Abstract
Die Erfindung betrifft ein Verfahren zur Erzeugung von Milchsäure, bei dem Stärkehydrolysats mit einem Zuckeranteil mit wenigstens 40 Gewichts-%, insbesondere wenigstens 50 Gewichts-%, und insbesondere größer 60 Gewichts-%, und einem Hülsenfruchtproteingemisch insbesondere aus Ackerbohne oder Erbse mit einem Anteil von weniger als 30 Gewichts-% bereitgestellt wird. Es wird ein Lactat produzierendes Bakterium oder Pilz hinzugefügt und das entstehende Lactat abgetrennt. Description
Die Erfindung betrifft ein Verfahren zur Erzeugung von Milchsäure aus einem Stärkehydrolysat.The invention relates to a method for producing lactic acid from a starch hydrolyzate.
HINTERGRUNDBACKGROUND
Die Verwendung proteinhaltiger Rohstoffe insbesondere auf Basis von Hülsenfrüchten für die Erzeugung von vegetarischen bzw. veganen Lebensmittelprodukten hat in letzter Zeit deutlich zugenommen. Dabei werden nicht nur konventionelle Ansätze auf Basis von Soja verfolgt, sondern es kommen zudem Hülsenfrüchte, beispielsweise Erbse und Ackerbohne vermehrt zum Einsatz. Jedoch besitzen Hülsenfrüchte einen relativ großen Stärkeanteil, der während der Prozessierung der Hülsenfruchtart zur Extraktion des Proteinanteils entfernt wird. Hierzu können unter anderem eine sogenannte Nass- bzw. eine sogenannte Trockenextraktion verwendet werden. Bei der Nassextraktion erfolgt die Trennung zwischen Stärkeanteil und dem Proteinanteil mittels einer wässrigen Lösung, wobei die Proteine durch geeignete Einstellung eines pH-Wertes ausgefällt und von der übrig gebliebenen Stärke und den Fasern getrennt werden. Anschließend kann die Stärke wiederum getrocknet und weiterverwendet werden. Bei der Trockenextraktion erfolgt die Verarbeitung über eine sogenannte Sichtermahlung, bei der die Hülsenfruchtart sehr fein zermahlen und anschließend über ein Sichtersieb in eine stärkehaltige bzw. eine proteinhaltige Fraktion aufgetrennt wird.The use of raw materials containing protein, in particular based on legumes, for the production of vegetarian or vegan food products has recently increased significantly. Eine zunehmende Schwierigkeit bei der Verarbeitung von Hülsenfrüchten bzw. auch anderen Früchten zur Erzeugung von Proteinen stellt der relativ hohe Stärkeanteil dar, der nach einer Extraktion des Proteinanteils übrigbleibt. Zwar wird dieser Stärkeanteil gegenwärtig in verschiedenen weiteren Prozessen verarbeitet, der Bedarf an Stärke bzw. Kohlehydraten für die Landwirtschaft, Kosmetik und Viehfutterherstellung ist jedoch gedeckt. Dadurch wird bei der zu erwartenden anwachsenden Verarbeitung von Hülsenfrüchten zusätzlich Stärke produziert, für die momentan kein Markt besteht, sodass es hier zu einem weiteren Preisverfall des an sich schon niedrigen Stärkepreises kommen kann. Damit besteht die Gefahr, dass die Verarbeitung von Hülsenfrüchten wirtschaftlich unrentabel wird. Dem entsprechend existiert das Bedürfnis, die Nebenprodukte, insbesondere stärkehaltige Nebenprodukte einer weiteren Verarbeitung zuzuführen, um so in der Wertschöpfungskette den Gesamtertrag bei der Verarbeitung von Hülsenfrüchten zu erhöhen.An increasing difficulty in the processing of legumes or other fruits to produce proteins is the relatively high proportion of starch that remains after extraction of the protein proportion. Die Erfinder haben erkannt, dass die stärkereiche Fraktion als Nebenstromprodukt bei der Trockenextraktion von Hülsenfrüchten neben dem Stärkeanteil einen nicht unerheblichen Proteinanteil besitzt. Bei einer Weiterverarbeitung dieses Anteils, insbesondere einer Verarbeitung der Stärke bleibt entweder der Proteinanteil zurück bzw. wird je nach Anwendung ebenfalls verbraucht. Nach dem vorgeschlagenen Prinzip soll nun ein Verfahren zur Prozessierung von Stärke geschaffen werden, bei dem dieser Proteinanteil weiterhin Verwendung findet, sodass sich im Ergebnis ein für die Weiterverarbeitung geeignetes Zwischenprodukt einstellt, welches zum einen einen verarbeiteten Stärkeanteil und zum anderen einen noch unverarbeiteten Proteinanteil aus den Hülsenfrüchten aufweist.The inventors have recognized that the starch-rich fraction, as a side-stream product in the dry extraction of legumes, has a not inconsiderable protein content in addition to the starch content. Bei diesem wird als erstes wenigstens eine Hülsenfruchtart bereitgestellt, die anschließend über einen Sichtermahlprozess gemahlen und in eine erste Fraktion und in eine zweite Fraktion aufgetrennt wird. Die erste Fraktion umfasst dabei einen höheren Proteinanteil als die zweite Fraktion und wird als proteinreiche Fraktion bezeichnet. Ebenso lassen sich auch medizinische Ausgangsstoffe und/oder Produkte herstellen, wie beispielsweise Insulin, Hyaluronsäure, Streptokinase und eine Vielzahl von Antibiotika (z. B. Penicillin). Es versteht sich hier, dass die Endstoffe, nämlich ein Proteingemisch bzw. Milchsäure durch die oben genannten Endprodukte ersetzt werden können. Dabei müssen je nach gewünschtem Endprodukt Parameter angepasst werden, allerdings ist der Ausgangsstoff, nämlich das hier erzeugte Hydrolysat jeweils das gleiche. Insofern kann das durch Sichtermahlung und anschließende Hydrolisierung erzeugte Zwischenprodukt als Ausgangsstoff für eine Vielzahl von weiteren industriell wertvollen Produkte dienen.In the following, both the further processing by means of a fungal mycelium and the fermentation with microorganisms for the production of lactic acid are explained in more detail. Gemäß einigen Aspekten des vorgeschlagenen Prinzips kann das so gewonnene Stärkehydrolysat mittels eines Pilzmycels aus der Abteilung der Ständerpilze, der Schlauchpilze und/oder auch Fusariumarten mit dem Stärkehydrolysat sowie einer zusätzlichen Stickstoffquelle kultiviert werden. Eine zusätzliche Stickstoffquelle ist zweckmäßig, da in diesem Fall das Pilzmycel aus der zusätzlichen Stickstoffquelle den für das Wachstum notwendigen Stickstoff erhält und hierfür nicht auf die noch vorhandenen Proteinbestandteile im Hydrolysats zurückgreifen muss. Nach einer Kultivierung wird das Resultat getrocknet und gemahlen und so daraus ein Pilzproteingemisch erzeugt. Alternativ kann es auch direkt ohne zusätzliche Trocknung weiterverarbeitet werden. Das Pilzproteingemisch umfasst neben einem Pilzproteinbestandteil auch noch restliche Bestandteile an Proteinen der Hülsenfruchtart. Mit dem derartig vorgeschlagenen Verfahren lässt sich somit die Verarbeitung von Hülsenfruchtprotein skalieren, sodass auch aus dem stärkehaltigen Anteil der Hülsenfruchtart im Endergebnis ein Proteingemisch gewonnen werden kann.According to some aspects of the proposed principle, the starch hydrolyzate obtained in this way can be cultivated using a fungal mycelium from the department of pillar fungi, sac fungi and/or Fusarium species with the starch hydrolyzate and an additional nitrogen source. Mit dem vorgeschlagenen Verfahren lässt sich somit auf kostengünstige Weise ein Proteingemisch aus einer Hülsenfruchtart mit einem sehr hohen spezifischen Gewichtsanteil erzeugen. Die beiden so erzeugten Anteile sind zum einen ein Proteingemisch mit einem hohen Anteil des Hülsenfruchtproteins sowie ein zweites Endprodukt, dass je nach Weiterverarbeitung aus dem zweiten Proteingemisch noch einen Restanteil an Protein aufweisen kann..By using classifier grinding and a dry extraction process, processing costs are significantly reduced compared to a wet extraction process. Dies kann zweckmäßig zur Herstellung eines Proteinisolats oder Konzentrat mit einem sehr hohen Proteinanteil beitragen. Einige weitere Aspekte beschäftigen sich mit der Möglichkeit, nach der Hydrolyse und Verzuckerung die unterschiedlichen Größen von dem entstandenen Zucker und den übrigen aus der stärkereichen Fraktion vorhandenen Proteinen und Fetten auszunutzen. Dazu ist in einigen Aspekten vorgesehen, Proteinanteile und/oder Fettanteile aus dem Hydrolysat zu separieren, um so noch eine hochangereicherte proteinhaltige und/oder fetthaltige Fraktion zu erhalten. Die übrige Zuckermasse kann für die Fermentation verwendet werden. Für eine Trennung lassen sich neben verschiedenen Filterverfahren auch mechanische Verfahren wie ein Dekantieren, ein Zentrifugieren oder andere mechanische Verfahren benutzen.In one aspect, the hydrolyzing step comprises additional filtering, in particular membrane filtering and/or precipitating the second fraction. In einem weiteren Aspekt umfasst das Verfahren ein Enthüllen der Hülsenfrüchte vor dem Schritt der Sichtermahlung. Zudem kann die zweite Fraktion vor dem Schritt des Hydrolysierens auch noch zusätzlich gesiebt werden, so dass Reststoffe mit einer Korngröße von größer als 100 um, insbesondere größer als 60 µm bis 70 µm aus der zweiten Fraktion entfernt werden. Hintergrund ist der auf dem Markt recht niedrige Preis für die erhaltene Stärke, da Stärke auch in Getreide und anderen Produkten als Haupt- oder Nebenstrom vorkommt und die auf dem Markt verfügbare Menge zum Teil den Bedarf übersteigt oder generell der Markt gesättigt erscheint. In einem praktischen Schritt wird eine Amylase verwendet, die bei relativ niedrigen Temperaturen ihr enzymatisches Maximum besitzt. - - Hydrolysieren der zweiten Fraktion zur Erzeugung eines Stärkehydrolysats, welches einen Proteinanteil im Bereich von 5 Gewichts-
% bis 30 Gewichts-% aufweist.

- - Dekantieren;
- - Ausfällen; und

- - Sieben der zweiten Fraktion zum Entfernen von Reststoffen mit einer Korngröße größer als 60 µm aus der zweiten Fraktion.

- 8. Verfahren nach einem der vorhergehenden Gegenstände, bei dem die hydrolysierte zweite Fraktion vor einem optionalen Schritt der Abtrennung eines noch vorhandenen Proteinanteils wenigstens einen der folgenden Bestandteile umfasst:
- - Asparaginsäure mit einem Gewichtsanteil von 1,5% bis 4%, insbesondere zwischen 2% bis 3% und insbesondere zwischen 2,5% bis 3,3%, jeweils bezogen auf die Trockenmasse der hydrolysierten Fraktion;
- - Glutaminsäure mit einem Gewichtsanteil von 2,7% bis 5,5%, insbesondere zwischen 3,0% bis 4,7% und insbesondere zwischen 3,6% bis 4,3% jeweils bezogen auf die Trockenmasse der hydrolysierten Fraktion;
- - Arginin mit einem Gewichtsanteil von 1,6% bis 2,6% und insbesondere zwischen 1,9 bis 2,2 Gewichts-% jeweils bezogen auf die Trockenmasse der hydrolysierten Fraktion;
- - Serin oder Alanin oder Phenylalanin oder Prolin oder Glycin, jeweils mit einem Gewichtsanteil zwischen 0,5% und 4,5% jeweils bezogen auf die Trockenmasse der hydrolysierten Fraktion;

- - Maltase;
- - Dextrinase;
- - Saccharase;
- - Glykosidase;
- - Glucoamylase; und
- - Pullulanase.
- 10. Verfahren nach einem der vorhergehenden Gegenstände, bei dem der Schritt des Hydrolysierens mittels einer Säure erfolgt, wobei nach Abschluss des Hydrolisierens ein Neutralisieren der Säure, insbesondere mit Ammoniak erfolgt.
- 11. Verfahren nach einem der vorhergehenden Gegenstände, wobei das Stärkehydrolysat wenigstens einen der folgenden Zucker jeweils bezogen auf die Trockenmasse umfasst:
- - Glukose im Bereich von 60 Gewichts-% bis 98 Gewichts-%;
- - Fructose im Bereich von 5 Gewichts-
% bis 30 Gewichts-%; - - Maltose im Bereich von 5 Gewichts-
% bis 30 Gewichts-%; und - - Saccharose im Bereich von 2 Gewichts-% bis 20 Gewichts-%.
- 12. Verfahren nach einem der vorhergehenden Gegenstände, weiter umfassend:
- - Kultivieren eines Pilzmycels aus der Abteilung der Ständerpilze und/oder Schlauchpilze mit dem Stärkehydrolysat sowie einer zusätzlichen Stickstoffquelle; sowie wenigstens einen der folgenden Schritte:
- - Trocknen und Mahlen des kultivierten Pilzmycels zur Erzeugung eines Pilzproteingemisches;
- - Kühlen des kultivierten Pilzmycels;
- - Nassverarbeiten des kultivierten Pilzmycels; und
- - Pasteurisieren des kultivierten Pilzmycels.

- 1. A method for producing a starch hydrolyzate, comprising the steps of:
- - Providing at least one type of legume;
- - Sifter grinding the provided at least one type of legume and separating it into a first fraction and a second fraction, wherein
- - the first fraction has a higher protein content than the second fraction; and
- - in the second fraction, optionally, a proportion of a starch contained in the at least one legume type provided is at least 40% by weight, in particular at least 50% by weight;
- - hydrolysing the second fraction to produce a starch hydrolyzate having a protein content in the range of 5% to 30% by weight.
- 2. The method of item 1, further comprising:
- - wet extraction of the first fraction to produce a protein isolate with a legume protein content in the range of 80% to 97% by weight, and in particular in the range of 85% to 95% by weight.
- 3. Method according to one of items 1 to 2, in which the hydrolysing step comprises filtering, in particular in membrane filters and/or precipitating the hydrolysed second fraction.
- 4. The method according to any one of the preceding items, in which after the hydrolyzing step at least one separation of the protein content and/or fat content that is still present is carried out by at least one of the following steps:
- - Filtering, in particular membrane filtering;
- - centrifugation;
- - decanting;
- - failures; and
- - combinations thereof.
- 5. The method according to any one of the preceding items, further comprising:
- - dehulling of the legumes before the classifying step; and or
- - Screening of the second fraction to remove residues with a particle size larger than 60 µm from the second fraction.
- 6. The method according to any one of the preceding items, in which in the second fraction, before an optional step of separating off a protein portion and/or fat portion that is still present, there is a protein portion in at least one of the following ranges:
- - 5% to 35% by weight;
- - 20% to 25% by weight;
- - 22% to 27% by weight;
- - 18% to 23% by weight;
- - 12% to 20% by weight;
- - 15% to 30% by weight;
- - 20% to 30% by weight;
- - 22% to 30% by weight;
- - 24% to 30% by weight.
- 7. The method according to any one of the preceding items, in which the hydrolysed second fraction, before an optional step of separating off a protein portion and/or fat portion that is still present, has a fat content of more than 0.5% by weight and in at least one of the following ranges:
- - 0.8% by weight to 1.6% by weight,
- - 1% to 1.4% by weight;
- - 1.5% to 6.0% by weight;
- - 2.25% to 5.5% by weight;
- - 0.5% to 5.0% by weight; 1.0% to 4.5% by weight; 1.5% to 4% by weight; 1.0% to 3.5% by weight.
- 8. The method according to any one of the preceding items, in which the hydrolyzed second fraction comprises at least one of the following components before an optional step of separating off a protein portion that is still present:
- - Aspartic acid with a weight fraction of 1.5% to 4%, in particular between 2% and 3% and in particular between 2.5% and 3.3%, in each case based on the dry matter of the hydrolysed fraction;
- - glutamic acid with a weight fraction of 2.7% to 5.5%, in particular between 3.0% to 4.7% and in particular between 3.6% to 4.3%, each based on the dry matter of the hydrolyzed fraction;
- - arginine in a proportion by weight of 1.6% to 2.6% and in particular between 1.9 and 2.2% by weight, each based on the dry matter of the hydrolysed fraction;
- - serine or alanine or phenylalanine or proline or glycine, each in a proportion by weight of between 0.5% and 4.5%, each relative to the dry matter of the hydrolysed fraction;
- - Lysine with a weight fraction of more than 4% based on the dry matter of the hydrolysed fraction.
- 9. The method according to any one of the preceding items, in which the step of hydrolyzing enzymatically, in particular with at least one enzyme from the group consisting of:
- - alpha-amylase;
- - beta-amylase;
- - maltase;
- - dextrinase;
- - sucrase;
- - glycosidase;
- - glucoamylase; and
- - pullulanase.
- 10. The method according to any one of the preceding items, in which the step of hydrolyzing takes place by means of an acid, with the acid being neutralized, in particular with ammonia, after the conclusion of the hydrolyzing.
- 11. The method according to any one of the preceding items, wherein the starch hydrolyzate comprises at least one of the following sugars, each based on the dry matter:
- - glucose ranging from 60% to 98% by weight;
- - fructose ranging from 5% to 30% by weight;
- - maltose ranging from 5% to 30% by weight; and
- - sucrose in the range of 2% to 20% by weight.
- 12. The method according to any one of the preceding items, further comprising:
- - Cultivation of a fungal mycelium from the department of pillar fungi and/or sac fungi with the starch hydrolyzate and an additional nitrogen source; and at least one of the following steps:
- - drying and grinding the cultured mushroom mycelium to produce a mushroom protein mixture;
- - cooling the cultivated mushroom mycelium;
- - wet processing of the cultivated mushroom mycelium; and
- - Pasteurization of the cultivated mushroom mycelium.
- 13. The method according to any one of the preceding items, further comprising:
- - fermenting the starch hydrolyzate with a lactic acid bacterium or a fungus to produce lactate, in particular Ca-lactate and
- - Further processing of the formed lactate to lactic acid; or
- - Fermenting the starch hydrolyzate with a microorganism to produce at least one end product substance selected from the group of

- - adding a nitrogen source, in particular in the form of ammonium, in particular ammonium sulphate, ammonia and/or nitrates; or
- - Adding at least one amino acid, in particular from the group comprising:

- leucine;
- isoleucine;
- threonine;
- methionine;
- phenylalanine and
- tyrosine.
- 16. The method according to any one of the preceding items, in which the provision of at least one type of legume comprises providing
