KAISERLICHESIMPERIAL
PATENTAMT.PATENT OFFICE.
B. Einhorn, Ber. d. deutschen ehem. Ges. 23, 2889, A. F. P. van Son, Nederl. Tijdschr. Pharm. It has already been tried in vain by various chemists (e.g. unicorn, Ber. d. German former Ges. 23, 2889, A. F. P. van Son, Nederl. Tijdschr. Pharm. 10, 270), tropidine with the aid of its hydrogen halide addition products to be converted into an alkamine, only Ladenburg has claimed (Ber. d. German former Ges. 23, 1780 and 2225) that he had obtained traces of tropine with the aid of the hydrobromide information in this regard has been refuted by Willstätter (Ber. d. German former Ges. 33, 1170 and 34, 143). Ladenburg states on page 1780, both in the cold, as well as having worked at a higher temperature, but he does not speak of heating to boiling, nor from heating under pressure, and generally abstains from any more precise Temperature indication. In the second communication he expressly says again, that he had prepared larger quantities with hydrobromic acid in the cold. It has to be So working in the cold finally gave him the best results. But these are still so inadequate that Ladenburg, although it is larger Quantum speaks, could only represent the platinum salt and examine it superficially.
;■Einhorn also received a base from tropidine hydrobromide with sodium acetate but which he only says that their platinum salt is at 200 °, the melting point of the tropine platinum double salt, melts. He expressly emphasizes that it is of course still more detailed Investigations are required to determine whether one is indeed dealing with a Tropine salt has to do. ; ■
In addition, Einhorn worked with the ß-tropidine hydrobromide, which only forms in traces and especially mentioned that the reaction with the α-tropidine bromohydrate, which is the main product, does not succeed.
However, it is now nevertheless able to transform the tropidine with considerable yield in pseudotropine, characterized that Halogenwasserstoffadditionsproduete DAF having hydrolytic reagents in closed Gefäfsen to a higher temperature are usually heated at 180 to 220 0 C.
The hydrogen halide addition products are also produced by heating tropidine with the acid in question in a closed vessel, e.g. B. with hydrogen iodide Heating 1 part by weight of tropidine with 6 parts by weight of concentrated hydriodic acid (spec. Weight 2.0) to 100 ° for 24 hours. This salt is very easily soluble in hot water, and very sparingly soluble and crystallized in cold water from the warm solution in water-free, colorless, strongly refractive, four-sided Panels with an approximately square outline, which are sharp at 197 ° C. with decomposition melt. The free tropidine hydroiodide is a water-soluble, colorless, heavy, highly refractive oil that decomposes when heated.
Am geeignetsten ist die Bromwasserstoffver-The hydrobromic acid hydrobromide was obtained by heating tropidine in an autoclave to 100 ° with 5 parts by weight of saturated glacial hydrobromic acid. From this compound the free hydrobromide was obtained as a oil which boils under 17 mm pressure at 109 0C. The most suitable is the hydrogen bromide
bindung, da die Jodverbindung nur geringe Ausbeuten liefert.bond, since the iodine compound gives only low yields.
As hydrolytic substances come into consideration: water, solutions of salts or Acids. With water alone or dilute hydrochloric acid z. B. is obtained by heating 220 ° C. the desired cleavage product. The yield is much lower than when dilute sulfuric acid is used, which leads to the most favorable results.
At the high temperature of 210 to 220 ° C., a large part of it occurs Material in the place of the halogen the hydroxyl group, according to the formulas:

On the other hand, if there is a considerable amount of the addition product, there is splitting off of Hydrogen halide takes place with the formation of tropidine, which is recovered. As a result of this, the pseudotropin can be removed from this volatile, oxygen-free base Separate low volatility with steam.
Zur Erläuterung des Processes dienen folgende Beispiele:The following examples serve to explain the process:
I. 1 kg of tropidine hydrobromide (base) is mixed with 6 kg of ioproc. Sulfuric acid heated for 4 hours in Autociayen at 200 ° to 210 0 C.. The cooled liquid is neutralized with caustic soda, and solid caustic soda is added until it is saturated. The oily mixture of bases is isolated by repeated extraction with ether and evaporation of the ethereal solution. To separate pseudotropin and tropidine, the halogen-free base obtained is diluted with ten times the amount of water and distilled with steam as long as the distillate has a strongly alkaline reaction. After neutralization with hydrochloric acid and evaporation, the distillate yields regenerated tropidine hydrochloride (on average 320 g).
On the other hand, the aqueous solution freed from tropidine with the aid of steam becomes processed on pseudotropin by having them excreted from a small amount The resin is filtered off, saturated with alkali, and often extracted with ether.
Die Ausbeute an dem reinen Product beträgt 120 g.The ethereal solution leaves behind raw pseudotropin as it evaporates. This alkamine is obtained by distillation and recrystallization, e.g. B. from ether or benzene Ligroinmischung, purified, and then displays the melting point of 108.5 ° C. and the boiling point of 240 0 C. (corr.). The yield of the pure product is 120 g.
■Example II. 1 kg of tropidine hydroiodide (base), freshly isolated from the hydroiodic acid salt, is mixed with 10 kg of 5 per cent. Hydrochloric acid is heated 2 hours at 185-190 0 C. in an autoclave. The cooled liquid contains the salts of tropidine and pseudotropin, but contaminated with another unsaturated by-product. It is possible in the same way as with the use of hydrobromide to obtain the pseudotropin in a pure state. The yield is about 80 g of alkamine; the regenerated tropidine is recovered almost quantitatively. ■
(D. i. ca. 120 g Alkamin.)Example III. 1 kg of tropidine hydrobromide is combined with 10 kg of 20 proc. aqueous solution of primary potassium sulphate (KHS OJ heated for 6 hours in an autoclave at 220 0th Work-up of Reactionsproductes done exactly the same as when using dilute mineral acid and the yields of pseudotropine and regenerirtem tropidine soft only slightly from the listed top. (D. i.approx. 120 g alkamine.)
Example IV. 1 kg of tropidine hydrobromide is combined with 20 kg of a 30proc. water Chlorammonium solution heated to 240 to 250 ° C in an autoclave for 6 hours. For isolation, the cooled liquid is acidified with hydrochloric acid and evaporated to dryness and separates the hydrochloric acid of the organic bases from the ammonia by extraction with alcohol. The purification of the pseudotropin from tropidine takes place as indicated above. The yield in this case is considerably less favorable.
