Die enzymatische Reaktion wurde durch Zugabe von 50 µl einer methanolischen Lösung von 2-PPA (50 mM) gestartet. Das Gesamtvolumen jedes Ansatzes betrug 0,5 ml. Die Konzentrationen von 2-PAA und Hydroxylamin waren 5 mM und 0,5 M. Es wurden 10 identische Ansätze gestartet und in Abständen von jeweils 10 min durch die Zugabe von 50 µl 1 N HCl gestoppt. Ausgefälltes Protein wurde durch Zentrifugation entfernt und aus dem Ansatz wurden 50 µl für HPLC-Messungen zur Konzentrationsbestimmung von 2-PPA und 2-PPHS entnommen. enzymatische Reaktion wurde durch die Zugabe von 3,725 g
(0,025 mol) (R,S)-Phenylpropionamid (gelöst in 500 ml Metha
nol) gestartet. Der Reaktionsverlauf wurde mittels Ionen
paar-Chromatographie verfolgt. Nach 120 min war der Umsatz
der S-Enantiomeren des Amids zur korrespondierenden Hydro
xamsäure beendet und die Zellen wurden durch Zentrifugation
entfernt, bzw. der Rohextrakt oder das gereinigte Enzym
