Die vorliegende Erfindung betrifft einen Gegenstand der aus einer polymerbasierten Zusammensetzung besteht und mit Laminin beschichtet ist. HINTERGRUNDBACKGROUND
Im klinischen Alltag werden heutzutage verschiedene Stentarten verwendet. Metallbasierte Stents zeichnen sich durch eine hohe mechanische Stabilität im Gefäß aus. Der Nachteil dieser Stents ist, dass diese nach dem Einpressen ins Gefäß dessen Schädigung induzieren. Eine mögliche Folge ist die intimale Hyperproliferation mit nachfolgendem Gefäßverschluss durch die Ausbildung einer Neointima. Ein weiteres Problem ist die langsame Endothelialisierung der Stentoberfläche, die mehrere Monate dauern kann. Somit besteht über diesen langen Zeitraum die Gefahr einer Ausbildung von Thromben an der Stentoberfläche aufgrund fehlender antithrombogener Eigenschaften des Stentmaterials. Das erfordert die Gabe hochdosierter Antikoagulantien über einen langen Zeitraum mit teils schwerwiegenden Nebenwirkungen. Ein weiterer Nachteil ist die schlechtere Darstellung des umliegenden Gewebes im Cardio-CT [Lapp, Harald et al., Das Herzkatheterbuch, 2014, S. 291-308].Various types of stents are used in everyday clinical practice. Um die Nachteile der Metallstents zu umgehen, wurden polymerbasierte Stents, mit oder ohne Arzneimittel in der polymerumgebenden Hülle, entwickelt. Der Vorteil dieser Stents ist, dass diese über die Zeit Substanzen freisetzen, welche eine intimale Hyperproliferation nach dem Einpressen des Stents reduzieren, gleichzeitig aber auch die Besiedlung des Stents mit Endothelzellen hemmen. Demzufolge erfolgt hier kein „Zusetzen“ des Stents durch Zellen aus der Gefäßintima, aber gleichzeitig eine verzögerte Endothelialisierung und damit auch wieder eine erhöhte Gefahr der Thrombenbildung an der Oberfläche. Die Komplexität der Beschleunigung der Endothelialisierung der Stentoberfläche in Anbetracht der Hämokompatibilität der Materialien bezüglich Minimierung von entzündlichen Prozessen und Thrombosen ist in mehreren aktuellen Reviews ausführlich dargestellt. Des Weiteren werden die spezifischen Eigenschaften von Endothelzellen, deren Wachstumsbedingungen und Trigger von Krüger-Genge et al. [Int. J. Mol. Sci. 2019, 20, 4411] beleuchtet. Das Einbringen eines Stents in das Gefäß induziert gewöhnlich eine Entzündung des umliegenden Gewebes und damit eine Irritation, welche wiederum mit einem erhöhten Risiko für den Patienten verbunden ist. Um diese Probleme zu umgehen, wurde von Abbot im Jahr 2017 der erste biologisch abbaubare Stent entwickelt. Der degradierbare Stent ermöglichte, dass Gefäßirritationen mit nachfolgendem Entzündungsgeschehen unterbunden wurden. Allerdings wurde dieser Stent kurz darauf wieder vom Markt genommen [Kipper, Matt J. et al. Materials Science & Engineering R 138, 2019, 118-152.], da sich zeigte, dass die Degradation des Stents in ungleichgroße Bruchstücke erfolgt, welche ins Gefäßsystem fallen und damit Todesfälle durch Thrombenbildung und Embolien induzieren. Der Stand der Technik in Bezug auf biologisch abbaubare Stents wird des Weiteren von Omar et al. Eine Stentimplantation wird durchgeführt, wenn eine dauerhafte Aufdehnung eines verschlossenen Gefäßes oder Hohlorgans durch eine bloße Ausweitung der Gefäße (Perkutane Transluminale Angioplastie, PTA) nicht mehr möglich ist. Zu diesen Fällen zählen:
- Durchblutungsstörungen in den Arm- und Beinarterien bei peripherer arterieller Verschlusskrankheit (pAVK), Erweiterung der Hauptschlagader (Aorten Aneurysma), Schlaganfall bei Verengungen der Halsschlagadern (Carotisstenose),
- Verengung der Nierenarterien (Nierenarterienstenose), Verengung von Gängen (z.B.
- Gallengangsstenose), Verengungen der Herzkranzgefäße bei koronarer Herzkrankheit (KHK), Gefäße verschließen sich dauerhaft durch Arterienverkalkung,
- der sogenannten Arteriosklerose.

Biologisch abbaubares PolymerBiodegradable polymer
Folgende Produktklassen sind u.a. im Gebrauch: aliphatische und aliphatische/aromatische Polyester, Polyesteramide, Polylactide, Stärke und StärkeDerivate sowie Blends hieraus und Cellulose-Derivate.
Der Begriff der biologischen Abbaubarkeit ist präzise in der harmonisierten EN Norm EN13432 beschrieben [Europäisches Norminstitut, DIN EN 13432:2000 2000].
Die biologische Abbaubarkeit von Polymeren wird durch verschiedene Faktoren bestimmt. Wichtig sind Sauerstoffgehalt, Wassergehalt, pH-Wert, Temperatur sowie die Vorbehandlung des Polymers [P. L. Nayak, j. Macromol. Sci., Rev. Macromol. Chem. Phys., 1999, C39(3), 481]. Bei gleicher Polymerart hängt die Geschwindigkeit des Abbaus entscheidend von der Kristallinität des Polymers ab. Aufgrund des größeren freien Volumens und der größeren Kettenbeweglichkeit in den amorphen Bereichen erfolgt die Erosion dort schneller als in den kristallinen Bereichen. Biologisch abbaubare Polymere werden in Biopolymere, die natürlich biologisch abbaubar sind, und synthetische biologisch abbaubare Polymere unterschieden. Zu den Biopolymeren zählen Polypeptide wie Proteine und bakteriell synthetisierte Polyester sowie Polysaccharide wie Stärke und Cellulose. Diese natürlichen Polymere werden hauptsächlich enzymatisch abgebaut [T. Hayashi, Prog. Polym. Sci., 1994, 19, 663.]. Synthetische biologisch abbaubare Polymere werden hingegen in erster Linie durch chemische Hydrolyse abgebaut. Dabei werden zunächst niedermolekulare Oligomere erhalten, die anschließend weiter zu den Monomeren und schließlich bis zu Kohlendioxid und Wasser hydrolysiert werden. Eine notwendige Eigenschaft ist die Resorbierbarkeit, d. h. die biologische Abbaubarkeit im gegenwärtigen Hauptanwendungsgebiet dieser Kunststoffe, dem medizinisch-pharmazeutischen Bereich. Eine wichtige Anwendung im Medizinsektor ist die Verwendung als wirkstoffhaltiges Implantat auch bezeichnet als drug delivery system. Der Wirkstoff soll durch den Abbau des Polymers innerhalb einer gewissen Zeit mit konstanter Geschwindigkeit an einer bestimmten Stelle abgegeben werden [M. R. Brunstedt, et al., Mater. Sci. Aromatische Polyester wie Polyethylenterephthalat (PET) sind dagegen biologisch nicht abbaubar, besitzen aber ausgezeichnete Materialeigenschaften. PET ist außerdem sehr gut recycelbar. Chitosan ist ein farbloser, amorpher, zäher Stoff. Die Bearbeitung erfolgt mit speziellen Werkzeugen auf automatisierten Fräsmaschinen, welche neben Kunststoff und Holz fast alle Metalle sicher fräsen. Anschließend wurden die Zellen für den Versuch 2 T-75 Zellkulturflasche (TrypLE) und die Zellzahl mit einem Zellzähler [CASY= 2,12×106 Zellen/mL benötigt werden 4×104 Z/mL für 16 mL; (302µL Zellen in 15,698 mL EGM-2)] bestimmt.
Die sterilen erfindungsgemäßen Beispiele oder Vergleichsbeispiele wurden jeweils in eine 24-Well-Platte eingebracht und ein steriler Glasring wurde jeweils auf die Plättchen gelegt.
Pro Well wurden 0.5 mL HUVEC mit 4 × 104 Zellen/ mL (also 2 × 104 Zellen/ Well) zu den Beispielen ausgesät.

Dazu wurden die jeweiligen erfindungsgemäßen Beispiele oder Vergleichsbeispiele mit einer sterilen Pinzette in PBS (-/-) eingetunkt und dann in die Färbelösung überführt, welche 1 mL Färbelösung (1 mL EGM™-2 + 2 µl FDA + 4 µl PI) enthielt. Nach einer 3-minütigen Inkubation bei 37 °C und 5% CO2 erfolgte die Aufnahme von Bildern (5x, 10x, 20x Vergrößerung) mit einem inversen Mikroskop unter Fluoreszenzanregung. Um einen Überblick über die Besiedlung (Konfluenz) des Materials und die Vitalität der Zellen zu bekommen wurden Bilder in 10x Primärvergrößerung aufgenommen. Zur Beurteilung der Zellmorphologie wurden Fotos in einer höheren Vergrößerung (20x Primärvergrößerung) aufgenommen.
