Die Kammer wurde vollständig mit der Silanlösung befüllt und mit einem Deckel verschlossen. Die Kammer wurde über Nacht bei 60°C in einem Trockenofen inkubiert. Danach wurde die Kammer aus dem Ofen genommen und nach dem Abkühlen wurden die Substrate aus der Kammer entfernt und für jeweils 10 min im Ultraschallbad nacheinander in Chloroform, Methanol und Wasser gespült. Die auf diese Weise silanisierten Glassubstrate wiesen Wasserkontaktwinkel von Θ ≈ 60° auf. Zur Lipidierung der mit APDMES aminosilanisierten Glasoberflächen wurden die Glassubstrate in einer frisch extrudierten Sulfolobus MPL Liposomensuspension (siehe Beispiel 2) über Nacht bei Raumtemperatur inkubiert. Im Anschluss daran wurden die Substrate nochmals zwei Stunden bei 70°C gelagert bevor sie im Ultraschallbad in Chloroform gereinigt wurden. Alle RifS-Analysen wurden mit einem BIAffinity System (Analytik Jena AG, Jena, Deutschland) durchgeführt. Bei der Funktionalitätsprüfung erfolgte die Detektion der komplementären Analyten mittels RifS, wobei das Verhältnis Unspezifität/Spezifität quantifiziert wurde. Zudem wurden die Chipmodifikationen im Hinblick auf Regenerierbarkeit getestet und die Zyklenstabilität der bindungsaktiven Oberfläche geprüft. Schließlich wurden die kinetischen Parameter, wie die Dissoziationskonstante KD ermittelt und mit den Literaturwerten verglichen.All RifS analyzes were performed with a BIAffinity system (Analytik Jena AG, Jena, Germany).
