The The invention relates to a flow measuring cuvette for transmission spectroscopic Examination of biological fluids in the mid-infrared spectral region (MIR) and a transmission spectrometer.

Bei den eingangs erwähnten bekannten Durchfluß-Meßküvetten wird dies dadurch erreicht, daß zwischen den beiden Fensterflächen der Küvette ein Abstandhalter einer entsprechend geringen Dicke vorgesehen ist.biological Liquids, such as blood, blood plasma, blood serum, hemolysate, Cerebrospinal fluid, urine, saliva, sperm, lymph fluid, synovial fluid, Amniotic fluid, tear fluid, Cyst fluid, sweat secretion or bile, are always aqueous solutions and Therefore, in the MIR range, the radiation of a wavelength of 2 to 20 μm includes, absorption coefficients with very large values. This leads to, that, for example at one wavelength of 10 μm 99.99% of a radiated infrared light intensity on one Transmission distance of 100 μm be absorbed. This results in the need to examine the biological fluid in the measuring cuvette in to arrange a layer thickness of only about 10 to 100 microns. At the beginning mentioned known flow measuring cuvettes is this is achieved in that between the two windows the cuvette Spacer is provided a correspondingly small thickness.
The with the study of native samples of biological liquids Problems associated with ME are discussed in WO 02/057753 A2. There, a transmission distance of less than 30 microns is required. However, no information is given as to how they are achieved can.
One special problem results from the fact that for filling a volume of formed two opposite with very small distance plates is due to the fluid dynamic laws a considerable pressure needed up to 50 bar becomes. Such a high pressure causes the walls of the Deform flow measuring cuvette and thereby the length the transmission distance between the two window areas changes. This leads to an unwanted change of the signal that is not spectroscopically or very difficult of to distinguish a concentration-related change in absorption is.
The invention is therefore to show a way as in a MIR transmission spectroscopic examination of a biological fluid the measuring accuracy increases can be and in particular influences of the initiation of the examined liquid required in a flow-through cuvette filling pressure, can be reduced to the spectroscopic measurements.
The The object is achieved by a flow-through cuvette with the characteristics of Claim 1 solved.
A flow-through cuvette according to the invention goes to avoid unwanted Influences of the filling pressure on the measuring signal not the way, by a particularly robust execution of the cuvette one pressure-related curvature to counteract the window, but reduces the initiation a biological fluid required filling pressure. In a flow measuring cuvette according to the invention the flow resistance of the flow channel almost independent from the length of the Transmission route. The flow channel can therefore be optimized in terms of its flow characteristics become. This not only greatly reduces the filling pressure, but also also the cleaning ability a flow measuring cell according to the invention advantageously improved.
Of the in the flow channel protruding light channel projection is of the liquid to be examined lapped on the side, so that one low total flow resistance of the flow channel is achieved, which makes a high inflation pressure superfluous. At the same time, the liquid to be examined lies on the transmission path between the free end of the light channel projection and the opposite Window in a sufficiently thin layer thickness in front. In particular, in a flow measuring cuvette according to the invention by the distance the window provided at the free end of the light channel projection to the opposite Window predetermined transmission distance in the range of 10 to 100 microns free according to Requirements of the fluid to be examined are selected, without that the flow resistance of the flow channel being affected.
Bevorzugt beträgt das Gesamtvolumen des Strömungskanals höchstens 1 ml, besonders bevorzugt höchstens 0,7 ml.Preferably, the flow channel at its narrowest point in each direction has a width which is at least five times, preferably at least ten times the transmission distance. In this way it is achieved that the liquid to be examined laterally of the light channel projection finds sufficient space to flow past with low filling pressure can. Particularly favorable flow conditions arise when the flow channel at its narrowest point - in particular the side of the transmission path - in each direction has a width of at least 0.1 mm, preferably 0.5 mm. An upper limit for the narrowest point of the flow channel does not exist under fluidic aspects, but increases with increasing cross-section of the flow channel and the chamber volume and thus required for an investigation amount of biological fluid. Preferably, the total volume of the flow channel is at most 1 ml, more preferably at most 0.7 ml.
Of the Light channel projection contains a light channel through which the radiated from a light emitter into the cuvette primary light up to the window located at its free end or after the Passing the transmission path resulting secondary light of the window at its free end towards one Detector is passed. The term "window" should not be understood as limiting in the sense may be that one Window must be present. Rather, the transparent ones Solid surfaces, the limit the transmission path on both sides, referred to as windows.
For the MIR area Suitable light guides can be produced, for example, as silver halide fibers. So that these of the to be examined aqueous biological fluids Not to be attacked, it is recommended to use the optical fiber on its peripheral surface through a suitable envelope, for example, a water resistant Protect paint. The free end of the fiber forming the window can pass through to be protected by a MIR-transparent coating.
A advantageous development of the measuring cuvette provides that the distance changeable between the two windows of the window pair is. This is preferably achieved in that the wall, the chamber encloses a first and a second wall element. On the wall element each one of the two windows is provided. The two wall elements are about Connected sealing means which are so flexible and / or elastic are that the first Wall element is movable relative to the second wall element and so the distance between the two windows can be changed.
Die Untersuchung einer biologischen Flüssigkeit in zwei Durchfluß-Meßküvetten mit unterschiedlichen Transmissionsstrecken bedeutet nicht nur einen erheblichen Aufwand, sondern auch, daß die gewonnenen Spektren, z.B. wegen unvermeidlicher Drifteffekte des Spektrometers, nur eingeschränkt miteinander vergleichbar sind.at the MIR transmission spectroscopic examination biological Liquids come it often before that the optical absorption in parts of the spectrum is very strong, while other areas have low absorption. For the signal-to-noise ratio are with strong absorption shorter Transmission distances and with less absorption longer transmission distances advantageous. By varying the distance of the window of a flow measuring cuvette according to the invention, the Transmissionsstrecke adapted to the occurring absorption structures and thereby the signal-to-noise ratio be optimized. It is advantageous not only an improved accuracy, but also, that yourself at the investigated Spetralbereich strongly changing absorption the sample must be introduced only once in a flow measuring cuvette and not about two or even three flow cuvettes with Different transmission distances must be prepared. The Examination of a biological fluid in two flow measuring cuvettes with different transmission distances does not just mean one considerable effort, but also that the acquired spectra, e.g. due to unavoidable drift effects of the spectrometer, only to a limited extent are comparable.
When Seal between the two wall elements is suitable, for example an elastic plastic which has a relative mobility allows the two wall elements to each other. As for a MIR transmission spectroscopic Analysis of a biological fluid As a rule, no transmission links outside of a range of 10 to 100 μm needed be enough a relatively low mobility of the two wall elements to each other, as easily ensured by elastomeric sealing materials can be. Preferably, the movable seal by means of a Bellows running, since this is u.a. offers the advantage, with a low force change the transmission path to be able to. This can be done for example with a servomotor.
For the examination of biological fluids, a method for transmission spectroscopic analysis is suitable in which radiation emitted by a light source is passed through a flow measuring cuvette in which the fluid is passed between two transparent windows through whose distance a transmission path is predetermined, wherein a portion of the light is absorbed by the liquid, and detects the resulting after passing through the transmission path intensity of the light passed through the sample and from a spectrum of the liquid is determined, which is characterized in that the length of the transmission path is changed in dependence on the detected intensity. This method allows a biological fluid over the entire spectral range examined with an optimal signal-to-noise ratio and thus high accuracy of measurement to investigate. If it is determined during a measurement that the absorption is so low or so high that the resulting increase or decrease of the detected secondary light intensity leads to a deterioration of the signal-to-noise ratio, the transmission distance can be increased or decreased immediately and thus the signal Improve noise ratio.
One corresponding transmission spectrometer has a flow measuring cell with two variable spacing Windows that specify a transmission path, and a motor for changing the Distance between the two windows. For evaluation by a detector detected intensity an evaluation and control unit is provided by pressing the Motors the distance between the two windows and thus the length of the transmission path automatically depending on detected Intensity changes.

The Adjustability of length the transmission path d leads in certain circumstances to that the absolute Length of Transmission line initially not is readily known. The absolute length of the transmission path d can, however, based on the law of absorption, according to which the light intensity with increasing Transmission distance d exponentially declines, calculated with little effort become. For this the must Absorption at at least two different transmission path lengths d, measured d + Δd which differ by a known distance Δd, (one step). The length or a change in length the transmission path d can also be independent of a transmission signal be determined, in particular by an interferometric measurement. For example, visible laser light, such as from a HeNe laser, blasted through the measuring cuvette and tested for interference effects become.
