In this meander bioreactor, the culture medium flows horizontally through the parallel meander channels, whereby the number of cells that can be cultivated is limited. In order to cultivate a large number of cells, especially T lymphocytes for therapeutic treatment, it is necessary to connect several bioreactors in series or in parallel. This requires a high level of equipment and a high level of effort to achieve the required sterility.

The Froude number describes the relationship between the flow velocity and the propagation speed of a shallow water wave. The wave spreads evenly in all directions, i.e. in a circle, and shows a parabolic pattern. It does not provide any information about backmixing and does not allow any statements to be made about whether and to what extent volume elements or substances, such as the overlay atmosphere and the culture medium within a reactor, are mixed by the prevailing currents, and therefore also does not provide any statement about whether the cells are sufficiently supplied with the overlay media for expansion.
With the solutions described in the prior art, it is also not possible to expand non-adherent cells, such as non-adherent T lymphocytes or invariant T lymphocytes and NK cells, but also other immune cells, such as tumor-infiltrated NK cells (TINK) and mucosa-associated invariant T cells (MAIT), in the quantities required for therapeutic applications, since they can only be insufficiently supplied with overlay media.
The object of the invention is to provide a perfusion meander bioreactor which eliminates the shortcomings of the prior art and is suitable for therapeutic To provide sufficient quantities of non-adherent growing cells, such as non-adherent growing T lymphocytes or invariant T lymphocytes and NK cells, but also other immune cells, such as tumor-infiltrated NK cells (TINK) and mucosa-associated invariant T cells (MAIT), for treatment.
The object of the invention is achieved by a perfusion plate meander bioreactor which consists of a bioreactor vessel 1 made of crystal-clear polymer material with a bottom 2, a lid 3, end walls 4 and 5, side walls 6, and feet 17, wherein in the bioreactor vessel 1 several cultivation supports 7 arranged one above the other are fastened by means of clamping nuts 9 and a clamping bracket 8, on which channels 10 formed by webs 13, one or more overflows 14 and in front of the overflows 14 over the entire width of the cultivation supports 7 an overflow web 33 with a height of 3 to 5 mm are arranged. Every second cultivation carrier 7 is rotated by 180° with its overflows 14 relative to the cultivation carrier 7 above it, and in the perfusion plate meander bioreactor, a lower collecting chamber for the culture medium 15 is arranged below the cultivation carriers 7 and an overlay collecting chamber 24 is arranged above the cultivation plates 7. The culture media is guided horizontally in the bioreactor vessel 1 in the channels 10 on the cultivation carriers 7 of the bioreactor vessel 1 and is guided through the overflows 14 and over the overflow web 33 onto the cultivation carrier 7 below, meandering vertically from cultivation plate 7 to cultivation plate 7 to the outlet 23 for the culture medium. The overlay media are guided in the channels 10 to the overlay collection chamber 24 for gas diffusion of the overlay media into the culture medium and for maintaining the concentration of the dissolved overlay media in the culture medium above the surface of the culture medium.
A supply channel 11 with a Luer-Lock connector 20 for overlay media is incorporated into the front wall 4 and is connected to the overlay spaces 16 above the culture medium 25 via inlets 12. A Luer-Lock connector 22 for the drainage of the overlay media is arranged in the front wall 5 and is connected to an overlay collecting chamber 24 located above the cultivation supports 7. A Luer-Lock connector 21 for the supply of the culture medium and a Luer-Lock connector 23 for the drainage of the culture media are arranged in the front wall 5, with the Luer-Lock connector 23 being connected to the collecting chamber 15 for culture media located under the cultivation plates 7. A light source with a wavelength of between 750 nm and 950 nm, in particular with a wavelength in the fluorescence spectrum for blood of 830 nm, is attached to a camera 32 on the outside of the bottom 2 of the bioreactor vessel 1 under a polished light window 19, wherein the bioreactor vessel 1 can be arranged on a vibrating table 30.
On the cultivation support 7, webs 13 for forming one or more channels 10 are arranged at a distance a from one another and at a distance b from end walls 4 and 5 and one or more overflows 14 as well as an overflow web 32, wherein the distance a of the webs 13 from one another is selected and the amount of culture media supplied and discharged is set so that a laminar flow with a Bodenstein number of 0.001 to 0.1 and the bottom flow remains close to a Bodenstein number of 0 in the flow thread in the channel 10 formed by the webs 13. The distance b of the webs 13 from the end walls is selected so that the culture medium can flow off via the overflows 14 without flow losses.
The Bodenstein number is a dimensionless number that describes the ratio of moles supplied by convection to those supplied by diffusion. The Bodenstein number thus characterizes the backmixing within a system (the larger the Bodenstein number, the lower the backmixing) and enables statements to be made about whether and to what extent volume elements or substances within the channel 10 are mixed by the prevailing currents.
Backmixing is greatest at a Bodenstein number of 0 or close to 0, which means that at a Bodenstein number close to 0 the overlay media are optimally mixed with the culture medium and thus the cells can be optimally supplied with the overlay media during their expansion.
The cultivation supports 7 with the webs 13 and the channels 10 formed thereby can consist of a hydrophilic plastic treated with low-temperature plasma with a cell-adhesive coating such as flbronectin, retronectin, collagen, gelatin, laminin, chitin and/or combinations thereof, wherein activating antibodies such as immunoglobulins, lymphocytes, anti CD 16, anti CD3, anti CD28, antiCD137, anti CD39, anti CD103 and/or combinations thereof, in particular 4-1 bb in combination with the cykines interleukin 12 or with a mixture of interleukin 12 and interleukin 2, can be incorporated into the cell-adhesive coating.
In one embodiment of the invention, instead of the Luer-Lock connectors 20 in the front wall 4, a three-way cock valve 28 and in the front wall 5 above the centrally arranged cultivation carrier 7, a Luer-Lock connector connected to the space for the culture medium 25 of this cultivation carrier 7 can be provided. binder 29, wherein the three-way cock 28 and the Luer lock connector 29 can be connected to one another via a bypass 26 and wherein an electromagnetic hose clamp 27 can be arranged around the bypass 26.
In a further embodiment of the invention, a shaking device in the form of a shaking table 30 and a pulse generator 31 is provided, on which the bioreactor is fastened in such a way that the plate plane of the cultivation carrier 7 lies horizontally and is mechanically, preferably electromechanically, shaken as required. The perfusion plate meander bioreactor is fastened to the shaking table 30 and the culture medium is subjected to a jolt in the direction of gravity by the pulse generator 31. The jolt introduced (change in acceleration per unit of time) should preferably be between 2-5 m/s 3 .
This reduces the segregation of cells as a result of sedimentation during flow in the channels, increases the proliferation rate and reduces the throughput time of the planned cell proliferation. The critical layer thickness within a cell cluster, from which the supply of the cells with nutrients becomes problematic, is approximately 1 mm. If the layer thickness is exceeded, the cells perish due to a lack of nutrients and overlay media. The compensation of these disadvantages of the critical layer thickness of the cell cluster is achieved by vertical shaking by the pulse generator 31.
This leads to an earlier start of treatment with the positive effect of higher chances of recovery, as well as lower costs.

The perfusion plate meander bioreactor consists of a bioreactor vessel 1 made of polystyrene, which is formed from a base 2, a lid 3, end walls 4, 5 and side walls 6. Several, in particular eight, cultivation supports 7 are fastened in the bioreactor vessel 1 via the clamping nuts 9 and a clamping bracket 8 with a distance of 11.5 mm. On the cultivation supports 7, channels 10 are formed by webs 13 through which the culture medium is guided and the culture medium is discharged via an overflow web 33 of 3 mm height and through one or more overflows 14 onto the cultivation support 7 below. This underlying cultivation The cultivation carrier 7, with its overflows 14 and its overflow web 33, is rotated by 180° with respect to the cultivation carrier 7 above it, so that the culture medium flows in a meandering manner from top to bottom into the collecting chamber 15 for the culture medium and is discharged from the bioreactor vessel 1 via the Luer-Lock connector 23. The distance a between the webs 13 is selected and the amount of culture medium supplied and discharged is adjusted so that a laminar flow with a Bodenstein number of 0.001 to 0.1 is formed in the flow thread, in the channel 10 formed by the webs 13, for the optimal supply of the expanding cells, and the bottom flow remains close to a Bodenstein number of 0. This is achieved primarily by the overflow web 33 of up to 3 mm.
A supply channel 11 for overlay media is incorporated into the front wall 4, which is connected via feeds 12 for overlay media to overlay chambers 16, which are located above the cultivation supports 7 with the flowing culture medium. Via the Luer-Lock connector 20, which is connected to the supply channel 11 for overlay media, the overlay media is guided in countercurrent over the cultivation medium into the collection chamber 24 for overlay media and is discharged from the bioreactor vessel 1 via the Luer-Lock connector 22 for the discharge of the overlay media.
The culture medium consists of a cell growth medium that is temporarily supplemented with a specific mixture of AB human serum, cytokines, antibodies and irradiated human feeder cells. The autologous tumor tissue pieces in a size of 1 to 2 mm 3 that are removed from the patient during an operation and chopped up are added to this culture medium and fed to the bioreactor vessel 1 with the culture medium via the Luer Lock connector 21 for the supply of culture medium. The chopped tissue pieces are evenly distributed in the bioreactor and cultivated in perfusion mode, with the culture medium being supplied with the appropriate overlay atmosphere in a controlled manner by the overlay media fed into the overlay spaces 16 via the supply channel 11 and the feeds 12 for overlay media. In a cultivation run over 7 to 14 days, cells (TIL or other cells) grow in sufficient quantities. As the number of cells increases, the supply of fresh medium is continuously increased during the proliferation of the number of cells (TIL or other cells), whereby the increased supply of fresh medium is automatically controlled by a corresponding algorithm.
The bottom 2 of the bioreactor vessel 1 has a light window 19, under which a light source 20 with a wavelength in the fluorescence spectrum for blood of 830 nm with a video camera 32 is mounted, with the aid of which the backmixing of the overlay media in the culture media can be monitored and controlled, wherein the culture media are provided with suitable means for detecting the backmixing.
Instead of the Luer-Lock connector 20, a three-way cock valve 28 can be arranged in the front wall 4 and a Luer-Lock connector 29 connected to the space for the culture medium 25 of this culture carrier 7 can be arranged in the front wall 5 above the centrally arranged cultivation carrier 7, wherein the three-way cock valve 28 and the Luer-Lock connector 29 are connected to one another via a bypass 26 and wherein an electromagnetic hose clamp 27 is arranged around the bypass 26.
This means that by switching the three-way cock 28 and the Luer lock connector 29, part of the culture media can be passed through the bypass 26 and a definable shear stress can be exerted on the expanding cells in the culture media by narrowing the cross-section of the bypass 26. The activation of cells, particularly T cells, is both a mechanical and a biochemical process. With the electromagnetic hose clamp 27, a reproducible, defined shear stress is exerted on the cells, particularly T cells, and thus the activation is mechanically stimulated.
The bioreactor vessel 1 is arranged on a vibrating table 30 with a pulse generator 31, wherein a light source 18 with a wavelength of between 750 nm and 950 nm, in particular with a wavelength in the fluorescence spectrum for blood of 830 nm, with a camera 32 is attached to the bottom 2 of the bioreactor vessel 1 under a polished light window 19.
Beispiel 2:Example 2:
In a first bioreactor (not shown) suitable for cells growing from tissue pieces, especially from non-adherently growing T lymphocytes, such as tumor-infiltrated NK cells (TINK) and mucosa-associated invariant T cells (MAlT), such as the one in the
Depending on the medium selected and the supplements added, the TILs are activated in this process phase and specifically primed by further contact with the initially still present mixture of mutated tumor cells from an individual tumor piece (obtained from a tumor resectate). As a rule, 1.5 to 2 × 10 9 TILs grow in the first bioreactor within a week. After shaking up the TILs that have grown in the first bioreactor, the TILs are fed through the outlet nozzle of this first bioreactor (not shown) provided with a sieve fabric via the Luer lock connector 29 arranged in the lid 3 of the bioreactor vessel 1 to the perfusion plate meander bioreactor according to the invention for the further expansion of TILs according to Example 1. The pieces of tissue from which the TILs have grown are completely retained by the sieve fabric of the first bioreactor (not shown) suitable for the outgrowth of cells.
