Abstract
Die Erfindung betrifft eine Vorrichtung sowie ein Verfahren zur Inkubation von Patientenproben, die für eine immunologische und/oder histochemische Untersuchung vorgesehen sind und die in Form von Gewebe oder Zellen vorliegen. Typischerweise werden, insbesondere auf dem Gebiet der Tumordiagnostik, die gesamten Proben oder ausgewählte Teilbereiche, deren Untersuchung von besonderem Interesse ist, zunächst einzeln oder in Gruppen auf Substratträgern arrangiert. Anschließend erfolgt eine Inkubation mit einer Flüssigkeit, die zumindest einen Reaktanten enthält, und/oder mit einer Waschflüssigkeit. Auf diese Weise werden die Patientenproben angefärbt und die entstehenden Färbemuster können im Rahmen einer Untersuchung der Proben ausgewertet werden. Die erfindungsgemäße technische Lösung zeichnet sich dadurch aus, dass einerseits die Substratträger einzeln oder zusammen mit weiteren Substratträgern jeweils in Haltern angeordnet werden und dass andererseits, insbesondere während der Inkubation, eine Relativbewegung zwischen den Patientenproben und der für die Inkubation verwendeten Flüssigkeit erzeugt wird. Demgegenüber gestaltet sich die Inkubation bei den aus dem Stand der Technik bekannten Verfahren oftmals problematisch. Bei der Immunhistochemischen Färbung mit „offenen Tropfen” liegt beispielsweise ein mehr oder weniger runder Tropfen über dem Substrat, etwa einem Gewebeschnitt. In der Mitte ist der Tropfen höher als außen, deshalb sieht man oft in der Mitte des Feldes stärkere Reaktionen, da hier mehr Antikörper zur Verfügung stehen. Manchmal findet man aber auch das Umgekehrte: Aufgrund der größeren Krümmung der Oberfläche am Rand des Tropfens verdunstet dort die Flüssigkeit schneller als in der Mitte und es entsteht ein Konzentrationsgradient der Reaktanten mit einem Maximum außen. Dann reagiert der Rand des Gewebeschnittes stärker als die Mitte. Deshalb fällt es bei der mikroskopischen Beurteilung oft schwer, starke von schwachen oder sogar negative von positiven Reaktionen zu unterscheiden, weil nicht eindeutig zu erkennen ist, welche Zone des Gewebeschnittes beurteilt werden soll.
