Aktivitätstest und Inhibitionstestactivity test and inhibition test
Zum Nachweis der Wirkung der identifizierten Peptide auf die SARS-CoV-2 3CLpro Aktivität wurden Inhibitionstest durchgeführt. Dabei wurde der Einfluss der Wirkstoffe einzeln und in Kombination auf die Enzym-Aktivität der SARS-CoV-2 3CLprountersucht.Inhibition assays were performed to demonstrate the effect of the identified peptides on the SARS-CoV-2 3CL per activity. Die Aktivitätstests wurden unter Verwendung eines fluorogenen Substrats DABCYL-KTSAVLQ↓SGFRKME-EDANS (Bachem, Schweiz) in einem Puffer mit 20 mM Tris pH 7,2, 200 mM NaCl, 1 mM EDTA und 1 mM TCEP [1-5] durchgeführt. Diese Reaktionsmischung wurde in eine Corning 96-Well-Platte (Sigma Aldrich), bestehend aus 0,5 µM Protein, pipettiert, und der Assay wurde mit der Zugabe des zuvor genannten Substrats in einer Endkonzentration von 50 µM gestartet. Die Inhibierung der SARS-CoV-2 3CLpro-Aktivität durch die identifizierten Peptide wurde mit dem oben beschriebenen Aktivitätsassay untersucht. 10 µM der Verbindungen wurden für einen vorläufigen Screening-Test verwendet (
Für die anschließenden Inhibitionstests wurden 0,5 µM des SARS-CoV-2 3CLpro Proteins mit 0-140 µM 3CVL-1 und -2, 0-120 µM 3CVL-4 und 0-100 µM 3CVL-7 inkubiert. Im Falle der retro inversen Peptide wurden folgende Konzentrationen getestet, 0-140 µM 3CVLri-2 und -4, 0-100 µM 3CVLri-7. Die Mischungen wurden 30 Minuten lang bei RT inkubiert. Nach Zugabe des Substrats mit einer Endkonzentration von 50 µM wurden die Fluoreszenzintensitäten in 60 s-Intervallen über 30 Minuten mit einem Plattenlesegerät Infinite 200 PRO (Tecan, Männedorf, Schweiz) gemessen. Die Temperatur wurde auf 37 °C eingestellt. Dazu wurde bei verschiedenen Endkonzentrationen der Inhibitoren und des zuvor genannten Substrats die Reaktionsgeschwindigkeit der SARS-CoV-2 3CLpro bestimmt. 0,5 µM SARS-CoV-2 3CLpro wurde dazu mit den jeweiligen Peptide Inhibitoren in verschiedenen Konzentrationen 30 Minuten lang bei Raumtemperatur (RT) inkubiert. Anschließend wurde die Reaktion durch Zugabe der entsprechenden Konzentrationsreihe des Substrats eingeleitet. Die Datenanalyse wurde mit Hilfe eines Lineweaver-Burk-Plots durchgeführt. Dabei wurde der Kehrwert der Reaktionsgeschwindigkeit (1/V) gegen den Kehrwert der Substratkonzentration (1/[S]) aufgetragen [4,5].
