Abstract
Description
Die Erfindung betrifft Verbindungen der allgemeinen Formel I,
The invention relates to compounds of the general formula I
B. Hexafluorophosphat oder Tetrafluoroborat darstellt.in which R 1 , R 2 , R 3 and R 4 are independently aryl or preferably alkyl groups, where R 1 and R 3 and R 2 and R 4 but also R 1 - R 4 can each be the same and, in addition, R 1 and R 2 and R 3 and R 4 can be linked to form a cycle, which may also contain heteroatoms, while X⁻ is a non-nucleophilic anion such as, for. B. hexafluorophosphate or tetrafluoroborate.
The invention also includes non-ionic isourea derivatives of the general formula II
The Compounds of type I, but also of type II, serve as condensation reagents Synthesis of acid amides, especially peptides, and also for the synthesis of esters.
B. H.-D. Jakubke, Peptide, Spektrum Akademischer Verlag 1996).Condensation reagents play a role in the synthesis of peptides and peptidomimetics Peptide conjugates play an outstanding role (overview, see e.g. H.-D. Jakubke, peptides, Spectrum Academic Publishing House 1996).
30 (1989), 1927). D.P. Curran, Chemtracts 9 (1996) 75; F. Balkenhohl et al., Angew. Chem. 108 (1996) 2437).Most of these condensation reagents activate the Carboxyl function. In addition to various forms of carboxylic acid anhydrides, azides and fluorides (L.A. Carpino, M. Beyermann, H. Wenschuh, M. Bienert, Acc. Chem. Res. 29 (1996), 268) activated carboxylic acid esters, i. H. those that easily undergo aminolysis are accessible, used as activated carboxyl compounds. As particularly useful and It has proven to be productive if these activated carboxyl compounds are not isolated have to be generated in situ in the presence of the amine component to be acylated can be. This principle of reaction is e.g. B. realized in that the Carboxyl compound by means of a carbodiimide in the presence of an α-nudeophilic additive, such as B. 1-hydroxybenzotriazole (W. Koenig, R. Geiger, Chem. Ber. 103 (1970), 788), is intermediately converted into the corresponding active ester. A further development This methodology consists in that carbodiimide and nucleophilic additive form one Combination reagent with isouronium structure can be connected, as is the case with N, N, N ', N'-tetramethyl-O- (1-benzotriazolyl) uronium tetrafluoroborate (TBTU) is the case (R. Knorr, A. Trzeciak, W. Bannwarth, D. Gillessen, Tetrahedron Lett. 30: 1927 (1989). The Significance of these reagents has recently emerged through the development of techniques combinatorial chemistry experience an additional expansion (see D.P. Curran, Chemtracts 9 (1996) 75; F. Balkenhohl et al., Angew. Chem. 108 (1996) 2437).
Als Aktivester mit besonders günstigen Eigenschaften haben sich die Pentafluorphenylester von Aminosäuren und Peptiden erwiesen (J. Kovacs, L. Kisfaludy, M. Q. Ceprini in "Peptides" 1966 (H. C. Beyerman, A van den Linde, W. Massen van den Brink, Hrsg.), North Holland, Amsterdam 1967, Seite 23; J. Kovacs et al., J. Am. Chem. Pentafluorophenyl esters have proven to be active esters with particularly favorable properties of amino acids and peptides (J. Kovacs, L. Kisfaludy, M. Q. Ceprini in "Peptides" 1966 (H.C. Beyerman, A van den Linde, W. Massen van den Brink, ed.), North Holland, Amsterdam 1967, page 23; J. Kovacs et al., J. Am. Chem. Soc. 89 (1967), 183), about a Adduct of dicyclohexylcarbodiimide and pentafluorophenol were obtained.
The Type I condensation reagents which are the subject of this invention combine the Advantages of the combination reagents with isouronium salt structure (see above) with those of the Pentafluorophenyl ester. They allow the pentafluorophenyl ester of carboxyl compounds, such as N-protected amino acids and peptides, in the presence of those to be coupled Generate anino component and the direct reaction to the desired Subject to condensation product.
The compounds of type I derived from pyrrolidine (R 1 , R 2 and R 3 , R 4 each = - (CH 2 ) 4 -, X = PF 6 ) dissolve z. B. excellent in dichloromethane, which is often not the case with other types of uronium coupling reagents. In addition, no additional additives are necessary in the condensation reagents according to the invention because the reactivity of the pentafluorophenyl esters formed as intermediates is sufficiently high. The free carboxylic acids also do not react, but rather the salts formed with tertiary amines, so that the method can be used even with very acid-labile reactants. The synthesis of condensation reagents according to the invention is shown below for two examples:
The pyrrolidine and phosgene are used to obtain the N, N-N ', N'-bisgetramethylene) urea, which can be converted to N, N-N', N'-bis with phosgene (or other suitable agents such as PCl 5 ). tetramethylene) chlorformamidium chloride is implemented. An anion exchange is carried out with potassium hexafluorophosphate and the chloroformamidinium hexafluorophosphate obtained is reacted with potassium pentafluorophenolate to give compound 1 according to the invention.
In an analogous manner, secondary amines can generally be converted into the corresponding ones condensation reagents according to the invention are converted.
The tetramethyluronium compound can be obtained from commercially available tetramethylurea going out.
This is obtained from tetramethylurea by reaction with phosgene Tetramethylchloroformamidinium chloride, which is the anion exchange to the corresponding Hexafluorophosphate is subjected. This chloroformamidium salt is with Potassium pentafluorophenolate transferred into the condensation reagent 2 according to the invention.
Both condensation reagents 1 and 2 are crystalline and therefore easy to handle. Particular advantages of these condensation reagents according to the invention are that they combine reactivity with very good solubility even in less polar solvents such as dichloromethane. The solubility in dichloromethane is important not least for solid phase syntheses including the synthesis of compound libraries according to the principle of combinatorial chemistry. The solubility can be modulated by varying the groups R 1 , R 2 , R 3 and R 4 . So z. B. reagent 1 derived from pyrrolidine is somewhat more soluble in dichloromethane than tetramethyl compound 2.
Die Wirksamkeit der Kondensationsreagenzien wird in Synthesen von Modellpeptiden
demonstriert:
The effectiveness of the condensation reagents is demonstrated in the synthesis of model peptides:
B. in Gegenwart von Ethyldiisopropylamin wesentlich schneller als bei Anwesenheit der schwachen Base sym-Collidin.1) Fluorenylmethoxycarbonyl-glycine (Fmoc-Gly-OH) reacts with valine tert-butyl ester (H-Val-OtBu) under the action of one equivalent of the reagent PfPyU 1 according to the invention in high yield to the protected dipeptide. The reaction rate depends on the choice of base. The reactions are e.g. B. in the presence of ethyldiisopropylamine much faster than in the presence of the weak base sym-collidine.
2) The condensation reagent 2 acts in the same way, as is shown for the condensation of Fmoc-alanine (Fmoc-Ala-OH) with valine tert-butyl ester to give the dipeptide derivative 4.
Das zeigt die Synthese des Tripeptids 5. 3) Even sterically very demanding coupling couplings such as those from valine to valine are smoothly accomplished by the reagents according to the invention. This is shown by the synthesis of tripeptide 5.
Another example of the peptide coupling brought about by a type I compound according to the invention, namely 1, embodies the linkage of the fmoc-asparagine (Fmoc-Asn (Trt) -OH) which is trityl-protected in the side chain with valine-tert-butyl ester to give the dipeptide derivative 6 .
5) Amide condensations with less basic amines can be achieved by the condensation reagents according to the invention thanks to their high reactivity. This is shown below by the synthesis of the N-glycosyl-asparagine derivative 10, which was prepared from the Fmoc-aspartic acid ester 7 deblocked in the side chain and the glycosylamine 9 obtained immediately before the reaction from the glycosyl azide 8 under the action of PfTU 2 in dimethylformamide.
In addition to the excellent solubility and high reactivity of the novel Compounds of type I according to the invention have the further advantage that the addition of another additive is not necessary, and that these reagents also in the absence of the couplable amino component also for the synthesis of Carboxylic acid pentafluorophenyl ester can be used.
For the synthesis of the amides but also for the synthesis of the pentafluorophenyl esters, this can continue related type II reagent, the 2,4,6-tris (pentafluorophenyloxy) -1,3,5-triazine 11, which is obtained from cyanuric chloride, are used. The reagent is preferred, even in to be n-hexane soluble.
The pentafluorophenyl esters can be stored in the customary manner and, as is known, themselves valuable activated carboxyl derivatives.
