risk of cross-contact mination cannot be excluded.
The digestion of biological cells by chemical means only chemicals individually adapted to the cells, which means that during the examination unknown cell types, such as the diagnosis of unknown patients pathogen is the difficulty of choosing the right chemical consists. An example of a cell disruption method using only The use of purely chemical digestion reactions is described in "Molecular Cloning - A Laboratory Manual, "Second Edition, Cold Spring Harbor Laboratory Press, 1989, Pages 1.25-1.31. The reak required for cell disruption times with the exclusive use of chemicals are several Hours despite the high concentrations of chemicals used that disrupt subsequent analysis steps, such as PCR reactions influencing factors. Also the digestion rate is with some microorganisms not sufficient; for example, it is extremely difficult with Micrococcus luteus to reach a rate higher than 20% by chemical means.
Finally, methods are known in which bio. Suspended in liquid media logical cells, such as bacteria or fungi, a strong electrical Field are exposed. Biological cells in the Ways of pasteurization and sterilization, such as in the Document US 5 235 905 is described. In this case, in order to a too large energy To avoid gie entry in the sample and its overheating, the sample with pulsed Most electrical fields each have pulse durations of a few micro seconds. Because a biological cell is considered electrically from an electrolytic and therefore there is electrically conductive content, that of an electrically insulating When the membrane is enclosed, the electrical charge carriers move inside ren when applying an electrical field from the outside. This way arise local potential differences between the cell interior and cell exterior, which lead to ren that the cell membrane experiences an electrical breakthrough, causing it to loses semi-permeable properties and increases in electrical conductivity. At low external field strengths and short pulse durations is the change in electrical conductivity of the cell membrane reversible, a phenomenon that occurs in the Electroporation (see E. Neumann, A. E. Sowers, C. A. Jordan, "Electroporati on and Electrofusion in Cell Biology ", Plenum Press, New York, 1989) Role play. This is used, for example, to deliver genetic material into the cells bring. A disadvantage of this application of electroporation is that typically the efficiency is in the range of 0.01%, d. H. only in one cell out of 10,000 the matter al is actually brought in.
With stronger fields and sufficient pulse duration, especially with a like repeated treatment of biological cells, these changes are permanent and ultimately lead to cell death. By default, this procedure becomes Killing microorganisms used in food.
So können das Ein- und Ausschleusen von Teilchen in Zellen nicht als äquivalente Vorgänge angesehen werden, zumal die Natur des Transportes asymetrisch ist.DE 35 37 261 A1 describes a method for field-induced infiltration of Ma described cromolecules in living cells, one for targeted removal of cell contents represents a reciprocal process and thus other process conditions is subject. This means that particles cannot enter and exit cells are regarded as equivalent processes, especially since the nature of the transport is asymmetrical.
The asymmetrical or directional transport of various substances through the Rather, cell wall is one of the most important functions of the cell membrane. So points on the one hand the cell membrane has an asymmetrical structure, on the other hand under the cell content differs significantly from the external environment of the cell. The size of the transported molecules is often ver with the cell size itself similar, which not least also means that the different transport dimensions mechanisms underlying the inward and outward transfer, not by simple ones Diffusion models can be described - including simple diffusion is not a purely symmetrical process. The transport mechanisms are complicated, be contain several steps and are very specific for the respective direction of transport.
The same applies to the transformation resulting from JP 04173093 A method of Bacillus stearothermophilus with a plasmid.
All known methods for disrupting biological cells are liable in addition to that for Part of very large equipment and technical outlay the disadvantage that the Procedures are not to be carried out fully automatically. It also comes that the cell opening closing rate is not always sufficiently large.
The invention has for its object a method for digesting biological To develop cells for extraction and analysis of the cell contents such that the Process fully automatically and without great equipment and technical effort can expire. In particular, the extracted portion of the cell to be analyzed consider to be high regardless of the cell type, the duration of the procedure for extracti on and isolation of the cell contents should be significantly reduced. Moreover, a pre direction to be specified, with a cost-effective, fast and safe Implementation of the procedure is possible.
According to the invention, it has been recognized that the disadvantages that exist in the known Ver drive up the exclusive chemical digestion of biological cells occur through a targeted sequence or overlay of electrical and chemical treatment of biological cells can be avoided.
According to the invention, the method for disrupting biological cells is characterized Extraction and analysis of the cell contents in that the cells an electri be exposed to the field and that the cells before or after the application of the electrical field can be brought into contact with a substance that finally supports.
The biological cells usually suspended in a liquid medium are a pulsed electric field with typical field strengths between between 5 and 100 kV / cm and a pulse length of approx. 0.5 to 50 microseconds set. The cells can also be found in cell assemblies, such as pieces of tissue are present or applied immobilized on a carrier, for example on a fil ter.
Due to the action of the electrical field and the electrical breakthrough There are pores in the layers of lypid that make up the cell membrane. But since she are very small, and because of certain cell types in addition to the cell membrane the cell wall the diffusion of large molecules such as nucleic acid treatment of the cells with an electric field is sufficient for the Release of the cell contents in many cases does not stop.
It is proposed according to the invention the formation of the pores in the cell membrane exploit bran to support the effect of chemicals for cell disruption zen and accelerate.
In a variant, chemicals are used that affect the cell content and esp act in particular on the molecules to be analyzed so that they are lighter and to be released quickly. These chemicals diffuse through the electri treatment created pores in the cell interior, and build for example Proteins cut and cut DNA molecules into smaller fragments, but for subsequent DNA analysis are suitable. The resulting DNA fragments can then diffuse freely through the pores. These processes will by adding a surface-active substance (detergent), such as Sodium dodecyl sulfate (SDS).
Another variant takes advantage of the fact that the electrical Treatment created pores represent weak spots that attack the Much easier and faster chemicals on the membrane. Chemical s that can be used to completely dismantle the cell walls are included in the for example chaotropic reagents such as urea, guanidinium hydrochloride or - thiocyanate and others. Dimethyl sulfoxide can also be used.
Because the biological cells suspended in a medium have an electrical field are exposed, the addition of the additional chemical substance is required not too high a concentration, causing negative effects due to the Presence of the chemical substance, in subsequent analysis steps can be closed.
To optimize and accelerate the desired chemical effect, who which the cells brought into contact with the chemicals, for example in the form of a Cell suspension, brought to a temperature level between 35 and 100 ° C.
According to the invention, a device for performing the method is thereby trained that one, the cells or cell aggregates suspended in a medium absorbing sample volume with two flat, opposing The electrodes provided is provided by a cover element, the Pro benvolum facing side is convex, on the top of the sample volume is lockable. Due to the convex cover element advantageously be avoided that with inclusion of the liquid medium to avoid trapping air bubbles within the sample volume to the uneven distribution of the electric field and therefore possibly lead to undesired electrical breakdowns in the treatment device would.
In Fig. 1 an advantageous device for performing the method for closing biological cells is shown, which shows a treatment cell having a plastic housing 2 , in which a lower electrode 3 is cast and provides an upper electrode 4 , which also represents the cover element . The biological cells to be disrupted suspended in a solution are located in the sample volume 1 between the electrodes. Alternatively, the cells can be immobilized on a carrier, for example a filter, the remaining volume of the device being filled with an electrolyte solution. The convexly curved surface of the cover element 4 ensures that no air bubbles are trapped in the sample volume 1 when the Dec element is put on. The lement by the Deckele when placing the lid member on the sample volume 1 1 displaced excess of the suspension 5 collects in a circularly provided around the lid member 4 recess 6 of the plastic housing. 2 The lower electrode 3 opposite the cover element 4 , however, is formed concave, so that the distance between the two electrodes is chosen to be constant. In addition, the design of the electrode shape facilitates the filling of a liquid into the sample volume 1 without air bubbles and a largely complete sampling after the cell disruption process has been carried out.
In order to contact the electrodes 4 and 3 and to transport and fix the treatment cell parts in particular in an automated device, the lower electrode 3 has a plug 7 and the upper electrode 4 has a recess 8 .
The electrode distance preferably set in the exemplary embodiment carries 2 to 5 mm, the electrode diameter approx. 1 cm. With these dimensions and with a still relatively easy to handle voltage of approx. 20 kV the Strength of the electric field between 40 and 100 kV / cm, a field strength that for cell disruption is quite suitable.
The electrode surfaces can preferably be made of aluminum, since this material is biologically inert and chemically stable. Furthermore it is easy to process and cheap, and therefore especially for disposable items suitable.
After treatment with an electric field, chemical treatment in the same volume can be continued by adding chemicals, or older natively, the cell suspension can be pipetted into another vessel.
The embodiment shown in Fig. 1 may be cumbersome due to the constructive design of the cover element for sampling, since the cover element is to be removed before emptying the sample volume 1 , to which a substantial part of the solution can get caught as drops. This in turn could lead to contamination of the environment and thus to cross-contamination of other samples, which must be avoided, however, especially when the samples are handled automatically.
In order to facilitate the taking of samples, in particular in an automated device, and to make them safe from cross-contamination, the device according to the exemplary embodiment in FIG. 2 provides a two-part design of the cover element 4 . The cover element 4 has a central bore 9 and is covered with an electrically conductive, preferably made of aluminum foil 10 enveloping the cover element 4 . After carrying out the method according to the invention, a pipette is inserted through the channel 9 , which pushes through the film 10 so that the suspension contained in the sample volume 1 can be sucked off. In this embodiment, the film must be replaced every time, but this appears to be advantageous, especially since the entire treatment cell can be designed as a disposable article to avoid cross-contamination.
FIG. 3 shows a further exemplary embodiment which has a cover element 4 comparable to the embodiment according to FIG. 1. However, this configuration provides a metal foil 11 which separates the sample volume 1 and which also represents the lower electrode. There is a cavity 12 below the electrode 11 , which is connected to a channel 7 . As in the exemplary embodiment according to FIG. 1, the treatment cell is filled with a suspension of biological cells and high-voltage treatment with chemical additives for cell disruption is carried out.
After completion of the electrical field application, the cavity 12 is evacuated through the channel 7 . As a result of the pressure difference, the film 11 tears open, so that the suspension being treated can be suctioned off through the channel 7 for further analysis.
This variant is particularly suitable for a fully automated Ver driving, in which the risk of cross-contamination is completely eliminated can be closed.
In all cases described, chemical after-treatment is necessary in order to to efficiently bring the cell contents outside. The chemicals can be the solution be added before the electrical field application; but in many cases Electrolytes are or should work optimally in electrolytic buffers after completion of the electrical field application of the cell suspension who added the.
In a variant of the chemical treatment, the cell suspension becomes a Mi added from 15 different restriction endonucleases (3 U / ml each), and incubated at 37 ° C for about 10 to 30 minutes. The reagents diffuse through the membrane pores formed in the cell interior during electrical field application and deliberately cut the DNA strands into pieces with a few thousand bases pairs that can later be used for PCR reaction. After the treatment The proteinase K suspension is added in a concentration of 1 µg / ml adds, and the temperature level is set to 60 ° C. Proteinase K diffuses likewise through the pores and causes the lysis of both cell proteins and of the enzymes added in the previous step, so that disruptive influences on the subsequent PCR reactions can be excluded. After incubation of 10 The temperature is brought to 95 ° C. for up to 30 minutes at 60 ° C. The denatures Proteinase K, and the DNA diffuses out through the pores in the membrane. By such treatment, the PCR inhibitors, both added and also cell's own, largely degraded.
In another variant of the chemical treatment, the elec tric field application of the cell suspension guanidinium hydrochloride or - thiocyanate (GdnHCl or GdnSH) added, so that a 0.3 to 3 M concentration tion is set. The suspension is ge to a temperature of 60 to 100 ° C. brought and incubated for 10 to 30 minutes. This will make the cell walls and membranes destroyed, releasing the DNA. This method is easier than the one above described, but has the disadvantage that guanidinium compounds, although in egg ner lower concentration than otherwise used for purely chemical cell disruption be able to inhibit the subsequent PCR reaction and therefore from the Solution must be removed.
In the embodiment according to FIG. 4, surface roughness is provided on the upper sides of the electrodes 13 and 3 , through which tissue pieces 16 brought into the sample volume 1 can be comminuted.
The upper electrode 13 and the housing 2 are designed according to the embodiment of FIG. 4 such that the electrode 13 is guided axially and rotatably. The remaining volume of the sample volume 1 is filled with a liquid 17 and the tissue sample 16 is pressed between the electrodes by pressure on the upper electrode 13 . In this case, the liquid is only necessary to rule out electrical discharges between the electrodes, since the sample itself contacts the electrodes directly. The liquid introduced between the electrodes is therefore not necessarily electrically conductive, so that pure water or oil can be used.
After the high-voltage treatment and the addition of appropriate chemical additives, the tissue sample 16 is crushed by applying a strong pressure to the upper electrode 13 , possibly combined with rotational movements. The upper electrode 13 and the housing 2 with the lower electrode 3 thus play the role of a stamp and a die. The electrode surfaces preferably have a rough profile 17 in order to make the disintegration of the tissue more efficient.
The liquid thereby released or the slurry that forms, which contains the nucleic acids, rises into the gap 14 between the upper electrode 13 and the housing 2 and can be sucked off through the channel 15 for further analysis.
