Abstract
The invention relates to a method for producing D-amino acids, a microorganism, and a vector. According to the invention, a DNA coding for an amino acid racemase is introduced into a microorganism which produces an L-amino acid and which racemizes an L-amino acid in vivo into a D-amino acid. The D-amino acid is then separated from the L-amino acid.
Description
The invention relates to a process for the preparation of D-amino acids, a microorganism, and a vector.
In contrast to the L-amino acids, they are naturally only formed in exceptional cases, or in concentrations which are not sufficiently high, in order to be able to use them in the pharmaceutical industry as a pure substance or for synthesis purposes.
Established is the production of L-amino acids, which can serve as a precursor for the recovery of D-amino acids. For example, the L-amino acids, L-lysine, L-arginine, L-serine or L-threonine are obtained from cheap sugars by bacteria such as Escherichia coli or Corynebacterium glutamicum. The L-amino acids are isolated by known methods. To prepare the D-amino acid, an in vitro conversion of the L-amino acid into the racemate then takes place for the D, L-amino acid.
The chemical and enzymatic syntheses are expensive and expensive to carry out. The enzymatic / chemical synthesis from precursors requires that isolated enzyme and the chemically synthesized starting stages are present. L-amino acid derived methods require the isolation of the L-amino acid and the isolation of an amino acid racemase to effect the enzymatic conversion. The methods mentioned also require a control of the concentrations of educt and the enzymes or catalysts. Also, the catalysts become inactive or less active over time. The methods mentioned are complicated and not cost-effective.
It is therefore the object of the invention to provide a process for the preparation of D-amino acids which is cheap and above all simple and thus better. The need for concentration control of starting material and product should be eliminated. Furthermore, a microorganism and means are to be made available, which allows a simple and inexpensive production of D-amino acids.
Starting from the preamble of claim 1, the object is achieved by the features specified in the characterizing part of claim 1.
Advantageous developments of the invention are specified in the subclaims.
According to the method of the present invention, in an L-amino acid producing microorganism, an amino acid racemase is more intensely expressed to the wild type of the microorganism which racemizes the L-amino acid in vivo, the racemate is separated and the D-amino acid is separated from the L-amino acid by known methods ,

As the host bacterium, an L-amino acid-producing microorganism is used.
In one embodiment, it is possible to use a microorganism which already contains in its wild form an amino acid racemase gene which is overexpressed compared with its wild-type or whose copy number is elevated compared to the wild-type.
The DNA coding for an amino acid racemase can be used with and without a signal sequence.
In a preferred embodiment, amino acid racemase genes are used without signal sequence so that the amino acid racemases are not transported into an undesired cellular compartment, such as e.g. As the periplasm, but that they remain in the cytosol where the L-amino acid is synthesized in the microorganism from the sugar.
Also DNA sequences of genes, in particular of the genes mentioned, can be isolated and used by DNA hybridizations according to known methods Aminosäureacemasegene, and also be obtained by reducing the stringency of the hybridization nonspecific Aminosäureacemasegene. Thus, a wider range of applications of amino acid racemases can be achieved. For the method according to the invention, it is therefore also possible to use amino acid racemase genes which have a homology of at least 90% compared to their natural form. In particular, amino acid racemase genes can be used after the sequences Nos. 1 to No. 14, which have at least a homology of 90%.
B. DIG Easy Hyb. von der Firma Roche Diagnostics GmbH, Mannheim, Deutschland, Catalog No. 1603558).For the hybridization reaction, for example, a buffer corresponding to 5 × SSC buffer at a temperature of about 50 ° C-68 ° C. may be used. In this case, probes can also hybridize with polynucleotides which have less than 90% identity to the nucleotide sequence of the probe used. Such hybrids are less stable and are removed by washing under stringent conditions. This can be achieved, for example, by lowering the salt concentration to 2 × SSC and optionally subsequently 0.5 × SSC (The DIG System User's Guide for Filter Hybridization, Boehringer Mannheim, Mannheim, Germany, 1995), a temperature of about 50 ° C. being reached 52 ° C-68 ° C, ca. 54 ° C-68 ° C, ca. 56 ° C-68 ° C C-68 [deg.] C, ca. 58 ° C-68 ° C, ca. 60 ° C-68 ° C, ca. 62 ° C 68 ° C, about 64 ° C-68 ° C, about 66 ° C-68 ° C. Temperature ranges of about 64 ° C-68 ° C or about 66 ° C-68 ° C are preferred. It may be possible to lower the salt concentration to a concentration corresponding to 0.2 × SSC or 0.1 × SSC. Optionally, the SSC buffer contains sodium dodecyl sulfate (SDS) at a concentration of 0.1%. By gradually increasing the hybridization temperature in steps of about 1-2 ° C. from 50 ° C. to 68 ° C., it is possible to isolate polynucleotide fragments which contain at least 90% or at least 91%, preferably at least 92%. or at least 93% or at least 94% or at least 95% or at least 96%, and most preferably at least 97% or at least 98% or at least 99% have identity to the sequence or complementary sequence of the probe used and encode a polypeptide having amino acid racemase activity. The nucleotide sequence of the polynucleotide thus obtained is determined by known methods. Further instructions for hybridization are available on the market in the form of so-called kits (eg DIG Easy Hyb. From Roche Diagnostics GmbH, Mannheim, Germany, Catalog No. 1603558).
The amino acid racemase genes are expressed in the host bacterium that produces the L-enantiomer of the desired D-amino acid.
Beispielsweise wird ein Serinracemasegen, Glutamatracemasegen, Prolinracemasegen, Lysinracemasegen, Alaninracemasegen, Ornithinracemasegen, Argininracemasegen, Threoninracemasegen, Tryptophanracemasegen, Valinracemasegen, Isoleucinracemasegen und/oder Methioninracemasegen gegenüber der Wildform verstärkt exprimiert. For example, a serine racemase gene, glutamate racemase gene, proline racemase gene, lysine racemase gene, alanine racemase gene, ornithine racemase gene, arginine racemase gene, threonine racemase gene, tryptophan racemase gene, valine racemase gene, isoleucine racemase gene and / or methionine racemase gene are expressed more prominently against the wild type.
Expression takes place via vectors, preferably plasmids, by the gene's own or vector's own promoters, or else plasmid-free by integration of the genes together with promoters into the host genome.
The activity of Aminsäureracemasegne can be enhanced by the fact that promoters are used against the wild form promoters. The use of wild-type promoters is possible both in chromosomal DNA and in a vector.

If the D-enantiomer, for example D-arginine, is preferably secreted, the D-amino acid can be separated from the L-amino acid by known methods exemplified below.
In the absence of preferential secretion of the enantiomers, the D-amino acid may be separated from the L-amino acid by known methods exemplified below.
A separation of the D-amino acid from the racemate can be achieved by prefential crystallization, chemical reaction of an enantiomer, eg. Alternativ oder zusätzlich können die Mikroorganismen Vektoren, enthaltend für eine Aminosäureracemase kodierende DNA enthalten.Alternatively or additionally, the microorganisms may contain vectors containing DNA encoding an amino acid racemase.
Der Mikroorganismus enthält bevorzugt ein Plasmid. The microorganism preferably contains a plasmid.
The microorganism can in its genome and / or a vector, or plasmid, at least one component from the group of genes of serine racemase, glutamate racemase, proline racemase, lysine racemase, alanine racemase, ornithine racemase, arginine racemase, threonine racemase, tryptophan Racemase, valine racemase, isoleucine racemase, and / or methionine racemase in a greater number than those corresponding to the wild form included.

However, particularly preferred is a microorganism which has DNA coding for an amino acid racemase in a wild-type increased number which does not have a signal sequence for the amino acid racemase gene, since thus the expressed amino acid racemases are not transported to an undesired cellular compartment, e.g. As the periplasm, but that they remain in the cytosol where the L-amino acid is synthesized in the microorganism from the sugar.
The microorganism of the present invention may contain amino acid racemase genes which are altered by hybridization, for example, by the above-mentioned methods. Thus, a wider range of applications of the expressed amino acid racemases can be achieved. The microorganism thus modified may be a coryneform bacterium, an enterobacterium, especially a corynebacterium glutamicum or an Escherichia coli.
The invention furthermore relates to vectors which comprise amino acid racemase genes.
In a preferred embodiment, the vector of the invention is a plasmid.

Preferably, the vector for the amino acid racemase gene does not contain a signal sequence.

Nr. 33.Particularly preferred according to the invention are the plasmids pEKEx3-ArgR according to Seq. No. 32 and / or the vector pEKEx2-bar-SP according to Seq. No. 33.
With the method according to the invention and using the microorganisms and vectors according to the invention, D-amino acids can be prepared in a particularly simple and inexpensive manner.
