Verfahren zur Herstellung von Anthranilsäureamiden Es ist aus
Journal of Organic Chemistry, Band 18, 1953, Seiten 1427 bis 1439, bekannt, daß
sich substituierte Anthranilsäureamide durch Umsetzung von Isatosäureanhydrid mit
primären oder sekundären Aminen herstellen lassen. Man trägt dazu bei Raumtemperatur
Isatosäureanhydrid in eine wäßrige Lösung des Amins ein, wobei sich unter Kohlendioxydabspaltung
das dem Amin entsprechende substituierte Anthranilsäureamin bildet. Die Umsetzung
von Isatosäureanhydrid mit Methylamin läßt sich wie folgt formulieren:
Die Ausbeuten an Anthranilsäureamid sind von der Art des verwendeten Amins, dessen
Konzentration sowie von den Mengenverhältnissen abhängig. Sie erreichen bei einem
Molverhältnis von Isatosäureanhydrid : Amin von 1:2,5 ein Maximum, das zwischen
30 und 90 % der Theorie liegt. Nach dem Verfahren der deutschen Patentschrift 1'091 120 kann man
auch so arbeiten, daß man das Isatosäureanhydrid unter Kühlen mit einer wäßrigen
Lösung des Amins übergießt.
Die so hergestellten Anthranilsäureamide
enthalten stets noch mehr oder minder große Mengen an 2-Ureidosäuren, besonders
bei der Umsetzung höherer Amine. Im Falle der Umsetzung von Isatoaäurean-.hydrid
mit n-Butylamin entsteht die 2-N'-n-Butylureido-benzoesäure:
Die Ureidobenzoesäuren lassen sich durch Herauslösen. des Anthrani-lsäureamids mittels
Salzsäure oder Schwefelsäure und nachfolgende Ausfällung mit Alkalilauge abtrennen
und reinigen. Für eine technische He--tellung von Anthranilsäureamiden ist dieses

oder Alkoxyreste mit 1 bis 4 Kohlenstoffatomen stehen, R3 einen aliphatischei_4
cycloaliphatischen, araliphatischen oder aromatischen Rest bedeutet und R4 Wasserstoff
oder einen Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeutet, durch Umsetzen von
Isatosäureanhydriden der allgemeinen Formel
in der R1, R2 und R4 die angegebene Bedeutung haben; mit einem
Amin
der allgemeinen Formel NH2R3 III, -in der R3 die angegebene Bedeutung hat, vorteilhaft
herstellen lassen, wenn man das Isatosäureanhydrid und das Amin gleichzeitig in
etwa etöchiometrischen Mengen in ein inertes Lösungsmittel ein-bringt. Das Verfahren nach der Erfindung ergibt bessere Ausbeuten an Anthranilsäureamiden
als die bekannten Verführen. Die Anthranilsäureamide feilen unmittelbar in sehr
reiner F a_F1 und praktisch frei von den entsprechenden 2-Ureido-benzoesäuren an.The process according to the invention gives better yields of anthranilic acid amides

In den bevorzugten Ausgangsstoffen II bedeuten R1 und R2 Wasserstoff,
die Nitrogruppe, ein Chlor- oder Bromatom, wobei die erwähnten Substituenten vorzugsweise
in 6- oder 8-Stellung stehen. Geeignete Iaatosäureanhydride sind beispielsweise:
6-Nitroisatosäureanhydrid, 6-Chlorisatosäureanhydrid, 6,8-Dichlorisatosäureanhydrid,
6-Nitro-N-methylisatosäureanhydrid und 6,8-Dibromisatosäureanhydrid. In den bevorzugten
Aminen III steht R3 für einen aliphatischen Kohlenwasserstoffrest mit 2 bis 18 Kohlenstoffatomen,
einen cycloaliphatischen Kohlenwasserstoffrest mit 5 bis 12 Wasserstoffatomen, einen
araliphatischen Kohlenwasserstoffrest mit 7 bis 12 Kohlenstoffatomen oder einen
aromatischen Kohlenwasserstoffrest mit 6 bis 10 Kohlenstoffatomen. Die erwähnten
Kohlenwasserstoffreste können auch inerte Gruppen oder Atome enthalten, wie Hydroxylgruppen,
Halogenatome und Ätherbrücken. Bevorzugt=.: Amine dieser
Art sind
beispielsweise: Äthylamin, Propylamin, Isopropylamin, sekundäres Butylamin, n-Butylamin,
Allylamin, Propargylamin, Laurylamin, Oleylamin, Stearylamin, Cyclohexylamin, Cyclododecylamin,
Benzylamin, ß-Phenyläthylamin, Anilin, ß-Naphthylamin, p-Toluidin, Dimethylamin,
Dibutylamin, N-Methylanilin und Dicyclohexylamin. Es ist ein Vorteil des Verfahrens
nach der Erfindung, daß man auch und gerade dann gute Ausbeuten und reine Produkte
erhält, wenn man das Isatosäureanhydrid@und das Amin etwa im stöchiometrischen Verhältnis
umsetzt. Hin und wieder ist ein geringer Überschuß ari Amin, beispielsweise bis
zu 10 Mol%, zweckmäßig. Die Verwendung noch größerer Mengen von Amin ist im allgemeinen
mit keinem besonderen Vorteil verbunden. Man führt das Verfahren zweckmäßig zwischen
0 und 130°C, insbesondere zwischen 50 und 120°C, durch. Bei niedrigeren Temperaturen
ist die Reaktionsgeschwindigkeit gering. Bei höheren Temperaturen treten in zunehmendem
Maße Neben- und@Folgereaktionen auf. Wenn es auch möglich ist, die Umsetzung ohne
Mitverwendung eines Lösungsmittels durchzuführen, indem man die Ausgangsstoffe im
stöchiometrischen Verhältnis miteinander vereinigt, so ist doch die Umsetzung in
Gegenwart eines inerten Lösungsmittels für die technische Durchführung der Reaktion
das Verfahren der Wahl. Geeignete inerte Lösungsmittel sind beispielsweise: Wasser,
Alkohole, Ketone" Äther, Kohlenwasserstoffe, Halogenkohlenwasserstoffe, .Carbonsäureester,
Dimethylformamid und Dimethylsulfoxyd. Im einzelnen seien beispielsweise Methanol,
Äthanol, Tsobutanol, Aceton, Methyläthylketon, Tetrahydrofuran, Dioxan, n-Octan,
Leichtbenzin,
Chloroform, Dichloräthan, Tetrachlorkohlenstoff, Äthylacetat
und Chlorbenzol genannt. Bevorzugte Lösungsmittel sind Wasser und polare, mit Wasser
mischbare organische Lösungsmittel, die die Aufarbeitung der Umsetzungsgemisch besonders
einfach machen. Man führt das Verfahren nach der Erfindung beispielsweise aus, indem
man die Ausgangsstoffe unabhängig voneinander, aber gleichzeitig in fester oder
flüssiger Form oder in einem der erwähnten Lösungsmittel gelöst oder suspendiert
unter gutem Durchmischen in ein vorgelegtes Lösungsmittel einbringt und dabei dafür
sorgt, daß die Ausgangsstoffe stets in etwa stöchiometrischen Mengen zugeführt werden.
Danach hält man das Umsetzungsgemisch zweckmäßig noch eine Zeitlang bei der Reaktionstemperatur.
Die Umsetzung nimmt imallgemeinen eine halbe bis eineinhalb Stunden in Anspruch.
Da die Anthranilsäureamide nicht zu Folgereaktionen neigen, kann man sie unbesorgt
auch längere Zeit bei der Reaktionstemperatur belassen, was bei technischen Ansätzen
hin und wieder nötig ist. Die Anthranilsäureamide kristallisieren manchmal aus dem
Umsetzungsgemisch aus, gegebenenfalls nach Abdestillieren eines Teiles des Lösungsmittels.
Wenn man mit einem wasserlöslichen organischen Lösungsmittel arbeitet, kann man
das Reaktionsprodukt durch Verdünnen des Reaktionsgemisches mit Wasser abscheiden.
Die nach dem neuen Verfahren erhaltenen Anthranilsäureamide sind wertvolle Zwischenprodukte
für die Herstellung von Pharmazeutika und Farbstoffen. Zum Teil haben sie auch selbst
eine therapeutische Wirkung (vgl. deutsche Patentschrift 1 091 120). Die in den
folgenden Beispielen genannten Teile sind Gewichtsteile.
Sie verhalten
sich zu den Raumteilen wie Kilogramm zu Liter. Beispiel 1
Eine Lösung von
163 Teilen Isatosäureanhydrid in 350 Raumteilen Dimethylformamid und eine Lösung
von 65 Teilen n-Propylamin in 300 Raumteilen Wasser läßt man unter schnellem Rühren
gleichzeitig und mit gleicher Geschwindigkeit in 200 Raumteile auf 80C erwärmtes
Wasser unterhalb von 30 Minuten einfließen. Man rührt das Gemisch 15 Minuten nach
und läßt die Lösung in 1 000 Raumteilen Wasser einfließen. Dabei scheidet sich das
gebildete Anthranilsäurepropylamid in weißen, nadeligen Kristallen ab, die abfiltriert
und bei 500C getrocknet werden. Die Ausbeute beträgt 161 Teile, entsprechend 91
% der Theorie. Die Vergleichsversuche (letzte Spalte der Tabelle) wurden nach a 'er Arbeitsweise
nach Journal of Organic Chemistry durchgeführt.
Beispiel 2
In
300 Raumteile siedendes Methanol läßt man gleichzeitig und unabhängig voneinander
eine Lösung von 32,5 Teilen Isopropylamin in 400 Raumteilen Wasser und eine Suspension
von 81,5 Teilen Isatosäureanhydrid in 400 Raumteilen Wasser gleichzeitig und mit
gleicher Geschwindigkeit einfließen, was 30 Minuten in Anspruch nimmt. Man hält
das Gemisch weitere 15 Minuten bei 70 bis 800C, kühlt es dann auf Raumtemperatur
ab und fügt 500 Raumteile Wasser zu. Das abgeschiedene Anthranilsäureisopropylamid
wird abgesaugt und@bei 500C getrocknet. Die Ausbeute beträgt 79,5 Teile, entsprechend
89 % der Theorie. Fp. 148 bis 14900. Beispiel 3
In 800 Raumteile 50%iges wäßriges
Methanol trägt man innerhalb von 45 Minuten gleichzeitig 163 Teile festes Isatosäureanhydrid
und 65 Teile Isopropylamin ein. Die Temperatur beträgt 600C. Man hält die erhaltene
klare Lösung weitere 30 Minuten bei 70 bis 750C und kühlt sie dann ab. Das ausgefallene
Anthranilsäureisopropylamid wird abgesaugt und bei 500C getrocknet. Die Ausbeute
beträgt 160 Teile Anthranilsäureisopropylamid, entsprechend 90 % der Theorie. Fp.
