Dies kann zum einen mittels einer Pick-and-place-Vorrichtung geschehen, bei der in jede Trenchzelle manuell und sequenziell ein poröser PTFE-Gewebematerialstopfen eingepresst werden kann. Es ist ebenfalls möglich, eine Matte von porösem PTFE-Gewebematerial vorzusehen, die vorstrukturiert ist, z. B. nach Art einer Eierkartonstruktur. Dabei passen Erhebung der Matte exakt zu der Anordnung der Trenchzelle, so dass beim Anpressen der Waferrückseite gegen die eierkartonartige strukturierte PTFE-Gewebematerialmatte die Erhebungen in die Trenchzellen eingepresst werden. Im Anschluss kann man den Überstand an PTFE-Gewebematerial bspw. mittels einer scharfen Klinge abtrennen, so dass die PTFE-Gewebematerialerhebungen in den Trenchzellen verbleiben und die restliche Matte abgeschnitten wird. Bei Vorliegen einer Verdeckelung erübrigt sich diese Maßnahme, weil der Chip ohnehin bereits durch den Deckel versiegelt ist.
- - Unterschichten des Mikroarraychips mit einem perfluorierten Medium, das in das PTFE-Gewebematerial von der Waferrückseite her eindringt und die Arrayzellen von unten her versiegelt.
- - Durchführen und Analyse der molekularbiologischen Analysereaktion, z. B. einer qPCR oder einer isothermalen PCR oder einer digitalen PCR. Dabei kann die exzellente Wärmeleitfähigkeit des perfluorierten Mediums von Vorteil sein.

- 4. Entfernen Fotolackmaske 1,
- 5. Fotolithographie: Fotomaske 2 (4 µm dicker Fotolack) mit Trenchzellen und zuführendem Kanalsystem bzw. Einlaufbereichen,
- 6. Tieftrenchen der Silizium-Trenchzellen durch den Wafer: Maskierung durch Fotolack 2 und thermisches Oxid, wobei der Fotolack 2 nicht aufgebraucht werden darf: bspw. mit dem sogenannten BOSCH-Deep-Reactive-Ion-Etching-Prozess zum anisotropen Hochratenätzen von Silizium mit SF6/C4F8 alternierend, Lackmaske und Silizium-Oxid, Kanalbereiche sind während des Tieftrenchens durch das Silizium-Oxid abgedeckt,
- 7. Übertragen der Fotomaske 2 (= zuführendes Kanalsystem) in das Restoxid durch selektives Plasmaätzen des Oxids (CHF3/Ar, CF4+C4F8/Ar) oder Nassätzen (BOE) mit Stopp auf Silizium: Jetzt ist das Silizium im Kanalbereich freigelegt, ggf. kurzzeitiges Anstrippen des Wafers mit einem O2-Plasma in einem O2-Plasmastripper zur Oberflächenreinigung.
- 8. Fluorinierung der Rest-Lackoberfläche mit CF4-Plasma, Fluorterminierung der Lackoberfläche 2 gibt hydrophobe PTFE-artige hochstabile Oberfläche, SiliziumOberfläche wird dabei mit F-Terminierung versehen, die sich nachfolgend durch Kontakt mit Wasser oder geeigneten wässrigen Medien in hydrophile OH-Terminierung konvertieren lässt, siehe
Schritt 10, - 9. Waferreinigung nass mit sogenanntem SC1-clean, das ist ein Gemisch aus Ammonika (NH3), Wasserstoffperoxid (H2O2) und Wasser, welches z. B. Fette und organische Verunreinigungen von der Waferoberfläche ablösen kann. Die PTFE-artige Lackoberfläche bleibt davon unbeeinflusst,
- 10. Finale Hydrophilisierung des Siliziums mit H2O2-Dampf, Ausbildung von OH-Gruppen an allen offenen Silizium-Flächen, d. h. evtl. noch verbliebenes Si-F oder Si-H wird vollständig in Si-OH konvertiert. Die PTFE-artige Lackoberfläche bleibt davon unbeeinflusst,
- 11. Anfügen einer Bodenmatte aus PTFE-Gewebe oder eines PTFE-Schwamms oder einer porösen PTFE-Gewebematerialstruktur an die Waferrückseite, z. B. durch Ankleben auf die Waferrückseite, oder alternativ Einpressen von Stopfen in die Trenchzellen von der Waferrückseite aus, z. B. ganzflächig mittels EierkartonStruktur und anschließendem Abschneiden des Überstands mit einer Klinge bzw. Abreißen über Sollbruchstellen,
- 12. PEG ggf. als selbst-aufgebaute Schicht (self-assembled Monolayer, SAM) aufbringen, bspw. durch PEG-Silane (Trichlorsilane, Trimethoxysilane, Triethoxsilane usw.) aus der Gasphase,

- 17. Einbau in LoC-Arraykammer,
- 18. Befüllen mit Eluat in Richtung gegen die Schwerkraft mithilfe von Druck und hydrophilen Kräften auf der Siliziumoberfläche. Fluorierte Fotolackoberfläche weist Wasser ab und schützt dabei vor Cross-Talk zwischen benachbarten Arrayzellen. Eluat stoppt am Boden der Arrayzellen am hydrophoben PTFE-Gewebe bzw. PTFE-Schwamm: Bis zu Drücken von 0,8 bis 1bar oder 8
bis 10 Meter Wassersäule weisen solche Gewebe jedes Eindringen von wässrigen Medien zurück und lassen nur Gase hindurch bzw. perfluorierte Flüssigkeiten eindringen, - 19. Spacern zwischen einem transparenten Deckel und der Chipoberfläche möglich, wenn der Deckel und die Chipoberfläche verklebt werden. Nach Befüllen der Arrayzellen mit Probenflüssigkeit sollte der Freiraum mit einem perfluoriertem Medium befüllt und die Arrayzellen damit versiegelt werden. SAM-Coating), da diese Verfahren Schichten nur auf den hydrophilen Siliziumflächen aufwachsen, nicht jedoch auf den PTFE-artigen Oberflächen. Somit kann z. B. eine selektive Beschichtung mit dauerhaft hydrophilen Überzügen bzw. - - Überschichten des Mikroarray-Chips mit einem perfluorierten Medium, wodurch der Mikroarray-Chip bzw. die Arrayzellen gegeneinander versiegelt werden.
- -Durchführen und Analysieren der molekularbiologischen Reaktion, bspw. einer qPCR, einer isothermalen PCR, einer rITA oder einer digitalen PCR.

Ein möglicher zweiter Prozessfluss zur Herstellung des Mikroarrays ist nachfolgend aufgezeichnet, wobei die ersten neun Schritte den ersten neun Schritten des zuvor dargestellten ersten Prozessflusses entsprechen, so dass diese nicht noch einmal hier aufgeführt sind:
- 10. PEG als selbstaufgebaute Schicht (self-assembled monolayer = SAM-coating), z. B. in Form eines PEG-Silans, aufbringen, wobei der Prozess selektiv nur hydrophile Siliziumoberflächen bedeckt, nicht jedoch die PTFE-artigen Oberflächen, auf denen keine Beschichtung erfolgt,
- 11. Biocontent in Arrayzellen bzw. Trenchzellen spotten,
- 12. PEG in Arrayzellen über Biocontent spotten,
- 13. Chips ggf. mit transparenter Abdeckung, bspw. Deckglas, Polymerkappe, abdecken,
- 14. Vereinzellung zu Einzelchips, bspw. durch Sägen, Brechen, Mahoh-Dicing usw.,
- 15. Einbau in LoC-Arraykammer,
- 16. Befüllen mit Eluat in Richtung gegen die Schwerkraft mit Hilfe von Druck und hydrophilen Kräften auf der Siliziumoberfläche bzw. im Netzwerk der Zufuhrkanäle und Einlaufbereiche. Fluorierte Foltolackoberfläche weist Wasser ab und schützt dabei vor Cross-Talk zwischen benachbarten Arrayzellen,
- 17. Auffüllen des Freiraums mit perfluorierter Flüssigkeit ebenfalls gegen die Schwerkraft, dadurch wird der Eluatüberschuss durch das Kanalsystem weggespült, ohne Cross-Talk zu anderen benachbarten Arrayzellen zu verursachen.

Durch minimale Totvolumina wird ein hocheffizientes Befüllen der Arrayzellen mit der wässrigen Probenlösung erreicht, d. h. die Verluste an Probenmaterial durch Totvolumina werden dergestalt minimiert und die Empfindlichkeit eines Nachweises des Probenmaterials durch die nachfolgenden PCRs in den Arrayzellen wird infolgedessen soweit verbessert, dass eine Voramplifikationsreaktion bzw. vorgeschaltete PCR entfallen kann. Der Mikroarray-Chip bzw. der Freiraum mit minimalem Totvolumen wird nach dem Befüllen mit einem perfluoriertem Versiegelungsmedium versiegelt, um einen Cross-Talk und eine Materialverschleppung zu unterdrücken.Embodiments and manufacturing methods for a microarray or a microarray chip and a sequence, for example for a PCR, a qPCR, a digital PCR or an rITA in a lab-on-chip are thus described above.
