Abstract
Wirkstoffkombinationen ausa) Ubidecarenon (Coenzym Q10) undb) UbidecarenolCombinations of active ingredients from a) Ubidecarenone (Coenzyme Q10) and b) Ubidecarenol
Description
The present invention relates to preparations containing ubiquinol and ubiquinone and their cosmetic or dermatological use for the care of aging skin, for the prophylaxis and treatment of senile xerosis, and for improved skin contouring or against cellulite.
Cosmetics can be used to summarize all measures that make changes to skin and hair for aesthetic reasons or are used to cleanse the body. Cosmetics therefore means caring for, improving and beautifying the exterior of the body in order to please others as well as yourself in a visible, tangible and smellable way.
Another aim of skin care is to compensate for the loss of oil and water caused by daily washing. This is especially important when the natural regeneration capacity is insufficient. In addition, skin care products should protect against environmental influences, in particular against sun and wind, and delay skin aging.
- a) dryness, roughness and formation of dryness wrinkles,
- b) itching and
- c) reduced lipid replenishment by sebum glands (eg after washing).
- d) Sichtbare Gefäßerweiterungen (Teleangiektasien, Cuperosis);
- e) Schlaffheit und Ausbildung von Falten;
- f) lokale Hyper-, Hypo- und Fehlpigmentierungen (z.B. Altersflecken) und
- g) vergrößerte Anfälligkeit gegenüber mechanischem Stress (z.B. Rissigkeit)
- h) nachlassende Vitalität und Regeneration der Haut.
- d) Visible vasodilatation (telangiectasia, cuperosis);
- e) slackness and wrinkles;
- f) local hyper-, hypo- and incorrect pigmentation (eg age spots) and
- g) increased susceptibility to mechanical stress (e.g. cracks)
- h) declining vitality and regeneration of the skin.
The present invention relates in particular to products for the care of naturally aged skin, and to the phenomena listed under a), e), f), g) and h).
Products for the care of sagging, especially aged skin are known per se. They contain, for example, retinoids (vitamin A acid and / or its derivatives) or vitamin A and / or its derivatives. However, their effect on structural damage is limited in scope. In addition, there are considerable difficulties in product development in sufficiently stabilizing the active ingredients against oxidative decay. In addition, the use of products containing vitamin A acid often causes severe erythematous skin irritation. Retinoids can therefore only be used in low concentrations.
Often, the sagging skin is also associated with obesity and / or the so-called cellullite that is often associated with it.
Consumers' body awareness has increased significantly in recent years. In addition to cleaning and caring applications, measures are increasingly being taken to improve the body silhouette. Cellulite - a widespread phenomenon - plays a central role in this. The visible picture of cellulite is based on an increase in fat deposits in the subcutis (subcutaneous fat tissue), a weak connective tissue and a reduction in the flow conditions in the blood and lymph vessels. The cause is thus a partially constitutional weakening of the connective tissue with the simultaneous occurrence of enlarged fat cell chambers as a result of obesity, an unbalanced diet, lack of exercise. The development of cellulite can also be traced back to an increased permeability of the hair vessel walls, which allows water to penetrate into the connective tissue.
The object of the present invention was therefore to find ways which avoid the disadvantages of the prior art. In particular, it was an object of the present invention to provide preparations which can bring about an advantageous tightening of slack skin.
One problem, however, is that coenzyme Q-10 eo ipso is not particularly easily absorbed into human skin. As a rule, sophisticated galenics are required to achieve good absorption.
One object of the present invention was to eliminate the disadvantages of the prior art. First and foremost, combinations of active ingredients and / or preparations based on coenzyme Q10 should be made available, with the help of which the full - possibly even improved effectiveness of coenzyme Q10 can be achieved.
In addition, active ingredients or preparations containing such active ingredients should be made available which, when used, stimulate collagen synthesis, strengthen the dermo-epidermal junction zone (skin tightening) and visibly improve the condition of the skin.
Weiter sollten Wirkstoffe bzw. Furthermore, active ingredients or preparations containing such active ingredients should be made available which can be used successfully for the care, prophylaxis and treatment of aging skin and / or senile xerosis.
The aim was to solve galenic problems, such as brownish discolouration of the formulation, degradation of the light-sensitive antioxidants contained, and skin intolerance, for example through oxidative degradation products.

In naturally occurring ubiquinols or their oxidized form, the ubiquinones, n can assume values from 0 to 50. In mammals, especially humans, n = 10 predominates (ubidecarenol), which according to the invention is the preferred ubiquinol.

The cosmetic or dermatological preparations according to the invention can have the usual composition and serve for the treatment, care and cleaning of the skin and / or hair and as a make-up product in decorative cosmetics. They preferably contain 0.001% by weight to 10% by weight, preferably 0.05% by weight to 5% by weight, in particular 0.01-2.0% by weight, based on the total weight of the preparations, of active ingredient combinations used according to the invention.

Complexing agents are known auxiliaries in cosmetology or medical galenics. By complexing interfering metals such as Mn, Fe, Cu and others, undesired chemical reactions in cosmetic or dermatological preparations can, for example, be prevented.
Complexing agents, in particular chelators, form complexes with metal atoms which, in the presence of one or more polybasic complexing agents, that is to say chelators, represent metallacycles. Chelates are compounds in which a single ligand occupies more than one coordination point on a central atom. In this case, therefore, normally stretched compounds are closed to form rings by complex formation via a metal atom or ion. The number of bound ligands depends on the coordination number of the central metal. The prerequisite for chelation is that the compound that reacts with the metal contains two or more atom groups that act as electron donors.

It is also possible and advantageous for the purposes of the present invention to incorporate active ingredient combinations used according to the invention into aqueous systems or surfactant preparations for cleaning the skin and hair.

Wirkungsnachweis:Proof of effectiveness:
Skin cells (keratinocytes) were incubated for 48 hours with ubidecarenone or ubidecarenol as well as the solvent control (50 µM concentration used each, n = 6 keratinocyte donors). The cells were then lysed and the total Q10 content (as the sum of ubidecarenone and ubidecarenol) using HPLC-MS / MS (high-performance liquid chromatography-mass spectrometry / mass spectometry) and the total protein content for normalization using the “Pierce BCA Protein Assay”.
Surprisingly, a comparison of the oxidized Q10 form ubidecarenone with the reduced Q10 variant ubidecarenol showed that the latter is on average 2.6 times better absorbed by the skin cells (2.6 times higher total Q10 content compared to the ubidecarenone supplemented cells) . In the skin cells (keratinocytes) the Q10 can be converted from the reduced to the oxidized form and vice versa. Nevertheless, the greater part of Q10 in the supplemented cells is present as anti-oxidative ubidecarenol.
Ubidecarenol sowie der Lösemittelkontrolle (je 50 µM Einsatzkonzentration, n=6 Spender).Q10 content in skin cells (keratinocytes) after 48 h incubation with ubidecarenone or ubidecarenol as well as the solvent control (50 µM application concentration each, n = 6 donors).
Surprisingly, the total Q10 content in the ubidecarenol-treated cells increases 2.6 times better than in the ubidecarenone-treated cells, which speaks for an improved bioavailability of the ubidecarenol for keratinocytes.
