mittels Borhydriden der Erdalkalien und des Magnesiums reduziert.reduced by means of borohydrides of alkaline earths and magnesium.
Diese Verbindungen können der Normal-Reihe (A: B = as) oder der Allo-Reilie (A: B = trans) gehören.These compounds can belong to the normal series (A: B = as) or the allo series (A: B = trans).
It is known that the reduction of the 3-keto steroids can be carried out using sodium borohydride (O. Mancera et al., Journ. Amer. Chem. Soc, 75, 1953, p. 1286). . However, the yield is low and the product obtained still has to be purified by laborious chromatography. In the case of reduction using sodium borohydride, the 20-position keto group is also partially reduced (EP Oliveto and E. W. Hershberg, Journ. Amer. Chem. Soc., 75, 1953, p. 488). Thus, the 3a-Oxypregnan-ll, 20-dione by reduction of pregnan-3,11,20-trione only after chromatography treatment using alumina in a conversion of 53/0 ° obtained. If one pregnane-3,20-dione is reduced, it Amer Chem passes without chromatographic purification of the reduction product in a yield of 40 ° / 0-3ct 0xypregnan-20-one (0. Mancera et al., Journ... Soc , 75, 1953, p. 1286). According to the USA. Patent 2,647,134 can reduce the 1 la-oxy-3,20-pregnanedione using sodium borohydride in a yield of 40 ° / 0th
So beträgt z. The reduction can be carried out selectively and with high yield by means of alkaline earth or magnesium borohydride. A pure product can be obtained from the crude product by recrystallization once, so that chromatographic work-up is unnecessary. So z. B. the yield in the reduction of the pregnane-3,20-dione using CaBH 4 75 to 80%. The reduction is expediently carried out in alcohol or an alcohol-containing solvent. The reduction can also be carried out advantageously in other solvents, e.g. B. in tetrahydrofuran. The temperature used in the reduction depends on the stability of the alkaline earth or magnesium borohydride in the particular solvent used. If calcium borohydride is used in ethyl alcohol, temperatures between -10 and -40 ° C. are advantageously used. B. cortisone, further processed in a known manner.
Die Lösung wird im Vakuum zur Trockne verdampft, der kristalline Rest mit Wasser verrührt, abgenutscht, dann7.5 g of pregnane-3,20-dione (melting point 120 to 122 ° C.) are reduced by means of calcium borohydride, which is prepared as follows: 0.9 g of CaCl 2 · 2 H 2 O are dissolved in 500 ml of absolute alcohol -2O 0 C cooled and mixed with 0.4 g of sodium borohydride. To this solution is the pregnane - 3,20-dione added and the reaction mixture stirred at -2O 0 C for 3 hours; Thereafter, the excess borohydride is decomposed by 10 ml of 35 ° / 0 acetic acid at this temperature. The solution is evaporated to dryness in vacuo, the crystalline residue is stirred with water, suction filtered, then

The mixture is stirred for 7 hours at -25 0 C, and then decomposed using 50 ml of 20% acetic acid and evaporated in "vacuo to dryness, the crystalline residue is mixed with water, filtered off under suction and washed with 1-η hydrochloric acid, the dried product is Recrystallized from 50 ml of benzene: Pregnan-3ct-ol-20-one is obtained in good yield, melting point 145 to 146 ° C; [a] D = + 112 "in 1% alcohol solution.
