Description
Die Erfindung betrifft ein Verfahren zum Ernten von Mikroorganismen aus einem flüssigen Medium. The invention relates to a method for harvesting microorganisms from a liquid medium.
Die Kultivierung von Mikroorganismen in industriellem Maßstab hat in den letzten Jahren vielseitige Anwendungen gefunden. So werden beispielsweise mit Hilfe von Mikroorganismen verschiedene Substanzen biologischen Ursprungs, beispielsweise für Nahrungsergänzungsmittel, Feinchemikalien oder Medikamente hergestellt. Außerdem werden Mikroorganismen gezüchtet, um Biomasse zur Stromgewinnung oder zur Produktion von Biodiesel zu produzieren. Bakterien und Hefen werden in der Regel in Fermentern oder Bioreaktoren kultiviert, wohingegen Algen und Cyanobakterien in offenen oder geschlossenen Aquakulturen gezüchtet werden. Die Kultivierung in großen Mengen erfolgt üblicherweise in einem flüssigen Medium, in welchem sich die Mikroorganismen vermehren und, gegebenenfalls, die gewünschten Substanzen produzieren. Anschließend werden die Mikroorganismen geerntet, wobei sie vom Kultivierungsmedium getrennt werden müssen. Herkömmlich geschieht dies durch Filtration, Sedimentation oder Zentrifugation. Daneben sind aber auch Verfahren bekannt, bei denen die Mikroorganismen mit Hilfe von Chemikalien ausflocken und anschließend sedimentieren. Dadurch, dass die Trennung der Mikroorganismen vom Medium automatisch bzw. passiv erfolgt, wird keine zusätzliche Energie benötigt. Zudem muss nicht das komplette Medium zum Ernten der Mikroorganismen umgewälzt werden, wodurch weitere Energieeinsparungen möglich sind. Das Verfahren bedarf auch keiner Zugabe von chemischen Stoffen, die vor allem bei der Produktion von Substanzen für Lebensmittelzusätze und Medikamente durch Mikroorganismen nachteilig wären. In einer besonders bevorzugten Weiterbildung der Erfindung werden die Mikroorganismen jedoch nicht zur Produktion von Flüssigkraftstoffen, wie beispielsweise Biodiesel, verwendet. Die Verwendung von Mikroorganismen, die auf Induktion hin ein Lipid bilden, ist insbesondere geeignet, zur Produktion von chemischen Substanzen und Naturstoffen. Die Mikroorganismen können kultiviert werden, bis ausreichende Mengen der Substanz oder des Naturstoffs gebildet wurden. Anschließend wird die Bildung des Lipids induziert, so dass die Mikroorganismen an die Oberfläche des Mediums steigen und geerntet werden können. In einer bevorzugten Weiterbildung der Erfindung wird die Bildung des Lipids durch einen externen Auslöser induziert. Durch ein solches Konstrukt kann die Lipidbildung einerseits gezielt induziert und andererseits zusätzlich gesteigert werden, indem ein Protein der Lipidbiosynthese überexprimiert wird. Als äußerer Auslöser wird dem Medium beispielsweise eine Substanz zugegeben, die mit dem Promotor derart interagiert, dass sie ihn aktiviert.
