Stand der TechnikState of the art
Einsatz definierter RNA exprimiert und in Aufreinigungsschritten angereichert werden.method for the efficient expression of defined proteins in a wide variety of Pro and eukaryotic organisms are known and require no further explanation. One significant problem in the expression of defined proteins in pro- and eukaryotic organisms and in cell-free expression lies in the purification and / or detection of the expressed defined Proteins. This is particularly problematic for defined proteins, for the there are no antisera or monoclonal antibodies. For simplification the purification and detection of such defined proteins these are expressed as so-called fusion proteins. in this connection the defined protein is N- and / or C-terminal an amino acid sequence added or between two protein domains inserted (internally), the fusion partner. This happens at the nucleic acid level, so that both the defined protein as well as the one for detection and / or purification attached During the merger a transcription / translation process together as a chimeric (fusion) protein consisting of the defined protein and the Fusion partner. This may be the attached fusion partner around individual amino acids, but also to act on peptides or proteins. For these attached fusion partners are available for purification immobilized binding partner, with where the fusion proteins can be isolated. Besides the possibility the purification of proteins can this also with for the fusion partner specific binding partner can be detected. These binding partners can for the Fusion partner specific antibodies or other proteins, peptides or chemical compounds, which specifically bind to the fusion partner.
Beispiele für solche Fusionspartner sind der Myc-tag (Munro & Pelham (1986) Cell 46, 291–300; Ward et al. (1998) Nature 341, 544–546), das Flag Peptid (Hopp et al. (1988) Bio/Technology 6 1204–1210), das KT3 Epitop Peptid (Martinet et al. (1990) Cell 63, 843–849; Martin et al. (1992) Science 255, 192–194) und das alpha-tubulin Epitop Peptid (Skinner et al. (1991) J. Biol. Chem. Examples for such Fusion partners are the Myc-tag (Munro & Pelham (1986) Cell 46, 291-300; Ward et al. (1998) Nature 341, 544-546), the flag peptide (Hopp et al. (1988) Bio / Technology 6 1204-1210), the KT3 epitope peptide (Martinet et al. (1990) Cell 63, 843-849; Martin et al. (1992) Science 255, 192-194) and the alpha-tubulin epitope peptide (Skinner et al., (1991) J. Biol. Chem. 266, 14163-14166), all successful for the detection and partly also for the purification of proteins were used. It could for some of the fusion partners, which are usually 3 to 12 amino acids long are shown do not affect the biological function of the defined proteins. The biological function of the defined protein is, however, with increasing length of the merger partner, since the addition expressed amino acids z. B. the formation of the secondary, tertiary and / or quaternary structure can influence. longer Fusion partners are therefore used to detect the proteins but less so suitable for the purification of proteins. On the other side longer Merger partners often higher affinity with their specific binding partners.
A significant disadvantage of the abovementioned fusion partners in the purification is that the binding to the binding partner is based on an antigen / antibody binding and the preparation and purification of the antibodies used as binding partners is complicated and expensive. Another disadvantage lies in the fact that the antigen / antibody binding a very strong bond between the binding partner, z. As immobilized to a matrix antibody, and the fusion partner. This has the consequence that in the elution of the bound via the fusion partner fusion proteins, sometimes extremely unphysiological conditions have to be created based on the defined protein. Under unphysiological conditions conditions are to be understood in this context conditions with z. As very high or extremely low salt concentrations, use of chaotropic salts and pH values that differ widely from the natural pH of the defined protein. This can u. U. influence the structure and / or functionality of the defined protein or irreversibly destroy. Accordingly, the purification of the defined proteins should be carried out under as mild, physiological conditions as possible in order to preserve the functionality of the proteins. Although three of the abovementioned fusion partners (Hopp et al., 1988) (Martin et al., 1990 (Skinner et al., 1991)) eluted under competitive conditions using competitive peptides, the problem remains the complex and expensive to produce and clean (serving as binding partner) antibodies and their binding to the matrix.

All known fusion partners that bind to the streptavidin the amino acid sequence HPQ, the so-called HPQ binding motif, which interacts with the biotin binding pocket of streptavidin occurs. Known peptides include the so-called Strep-tag I: AWRHPQFGG with a dissociation constant Kd of 10-37 μM, the so-called Strep-tag II: WSHPQFEK with a dissociation constant Kd of 18-72 μM and the so-called SBP-tag: MDEKTTGWRGGHVVEGLAGELEQLRARLEHHPQGQREP with a dissociation constant Kd of 2.5 nM. In contrast to the two short Strep-tags I and II have the longer SBP-tag a much stronger one Binding to streptavidin. However, as stated above, the Function of the defined protein, in particular with increasing length of the Affiliated with the merger partner. Also disturb especially long fusion partners in the crystallization of the defined proteins.

The binding affinity of the the expression of the expression described in Example 1 proteins to streptavidin was measured according to the manufacturer's instructions in the Biacore device. This is the protein to be measured on the sensor chip and a streptavidin solution with defined concentration is injected. The interaction (Binding) between the two molecules is measured by the device and referred to as so-called Resonance Units (RU). For measurement, the used by the manufacturer.

In times the usual Example, the FAB protein, which at the N-terminus one of fifteen amino acids existing additional Peptides with the amino acid sequence D V E A W L D E R V P L V E T (motif 3) was a fusion partner, in vitro in a cell-free protein biosynthesis by means of a coupled Transcriptional translation system expressed.
Für die Aufreinigung wurden folgende Puffer verwendet: Waschpuffer (100 mM Tris-HCl (pH 8.0), 150 mM NaCl, 1 mM EDTA).to Purification of the overexpressed the protein was coupled to the streptavidin on a solid phase. The solid phase was a Sepharose. The expressed FAB protein was subsequently in usual practice wise affinity-chromatographic over a Containing column Streptavidin-Sepharose or StrepTactin-Sepharose purified. For the Purification, the following buffers were used: wash buffer (100 mM Tris-HCl (pH 8.0), 150 mM NaCl, 1 mM EDTA).
For the elution of streptavidin, the washing buffer contained 2 mM biotin and for elution of StrepTactin, the wash buffer contained 2.5 mM desthiobiotin.
