Description
The present invention relates to an inhibitor for the prophylaxis and treatment of an infection of a living being with a hantavirus and a pharmaceutical composition containing the inhibitor.
Viral infections represent one of the greatest challenges of modern medicine. Despite intensive research, the current possibilities of targeted therapies are limited.
Hantaviruses of the family Bunyaviridae include i.a. the human pathogenic species Hantaan virus, Puumala virus, Dobrava-Belgrade virus, Seoul virus, Korea fever virus and Sin-Nombre virus. These single-stranded (-) RNA viruses [ss (-) RNA] cause various diseases depending on the type of virus. These include severe lung disease, acute kidney failure or hemorrhagic fever. The viruses are distributed worldwide and also occur in Central Europe. They are transmitted to humans through the faeces or urine of infected rodents, which are inhaled as dust. The infected rodents themselves show no disease symptoms. The human diseases have different degrees of difficulty. While Puumula and Dobrava-Belgrade viral infections occur in less than 1% of clinically abnormal cases in Central Europe, lethality of Hantaan virus and Balkan Dobrava-Belgrade infections in East Asia Virus to 5% and in the American hantaviruses, such as the Sin-Nombre virus and others, about 30 to 40%.
There is currently no proven effective antiviral therapy against hantavirus infection. A vaccine is only in the developmental stage.
Against this background, it is an object of the present invention to provide an active ingredient which is suitable for the prophylaxis and / or treatment of an infection of a living being with a hantavirus.
This object is achieved by providing an inhibitor of the NF-κB signal transmission pathway.
According to the invention, an inhibitor is understood to mean an active ingredient or a composition containing such an active ingredient which specifically reduces or inhibits the signal transmission in the NF-κB signal transmission pathway or a component thereof, preferably blocks it. The active ingredient may according to the invention have any desired configuration, for example, in the form of a chemically defined molecule, a protein, a nucleic acid, etc.
Inhibitors of the NF-κB signal transduction pathway are fully described in the prior art. They can also easily be identified by means of a simple test system.
P105), NF-κB2 (P52 bzw. P100), Rel A (P65), Rel B und C Rel. The NF-κB signal transmission path is a significant signal transmission path in the cell. Central component of this transmission pathway is the heterodimeric NF-κB transcription factor-protein complex. The NF-κB transcription complex consists of a group of homo- and heterodimers belonging to the so-called Rel family. The five or seven subunits of NF-κB currently known in mammals are: NF-κB1 (P50 or P105), NF-κB2 (P52 and P100), Rel A (P65), Rel B and C Rel. NF- κB occurs in virtually all cell types and tissues. Dimers of these NF-κB subunits bind to regulatory regions of the DNA called κB sites. Dimers containing Rel A, Rel B or Rel C are transcriptional activators, while homodimers of p50 and p52 lack a transcriptional activation domain. They act as repressors. NF-κB is of great importance for the regulation of immune response, cell proliferation and cell death. The activation of NF-κB is considered critical for the development of inflammation. Finally, NF-κB fulfills important functions in the area of development of the immune system and the lymphatic organs.
The activation of NF-κB takes place for example by growth factors, cytokines, but also bacterial and viral antigens and chemical-physical Noxen such as UV radiation or free radicals. The stimulus usually beginning at the cell exterior is relayed to the NF-κB transcription factor in the sense of a signaling pathway via intracellularly interacting proteins that represent nodal points.
According to the invention, a living being comprises every living being, preferably a mammal, including humans.
The findings of the inventors were surprising. Thus, the state of the art even points in the opposite direction.
The findings of the inventors, however, prove the opposite. The inventors have recognized that inhibition of the NF-κB signal transduction pathway actually leads to a reduction in hantavirus burden.
Using the example of non-steroidal anti-inflammatory substances, such as salicylates, the inventors were able to demonstrate impressively that the proliferation of hantaviruses can be significantly inhibited in cell culture experiments. They were also able to show that in cells in which the NF-κB signaling pathway is inhibited by retroviral transfection, an increase in hantaviruses no longer takes place. This effect is highly specific and is not observed in the inhibition of other signal transduction pathways, such as the Raf / MEK / ERK signaling pathway.
The invention is completely solved by providing an inhibitor of the NF-κB signal transmission pathway.
Dabei ist die Komponente vorzugsweise ausgewählt aus der Gruppe bestehend aus: Tumor necrosis factor receptor associated factor (TRAF), NF-κB inducing kinase (NIK), Mitogen-activated protein kinase kinase kinase 1 (MEKKK1), Mitogen-activated protein kinase kinase kinase 3 (MEKKK3), AKR mouse thymoma kinase (AKT), TGFβ activated kinase (TAK1), Inhibitor of NF-κB kinase alpha (IKKalpha)‚ Inhibitor of NF-κB kinase beta (IKKbeta), NEMO, Inhibitor von κB (I-κB), RELA (p65), C-REL, RELEB, NF-κB1 (p105), NF-κB2 (p100), p50, p52.It is preferred if the inhibitor inhibits a component of the NF-κB Signalübertragungsgweges. In this case, according to the invention, a component is understood to mean a node in the NF-κB signal transmission pathway, such as a molecule or preferably a protein which is involved in signal transmission in the NF-κB signal transmission pathway. The component is preferably selected from the group consisting of: tumor necrosis factor receptor associated factor (TRAF), NF-KB inducing kinase (NIK), mitogen-activated protein kinase kinase kinase 1 (MEKKK1), mitogen-activated protein kinase kinase kinase 3 (MEKKK3), AKR mouse thymoma kinase (AKT), TGFβ activated kinase (TAK1), inhibitor of NF-κB kinase alpha (IKKalpha), inhibitor of NF-κB kinase beta (IKKbeta), NEMO, inhibitor of κB (I- κB), RELA (p65), C-REL, RELEB, NF-κB1 (p105), NF-κB2 (p100), p50, p52.
This measure has the advantage that the inhibitor according to the invention attaches to important nodes of the NF-κB signal transmission pathway and effectively inhibits it. Inhibitors of these components are thus particularly suitable for the realization of the invention.
According to a preferred development, the inhibitor according to the invention is selected from the group consisting of non-steroidal anti-inflammatory substances (NSAIDs), pyrrolidinedithiocarbamate (PDTC), MG132, BAY11-7085 or derivatives thereof. By a derivative is meant a chemically related and derived from the parent compound which is equally capable of specifically inhibiting the NF-κB signaling pathway.
This measure has the advantage that such inhibitors according to the invention are provided which, according to findings of the inventors, are particularly well suited to the realization of the invention and are distinguished by a high therapeutic safety.
According to the invention, non-steroidal anti-inflammatory substances comprise non-selective NSAIDs, such as acetylsalicyl derivatives, for example aspirin, and arylpropionic acid derivatives, for example ibuprofen, and arylacetic acid derivatives, for example diclofenac, etc. According to the invention, NSAIDs furthermore comprise selective COX-2 inhibitors, such as parecoxib, etoricoxib or celecoxib.
In this case, it is preferred according to the invention if the inhibitor of the NF-κB signal transmission path is an acetylsalicylate or a derivative thereof.
This measure has the advantage that such substances are used, which are have been distinguished by their special effectiveness and low side effects. Particularly suitable acetylsalicylate are salicylic acid, acetylsalicylic acid and acetylsalicylates modified by amino acids, such as LASAG. By a derivative is meant a chemically related compound obtained from the matrix which is equally capable of inhibiting the NF-κB signaling pathway.
It is particularly preferred when the inhibitor of the NF-κB signal transduction pathway is D, L-lysine acetylsalicylate ∙ glycine (LASAG).
As the inventors have shown, LASAG has a particularly high efficacy against hantaviruses.
Nach einer bevorzugten Weiterbildung der Erfindung ist das Hantavirus ausgewählt aus der Gruppe bestehend aus: Hantaan-Virus, Dobrava-Virus, Seoul-Virus, Saaremaa-Virus, Amur-Virus, Puumala-Virus, Sin-Nombre-Virus, New-York-1-Virus, Monongahela-Virus, Bayou-Virus, Black-Creek Canal-Virus, Andes-Virus, Laguna-Negra-Virus, Araraquara-Virus, Choclo-Virus, Juquitiba-Virus, Castelo dos Sonhos, Bermejo-Virus und Lechiguanas-Virus.According to a preferred embodiment of the invention, the hantavirus is selected from the group consisting of: Hantaan virus, Dobrava virus, Seoul virus, Saaremaa virus, Amur virus, Puumala virus, Sin Nombre virus,
This measure has the advantage that, by means of the inhibitor according to the invention, infections by such hantaviruses can be treated or prevented, which have a particular clinical relevance.
It is preferred if the inhibitor of the NF-κB signal transmission path is provided for the prophylaxis and / or treatment of a disease selected from the group consisting of: hemorrhagic fever with renal syndrome (HFRS), hantavirus-induced cardiopulmonary syndrome (HCPS ) and Nephropathia epidemica (NE).
This measure has the advantage that can be treated with the invention targeted by hantavirus-induced diseases.
Against this background, another object of the present invention relates to a pharmaceutical composition comprising the inhibitor of the invention.
Auflage, American Pharmaceutical Association and Pharmaceutical Press, verwiesen. It is understood that the pharmaceutical composition of the invention may have a pharmaceutically acceptable formulation. Pharmaceutically acceptable formulations are well known in the art. For example, reference is made to the paper by Kibbe A. (2000), Handbook of Pharmaceutical Excipients, 3rd Ed., American Pharmaceutical Association and Pharmaceutical Press. The pharmaceutical composition of the invention may further contain additives. These include any compound or composition which is advantageous for use in the invention, including salts, binders, solvents, dispersants and other substances commonly used in the formulation of pharmaceuticals.
It is further understood that in the composition according to the invention the inhibitor can be provided as the sole active ingredient. However, according to an alternative embodiment, the pharmaceutical composition may contain other active ingredients.
It is particularly preferred if the composition according to the invention is used as aerosol or for the inhalative, i. nasal or oral or bronchial administration is configured. An aerosol according to the invention is understood as meaning a mixture of a solid or a liquid and a gas. The disperse phase, also referred to as a discontinuous phase, consists of multiple solid or liquid particles. In general, the particle size of the disperse phases is less than 100 microns. According to the invention, both basic types of aerosols, i. solid and liquid dispersions in a gas phase, can be used as pharmaceutical aerosols. Examples of solid particle gas phase aerosols are those emitted by dry powder inhalers (DPIs). In contrast, metered-dose inhalers, soft-mist inhalers or nebulizers generate aerosols whose disperse phase is fluid. Inhalation administration is achieved through the use of a suitable inhalation device that generates an aerosol that is inhaled by the animal, for example via an oral or nasal piece.
It is preferred if the pharmaceutical composition according to the invention comprises the inhibitor in a concentration of about 0.1 to 4 mM.
This measure has the advantage that with such amounts sufficient concentrations can be achieved in the animal, preferably in the human organism, in particular in the lungs, which ensure an effective control of a hantavirus infection or a prevention thereof.
Against this background, a further subject of the present invention relates to the use of the inhibitor according to the invention and the pharmaceutical according to the invention A composition for the prophylaxis and / or treatment of an infection of a living being with a hantavirus.
The properties, features, advantages and developments of the inhibitor according to the invention apply equally to the pharmaceutical composition according to the invention and to the use according to the invention.
It is preferred if the total amount of the inhibitor according to the invention per administration unit is about 0.1 to 70 mg.
This measure has the advantage of providing such amounts of inhibitor that ensure optimum therapy. When administered by inhalation, the total dose per day in humans is expediently in the range of 0.1 to 30 mg, in the case of bronchial administration 0.1 to 70 mg. Depending on the application, however, the lower limit may also be between 0.1 and 20 mg or 50 mg. Depending on the application, however, the upper limit may be between 1 mg and 2 mg and the specified maximum values. A daily dose is advantageously administered in one to eight doses, which are desirably evenly distributed over a period of vigilance of 16 hours. A treatment is expediently carried out over a period of 1 to 7 days and longer. The invention thus also includes galenically prepared delivery units, wherein the amount of the inhibitor present in a delivery unit can be easily calculated in accordance with the above ranges of a treatment plan.
Against this background, it is preferred that the daily dose of the inhibitor in humans does not exceed 70 mg.
This measure has the advantage that side effects are largely avoided or at least significantly reduced.
Another object of the invention therefore also relates to the use of the inhibitor of the invention or the pharmaceutical composition of the invention for the treatment of a disease selected from the group consisting of hemorrhagic fever with renal syndrome (HFRS) hantavirus-induced cardiopulmonary syndrome (HCPS) and nephropathy epidemica (NE).
It is understood that the features mentioned above and those yet to be explained below can be used not only in the particular combination given, but also in other combinations or in isolation, without departing from the scope of the present invention.
Effective concentration of LASAG, which inhibits the proliferation of 50% of hantaviruses, and which is toxic to 50% of infected Vero cells
In this experiment, the EC 50 and TC 50 are to be determined by LASAG.
Vero cells are seeded at a concentration of 8.0 x 10 4 cells / well in 100 μl of growth medium (IMDM-10% FCS-penicillin-streptomycin-L-glutamine) in a 96-well plate. There are three approaches per substance dilution made. Subsequently, the plate is incubated overnight at 37 ° C and 5%. This results in a doubling of the cell count per well. The next day, the seeded Vero cells are washed with IMDM without FCS and then infected with Hantavirus Puumala strain Sotkamo at an MOI of 0.05 (8.5 x 310 ffu - 50 μl) for one hour at 37 ° C. The virus dilution is then removed from the 96-well plate and the respective test substance is added in 200 μl.
LASAG (D,L-Lysinacetylsalicylat·Glycin; Fa. Bayer; Ch.B.BXNNEV1; MW: 363,50): Es wird eine 1M Stammlösung in IMDM/10% FCS hergestellt und mit 1N NaOH auf pH 7,4 eingestellt und anschließend auf 1 M bis 0,01 µM in 10er Schritten in IMDM/10% FCS verdünnt.LASAG (D, L-lysine acetylsalicylate · glycine; Bayer; Ch.B.BXNNEV1; MW: 363.50): Prepare a 1M stock in IMDM / 10% FCS and adjust to pH 7.4 with 1N NaOH and then diluted to 1 M to 0.01 μM in 10-step increments in IMDM / 10% FCS.
The virus growth control consists of IMDM / 10% FCS. The plates are incubated at 37 ° C and 5% CO 2 for ten days. The supernatants are then removed and plated on Vero cells in a 96-well tissue culture plate titrated. After dilution of the supernatant to be tested, the tissue culture plate is re-incubated for 10 days at 37 ° C / 5% CO 2 . The cells are then fixed, permeabilized and subjected to immunocytological staining with an anti-nucleocapsid protein primary antibody (Anti-Puumala NP, Progen, 1: 200 in PBS / 10% FCS / 0.1% Tween). After incubation with a secondary antibody (anti-mouse POD-coupled antibody, Dianova), the substrate is added (TrueBlue, KPL / Medac). Virus-infected cells are stained blue. The plate is dried and then the virus foci are counted in each dilution. The number of focus-forming virus particles is given in ffU / ml (focus forming to / ml).
There are three approaches per substance dilution made. Subsequently, the plate is incubated overnight at 37 ° C and 5% CO 2 , so that the next day a dense cell lawn is created. Then the seeded Vero cells are washed with IMDM without FCS. Thereafter, the already described above dilutions of the test substance in 200 ul medium is added per well. The control consists of medium with 1% DMSO. The plate is incubated for 24 hours at 37 ° C and 5% CO 2 . The next day, the measurement of viability is carried out using the WST assay (Roche). The test is carried out according to the manufacturer's instructions. For this purpose, 20 μl of WST-q-1 reagent are added to each well (N dilution 1:10) and incubated for four hours at 37 ° C./5% CO 2 . The absorption of the samples at a wavelength of 460 nm is then measured in an ELISA reader. With the measured absorptions, the

It follows that at a concentration of 0.62 μM LASAG, the proliferation of 50% of hantaviruses is inhibited (EC 50 = 0.62 μM) (A). A concentration of 13.33 mM LASAG from is toxic to 50% of the infected Vero cells (TC 50 = 13.33 mM) (B). This results in a selectivity index of 21,500, which describes the difference between effect and toxicity.

As a result, the anti-viral effect of LASAG (A), ASA (B) and SA (C) against Hantaviruses is comparably good, with LASAG showing the best effect. LASAG, ASA and SA lead to a reduction in viral load in the concentrations of 10.1 and 0.1 μM (gray bars) compared to the control (black bar).
MEK inhibitors are unable to inhibit the proliferation of hantaviruses
