The benzimidazole derivatives prepared according to the process have coronary vasodilator activity. From the group specified by the general formula reproduced new coronary vasodilator compounds protrude, in particular the 2-substituted 1- (3 ', 4'-diethoxybenzyl) -5,6-diethoxybenzimidazoles because they are characterized by lasting effectiveness.
Die coronargefäßerweiternden Substanzen dienen dazu, die Stoffwechselbedingungen im Herzmuskel zu verbessern; ein Maß für den Nutzeffekt solcher Substanzen ist die Zunahme der SauerstpffsättigungThe coronary vasodilator substances serve to improve metabolic conditions in the heart muscle; a measure of the efficiency of such Substances is the increase in oxygen saturation

des Blutes im Sinus 00Γ0Μπμ5. Bei geringer oder fehlender Zunahme der Sauerstoffsättigung bewirkt nämlich die alleinige Coronargefäßerweiterung keine Verbesserung der Sauerstoffversorgung und der Stoffwechselbedingungen des Herzens.of the blood in the sinus 00Γ0Μπμ5. With less or If there is no increase in oxygen saturation, coronary vasodilation alone does not cause any Improvement of the oxygen supply and the metabolic conditions of the heart.
The coronary effect was demonstrated in a circulatory test on dogs under anesthesia that is bromoalkyl-sec.-butyl-barbituric acid determined with the thorax closed. The preparations were administered intravenously into the saphenous vein applied. The increase in Sauer- <o Substance saturation of the coronary sinus blood compared to the oxygen saturation in the. Femoral artery blood; at the same time the blood pressure was recorded. The experimental set-up used to measure oxygen saturation is the publication of H.J. '5 B etschnei der and coworkers in the »Negotiations of the German Society for Circulatory Research ", 22nd meeting (1956), pp. 300 to 305, taken. The oxygen saturation of the hemoglobin in the coronary sinus was measured with the aid of a photocell cell measured photometrically, as in "Oximetry" by Kurt K r a m e r, Thieme-Verlag, Stuttgart, 1960, pp. 1 to 10, is described.
To be sure that the coronary effects of the new benzimidazoles are not due to metabolic inhibitors Properties is based on the arteriovenous lactic acid difference of the heart after the dog Administration of the long-acting l- (3 ', 4'-diethoxybenzyl) -2-diethylaminomethyl - 5,6 - diethoxybenzimidazole hydrochloride. It was found that the increase in oxygen saturation in myocardial blood is actually a result of the improved blood flow to the heart muscles.
Some already known benzimidazoles also show coronary vasodilator effects; however, they are therapeutically useless. For example, with 2-benzylbenzimidazole (Walther v. Pulawski, "Journal for practical chemistry", 2nd part, vol. 59 [1899], pp. 235 to 255) only at 5 mg / kg in dog experiments notice a short-term coronary dilation (increase in oxygen saturation 18%), which but at the same time is accompanied by a sharp decrease in blood pressure (by 30 mm Hg).
The 1-benzyl-1-dimethylaminomethylbenzimidazole known from German patent 890 644 also gives only a weak coronary effect at 1 mg / kg in dog experiments (increase in oxygen saturation in the coronary sinus 9% for I 1 Z 2 minutes).
B. aus B. H e 1 w i g, »Moderne Arzneimittel«, 1961, 2. Auflage, S. 767 und 768, bekannt sind, herangezogen und den oben beschriebenen Untersuchungsmethoden unterworfen.2-Ethyl-3- (3 ', 5'-diiodo-4'-hydroxybenzoyl) -benzofuran were used for further comparative tests (XXXI) and N- [3'-phenylpropyl- (2 ')] -1, l-diphenylpropyl- (3) -amine (XXXII), e.g. B. from B. H e 1 w i g, "Moderne Arzneimittel", 1961, 2nd edition, pp. 767 and 768, are known, used and the above are subjected to the investigation methods described.
Compound XXXI has little effect on blood pressure (slight decrease), but it is also only very weakly effective as a coronary expander. Its effect on myocardial blood flow is only for a short time measurable (oxygen saturation in the coronary sinus). It only shows when 5 mg / kg are applied in dog experiments an increase in oxygen saturation of 8% for 2 minutes.
Compound XXXII has better myocardial benefit compared to XXXI. Your activity But it also subsides quickly and is accompanied by a sharp drop in blood pressure, which is unfavorable significantly longer than the increase in oxygen saturation in the coronary sinus continues. So done at a dose of 1 mg / kg in a dog experiment an increase in oxygen saturation of 15% for 4 minutes and a 44% blood pressure drop for over 12 minutes. As a result of toxicity of 18 mg / kg (Mouse i.v.) the therapeutic range is also not very large.
The following table gives the effects of the benzimidazoles prepared according to the process, the compounds XXXI and XXXII again; it shows that the compounds clearly match the comparison substances are superior.

Die Maximalwirkung des bekannten 3,5-Dioxo-l ,2-diphenyl-4-n-butylpyrazolidins beträgt in diesem Test —69%.Furthermore, individual representatives of the new group of compounds also show a good anti-inflammatory effect Activity, such as B. 1-p -chlorobenzyl-5,6 - diethoxybenzimidazole hydrochloride (X) and 1 - (3 ', 4' - dimethoxybenzyl) - 2 - dimethylaminomethyl-5,6-dimethoxybenzimidazole hydrochloride (XV). The compound XV results in the kaolin edema test on the rat paw with 2 × 60 mg and 2 × 120 mg / kg i. p. a dose-dependent anti-inflammatory effect of a maximum of -67%. The maximum effect of the known 3,5-Dioxo-1,2-diphenyl-4-n-butylpyrazolidine is -69% in this test.
So betragen z. B. diese Werte fürIn addition, the compounds produced according to the process also have sedative properties to. This sedative component should be emphasized because the coronary vasodilators known so far do not have this quality, but on the contrary often show a central stimulating effect. One However, the pathogen effect is completely undesirable in the treatment of heart diseases such as angina pectoris. Also, the compounds produced according to the method are well absorbed, what from the Comparison of intravenous and oral toxicity can be seen. The smaller the difference this Toxicity values, the better the substances in question are absorbed. For example B. these values for

While stirring and cooling with ice water with exclusion of moisture, a solution of 8.4 g of diethanolamine in 20 cm 3 of chloroform was gradually added. The batch is left to stand overnight at room temperature and then heated under reflux for 3 hours. After cooling, the batch is washed with water and evaporated the chloroform phase in vacuo at 4O 0 C maximum. The residue is then taken up in benzene while warming, cooled to room temperature and the benzene solution is washed once with a little water. The benzene phase is then extracted with dilute hydrochloric acid. The acidic, aqueous extract is washed with ether, treated with decolorizing charcoal and made alkaline with soda solution. The l- (3 ', 4'-diethoxybenzyl) -2-diethanolaminomethyl-5,6-diethoxybenzimidazole is taken up in benzene and dried over potassium carbonate. The benzene is evaporated off in vacuo; the base which soon crystallizes remains behind. It has a melting point of 85 to 89 ° C after drying in a vacuum desiccator. The monohydrochloride is treated with decolorizing charcoal in an aqueous solution and then evaporated in vacuo. After recrystallization from methanol-ether and drying in vacuo at 60 ° C, it has a melting point of 171 to 172 ° C; Yield about 30% of theory.
