Preferably, the oxo group is attached with respect to the benzene rings of the starting material at the free ortho-position of the Rl groups.
The ring cleavage of the compounds of the formula II can be carried out in various ways ; .eise done. For example, the cleavage can take place under alkaline conditions under use a caustic alkali, for example an alkali metal hydroxide, such as sodium or potassium hydroxide. On the other hand, the split can can be carried out using iydrazine. The cleavage is preferred in an aqueous medium and with heating, for example at reflux in a inert atmosphere such as nitrogen. After the cleavage, the received Compound of formula III obtained by acidification and, if desired, by conventional means Crystallization processes are purified.
The compound of formula III or a salt or alkyl ether of the phenolic OB group can be converted to the compounds of formula I or derivatives thereof by various methods. Thus, the compounds of the formula IU or a salt or alkyl ether thereof can be used in compounds of the general formula where R1 is an alkoxy group and M is hydrogen, an alkali metal cation or an alkyl group; in which M1 is hydrogen or an alkali metal cation and R "is an OH group or an alkoxy group, and then these intermediates are converted into compounds of the formula I or derivatives of the formula I; the conversion of the compounds of the formula ITI can also be via a chromanone compound or a chromone compound ,, which in positions 2 and / or 21 carrying groups which can be converted into the desired COOIf group or derivatives thereof, take place. * the compounds of formula IV can .werden prepared by reacting a compound of formula III or a salt or ether thereof, in which Ri is acetyl , with a compound of the formula R7Co CZ R8, in which 1t7 is a halogen atom or an alkoxy group, Z is a (lial_) z group, where Hal is lloccii, and R8 denote an alkoxy group or Z, a carbonyl oxygen atom and R8 represent a halogen atom or an alkoxy group, is condensed ii; ilot @ en compound in which R @ is hydrogen, hold @@@ =. rden, Examples of suitable compounds of the formula R7CU CL R8 are dialkyloxaliite esters, such as diethyloxality; Oxaline mono- or dihalogerides, such as ethoxalyl chloride or oxalyl chloride i (]; and alkoxy dihaloacetates such as ethoxy dichloride acetate.
Dic thereof can condensing the compound of formula III wherein R1 is acetyl, or a salt or ether by merely: #ischen of the reactants and, if desired, heating to a temperature of 25 to 1500C, preferably about 800C .s 7e L,? be performed. In the case of the oxalate esters, the reaction is expediently in the presence. (a KondensationsinIttels performed Suitable agents are, for example, metal alkoxides such as Natriumäthoxyd, Natr iiii:.... ihvdrid, sodium amide or sodium metal The honderisiitionsmittel can be formed in situ, for example by use of ethanol as a reaction medium and addition of Natriuinmetall In MAN above cases, the alkali metal salt of the compound of the formula III act as part of the required condensing agent.
When a substituted dihaloacetate is used, the reaction is preferably carried out in the presence of a finely divided metal catalyst such as finely divided platinum group metals.
If desired, the reaction can be carried out in an inert solvent or diluent such as 1) ethyl ether, dioxane, ethanol, benzene, tuluene, tetrahydrofuran or mixtures thereof .
The reactants are usually employed in essentially stoichiometric proportions. For example, if desired, each reactant can be employed in a 100 to 30 molar excess of molar oil . If a condensing agent is used, it is expediently used in an amount of from zoo to 750 mol%, based on the amount of the compounds of the formula III used, preferably from zoo to 500 mol% . It goes without saying that the condensation is expediently carried out under essentially anhydrous conditions , ie essentially in the absence of originally present or added water.
When the compounds of the formula R7 CO CZ Ra. Oxalylhalogenide Bind, the condensation can be carried out in a manner similar to that described above with respect to the use of the other compounds of the formula 87C0 CZ R a , with the exception that an acid binder is used in place of the condensing agent and an anhydrous organic solvent is used; is not worth it. Suitable acid binders are alkalis, such as sodium or potassium carbonate. and alkali metal oxides and organic amines such as pyridine or triethylamine. The acid binder is present in at least the theoretical stoichiometric amount; to bind all of the halogen in the Diy carbonyl compound. It may be desirable to use an excess of the acid binder and, if desired, the acid binder can be added to the reaction mixture in a variety of additions over a period of time. In some cases, the acid binder can be used as the reaction medium. The reaction mixture in this process usually contains the intermediate of the formula: where R @ those given above. Has meaning or represents halogen, or a precursor or a derivative thereof. The rearrangement of this intermediate product can be achieved in situ by adding alkali and heating. It will. however, it is usually preferred to recover the intermediate product from the reaction mixture and rearrange it after purification, which can be carried out as desired, and then rearranged in a separate process step. The recovery and purification of the intermediate product can be achieved by conventional methods . The rearrangement of the intermediate can be carried out using a Baker Venkatamann rearrangement method.
Gewöhnlich wird es vorgezogen, die Verbindung der Formel IV aus der Reaktions- mischung zu gewinnen und sie in Anwesenheit eines Zyklisierungsmittels und gegebenenfalls einer kleinen Menge Wasser, wie im nachstehenden beschrieben wird, zu zyklisieren.pie reaction mixtures of the above reactions usually contain the compound of formula IV or a precursor thereof, although in some cases the cyclization of this pro-. ducting to the compound of the formula I or a derivative thereof can take place spontaneously. The Zvklisierung the compound of the formula IV to. The compound of the formula -I or a derivative thereof can be obtained by acidifying the reaction mixture in situ . It is usually preferred to recover the compound of formula IV from the reaction mixture and to cyclize it in the presence of a cyclizing agent and optionally a small amount of water, as will be described below .
The compound can be recovered from the crude reaction mixture in which it was prepared by conventional methods .
. The compounds of the formula IV including their salts and ethers can easily be cyclized , for example by heating under non-basic conditions, the analogous compounds of the formula I or their derivatives being obtained. The cyclization is preferably carried out in a non-reactive solvent such as ethanol or oxoane; 6s is also preferred, the cyclization in the presence of a cyclization catalyst, preferably an acid cyclization catalyst such as a polyphosphoric acid, sulfuric acid, hydrochloric acid, acetic acid or mixtures thereof. to perform. When an ether of a compound of the formula IV 'is used, cyclization and dealkylation can be achieved simultaneously by using hydrogen iodide or hydrogen bromide as cyclization catalyst. .
The cyclization can be carried out from ambient temperature to about 1000 ° C. , for example by heating the reaction mixture on a steam bath and, if the nature of the reaction medium permits, under atmospheric reflux. The compounds of the formula I or their derivatives can be obtained from the cyclization Reaction mixture can be obtained by conventional methods.
The compounds of the formula Y kbanen * ftrbh reacting the compound of the formula III, in which R: i is hydrogen, or an alkali metal metal thereof with a koetylenedicarboxylic acid or an ester thereof, are obtained under alkaline conditions, the compound of the formula V being obtained after hydrolysis. The kcetrlenedicarboxylic acid esters can be derived from alcohols with 1 to 10 carbon atoms . However, since the ester portion has to be removed, it is preferable to use simple esters, such as methyl, ethyl, propyl or butyl alcohols. It is further preferred that both carboxylic acid groups of the acetylenedicarboxylic acid are esterified. In this process, the ester and the phenol compound III are preferably reacted in approximately -atoichiometric amounts under alkaline conditions. These can be caused by the presence of an organic base,. such as benzyltrimethylammonium hydroxide, or an alkali metal hydroxide. However, it is expedient if the alkali present is in the form of an alkali metal salt, for example the sodium salt, of the compound of the formula III, such a salt being regarded as free phenol when the amount of phenol compound present in the reaction mixture is determined. When this happens, the alkali metal salt can conveniently be prepared in situ in the reaction mixture by adding sodium metal. The alkali is believed to be catalytic and it is possible to use less than 100 mole percent thereof based on the phenolic compound present. Preferably, essentially 10 mole percent are used. It can be seen that the reaction, particularly when the alkali metal salt is formed in situ, is conveniently carried out under essentially anhydrous conditions. The reaction is preferably also carried out in a solvent. or. Diluent carried out. Suitable media are, for example, an excess of the compound of the formula III, diphenyl ether, dioxane or anisole. These media have the advantage that the reaction can be carried out at elevated temperatures and at atmospheric pressure. In general, it is preferred to carry out the reaction at temperatures of from 50 to 1,000.
Instead r d $ s Acetylendioarbonsäureesters used in the above process for the preparation of the compound of V rormel an ester of a Konohalogenfumarsäüre or a precursor thereof are used, that is, esters of the acids. general formula: wherein R12 is halogen and R 13 is hydrogen when Rlo and R 11 form a carbon-carbon bond with one another; or any two of the groups R1Q, R11, R12 and R13 are halogen and the other two are hydrogen. in this case the reaction is not an addition reaction but a condensation reaction which at the same time brings about the splitting off of hydrogen halide between the phenol compound of the formula III and the halofumaric acid ester. This acid must be removed from the system and the reaction is therefore carried out in the presence of at least a sufficient amount of an acid binder to remove the hydrogen halide which would be formed during the overall process. The hydrogen chloride does not necessarily have to be removed in one step, but can first be removed as a proton and then as a halogen anion. In this context, the term "acid binder" is used both for # conventional acid binders, such as pyridine and triethylamine, as well as for materials which, for example, first remove the hydrogen from the phenolic OK group (where = # in a phenate salt is formed, and then are displaced from the phenate salt, forming a salt with the halogen of the halo tamaric acid ester. Apart from the use of an other acid ester reactant and the presence of the acid binder, the process can be carried out in a manner similar to that used when an acetylenedicarboxylic acid ester is used If the acid binder is usually also a strong alkali, it is generally not necessary to include a particularly strong alkali in the reaction mixture.
As stated above, it is also possible to use connections which under the conditions of the reaction with phenol the desired halofumaric acid ester result. Such other compounds or precursors are halogen maleic acid esters and dihalosuccinic acid esters. if can be used it be necessary to provide further alkali to convert the precursor into to ensure the desired halofumaric acid ester. Such an alkali can be a mere excess of the acid binder.
The products obtained from the reactions just mentioned usually contain the compounds of the formula V in the form of their esters. The compounds of the formula can be prepared from these products by acidifying the reaction mixture, followed by hydrolysis of the esters by boiling with alkali and acidifying to release the free acid, Removal of the organic solvent or diluent (if present) and extraction of the aqueous solution, for example with ether, which can then be evaporated. The solvent extraction of the acid can be carried out as indicated or, if desired, can be carried out after hydrolysis of the ester of the compound of the formula Y. If necessary, the product can be subjected to further purification, for example by extracting the ethereal solution with sodium bicarbonate and then precipitating the acid of the formula V by adding dilute sulfuric acid.
The compounds of the formula V can be cyclized by treating the compound with a cyclizing agent at ambient temperature or above. Suitable cyclizing agents are dehydrating agents such as phosphorus pentoxide, polyphosphoric acid, sulfuric acid, chlorosulfonic acid and other Lewis acids. In certain cases it is also advisable to use glacial acetic acid, which contains a small amount of hydrogen chloride or hydrogen bromide. It can be seen that since cyclization in these cases is achieved through the use of dehydrating agents, the presence of added or primal water in the reaction mixture is undesirable. It is usually preferred to subject the compounds of Formula V to an initial drying step and to carry out the cycling under essentially anhydrous conditions.
On the other hand, the cyclization can be achieved by converting the free carboxyl groups of the compound of the formula V into acyl chloride groups, for example by treatment with PC13 or PC15, and subjecting the acyl chloride thus obtained to an internal = Friedel-Crafts reaction .
I. The compounds of the formula VZ can be prepared by reacting a compound of the formula III, in which R 1 is acetyl, or an alkali metal salt thereof with olvoxalic acid or an enter thereof in the presence of a base (e.g. aqueous sodium hyaroxide) or a mineral acid. A water-miscible solvent such as alcohol may be added to facilitate the reaction. The residue, which contains the compound of the formula VI in which R "is OH means, can be recovered using conventional methods and then purified by, for example, recrystallization. The reaction can, however @ proceed, wherein a bis-2-carboxyohromanonverb-indung is directly obtained without isolation of an intermediate, which is then, as described in post-_ a stationary, can 'be converted into a compound of formula L, the compounds of Fvrmel VI can be treated. with an alkali or an organic base in a suitable inert solvent medium be cyclized to the compounds of formula 1, or derivatives thereof. The compounds of Formula IX can be cycled in a similar manner which is used in the compounds of formula V.
As with the compounds of the formulas IV and VIII, the preparation of the compounds of the formulas V and IX. zuatcanen largely be described as a process in which a compound of the formula III, wherein R1 is hydrogen, or a salt heave with a compound of the formula or an ester thereof, in which formula Rlo, R11 'R12 R 1 3 and D have the meanings indicated, and when Rlo and @ R11 together form a carbon-carbon bond, R12 and R 1 3 also can form a carbon-carbon bond, is reacted will. . The compounds of .Formula X can likewise be prepared and.cyclized in a manner similar to that used for the preparation and cyclization of the compounds of formula VI. a compound of the formula III, in which R 1 is acetyl, or a salt thereof with an aldehyde of the formula OHCV, for example cinnamaldehyde, can be used under essentially the same conditions as are used to prepare the compound of the formula VI from glyoxalic acid, implemented. However, it is preferred to use ether. of compounds of the formula III to apply and to @dealkylate the reaction product, a compound of the formula X being obtained.
As with the other intermediate products, certain procedures for the preparation of compounds of the formulas VI and X7 together can largely be described as a process in which a compound of the formula III, in which R 1 is acetyl, or a salt or ether thereof, with a compound of the formula OHOD, in which D. has the meaning given above, is implemented . In addition to the processes described above for the preparation of the compounds of formula VII, a number of other processes which do not necessarily lead to the intermediates of formulas VIII, IX or X can readily be given. Thus, the 2-formyl compound can be prepared by reacting a compound of the formula III, in which R1 is acetyl, or a salt or ether thereof with a substituted acetic acid or an ester thereof of the formula (R60) 2CH-COOR6, in which R6 is an alkyl group, for example ethyl diethoxyacetate , getting produced.
In this case, an acetal compound is used as an intermediate product, which can be hydrolyzed, for example, with a dilute mineral acid, the desired -CHO group being obtained. On the other hand, an A1-Ikylaeylacetat can be condensed with a compound of the formula 11I, in which R1 is hydrogen, optionally in the presence of a dehydrating agent such as phosphorus pentoxide.
In addition to the direct conversion of a compound of the formula VIII into the desired one. Compound of the formula I can be the group V in the compounds. of formula VII can be converted in a known manner from one form of the substituent to another, more preferred substituent. -That is how she serves. Compound of formula VII! where V represents a methyl group, also as an intermediate product in the production of a number of other oxidizable derivatives. For example, the methyl group can be converted into a halomethyl group, for example by reaction with hydrogen chloride and manganese dioxide in boiling acetic acid, whereby a chloromethyl group is formed, or by reaction with bromine in acetic acid, a bromoethyl group being obtained.
The halomethyl compound may be accomplished using, for example, chromium trioxide y as an oxidizing agent in the presence of acetic acid = in the corresponding carboxylic acid are oxidized.
The methyl compound can also be reacted with p_nitrosodimethylaniline and the reaction product hydrolyzed with dilute mineral acid, the corresponding formyl compounds being obtained, which can be oxidized into the corresponding carboxylic acid using, for example, chromium trioxide as the reactant.
By condensing the xethyl compound with an eenzaldehyde in the presence of a condensation catalyst, the styryl compound is obtained, which can be oxidized into the corresponding carboxylic acid using, for example, potassium permanganate. The formyl compound can also serve as a starting material for the preparation of the cyano compound . For example , the formyl compound can be reacted with hydroxylamine , the oximino compound being obtained which, after dehydration to decorate the cyano compound , can be hydrolyzed under acidic conditions to give the carboxylic acid or an amide thereof. As indicated above, the compounds of the formula I can also be prepared via a compound in which A1 and A2 form the chain -CO-CH2-CH (D) -0-. The conversion of this intermediate product can take place via a compound of the formula VII, if the group D is a group V, or lead directly to the compound of the formula I or a derivative thereof. The conversion can be achieved by dehydrogenation and subsequent oxidation, or hydrolysis of one of the substituents in the 2 and 21 positions, if necessary. The dehydration can be effected, for example, by using selenium dioxide, palladium black or chloranil. On the other hand, the dehydrogenation can be carried out by bromination and subsequent dehydro- bromination. For example, the intermediate in which A1 and A2 are present as COCH2-CH (D) -O chains can be obtained using N-bromosuccinimide in an inert solvent or by treatment with pyridine perbromide in an inert solvent such as chloroform in the absence of a free radical catalyst as-benzoyl peroxide, which can be dehydrobrominated then be brominated to give the 3-bromo-äerivat_erhalten is. The watchomanone intermediates can be prepared by the action of a w-substituted w-chloropropionic acid or a derivative thereof on a compound of the formula II.I ,, in which R1 is hydrogen, in the presence of a basic reactant and subsequent conversion of the acid into the acid chloride and treatment with aluminum halide in Presence of a suitable solvent (e.g. nitrobenzene); or by the action of a compound of the formula III, in which R 1 is hydrogen, on a W -substituted acrylonitrile, with hydrolysis and cyclization of the product. As indicated above, the cyclization of the intermediates of the formulas VI and X can lead to the preparation of a corresponding bischromanone compound which, as indicated above, can then be converted into the corresponding bis-chromone compound.
In addition to the processes given above for preparing the compounds of the formula I from the compounds of the formula III via the intermediates IV to VI and VII to X, other methods can be given which do not necessarily lead to the formation of one of these intermediates. Thus, a compound of the formula III, in which R 1 is hydrogen, can be reacted with a suitable monohalo oxalate, the analogous oxalate ester of the formula R1500CCOORI4 being obtained, in which formula R150 is the remainder of the compound der-Fommel 'III and R14 is cne Denotes alkyl or aryl group. This oxalate ester can then be condensed with acetyl halide, acetic anhydride or acetic acid in the presence of a Lewis acid, an ester of a compound of the formula I being obtained. optionally in the presence of a dehydrating agent such as phosphorus pentoxide, heated.
It can be seen that in the above methods it is possible ditto Groups R1 and 011 on each benzene ring by vc-rscIiiedc: ne, method into the compound - to convert formula III. In the above at;, egebexien general procedure is it hence it is not necessary that every pair of groups A3 and A4 be the same. . By the methods given above can use the free acids of the formula I or derivatives are made from it. It is also within the scope of the present invention, either product obtained according to the invention after any isolation and purification steps, which may be desired to be treated to release the free acid or convert one form of derivative into another. The procedures used to Release of the acid to convert one derivative into another and to isolate as well as cleaning a product can be those conventionally used for keeping animals of, for example, alkyl esters, ammonium, alkali metal or alkaline earth metal salts, Amides derived from ammonia, primary or secondary aliphatic or aromatic Amines. or A: ninosäureii can be used. Furthermore, these methods can also used to make derivatives of other functional groups in the molecule to build.
