Die vorliegende Erfindung betrifft synthetische Biofilm-Hydrogele mit Mikroorganismen, welche in sprühbarer Form appliziert werden, sowie ein Verfahren zur Herstellung eines synthetischen Biofilms.The present invention relates to synthetic biofilm hydrogels with microorganisms, which are applied in sprayable form, and a method for producing a synthetic biofilm.

Bei Biofilmen handelt es sich um eine Schleimschicht (einen Film), in den Einzel- oder Mischpopulationen von Mikroorganismen (z. B. Bakterien) eingebettet sind. In Biofilmen aggregierte Mikroorganismen können so mit hoher Zelldichte immobilisiert werden und auf Oberflächen anhaften. In Biofilmen aggregierte Bakterien haben gegenüber im planktonischen Zustand, also in freier Suspension, vorliegenden Bakterien einige Vorteile. Die Biofilm-Matrix, in der die Bakterien eingebettet vorliegen, bietet mechanische Stabilität und erlaubt es den Biofilm-Organismen, langfristige synergistische Wechselwirkungen aufzubauen, Hungerperioden zu überstehen und verhindert das Abschwemmen extrazellulärer Enzyme. Die Aggregation im Biofilm ermöglicht den Bakterien darüber hinaus, durch spezielle physiologische Wechselwirkungen, wie z.B. über Signalmoleküle, sich in einem untereinander agierenden, flexiblen, leistungsfähigen Verbund zu organisieren. Der Biofilm bietet dem einzelnen Mikroorganismus darin optimierte Lebensbedingungen sowie einen erhöhten Schutz vor Veränderungen der Umgebungsbedingungen, wie pH- und TemperaturSchwankungen, Schadstoffen (z. B. Bakteriziden), oder UV- und Röntgenstrahlung, sowie Nahrungsmangel. Biofilme eignen sich damit besonders gut zur Stabilisierung und Immobilisierung hoher Zelldichten von Mikroorganismen auf Flächen.Biofilms are a layer of mucus (a film) in which single or mixed populations of microorganisms (e.g. bacteria) are embedded. Biofilms are therefore particularly suitable for stabilizing and immobilizing high cell densities of microorganisms on surfaces.

So zeigten beispielsweise Zadjel et al. (Zajdel et al. „PEDOT: PSS-based multilayer bacterialcomposite films for bioelectronics“, Scientific Reports 8.1 (2018), 1-12), dass ein leitfähiges Hydrogel, in welches S. oneidensis eingebettet wurde, eine erhöhte Stromdichte im BES ermöglicht. Weiterhin gibt es den Ansatz, bereits bestehende, natürliche Multispezies-Biofilme in Hydrogele einzubetten, um die Resistenz gegenüber harschen Umgebungsbedingungen im Prozess zu steigern (Chen et al. „Encapsulation of a living bioelectrode by a hydrogel for bioelectrochemical systems in alkaline media“ Journal of Materials Chemistry 8 3.23 (2015), 4641-4646). Weiterhin wurde Agarose als Hydrogel von Suruvaram et al. (Suravaram et al. „Conductive gels based on modified agarose embedded with gold nanoparticles and their application as a conducting support for Shewanella oneidensis MR-1“ ChemElectroChem 6.23 (2019), 5876-5879) so modifiziert, dass Goldnanopartikel gleichmäßig eingebettet und eine erhöhte Stromdichte mit planktonischer Beimpfung mit S. oneidensis nachgewiesen werden konnte. Insbesondere bestand die Aufgabe der vorliegenden Erfindung darin, eine Möglichkeit zur Herstellung verbesserter synthetischer Biofilme mit Mikroorganismen bereitzustellen, welche einen oder mehrere der vorgenannten Vorteile aufweisen. So ist ein Aspekt der Erfindung auf die Aufgabe gerichtet, eine Möglichkeit zur Herstellung synthetischer Biofilme mit exoelektrogenen Mikroorganismen oder Gas-fermentierenden Mikroorganismen bereitzustellen, die eine erhöhte Effizienz in der Anwendung ermöglichen. Ein weiterer Aspekt der Erfindung ist auf die Aufgabe gerichtet, synthetische Biofilme schnell, einfach und kostengünstig verfügbar und applizierbar zu machen. Dadurch wird auch die Biofilm-basierte Katalyse aus Abgasen und weiteren zur Verfügung stehenden Abfallstoffen als Edukte möglich. Jedes BES wurde unter sterilen Bedingungen aufgeschraubt und mittels Sprühapplikator (gemäß1 ) wurden 8 mL der LMA-Bakterien-Suspension im flüssigen Zustand auf die Anodenfläche aufgesprüht. Nach erfolgter Gelierung des Hydrogels wurde die Anode erneut in das System eingebracht und das System erneut mittels Schrauben verschlossen.
