Es wurden zwei kompetitive Assays für verschiedene Analyte auf derselben Oberfläche durchgeführt. Das Prinzip ist in der Abb. 1 wiedergegeben. Es wurde eine Sensoroberfläche (Tantaloxid-Lichtwellenleiter, Gitterkoppler) mit einem Oligonucleotid (DNA 35-mer, Template) durch kovalente Kopplung am 5'-Ende modifiziert. An das 3'-Ende wurde ein 14-mer (DNA) hybridisiert, das mit 2,4-Dichlorophenoxyessigsäure (2,4-D) am 5'- Ende modifiziert war. Hierdurch entsteht eine Oberfläche, die mit 2,4-D belegt ist. Auf dieser läßt sich mittels Monoklonaler Antikörper (MAk) ein kompetitiver Immunoassay durchführen. In der Abb. 2 sind Bindung eines MAk gegen 2,4-D (a) und unterdrückte Bindung (b) durch einen Überschuß an 2,4-D wiedergegeben. Zwischen allen Schritten wurde regeneriert, in dem der Doppelstrang durch NaOH gelöst wurde. Danach wurde die Oberfläche neu mit markiertem Oligomer durch Hybridisierung belegt. Anschließend wurden dieselben Schritte mit einem Atrazin-modifizierten Oligomer (14-mer mit derselben Basenzusammensetzung wie das 2,4-D markierte) wiederholt. (c) und (d) gibt die Bindung des Anti-Atrazin-Antikörpers in verschiedenen Konzentrationen wieder, (e) zeigt die Inhibition der Bindung in Gegenwart von Atrazin.Two competitive assays for different analytes were performed on the same surface.
