Für die Enrichment-PCR wurde nun isolierte, genomische DNA der transgenen Weizenlinien mit dem Enzym RsaI verdaut. Mutagenisierte DNA sollte nun nicht verdaut werden, so dass bei einer anschließenden PCR über den zu mutagenisierenden Bereich des Gens nur solche DNA amplifiziert werden sollte, die nicht verdaut wurde, also durch die Wirkung des TALEN mutagenisiert wurde. Da ein Restriktionsverdau auf genomische DNA niemals vollständig ist, wird es jedoch auch immer zur Amplifikation der nicht mutagenisierten DNA kommen. In einem zweiten Schritt wurde daher das amplifizierte PCR-Produkt noch einmal mit dem Enzym RsaI verdaut. Sollten hier unverdaute Fragmente entstehen, liegt die Vermutung nahe, dass der amplifizierte Genlocus tatsächlich durch die Wirkung des TALEN mutagenisiert wurde. Um diese Vermutung zu bestätigen, wurden die generierten, unverdauten PCR-Produkte in den Klonierungsvektor pGEM-T kloniert und sequenziert. Die aus den Körnern der T0-Pflanzen hervorgegangenen Pflanzen (T1-Generation) wurden erneut mittels PCR auf Vorhandensein der Mutation getestet. Dabei wurde zunächst eine PCR auf die einzelnen Weizenpflanzen gemacht, um dann die PCR Produkte anschließend mittels RsaI-Verdau auf Mutagenese des TaTDF-Gens zu testen. Ein vorheriger Verdau der genomischen Weizen-DNA vor der PCR Reaktion war nun nicht mehr nötig, da davon ausgegangen werden konnte, dass eine evtl. vorhandene Mutation nun in allen Zellen der Pflanze vorliegt.The transgenic parent wheat lines, for which mutations in the region of the TaTDF gene could be detected, were therefore transferred to the next generation by selfing. Diese Pflanzen wären dann homozygot für die Mutation. Erstaunlicherweise war dies jedoch nicht der Fall, sondern es wurde in allen Pflanzen sowohl unverdaute als auch verdaute DNA Fragmente identifiziert (

Durch Amplifikation von ca. 1 kb langen Fragmenten aus dem Genom der Weizensorte „Taifun” im Bereich des TaTDF-Gens und anschließender Sequenzierung von 8 klonierten PCR-Fragmenten, konnten die Sequenzen des TaTDF-Locus in den einzelnen Subgenomen identifiziert werden (

Auf Grundlage der Nachkommenschaftsanalyse der Linie WA500-T-003 wurden zwei Einzelpflanzen ausgewählt, WA500-T-003-003 bzw. WA500-T-003-021. WA500-T-003-003 war homozygot für das A- und D-Subgenom, und heterozygot für das B-Subgenom. WA500-T-003-021 hingegen war homozygot für das A-Subgenom und das B-Subgenom, aber nur heterozygot für das D-Subgenom.Based on progeny analysis of line WA500-T-003, two single plants were selected, WA500-T-003-003 and WA500-T-003-021, respectively. Die Ähren dieser Pflanzen (der Nachkommen von WA500-T-003-003 und WA500-T-003-021) wurden dann mit einer Tüte isoliert, um zu prüfen ob doch noch eine Selbstbefruchtung möglich war. In keiner der isolierten Weizenähren konnten Samen gefunden werden. Somit konnte eine 100%-ige männlichen Sterilität etabliert werden.The ears of these plants (the progeny of WA500-T-003-003 and WA500-T-003-021) were then isolated with a bag to check if self-fertilization was still possible.
