Description
Process for the preparation of an acid-fast digestive enzyme preparation The invention relates to a method for producing an acid-resistant digestive enzyme preparation with proteolytic and amylolytic effects through; cultivation of Aspergillus oryzae on wheat bran in an acidic environment.
The inventive method is characterized in that one breeds the Aspergillus oryzae (TPR-18) JAM 2800 strain on acidified wheat bran, the enzymes formed are extracted from the fermentation mass with water, the enzymes stabilized by the addition of zinc chloride or cobalt chloride, the extract narrows and the enzyme complex precipitates with isopropanol.
According to the invention, acid-fast digestive enzymes are thus obtained, the protein and starch in the stomach without the use of acid neutralizing agents digest.
So far, pancreatin and malt diastasis are commonly used as digestive remedies been administered. Pancreastrypsin and amylase are in. Neutral and weakly alkaline Media very effective. However, these effects are brought about by touching Destroyed for the most part with gastric acid. A reactivation of these enzymes does not enter the intestinal tract (K. O k a z a k i: Yakuxaigaku (Japan), vol. 16, P.15 [1957], Chemical Abstracts, vol.51, p.9738 [1957]). Malt diastasis is opposite more sensitive to this acid. This is why these enzymes work together with an acid neutralizing agents, such as sodium bicarbonate.
Although by some doctors many beneficial properties of the acid Neutralizing agents are recognized as side effects of these Means, such as heartburn, alkalosis, constipation, diarrhea, suppression of the bactericidal Effect of the stomach, etc., among other things, has been discussed. (C. F u c h s: Drug and Cosmetic Industry, June, p. 692 [1949]). The acid neutralizing agent is for not suitable for those patients suffering from high blood pressure, kidney disorders etc. and who are therefore not allowed to consume the sodium salt. From this point of view from studies for the purpose of producing acid-fast proteolytic ones and amylolytic enzymes.
The first step in this research was to examine fungi to determine which of them are naturally capable of producing acid-fast digestive enzymes. Aspergillus mushrooms have long been used to make saki, shochu (Japanese distilled wine), miso (bean paste) and shoyu (soya) in Japan. Among them, a number of strains have been known which produce the highly proteolytic or amylolytic enzymes. As a remedy, the Aspergillus oryzae class of mushrooms has been used in the manufacture of digestive preparations since Dr. T akamine produced the "takadiastase". It has been found that the optimal pH for these enzymes is in a pH range of 6-8. Thus, the acid-fast enzymes which can be prepared according to the invention cannot be obtained from these known fungi. In order to isolate the fungi which produce the acid-fast enzymes, wild fungi were cultured at a temperature of 30 ° C. on a koji agar culture medium at an adjusted pH of 2 to 3 and then a pure Aspergillus genus was mainly grown from this medium . From these strains, a new strain was obtained which was satisfactory for the purposes of the invention, as shown by the tests for its amylolytic and proteolytic activities.
Nach dem Klassifizierungsverfahren von T h o m und R a p e r und dem Klassifizierungsverfahren von Sakaguchi-Yamada(K.Sakaguchiund Y a m a d a: J. of Agri. Chem. Soc. According to the classification procedure of T h o m and R a p e r and the Classification method of Sakaguchi-Yamada (K.Sakaguchi and Y a m a d a: J. of Agri. Chem. Soc. [in Japan], vol. 20, Pp. 65, 141 [1944]) shows the new strain as a modification of the Aspergillus oryzae.
Inst. Sci. and Ind. Research, Osaka University, Bd. 9, S. 194 [1952]; Chem.- Abst.; Bd. 47, S.10568 [1953]; H. O k a s a k i: Symposia an enzyme Chem. [in Japan], Bd. 9, S. 43 [1954]; K. K i t a h a r a: J. Fermentation Technology, Bd. 27, S. 254 [1949]) und daß jeder Pilz des Aspergillus oryzae, Aspergillus usamii, Aspergillus awamori, Rhizopus tonkinensis und Rhizopus peka das Enzym bildet, das an die drei verschiedenen Aktivitäten der Amylase,_ der Amyloglucosidase und der Transglucosidase aufweist, und der Stärke verzuckernde Typus jedes Pilzes in Abhängigkeit von dem anteilmäßigen Verhältnis der obigen drei Enzyme schwankt. It was also found that the enzyme from the strain many interesting shows enzymological properties. It has been shown in the past few years that amylase obtained from mushrooms contains the following substances: amylase, amyloglucosidase and transglucosidase (J. Ni k u n i: Mein. Inst. Sci. and Ind. Research, Osaka University, Vol. 9, p. 194 [1952]; Chem. Vol. 47, p. 10568 [1953]; H. O k a s a k i: Symposia an enzyme Chem. [in Japan], Vol. 9, p. 43 [1954]; K. K i t a h a r a: J. Fermentation Technology, Vol. 27, p. 254 [1949]) and that every fungus des Aspergillus oryzae, Aspergillus usamii, Aspergillus awamori, Rhizopus tonkinensis and Rhizopus peka forms the enzyme involved in the three different activities of the Amylase, _ the amyloglucosidase and the transglucosidase, and the starch saccharifying type of each mushroom depending on the proportion of the above three enzymes varies.
O k a z a k i (H. O k a z a k i: J. Agr. Chem. §0C. [in Japan], Bd. 24, S. 88 [1950]; Chem. Abstr., Bd. 45, S. 9115 [1951]) berichtete, daß fünf Verzuckerungstypen in Pilzen beobachtet wurden, und gemäß einem weiteren Bericht von ihm (H. O k a z a k i: Arch. Biochem. and Biophys., Bd. 63, S. 322 [1956]; Chem. Chem. §0C. [In Japan], Vol. 24, p. 88 [1950]; Chem. Abstr., Vol. 45, p. 9115 [1951]) reported that five types of saccharification observed in mushrooms, and according to a further report by him (H. O k a z a k i: Arch. Biochem. and Biophys., Vol. 63, p. 322 [1956]; Chem. Abstr., Vol. 50, S.15652 [1956]) Aspergillus oryzae forms large amounts of amylase, so that the saccharification rate increases in the first stage and does not increase with time. In general was - the amylolytic effect of the Aspergillus oryzae enzyme in a medium with a pH value below 3 bad, because the amyloglucosidase is acid-fast and the amylase is not acid-proof. It was found that the new strain isolated from nature Morphologically belongs to Aspergillus oryzae, but a saccharifying one in a neutral medium Behavior shows that the person in F i g. 1 behavior of the Aspergillus awamori type shown is similar, and in acidic medium the enzyme digests starch better than ordinary Aspergillus oryzae enzyme as shown in FIG. 2 shown does.
Im Jahre 1954 berichtete Y o s h i d a (F. Y o s h i -d a: J. Agr. Chem. Soc. In 1954, Y o s h i d a (F. Y o s h i-d a: J. Agr. Chem. Soc. Japan, Vol. 28, p. 66 [1954]) that the optimal pH value for the effect the protease near pH 6.5 with respect to yellowish green Aspergillus such as Aspergillus oryzae, and near pH 2.5 for blackish Aspergillus, such as Aspergillus niger, based on the determination of the proteolytic effects of various Aspergillus enzymes obtained from wheat bran koji extracts In 1955, Kageyama (K. Kageyama: J. Fermentation Technology, Vol. 33, p. 109 [1955]; Chern. Abstr., Vol. 49, p. 13589 [1955]) that strains of Aspergillus oryzae mainly form acid-active protease from Reiskoji, but form the same Also from wheat brankoji strains alkaline-active protease.
As explained above, the stems are made from wheat brankoji the Aspergillus oryzae group mainly alkaline-active protease and the Aspergillus niger group acid active protease. However, the new fact was found that the enzyme of the strain isolated according to the invention in addition to strongly acid-active protease contains protease active in neutral medium, as shown in FIGS. 3 and 4 shown. By the facts listed above, namely the in-depth investigations regarding the classification based on morphological studies regarding the characteristic Behavior in enzyme activity, the novel strain was identified as new Tribe found under the group Aspergillus oryzae and the same the name Aspergillus oryzae TPR-18 given. This Aspergillus oryzae TPR-18 is in the Applied Microorganism Institute of Tokyo University under the designation JAM 2800.
The second stage in the investigations carried out consisted of determine the optimal conditions of the medium and the culture process the Aspergillus oryzae (TPR-18) JAM 2800 can be made to grow sufficiently so as to obtain a sufficient amount of the acid-fast enzyme. It was each wheat bran, oil cake, soybean cake and copra flour for the culture medium and wheat bran was found to be the most suitable for this purpose Substance is; it was further found that the yield of acid-active Protease is increased by adding a small amount of acid to the culture medium.
The culture medium used was initially suitable Way with 0.2 n-HCl, corresponding to 20 to 50 percent by weight of the wheat bran, moistened and then sterilized at 100 ° C for 1 to 2 hours. Left after sterilization to cool, and the nutrient medium was then inoculated with fungal spores. They left the Grow culture at 30 ° C for 40 to 50 hours.
At the third stage of the investigations carried out, the procedure was for the production of the acid-resistant produced by Aspergillus oryzae (TPR-18) JAM 2800 Enzyme examined with regard to the cost factor and the expediency.
In the method according to the invention, a stabilization of the Enzyme solution caused by the addition of zinc chloride and cobalt chloride. It came that the following methods are used: That in a sufficient amount Aspergillus oryzae (TPR-18) JAM 2800 formed culture medium was extracted with water and then the extract to which zinc »chloride or cobalt chloride with a concentration of 10-3 moles had been added, adjusted to a pH range of 5 to 6 and then concentrated to half its volume under reduced pressure at 30 ° C. In the above procedure, the enzyme was not destroyed when the extracts were concentrated, and the reduced volume of the extract solution caused the subsequent Procedure an expediency both in terms of procedure and with regard to the Consumption of reagents.
The next step in the process was that of the narrowed Extract solution to precipitate the enzyme present. This was using in place of ethanol 11/2 times the volume of isopropanol and the enzyme after centrifugation dried under reduced pressure. This product was called Wernasea.
The following is a description of the digestive ability of the acid-fast Digestive enzyme (Veraase) described that according to the method according to the invention has been made.
The enzyme made a powder slightly gray or brownish-white Color, was almost odorless, possessed a characteristic taste, was in Soluble in water and in almost all organic solvents such as alcohols, ethers, Acetones, insoluble.
The pH-activity curve of protease and amylase at 40'C and their Dependence on the enzyme concentration, the reaction time at different pH and at the optimal pH of an acid-stable digestion enzyme, Vernase and Aspergillus saitoi enzyme compared.
The F i g. 5, 6 and 7 illustrate the paramount enzymatic Vernase activity.
(a) Protease-Aktivität bei verschiedenen pH-Werten Vernase wirkt bei einem pH-Wert von 3 bis 6 optimal und hat im Vergleich zum Aspergillus-saitoi-Enzym bei einem pH-Wert von 3 die gleiche, bei einem pH-Wert von 6 jedoch eine zwei- bis dreimal höhere Aktivität.(a) Protease activity at different pH values Vernase is involved a pH value of 3 to 6 and has in comparison to the Aspergillus saitoi enzyme at a pH value of 3 the same, at a pH value of 6 a two to one three times higher activity.
Vernase from Aspergillus oryzae (TPR-18) JAM 2800 has two activity peaks with regard to the proteolytic effect at pH 3 and pH 6 and lies with his Values with increasing pH above the values of the other curves.
On the other hand, the Aspergillus saitoi enzyme has only one peak of activity at a value of pH 3 and also shows the disadvantageous for a digestion enzyme Tendency to decrease rapidly in its activity at pH values above pH 3 (Fig. 5).
(b) Amylaseaktivität bei verschiedenen pH-Werten F i g. 8 und die untenstehende Tabelle dienen zur Erläuterung der Einheit-pro-Gramm-Werte von Vernase und Aspergillus-saitoi-Enzym bei pH-Werten von 3, 5 und B.(b) Amylase activity at different pH values F i g. 8 and the The table below is used to explain Vernase's unit-per-gram values and Aspergillus saitoi enzyme at pH values of 3, 5 and B.
It is well known that an enzyme has a specific effect against it having a specific substrate. Because the differences in specificities are the activities and change enzymatic action, therefore, the applicability of an enzyme is due requires a specificity.
I)).Vernase protease shows a lower disintegration effect than that of Aspergillus saitoi with an enzyme content of 0.010 / "at a pH of 3 (Fig. 6 (I)).
However, Vernase shows a higher disintegration effect than Aspergillus saitoi enzyme at all other concentrations with the exception of 0.010 /,.
As shown in FIG. 6 and 7, the reaction mode and the Specificity superior to the Aspergillus saitoi enzyme.
. . . . . ... . . . . . . . . 1,2 °/o und 2,1 °/o Milch-Kasein Enzymkonzentration . . . . . 0;01, 0,05 und 0,1 °/o pH-Wert . . . . . . . : : : . . -. .-. It is expected that Vernase will break down and react numerous peptides and carbohydrates and have a higher affinity for the peptides and carbohydrates than the other enzyme. To the solution, 6.2 g of cobalt chloride was added and added with acetic acid or sodium acetate adjusted to a pH range of 5 to 6 and then concentrated to 141 under reduced pressure at a temperature of 30 ° C.
Die Ausbeute an Enzym betrug 405 g.The concentrated solution was cooled to 0 ° C. in the refrigerator. 211 99% Isopropyl alcohol that had previously been cooled slowly became the enzyme solution poured. The precipitate formed was centrifuged off and treated with 11 isopropyl alcohol washed in the mixer, separated and dried in a vacuum dryer. The yield of enzyme was 405 g.
