Reaktion so"rie einen negativen Eisen(II)-chlorid- und
Millon Test. Die Substanz reagiert Ninhjdrin-positiv und

Die Toxizität des Tetrauycins ist bei oraler Verabrei-
chung-wie allgemein bei Polyenantibiotika gering. Bei
i. v.-Applikation wurde für die Maus die LD50 =
83,5mg/kg bestimmt.
Tetramycin wird durch Züchtung des Streptomyces-Stammes,
Strept'omyces noursei HAZEN et BROWN, 1950 var. jenensis
nov. var. JA 3789 oder seiner Mutanten oder Varianten
unter aeroben Kulturbedingungen in einem flüssigen Nähr-
medium gebildet Die Züchtung wird in geeigneten Nähr-
medien bei Temperaturen von 24 bis 32 00 während 2 bis
4 Tagen unter Belüftung und Rührung durchgeführt und
das gebildete Antibiotikum durch Extraktion aus der
Kulturlösung oder getrennt aus dera Kulturfiltrat und
dem Mycel gewonnen.
Der Stamm JA 3789 wurde im Jahre 195? in humösem Lehm.
aus Vietnam gefunden. In der Tabelle I sind die morpho-
logischen und physiologischen Merkmale dieses Stammes
zusammengefaßt:
Durch Vergleich der morphologisch-Physiologischen Eigen-
schaftendes Stammes JA 3789 mit in der Literatur be-
schriebenen Tetraenantibiotika-Bildnern und anderen _
Streptomyceten:Arten wurde Ähnlichkeit mit Actinomyces
rochei BLRGER et a1.. , 1949 emend. GAÜSE et al. , 1957
festgestellt (fÜGIMR u. BRADT.ER, 1963). Aus später mög-
lichen Direktvergleichen mit Referenzstämmen der Art
Streptomyces rochei BERGLR et a1., 1949-(ML B-1599,
ATCC 10739) und Streptomyces noursei HAZEN et 'BROWN,
1950 (ATCC 11455) ergab sich, daß die morphologischen
und physiologischen Merkmale des Stammes JA 3-89 weit-
gehend mit denen der Species Streptdmyces üöürsei über-
einstimmen.:
Bei Streptömyces rochei ATCC 10739 wL.rde im Gegensatz
zu JA 3789 eine glatte Oberfläche der.,Sporeüfestge-
stellt. Die Sporophoren bilden: offene, sehr läng gezo-
gene Spiralen oder unregelmäßige Wellen mit einem an-
deren Abzweigungsmodus als JA 3789. Auch düs Spektrum
der Kohlenhydrätverwertung ist verschieden.
Nach dem Vergleich mit Streptomyces noursei ATCC 11455 wird auf
Grund von Abweichungen in der Kohlenhydratverwertung und der speziellen Bildung
von `letramycin der Stamm JA 3`e39 als Varietät der Species Streptom#ces noursei
Streptomyces noursei HAZEN et BROVJN, 1950 var. jenensis nov. var. JA

Filtrat versetzt man tropfenweise unter Rühren mit Aceton. Dabei fällt das Triäthylammoniumsulfat-Doppelsalz
amorph an und ist hellgelb gefärbt. Eine Suspension des Doppelsalzes in Methanol/Wasser
wird mit Natriumhydroxid auf pH 9 bis 10 eingestellt, filtriert, neutralisiert und
unter Röhren -tropfen @ezse mit Wasser versetzt. Das Antibiotikum fällt als gelblich-weißer,
schleimiger Niederschlag aus, der erbfiltriert und öetrocknet wird. Zur weiteren
Reinigung wird mehrfach mit heißem absoluten Methanol ausgezogen und der verbleibende
Rückstand aus.807'öigem Methanol umkristallisiert. Die Reinigung des Rohproduktes
kann auch durch Lösen in Pyridin und Ausscheidung in. der Kälte und/oder Ausfällen

150 g Rohprodukt werden unter Rühren in 1 1 20;öiger
methanolischer Triäthj-lammoniumßulfat-Lösung suspen-
diert und anschließend weitere 30 blin. gerührt. Dann
filtriert man den unlöslichen Rückstand ab (20 g) und
enrit das Filtrat i.m. Vakuum auf 500 ml ein. Das einge-
engte Filtrat wird dann langsam. unter Rühren in 2 1
Aceton eingegossen und zur Vervollständigung der Fäl-
lung über Nacht im Kühlschrank stehengelassen, bevor
der Niederschlag abfiltriert und getrocknet wird. Das
Triäthjlaiiimoniumsulfat-Doppelsalz fällt amorph an und
ist hellgelb gefärbt.
Ausbeute: 35 g
Eine Suspension von 35 g Doppelsalz in 500 ml Methanol/
Wasser (1:1) wird mit 2,5 n methanolischer Natronlauge
auf pH 10 eingestellt, danach filtriert und mit metha-
nolischer Phosphorsäure wieder neutralisiert. Diese
Lösunö fügt man tropfenweise unter Rühren zu 1500 ml
Wasser, läßt den Niederschlag über Nacht bei 0 00 ste-
hen, filtriert das schleimig anfallende Antibiotikum
ab und trocknet es zuletzt über Galciumehlorid.
Ausbeute: 15 g gelblich gefärbtes, amorphes Rohprodukt
Diese 15 g werden mehrmals mit heißem absolutem Methanol
extrahiert. Die Extrakte sind gelb gefärbt. Der metha-
nolfeuchte Rückstand wird in der notiendigen Menge
Pyriüin gelöst und 24 Std. bei - 5 00 stehengelassen.
Der ausr-».;fallene breiartige Niederschlag -.jird abfil-
triert und so--Lange mit Äther gewaschen, bis das Fil-
trat farblos erocheint.
Anschließend kristallisiert man den fast .reißari Filt-a--
kuchen' aus 70;o-igem P:iethanol um und erhält feine, @.ei@3e
