Das allgemein anerkannte Mehrschrittmodell der Krebsentstehung geht davon aus, daß durch Anhäufung von mehreren Mutationen in einer einzelnen Zelle diese so in ihrem Proliferations- und Differenzierungsverhalten geändert wird, daß letztendlich über benigne Zwischenstufen ein maligner Zustand mit Metastasierung erreicht wird. Hinter dem Begriff Krebs oder Tumor verbirgt sich ein Krankheitsbild mit mehr als 200 verschiedenen Einzelerkrankungen. Tumorerkrankungen können gutartig oder bösartig verlaufen. In Tabelle 2 ist die zytotoxische bzw. wachstumshemmende Aktivität der beschriebenen Verbindung mit/ohne Expression von p27kip1 gezeigt. Die getestete Verbindung zeigte im induzierten Zustand von p27kip1 keine zytotoxische Aktivität. Die Substanzen in Kulturmedium werden dann zu den Zellen gegeben und für 45h im Begasungsbrutschrank inkubiert. Als Kontrolle dienen Zellen, die nicht mit Testsubstanz behandelt werden. Für das XTT-Assay werden 1 mg/ml XTT (Natrium 3'-(1-(phenylaminocarbonyl)-3,4-tetrazolium]-bis(4-methoxy-6-nitro)benzensulfonsäure) in RPMI-1640 Medium ohne Phenolrot gelöst.
