Abstract
Description
The invention relates to a flow-through system with a channel structure for a fluid, in particular a liquid containing particles, wherein the flow-through system has a retention structure for the particles, which is formed by constrictions in the channel structure serving as obstacles for the particles. Moreover, the invention relates to a use of such a flow system.
Flow systems to filter particles from liquids are well known. For example, a glass frit can be inserted into a channel structure. This then represents a flow obstacle on which the particles get stuck. However, this can lead to blockages, since the particles accumulate in front of the glass frit or a comparable filter structure. In addition, there is a risk, especially with sensitive particles, that they will be damaged by the retention structure. For example, cells are very sensitive to mechanical stress, which can be caused by shear forces or excessive pressure. Therefore, the replacement of media or the separation of cells from a liquid such. B. Blood is limited to methods that do not endanger the cells to be separated. Centrifuges are currently predominantly used. Another possibility is filtering. However, such methods can only be used to a limited extent in flow systems if a continuous liquid exchange is required.
The object of the invention is to specify a flow-through system in a retaining structure, with which on the one hand a continuous separation of particles from the liquid is made possible and on the other hand this separation process is to exert on the particles to be separated as little mechanical stress as possible.
This problem is solved according to the invention with the flow-through system given at the outset in that the bottlenecks are formed by wall parts of the channel structure which are coupled in the flow-through system to an actuator in such a way that the flow cross-section of the bottlenecks can be changed by actuation of the actuator. This means according to the invention that over the setting range over which the flow cross-section of the bottlenecks can be changed by means of the actuator, it is defined how large the retainable and transportable particles can be in the relevant flow-through system. The smallest particle size, which can be retained by the retention structure, thus corresponds to the smallest possible distance of the wall parts in the bottleneck. Here, the smallest possible flow cross section is achieved. It should be noted that it is sufficient if the distance of the wall parts in one dimension (for example, the height of the channel structure) is changed. The width of the flow cross-section depends on the width of the channel structure as such, which is not changed according to an advantageous embodiment of the invention. This makes it possible to realize a simpler adjustment mechanism. It is thus at bottlenecks to a slot whose width can be adjusted. On the other hand, the limit for the largest particles that can still be transported is given by the fact that the adjustment range is also limited in the direction of increasing the distance of the wall parts. When the upper limit is reached, the slot width then determines which particles pass through there.
The adjustability of the flow cross-section of the bottlenecks additionally has the advantage that a blockage of the flow-through system can be counteracted by a change in cross-section being possible by adjusting the wall parts. The particles that were previously retained can be freed in this way and, for example, selectively removed from the flow system (more on this in the following). The adjustment can be used as appropriate for cleaning the flow-through system or for removing the particles, if the method carried out with the flow-through system has targeted the separation of these particles.
According to an advantageous embodiment of the invention, it is provided that several bottlenecks are arranged one behind the other in the direction of flow of the channel structure and the channel structure forms retention spaces between the bottlenecks. In this way, it is advantageously possible to increase the capacity of the retention structure without the volume of the retention spaces individually being too large. This also advantageously counteracts blockages, since only a certain number of retained particles can remain in each retention space. In addition, the bottlenecks in this case can advantageously be brought into a position in which they provide differently sized flow cross sections. This has the additional advantage that the individual bottlenecks are suitable for retaining different particle sizes. Smaller particles will therefore be able to pass through some bottlenecks in the direction of flow before being trapped by a bottleneck. In this way, it is advantageous to classify the particles in different sizes. The classification is advantageously particularly simple if the flow sections of different size in the flow direction of the channel structure are staggered such that they represent an obstacle to ever smaller particles. On In this way, the large and then smaller and smaller particles are sequentially segregated first as the fluid flows through it.
The flow-through system can advantageously be used particularly flexibly if the bottlenecks with several actuators can be adjusted independently of one another. In extreme cases, each bottleneck is assigned an actuator, so that each bottleneck can be provided with a desired cross-section. With such a flow-through system, the above-described methods can be advantageously carried out without having to select a different flow-through system for each of these methods. Thus, both uniform bottlenecks can be produced with a cross section limited only by the adjustment range of the actuators. It is also possible that a staggering of the cross-sectional areas of the bottlenecks is set in order to achieve a classification of the particles.
A particular embodiment of the invention provides that at least a part of the retention spaces is equipped with a lock mechanism through which the particles can be discharged from the flow system. The lock mechanism must therefore provide a sufficiently large cross-section so that the retained particles fit through the lock opening. Only then can they be ejected. In addition, the lock mechanism may be used to create a cross flow within a retention space to allow the retained particles to be transported. For this purpose, a suitable inflow is required, which itself need not have the cross section, so that the retained particles pass through. The lock mechanism can be used advantageously for cleaning purposes of the flow-through system. This can always happen when the flow system is to perform a fluid change. In this case, it is possible, for example, to rinse with a neutral liquid. Another possibility is to use the lock mechanism to eliminate blockages. If the pressure drop in the direction of flow of the flow-through system becomes too great, this indicates a blockage, so that the retention spaces can be flushed by crossflows. Last, of course, the lock system is also suitable for a targeted removal of restrained particles. For example, retained cells could be removed from the flow system for further investigation.
Another embodiment of the invention provides that the volume content of the retention areas is simultaneously changed by actuation of the actuator with the change in the flow cross section of the bottlenecks. This can be done by a suitable mechanical coupling of the wall parts of the flow system, each limiting the bottlenecks and the retention areas. This is most easily possible if the bottlenecks as well as the retention areas are formed by a single wall structure. The change in the volume of the retention spaces can be used specifically for the fact that not only the cross section of the retention structure is changed with the actuation of the actuators in the flow system, but also the volume of the channel structure. As a result, the actuator can also be used specifically for a pump mechanism realized in the flow-through system. In particular, it can be achieved by periodically actuating the actuators that particles do not finally catch on a flow obstacle formed by the retention structure but are transported through the flow-through system. However, the fluid or non-retained particles in this fluid flows faster than the retained particles, which still allows separation of the particles from the fluid.
For example, the channel system may advantageously be formed by two mutually opposite, relatively movable housing structures, wherein the bottlenecks are formed by the profile of the inner surface of at least one of the housing structures. The relative movement between the housing structures is ensured by the actuator. There are various configurations conceivable. Both housing structures can be profiled; However, at least it is necessary that one of the two housing structures is profiled in order to realize the change between retention structures and retention spaces geometrically. In order to achieve a variability of the cross-section provided by the retaining structures, it is basically sufficient to make one of the two housing structures movable. The mobility can be ensured that the housing structure is substantially rigid and, for example, moved toward the other housing structure and can be moved away from this. However, it is also possible that the housing structure has an elastic wall in itself, so that the profile provided by this housing structure is variable as such.
It is particularly advantageous if the profile of the inner surface is wave-shaped, with the corrugations forming the bottlenecks and extending at right angles or obliquely to the flow direction of the channel structure. It goes without saying that the wave ridges can not run along the direction of flow, because then no flow obstacles would form. Advantageously, the angle between the wave back and the flow direction is greater than 45 °, more preferably at least substantially at right angles. To this In this way, the above-mentioned transverse flow can be realized particularly easily, wherein it should be noted at this point that the transverse flow does not necessarily have to lie at right angles to the flow direction in the flow-through system. Preferably, the angle between the cross flow and the actual flow in the flow system should also be greater than 45 °, in particular at right angles. The transverse flow advantageously extends in particular along the wave ridges, ie with their course parallel.
It is particularly advantageous if the housing structures both have a corrugated profile of the inner surface, with the corrugation ridges of both profiles running parallel. As a result, the ratio of the cross sections between the restraint structures and the retention areas can advantageously be expressed to a comparatively high degree, without generating sudden cross-sectional jumps and thus edges in the respective profile. In general, it is advantageous if it is dispensed with sudden cross-sectional jumps. The particles can, as already mentioned, also consist of biological material such as cells. These are particularly sensitive and can be injured by sharp-edged cross-sectional jumps. If both housing structures are provided with a wave-shaped profile, the cells can be clamped in the area of the constriction, whereby the retention structure fulfills its function. However, the retained cells are not damaged and can be removed, for example, via a flushing process from the flow system.
It is particularly advantageous if the opposing groups of wave spines of the housing structure are movable parallel to the direction of flow and at right angles or at an angle to the course of the wave spines. Here one can make use of a peculiarity of this geometry of the retention structures and retention spaces. In fact, the cross-sectional area of the retention structures can not only be increased and decreased by moving the housing structures towards or away from each other transversely to the flow direction. Rather, it is also possible for a wavy structure of the two housing structures to move them parallel to the flow direction against each other. In doing so, the backs of the waves travel along (ie in the direction of or against) the flow direction, so that alternately two opposite wave backs with a constriction arise and then a wave back is again arranged opposite a wave trough, whereby the previously formed constriction is widened. This makes it particularly advantageous to produce the above-mentioned transport system for the particles. These can switch in this way between adjacent retention spaces.
Furthermore, it can be provided that the actuator is a polymer actuator. The use of polymer actuators has the advantage that they can realize relatively large strokes in the smallest space. In addition, polymer actuators can be advantageously installed in elastic wall parts of housing structures to change their example, wavy shape.
The above object is also achieved by the use of a flow-through system of the type described, wherein in use liquids are passed through which contain cells as particles. It has already been noted that the use of the flow-through system is particularly advantageous for particles such as cells, since these are particularly sensitive to mechanical action. The flow-through system according to the invention enables a comparatively gentle treatment of such particles, which can also be removed with the aid of the flow-through system from, for example, a liquid such as blood.
