- • Die beschriebene Erfindung kann auf einer bereits bestehenden LoC-Plattform angewendet werden. Somit ergeben sich weitere fluidische Möglichkeiten, ohne dass der Kern der Fluidik angepasst werden muss.
- • Einige der erfindungsgemäßen Funktionen sind durch eine Anpassung der Probeneingabekammer einfach umsetzbar. Die Probeneingabekammer kann durch einen Einsatz oder direkt im Spritzgussteil angepasst werden.
- • Zur Integration der beschriebenen Funktionen sind ansonsten keine zusätzlichen Fertigungsschritte oder Materialien notwendig.
- • Durch eine Verbindung der Kammern, die sich auf einer gewissen Höhe befindet, können die Volumina sowohl getrennt prozessiert werden, als auch miteinander vereinigt werden.
- • Durch die Verbindung lässt sich auch ein Durchfluss durch die Probeneingabekammer erzeugen, zum vollständigen Probeneinzug oder als Spülschritt.
- • Die Proben können je nach Bedarf über verschiedene Eingabestränge in die Fluidik eingezogen werden.
- • Durch die erfindungsgemäßen Prozesse ist es außerdem möglich auch auf dem Chip vorgelagerte Reagenzien in die Probeneingabekammer zu spülen.
- • Durch das Einpumpen von Öl aus der Fluidik in die Eingabekammer, kann die Wahrscheinlichkeit des Einzugs von Luftblasen in das fluidische System verringert werden.
- • Durch die beschriebenen Prozesse der Erfindung können auch Kammern in die Probeneingabe integriert werden, die keine direkte Verbindung zur Fluidik besitzen. Der Probeneinzug erfolgt durch Durchfluss oder mit Hilfe eines Unterschichtungsprinzips.
- • In der Probeneingabekammer können Reagenzien und Lysepuffer unabhängig voneinander vorgelagert werden und werden durch die erfindungsgemäßen Abläufe in die Analyse integriert und prozessiert.
- • Die Erfindung beschreibt Mischvorgänge, die in der Probeneingabekammer mit Verbindung zur Atmosphäre durchgeführt werden können. Dies verringert die Wahrscheinlichkeit der Entstehung von Luftblasen im fluidischen System.
- • Die Aufteilung der Probeneingabekammer in mehrere Reservoire ermöglicht Lysevorgänge die mehrere Schritte beinhalten. Dabei werden die Komponenten der Lyse bis zum Zeitpunkt der Verwendung voneinander unabhängig gelagert und dann in der benötigten Reihenfolge zugeführt. Z.B. wird eine alkalischen 3-Komponenten Lyse ermöglicht, bei der nacheinander zunächst destilliertes Wasser zum Aufbrechen der Zellen, dann Kaliumhydroxid zur Lipidentfernung und Proteindegradierung, gefolgt von der abschließenden Neutralisation mit Chlorwasserstoffsäure zugegeben werden.
- • Durch die erfindungsgemäßen Prozesse können von einer Probe in der Eingabekammer verschiedene Verdünnungsstufen hergestellt und nacheinander in die Fluidik eingezogen werden.
- • In der Probeneingabekammer kann ein Lyophilisat vorgelagert und transportiert werden. Dies ermöglicht es, ein Lyophilisat nach dem Einlegen des Chips in das System zur Probe zu geben und aufzulösen.
- • Das Lösen des Beads vor dem Einzug der Probe in die Fluidik hat zudem den Vorteil, dass entstehende Luft entweichen kann. Die Gefahr von Schaum- und Blasenbildung wird dadurch verringert.
- • Die Gefahr des Zerfalls von fragilem Probenmaterial ist verringert, da Schritte zur Probenaufbereitung in die Funktionseinheit eingeführt werden können und somit die zeitintensiven, händischen off-Chip-Schritte entfallen.
- • Die Eingabe der Probe kann für viele Proben standardisiert werden, differierende Schritte werden on-Chip, bzw. in der Probeneingabekammer durchgeführt. Dadurch ist für viele verschiedene LoC-Anwendungen keine zusätzliche Schulung des Personals notwendig.
- • Trennmethoden wie Sedimentation und Extraktionen können im beschriebenen Verfahren ermöglicht werden.
- • The described invention can be applied to an already existing LoC platform. This results in further fluidic possibilities without having to adapt the core of the fluidics.
- • Some of the functions according to the invention can be easily implemented by adapting the sample input chamber. The sample entry chamber can be customized through an insert or directly in the injection molded part.
- • No additional manufacturing steps or materials are otherwise necessary to integrate the functions described.
- • By connecting the chambers, which is at a certain height, the volumes can be processed separately as well as combined with one another.
- • The connection can also be used to create a flow through the sample entry chamber, for complete sample insertion or as a rinsing step.
- • Depending on requirements, the samples can be drawn into the fluidics via various input lines.
- • The processes according to the invention also make it possible to rinse reagents upstream on the chip into the sample entry chamber.
- • By pumping oil from the fluidics into the input chamber, the likelihood of air bubbles being drawn into the fluidic system can be reduced.
- • The described processes of the invention can also be used to integrate chambers into the sample input which have no direct connection to the fluidics. The sample is drawn in through flow or with the help of an underlaying principle.
- • Reagents and lysis buffers can be stored upstream in the sample entry chamber and are integrated and processed in the analysis by the processes according to the invention.
- • The invention describes mixing operations that can be performed in the sample entry chamber with connection to the atmosphere. This reduces the likelihood of air bubbles forming in the fluidic system.
- • The division of the sample entry chamber into several reservoirs enables lysis processes that involve several steps. The components of the lysis are stored independently of one another until they are used and then supplied in the required order. For example, alkaline 3-component lysis is made possible, in which first distilled water is added to break up the cells, then potassium hydroxide to remove lipids and protein degradation, followed by the final neutralization with hydrochloric acid.
- • Using the processes according to the invention, different dilution levels can be produced from a sample in the input chamber and can be drawn into the fluidics one after the other.
- • A lyophilisate can be stored and transported in the sample entry chamber. This makes it possible to add and dissolve a lyophilizate after inserting the chip into the system.
- • The detachment of the beads before the sample is drawn into the fluid system also has the advantage that air can escape. This reduces the risk of foam and blistering.
- • The risk of the breakdown of fragile sample material is reduced, since steps for sample preparation can be introduced into the functional unit and thus the time-consuming, manual off-chip steps are eliminated.
- • The sample input can be standardized for many samples, different steps are carried out on-chip or in the sample input chamber. As a result, additional training of the personnel is not necessary for many different LoC applications.
- • Separation methods such as sedimentation and extraction can be made possible in the described process.
