Abstract

Die vorliegende Erfindung betrifft ein optisches Verfahren zur Gehaltsbestimmung von Hyaluronsäure in Pharmazeutika, Medizinprodukten, Kosmetika und Rohstoffen, umfassend die Schritte:
- a) Herstellen einer Testlösung, die eine Probe enthält, deren Gehalt an Hyaluronsäure zu bestimmen ist,
- b) Messen der Trübung oder Absorption der Testlösung,
- c) Auswerten der gemessenen Trübung oder Absorption unter Zuhilfenahme einer Kalibrierkurve.

Verfahren zur Gehaltsbestimmung von Hyaluronsäure sind aus dem Stand der Technik bekannt. Drei wichtige Verfahren bestimmen hierbei quantitativ die Abbauprodukte der Hyaluronsäure: Bei der als Carbazol-Methode bekannten Reaktion wird Hyaluronsäure durch Schwefelsäure in seine Monomere hydrolysiert. Diese reagieren dann weiter zu Aldehyden, die mit Carbazol einen farbigen Komplex bilden. Der genaue Reaktionsmechanismus ist unklar, ebenso mögliche Störfaktoren, so dass es großer Erfahrung bedarf, nach dieser Konventionsmethode zu arbeiten. Nachdem diese Methode in einem Labor validiert wurde, gestaltet sich der Transfer dieser Methode in ein anderes Labor als schwierig. Aus der

Nach dem Abkühlen auf Raumtemperatur wird gegebenenfalls mit einem Magnetrührer so lange gerührt bis die Hyaluronsäure vollständig gelöst ist und keine Klumpen mehr sichtbar sind.

Durch graphisches Auftragen der gemessenen Trübung bzw. Absorption der Standardlösungen gegen die Hyaluronsäurekonzentrationen der Standardlösungen wird eine Kalibrierkurve erstellt. Unter zu Hilfenahme der Kalibrierkurve wird der Gehalt an Hyaluronsäure der Probe aus der gemessenen Trübung bzw.
